オリゴヌクレオチドの合成

技術分野
本発明は、35S標識3H−1,2−ベンゾジチオール−3−オン−1,1−ジ
オキシド(1)の合成、および部位特異的な35S標識オリゴヌクレオチドを作製
する際のその合成方法に関する。
従来の技術
ZamecnikおよびStephenson、Proc.Natl.Acad.Sci.USA 75,280-284(1978
)が、合成オリゴヌクレオチドによるウイルス複製阻害を最初に示してから、治
療剤としてのオリゴヌクレオチドに大きな関心がよせられた。最近、治療剤とし
て、また遺伝子発現調節剤としてのオリゴヌクレオチドの開発が急増した。最も
大きな開発は、標的mRNAでワトソン‐クリック二重鎖を形成する、いわゆる
アンチセンスオリゴヌクレオチドの用途におけるものである。Agrawal、Trends
in Biotechnology 10, 152-158(1992)では、抗ウイルス剤としてのアンチセン
スオリゴヌクレオチドの開発を広く検討する。Uhlmannおよびpeymann、Chem.Re
v.90,543(1990)も参照さ
れたい。
このような目的のオリゴヌクレオチドの合成に関し種々の方法が開発された。
概して、Methods in Molecular Biology,20巻、protocols for Oligonucleotid
es and Analogs (S.Agrawal編、Humana Press,1993)、Oligonucleotides and
Analogues: A Practical Approach(F.Eckstein編、1991)、UhlmannおよびPey
manの上記文献を参照されたい。初期の合成アプローチには、リン酸ジエステル
およびリン酸トリエステル化合物が含まれた。オリゴヌクレオチド合成のための
リン酸ジエステル化合物が、Khoranaら、J.Molec.Biol.72,209(1972)に、
オリゴヌクレオチドおよびポリヌクレオチドの合成のためのリン酸トリエステル
化合物がReese、Tetrahedron 34,3143-3179(1978)に開示されている。これら
の初期のアプローチは、合成により効果的なホスホロアミダイトおよびH−ホス
ホネートアプローチに大部分取って代わられた。BeaucageおよびCaruthers、Tet
rahedron Lett.22,1859-1862(1981(BeaucageおよびIyer、Tetrahedron 48,
2223(1992)にてレビュー)は、ポリヌクレオチド合成中にデオキシヌクレオシ
ドホスホロアミダイトを使用することを開示する。AgrawalおよびZamecnikの米
国特許第5,149,798 号(1992)には、H−ホスホネートアプローチによるオリゴ
ヌクレオチドの最適化された合成が開示されている。
これらの現代的アプローチは、両方とも、種々の修飾
ヌクレオチド間連結を示すオリゴヌクレオチドを合成するのに使用されている。
AgrawalおよびGoodchild、Tetrahedron Lett.28,3539-3542(1987)は、ホス
ホロアミダイト化合物を用いたオリゴヌクレオチドメチルホスホネートの合成を
報告する。Connollyら、Biochemistry 23,3443(1984)には、ホスホロアミダ
イト化合物を用いたオリゴヌクレオチドホスホロチオエートの合成が開示されて
いる。Jagerら、Biochemistry 27,7237(1988)には、ホスホロアミダイト化合
物を用いたオリゴヌクレオチドホスホロアミダイトの合成が開示されている。Ag
rawalら、Proc.Natl.Acad.Sci.USA 85,7079-7083(1988)には、H−ホス
ホネート化合物を用いたオリゴヌクレオチドホスホロアミダイトおよびホスホロ
チオエートの合成が開示されている。
ホスホロアミダイト化合物(BeaucageおよびCaruthers、Tetrahedron Lett.2
2,1859-1862(1981)、およびBeaucageおよびIyer、Tetrahedron 48,2223-231
1(1992))を用いた接合の際の硫化剤(Iyerら、J.Am.Chem.Soc.112,1253
-1254(1990))としての3H−1,2ベンゾジチオール−3−オン−1,1ジ
オキシド(1)の使用は、今日では、種々のオリゴヌクレオシドホスホロチオエ
ートの慣例の合成および大量生産として十分に確立されている。メチルホスホノ
チオエートおよび他の類似体の合成に、この試薬を使用することが報告されてい
る。Padmapriyaら、Antisense Res.& Dev.4,185-1
99(1994)およびAndradら、Bioorg.& Med.Chem.Lett.pp.2017-2022(1994
)。生物学上の研究において、35S標識オリゴヌクレオシドホスホロチオエート
は、代用化合物、すなわちH−ホスホネート化合物を使用して製造される。Gare
ggら、Chem.Scr.25,280-282(1985)。しかしながら、H−ホスホネート化合
物を用いてホスホロチオエートの部位特異的標識を行うのは困難であり、(a)
混合リボヌクレオチド−デオキシリボヌクレオチド集団(「ハイブリッドオリゴ
」)、(b)不均一な骨格、例えばデオキシリボヌクレオチド−メチルホスホネ
ート(「キメラオリゴ」)および(c)混合ホスホロジエステル−ホスホロチオ
エート(PO−PS)骨格等との35S標識オリゴヌクレオシドホスホロチオエー
ト構築物の作製を行うには不都合である。立体化学的に「一様な」生成物を確実
