植物アデニロコハク酸シンテターゼおよびそれをコードするDNA
本発明は一般的には、植物におけるアデノシン5’−一リン酸生合成に関係す
る酵素活性に関する。特に、本発明はアデニロコハク酸の合成を触媒する植物酵
素および該酵素をコードする遺伝子に関する。一つの面において、本発明は異種
宿主における上記酵素の組換え産生に関する。別の面において、本発明は新規除
草剤の同定に適用される。さらに別の面において、本発明は植物における遺伝標
識形質の開発に関する。
アデノシン5’−一リン酸(AMP,アデニル酸としても知られている)は、
全ての生命系のための重要エネルギー伝達分子であるアデノシン5’−三リン酸
(ATP)の前駆体である。AMPの生合成における第1の明らかにされた酵素
段階はイノシン5’−一リン酸(IMP,イノシン酸)およびアスパラギン酸塩
からアデニロコハク酸の合成である。この段階を触媒する酵素はアデニロコハク
酸シンテターゼ(IMP:L−アスパラギン酸リガーゼ(GDP形成),EC6
.3.4.4,本明細書では「ADSS」と記載)として知られている。
大腸菌(E.coli)において、ADSSは同一な48kDサブユニットの二量
体である。その三次元構造は分解能2.8Åまで決定されている(Poland等,J
.Biol.Chem.268: 25334-25342(1993))。哺乳細胞において、
ADSS酵素は2種のイソ体として存在する。非筋肉組織に存在する酸性体はA
MPのデノボ(新規)産生に関係すると考えられている。筋肉組織に存在する塩
基性体は、アスパラギン酸塩のフマル酸塩への脱アミノ化の最終的な結果を伴う
IMPとAMPの相互変換を包含するプリンヌクレオチド回路の一部として作用
すると考えられている(Lehninger,Biochemistry.Worth Publishers,NY(1975
),p.743; Lowenstein,Int.J.Sports Med.11: S36-S46(1990)。
ADSS酵素をコードする遺伝子は以下のものを包含するさまざまな種から単
離されている:大腸菌(Wolfe およびSmith,J.Biol.Chem.263: 19147-19153
(1988))、ディー.ディスコイデウム(D.discoideum)(Weismuller等,J.
Biol.Chem.266: 2480-2485(1991))、マウス(Guicherit 等,J.Biol.Chem
.266: 22582-22587(1991); Guicherit 等,J.Biol.Chem.269: 4488-4496(19
94))、バチラス・サブチリス(枯草菌,Bacillus subtilis)(Maentsaelae お

しながら、ADSS酵素をコードする遺伝子はこれまでいずれの植物種からも単
離
されていない。
現在、植物ADSS酵素および他の生物から単離されたADSS酵素/遺伝子
と上記植物ADSS酵素との関係について知られていることはあまりに少ないの
で、公知の研究方法を用いていずれかの植物種からADSSコード遺伝子を単離
することはできない。
遺伝子を単離する方法はそれらがコードするタンパク質の構造の知識に基づい
ているが、植物ADSS酵素には適用され得ない。なぜならば、現在、植物AD
SS酵素について知られていることがあまりも少ないからである。代謝系酵素、
例えばADSSは、極端に低い存在量の故に、植物から精製することが一般的に
非常に困難である。さらに、植物内の種々のフェノール系および炭化水素化合物
の存在は天然の活性を有する純粋な酵素の単離を妨害し得る。
直接的な構造に関する情報がない状況下で、多くの標準的技術がタンパク質お
よびそれらの相当する遺伝子の単離に利用可能である。そのような標準的技術は
核酸ハイブリダイゼーションおよび保存されたアミノ酸配列モチーフに相当する
オリゴヌクレオチドプライマーを用いるポリメラーゼ連鎖反応(PCR)による
増幅を包含する。残念なことに、これらの技術は、それらが同じ機能を有する公
知タンパク質および遺伝子との顕著な構造的類似性(すなわちアミノ酸配列およ
びDNA配列)の存在に依存するので、植物ADSS遺伝子の単離のために
有用であることは期待されないであろう。植物以外の生物からの公知ADSS遺
伝子およびタンパク質の間でさえも、顕著な構造的類似性がないので(例えばPo
well等,FEBS Lett.303: 4-10(1992)参照)、これらのタンパク質が植物AD
SSタンパク質と顕著な構造的類似性を共有することは考えられない。
高等真核生物から他の代謝経路における生合成遺伝子を単離するために使用さ
れてきた別の研究方法は当該活性を欠失している微生物突然変異体の補足性であ
る。この研究方法のために、微生物宿主においてcDNAの発現を導き得るベク
ター内で高等真核生物からのcDNAのライブラリーがクローン化される。前記
ベクターは次に形質転換されるか、またはさもなければ突然変異微生物中に導入
され、そして表現型がもはや突然変異体ではないコロニーが選択される。
この戦略は以下のいくつかの代謝経路に関係する高等真核生物からの遺伝子を
単離するために行われている:ヒスチジン生合成(例えば、参照により本明細書
にその全体が編入される国際特許出願WO94/26909参照)、リジン生合
成(例えばFrisch等,Mol.Gen.Genet.228: 287(1991))、プリン生合成(例
えばAimi等,J.Biol.Chem.265: 9011(1990))およびトリプトファン生合成
(例えばNiyogi等,Plant Cell 5: 1011(1993))。この戦略はまた、トウモロ
コシグルタミンシンターゼ(Snustad 等,Genetics 120: 1111-1114(1988
))、ダイズ−ピロリン−5−カルボキシレートレダクターゼ(Delauney等,Mo
l.Genet.221: 299-305(1990))、トウモロコシジヒドロジピコリネートシン

.2; 417-422(1992))をコードするものを包含する植物遺伝子を単離するために
使用されてきた。
ADSS活性を欠失していると考えられる微生物突然変異体は利用可能である
(例えばCGCS5408と表記される大腸菌PurA突然変異体およびエール大学
の大腸菌ジェネティック・ストック・センター(E.coli Genetic Stock Center
)からの大腸菌CGCS4431および7039株; Dorfman,Genetics 61: 3
77-389(1969))に報告されている酵母ade12 突然変異体)。しかしながら、こ
れらの突然変異体が利用できるにもかかわらず、ADSS酵素活性をコードする
cDNAを単離するための補足性技術の適用は、鳥(Powell等,FEBS Lett.303
: 4-10(1992))およびバチラス・サブチリスADSS(Maentsaelae およびZa
lkin,J.Bacteriol.174: 1881-1890(1992))に対して不成功であることが示
されている。
ADSS、特に真核生物ADSS遺伝子に適用される場合に、この補足性戦略
の失敗を説明し得るいくつかの理由がある。第1に、例えば微生物宿主の利用優
位性と一致しないコドン利用性のために、真核生物ADSScDNA配列が突然
変異微生物において適度なレベルまで発現され得ないことである。第2に、例え
ば微生物により行われない翻訳後の修飾、例えばグリコシル化を活性が必要とす
る場合、クローン化された真核生物のコード配列からの一次的翻訳産物が機能的
ポリペプチドを産生し得ないことである。第3に、大腸菌内に発現される異種タ
ンパク質が発現される細胞に致死的であり得、結果的にその単離を不可能にする
ことである。第4に、例えば微生物宿主が特異的に欠いている特定のオルガネラ
膜系にタンパク質が通常標的化されている場合、真核生物タンパク質が微生物宿
主においてその活性コンホメーションを呈し得ないことである。この最後の可能
性は、補足性アッセイにおいて使用される微生物宿主に存在しないオルガネラを
有する植物細胞に存在しており(Schubert,Annu.Rev.Plant Physiol.37: 53
9-574(1986))、そしてN−末端の導入配列および成熟ポリペプチドの固有の特
性の両方を包含する翻訳後の標的化機構の結果としてオルガネラ系におそらく達
する(例えばKohormおよびTobin,Ptant Cell 1: 159(1989); Li 等,Plant Cel
l 3: 709(1991); Li等,J.Biol.Chem.267: 18999(1992))植物ADSS酵素
に対して特に当てはまりそう
である。さらに、植物から単離された2つのその他のプリン生合成遺伝子、5’
−ホスホリボシル−5−アミノイミダゾールシンテターゼ(SenecoffおよびMaeg
her,Plant Physiol.102; 387-399(1993)およびグリシンアミドシンテターゼ
(Schnorr 等,Plant J.6: 113-121(1994)はまた葉緑体に標的化されるタンパ
ク質をコードすると考えられている。
従って、植物供給源からのアデニロコハク酸シンテターゼ(ADSS)酵素を
コードするDNA分子を同定および単離することが本発明の主な目的の一つであ
る。この目的は本発明の範囲内で達成され得る。
従って、本発明は、植物供給源からのアデニロコハク酸シンテターゼ(ADS
S)酵素をコードする単離されたDNA分子を提供する。
アラビドプシス・タリアナ(Arabidopsis thaliana)、ゼア・メイズ(Zea ma
ys)およびコムギにおけるADSS酵素のためのDNAコード配列は、それぞれ
配列番号:1,3および5に示されている。本発明により提供される情報を用い
て、あらゆる植物供給源からのアデニロコハク酸シンテターゼ(ADSS)酵素
のためのDNAコード配列は標準的方法を用いて今では得られることができる。
本発明は従って、アデニロコハク酸シンテターゼ(ADSS)活性を有する植
物から、好ましくは双子葉または単子葉植物から、より好ましくはアラビドプシ
ス種、
トウモロコシまたはコムギ植物からのタンパク質をコードする単離されたDNA
分子に関する。
特に、本発明は、アラビドプシス・タリアナ、ゼア・メイズおよびコムギにお
けるADSS酵素のためのコード配列を含む単離されたDNA分子に関し、該酵
素はそれぞれ配列番号:2,4および6に示されたアミノ酸配列からなる。
本発明はさらに、アデニロコハク酸シンテターゼ(ADSS)活性を有する植
物から、好ましくは双子葉または単子葉植物から、より好ましくはアラビドプシ
ス種またはトウモロコシおよびコムギ植物からのタンパク質をコードするDNA
分子に操作可能に連結されたプロモーターからなる発現カセットに関する。
本発明のその他の対象は、上記発現カセットを含有する組換えベクターであり
、該ベクターは宿主細胞中に安定に形質転換され得る。また、上記ベクターで安
定に形質転換された宿主細胞もまた包含され、該宿主細胞は好ましくはバクテリ
ア細胞、酵母細胞および昆虫細胞からなる群から選択される細胞であり、そして
さらに本発明に係るDNA分子を発現することができる。
本発明はまた、ADSS酵素の組換え産生を包含する。
特に、本発明は、
(a)選択された宿主のために設計された発現カセット内にアデニロコハク酸シ
ンテターゼ(ADSS)活性を有するタンパク質をコードするDNA配列を挿入
し;
(b)適当な読み枠内に連結された個々の要素を含有する得られた分子を、宿主
細胞内に形質転換され得るベクター内に挿入し;
(c)このようにして形質転換された宿主細胞を適当な培養培地中で増殖させ;
そして
(d)形質転換された細胞もしくは培養培地またはそれらの両方からタンパク質
産生物を単離し、そしてそれを精製する
ことからなるアデニロコハク酸シンテターゼ(ADSS)活性を有するタンパク
質を宿主生物内で製造する方法に関する。
また、組換えにより産生されたADSSを使用する方法も本発明に包含される
。特に、本発明は、ADSSの活性に影響を及ぼす新規除草剤をスクリーニング
するために精製されたADSSの使用方法を提供する。
好ましいのは、植物からのADSS酵素の活性を阻害する性質に対して化学物
質をアッセイするための方法であって、
(a)第1の反応混合物中に上記ADSS酵素を、上記ADSS酵素がアデニロ
コハク酸の合成を触媒し得る条件下で一緒にし;
(b)第2の反応混合物中に上記化学物質および上記ADSS酵素を、上記第1
の反応混合物の場合と同じ条件下で一緒にし;そして
(d)上記第1および第2の反応混合物中で産生された
アデニロコハク酸の量を比較する
ことからなる方法であり、上記第2の反応混合物中のアデニロコハク酸の量が上
記第1の反応混合物中のアデニロコハク酸の量に比べ顕著に少ないならば、上記
化学物質は上記ADSS酵素の活性を阻害し得る。
本発明はさらに、植物アデニロコハク酸シンテターゼ遺伝子またはmRNAに
特異的にハイブリダイゼーションし得るプローブに関し、該プローブは、長さが
少なくとも10ヌクレオチドの植物からのアデニロコハク酸シンテターゼ酵素に
対するコード配列の隣接部分を含有する。
本発明のその他の態様は、アデニロコハク酸シンテターゼ(ADSS)活性を
有するタンパク質をコードするDNA部分を含有するDNA分子を製造する方法
であって、
(a)植物アデニロコハク酸シンテターゼ遺伝子またはmRNAに特異的にハイ
ブリダイゼーションし得るヌクレオチドプローブであって、該プローブは、長さ
が少なくとも10ヌクレオチドの植物からのアデニロコハク酸シンテターゼ酵素
に対するコード配列の隣接部分を含有する上記プローブを調製し;
(b)段階(a)に従って調製されたヌクレオチドプローブを用いて、選択され
た生物からのクローン化ゲノムDNA断片またはcDNA断片の集合体中で、そ
の他のADSSコード配列をブローブ(探針)し;そして
(c)アデニロコハク酸シンテターゼ(ADSS)活性を有するタンパク質をコ
ードするDNA部分を含有するDNA分子を単離する
ことからなる。
本発明はさらに、ADSS遺伝子の存在およびその形態を検出する方法、およ
び生物(器官)中のADSS転写物のレベルを定量する方法を具体化する。これ
らの方法はADSS酵素の変更された形態またはADSS酵素発現の変更された
レベルと関連する疾病状態を診断するために使用され得る。
一つの面において、本発明は、IMPからアデニロコハク酸の合成を触媒する
酵素であるアデニロコハク酸シンテターゼ(本明細書では「ADSS」と記載)
の真核生物の形態をコードする単離されたDNA分子に関する。アラビドプシス
・タリアナからのADSS酵素に対するDNAコード配列および相当するアミノ
酸配列はそれぞれ配列番号:1および2として提示されている。トウモロコシ(
メイズ)ADSS酵素に対するDNAコード配列および相当するアミノ酸配列は
それぞれ配列番号:3および4として提示されている。コムギADSS酵素に対
するDNAコード配列および相当するアミノ酸配列はそれぞれ配列番号:5およ
び6として提示されている。
ADSS酵素をコードするDNAは本発明に従って望ましいあらゆる植物種の
ゲノムから単離され得る。植物ADSSコード配列を単離するために教示される
一方法
は実施例1に提示されている。この方法において、ADSS酵素をコードするc
DNAクローンは、ADSS酵素活性を該活性を欠失している突然変異宿主生物
に供給する能力に基づいて当該真核生物から誘導されるcDNAクローンのライ
ブラリーから同定される。この方法に使用するための適当な宿主生物は、cDN
A発現ライブラリーをスクリーニングするために使用され得るものであり、その
ためのADSS活性を欠失する突然変異体は利用可能であるか、または一般的に
作出され得る。そのような宿主生物は大腸菌および酵母を包含するが、これらに
限定されない。
また、植物ADSSコード配列は、本発明により教示されるアラビドプシス・
タリアナ(配列番号:1)、ゼア・メイズ(配列番号:3)またはコムギ(配列
番号:5)ADSSコード配列に対するそれらの配列相同性に基づいて十分に公
知の技術に従って単離され得る。これらの技術において、公知ADSSコード配
列の全てまたは一部は、選択された生物からのクローン化ゲノムDNA断片また
はcDNA断片の集合体(すなわちゲノムまたはcDNAライブラリー)中に存
在するその他のADSSコード配列に選択的にハイブリダイゼーションするブロ
ーブとして使用される。そのような技術は平板培養されたDNAライブラリーの
ハイブリダイゼーションスクリーニング(プラークまたはコロニー;例えばSamb
rook等,“Molecular Cloning”,Cold Spring Harbor Labo
ratory Press編,(1989)参照)および公知ADSSアミノ酸配列の中で保存され
た配列ドメインに相当するオリゴヌクレオチドプライマーを用いるPCRによる
増幅(例えばInnis 等,“PCR Protocols,a Guide to Methods and Applicatio
ns”,Academic Press刊(1990))を包含する。これらの方法は、プローブ配列
が誘導される生物に密接に関連する生物からのADSSコード配列の単離に特に
よく適している。従って、アラビドプシス、ゼア・メイズまたはコムギのコード
配列をプローブとして用いる上記方法の適用は、その他の植物種からADSSコ
ード配列を単離するために特によく適していることが期待される。
本発明により教示される単離された植物ADSS配列はあらゆる所望の目的に
適合する標準的遺伝子操作技術に従って操作され得る。例えば、全体のADSS
配列またはそれらの一部はADSSコード配列およびメッセンジャーRNAに特
異的にハイブリダイゼーションし得るプローブとして使用され得る。様々な条件
下で特異的なハイブリダイゼーションを達成するために、上記プローブはADS
Sコード配列の中でユニークである配列を包含し、そして少なくとも10ヌクレ
オチドの長さ、好ましくは少なくとも20ヌクレオチドの長さ、そして最も好ま
しくは少なくとも50ヌクレオチドの長さである。上記プローブはポリメラーゼ
連鎖反応(複製連鎖反応,PCR)の十分に公知の方法によりADSSコード配
列
を増幅および/または分析するために使用され得る。この技術は所望の生物から
追加のADSSコード配列を単離するために、また生物内にADSSコード配列
の存在を決定するための診断アッセイとして有用であるかもしれない。この技術
はまた、ある特定の条件、例えば除草剤耐性、AMP欠乏、健康不良等と関連す
る植物において、変更されたADSSコード配列の存在を検出するために使用さ
れ得る。
ADSS特異的ハイブリダイゼーションブローブはまた、ゲノムADSS配列
に対するプローブの選択的ハイブリダイゼーションに基づく標準的技術を用いて
選択された植物のゲノム内の天然ADSS遺伝子の位置をマップ(地図作成)す
るために使用され得る。これらの技術はADSSプローブ配列内に同定または包
含されるDNA多形の同定、および2種の多形の親系列のハイブリッドの自家受
粉から誘導されるマッピング集合体における公知マップ(地図)位置のその他の
マーカー(標識)に対するADSS遺伝子の分離を追跡するための上記多形の利
用を包含するが、それらに限定されない(例えばHelentjaris 等,Plant Mol.B
iol.5: 109(1985); Sommer等,Biotechniques 12: 82(1992); D'Ovidio 等,Pl
ant Mol.Biol.15: 169(1990))。あらゆる植物ADSS配列がADSS遺伝
子をマップするためのプローブとして有用であると考えられているが、好ましい
プローブは選択された植物種により密接に関連する植物種から
のADSS配列であり、そして最も好ましいプローブは選択された植物種からの
ADSS配列である。この方法でのADSS遺伝子のマッピングは育種目的に特
に有用であると考えられる。例えば、除草剤耐性を付与する突然変異ADSS遺
伝子の遺伝子地図上の位置を知ることにより、フランキングDNAマーカーが参
照遺伝子地図から同定され得る(例えばHelentjaris,Trends Genet.3: 217(19
87))。除草剤耐性の形質を新規育種系列に遺伝子移入する間に、これらのマー
カーは戻し交雑の各回の後に反復親に依然として存在するADSS連結フランキ
ング染色体DNAの範囲を追跡するために使用され得る。
ADSS特異的ハイブリダイゼーションプローブはまた、標準的技術、例えば
ノーザンブロット分析を用いて植物中のADSSmRNAのレベルを定量するた
めに使用され得る。この技術は、特定の状態、例えばアデニロコハク酸の欠乏に
関連し得るADSS発現の変更されたレベルまたはAMPレベルまたはADSS
を標的とする除草剤に対する高められた許容性を検出するための診断アッセイと
して有用であり得る。
宿主生物内での酵素の組換え産生のために、植物ADSSコード配列は選択さ
れた宿主のために設計された発現カセット中に挿入され、そしてそれが組換えに
より産生される宿主中に導入され得る。特定の制御配列、例えば選択された宿主
に適当であるプロモーター、シグナル
配列、5’および3’非翻訳配列およびエンハンサーの選択は当業者の通常の知
識のレベルの範囲内である。適当な読み枠内に連結されて個々の要素(エレメン
ト)を含有する生成分子は宿主細胞中に形質転換されることができるベクター中
に挿入されてもよい。適当な発現ベクターおよびタンパク質の組換え産生のため
の方法は宿主生物、例えば大腸菌(例えばStudier およびMoffatt,J.Mol.Bio
l.189: 113(1986);Brosius,DNA 8: 759(1989)参照)、酵母(例えばSchneide
r およびGuarente,Meth.Enzymol.194: 373(1991)参照)および昆虫細胞(例
えばLuckowおよびSummers,Bio/Technol.6: 47(1988)参照)に対して十分に公
知である。特定の例はプラスミド、例えばpBluescript(Stratagene
,La Jolla,CA)、pFLAG(International Biotechnologies,Inc.,New H
aven,CT)、pTrcHis(Invitrogen,La Jolla,CA)およびバキュロウ
イルス発現ベクター、例えばオートグラフィカ・カリフォルニカ核多角体病ウイ
ルス(AcMNPV)のゲノムから誘導されるものを包含する。好ましいバキュ
ロウイルス/昆虫系はpVI11392/Sf21細胞(Invitrogen,La Jolla
,CA)である。
組換えにより産生された植物ADSS酵素は種々の標準的技術を用いて単離お
よび精製され得る。使用され得る実際の技術は使用される宿主生物、ADSS酵
素が分泌のために設計されているか否か、および当業者に周知
のその他のそのようなファクターに応じて変更し得る(例えばAusubel,F.等,
“Current Protocols in Molecular Biology”,John Wiley & Sons,Inc.刊(
1994)の第16章参照)。
組換えにより産生される植物ADSS酵素は様々な目的のために有用である。
例えば、アデニロコハク酸を試験管内で合成するためにADSS酵素活性を供給
するために使用され得る。アデニロコハク酸の試験管内合成はIMP,GTPお
よびアスパラギン酸塩を、2価カチオン、例えばMg2+を含有する適当な緩衝液
中ADSS酵素の存在下で反応させることにより行われ得る(例えばBaugher 等
,Biochem.Biophys.Res.Commun.94: 123-129(1988); Stayton等,Curr.Top
.Cell.Regul.22: 103-141(1983); Bass等,A-rch.Biochem.Biophys.256:
335-342(1987))。産生されるアデニロコハク酸は他の供給源からの従来市販
の精製アデニロコハク酸の代替物として使用され得る有用な試薬である。
組換えにより産生される植物ADSS酵素は、その標的が同定されておらずA
DSSを阻害するか否かが決定されていない公知除草性化学物質をスクリーニン
グするための試験管内アッセイにおいても有用であり得る。そのような試験管内
アッセイはまた、ADSS活性を阻害し、そしてそれ故に除草剤の候補体である
化学物質を同定するためのより一般的なスクリーニングとして使用され得る。ま
た、組換えにより産生されるADSSはこの
酵素の複雑な構造を解明するためにも使用され得る。ADSS酵素の構造に関す
るそのような情報は、例えば新規な阻害性除草剤の合理的な設計に使用され得る
。
典型的には、ADSSに対する阻害効果は試験管内アッセイにおけるアデニロ
コハク酸合成の低下または完全阻害により決定される(例えばBaugher 等,Bio
chem.Biophys.Res.Commun.94: 123-129(1980); Stayton等,Curr.Top.Ce
ll.Regul.22: 103-141(1983); Bass等,Arch.Biochem.Biophys.256: 335-
342(1987)参照)。そのような決定は候補阻害剤の存在下および不在下での試験
管内アッセイにおいて合成されるアデニロコハク酸の量を単に比較することによ
りなされ得る。化学物質の存在下で合成されるアデニロコハク酸シンテターゼの
量が不在下で合成される量に比べ顕著に少ないならば、その化学物質はADSS