にするためには、合成および生物学的評価の両方に同一の化合物を用いることが
望ましい。硫黄元素を使用することの欠点も認識されている。Iyerら、J.Am.C
hem.Soc.112,1253-1254(1990)およびIyerら、J.Org.Chem.55,4693-469
8(1990)。
薬理学上の化合物、例えばアンチセンスオリゴヌクレオチドホスホロチオエー
トのin vivo薬物動態学の研究(Agrawalら、Proc.Natl.Acad.Sci.U.S.A.88
,7595-7599(1991))では、検出可能にするために化合物を標識することが必
要である。35S標識は、確立された広く知れ渡った技術である。35S標識オリゴ
ヌクレオシド
ホスホロチオエート構築物を合成する上での前述の難点から、改善された方法が
望まれている。
発明の概要
本発明は、35S標識オリゴヌクレオシドホスホロチオエートを合成するための
新規化合物および改善方法を提供する。本発明は、いくつかの態様からなる。第
1の態様では、本発明は、35Sで標識されたオリゴヌクレオチドホスホロチオエ
ートを合成するために有用な新規化合物を提供する。この化合物の35S−3H−
1,2−ベンゾジチオール−3−オン−1,1ジオキサイド(1)は、構造式
(式中、アステリスクは35S標識を示す。)で表される。
本発明の第2の態様では、35S−3H−1,2−ベンゾジチオール−3−オン
−1,1ジオキサイド(1)を合成する新規方法を提供する。この方法の重要性
は、種々の35S標識オリゴヌクレオチドホスホロチオエートが製造でき、それに
よりこれらの化合物の薬物動態学の研究が促進できることにある。
35S−3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサイド(1
)を合成する方法を図2に示すが、この方法は、まず35S−チオ安息香酸(4)
をチオ
サリチル酸(o−メルカプト安息香酸)(5)と接触させて、縮合物である35S
−3H−1,2−ベンゾジチオール−3−オン(2)を得ることからなる。次い
で、35S−3H−1,2−ベンゾジチオール−3−オン(2)を酸化させて所望
の生成物、すなわち、35S−3H−1,2−ベンゾジチオール−3−オン−1,
1ジオキサイド(1)を得る。
本発明の第3の態様では、35S標識オリゴヌクレオチドを合成する新規方法を
提供する。この方法は、35S−3H−1,2−ベンゾジチオール−3−オン−1
,1ジオキサイド(1)を、酸化的硫化されやすいオリゴヌクレオチドと接触さ
せることからなる。ホスホロアミダイト法(phosphoramidate method)を介して
合成されたオリゴヌクレオチドを35S標識化する方法を図3に示す。しかしなが
ら、アルキルおよび/またはアリールホスファイトを酸化的硫化により対応する35
S標識アルキルおよび/またはアリールホスホノチオエートを得るというよう
な他の方法は検討中である。
当業者であれば、35S−3H−1,2−ベンゾジチオール−3−オン−1,1
ジオキサイド(1)が、その未標識の類似体である3H−1,2−ベンゾジチオ
ール−3−オン−1,1ジオキサイドが使用できるいかなる目的および方法にも
、使用できることが理解されよう。
前述の記載は、本発明のいくつかの態様を単に要約するものであって、本発明
をいかなる様にも限定すること
を意図するものでなく、また解釈されるべきものでもない。
本明細書で引用される全ての特許および他の文献はここで全体にて参照により
組み込まれる。

好ましい実施例の説明
治療剤としてのアンチセンスオリゴヌクレオチドへの関心が相変わらず増加し
ているため、これらの化合物の薬物動態学上の特性が試験できる方法を提供する
必要がある。バイオ分布とともに半減期および分解産物を測定する必要がある。
これらの作業を成し遂げる1つの方法は、オリゴヌクレオチドを、生物学的化合
物を追跡したり検出したりするのに使用される一般的なアイソトープ標識である35
Sで標識することである。
本発明は、35S標識アンチセンスオリゴヌクレオチドを合成するのに有用な新
規化合物、その化合物を合成するための新規方法、およびオリゴヌクレオチドを35
S標識するための新規方法を提供する。
本発明の第1の態様は、以下の構造式
(式中、アステリスクは35S放射性核種の位置を示す。)を示す新規化合物35S
−3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサイド(1)から
なる。
非放射性標識類似体である3H−1,2−ベンゾジチオール−3−オン−1,
1ジオキサイドは公知であるが(例えば、Beaucage,ReganおよびIyerの米国特
許第5,003,097号(Beaucageら、'097)およびIyerら、J.Am.Chem.Soc.および
J.Org.Chem.,上述文献)、35S標識化合物は、まだ合成されていない。
当業者であれば、35S−3H−1,2−ベンゾジチオール−3−オン−1,1
ジオキサイド(1)が、その非放射性標識の対応物と同じ目的および方法で使用
できることが理解されよう。本発明の第2の態様は、35S−3H−1,2−ベン
ゾジチオール−3−オン−1,1ジオキサイドを合成する新規方法からなる。こ
の方法は、例えば、Beaucageら、'097の方法の応用例である。この方法の重要な
利点は、35S−3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサイ
ド(1)の生成を可能にすることである。先行技術の方法と対比的に、本発明の
方法は、35S標識が容易に組み込まれる反応物を使用する。
先行技術では、硫酸中で2−チオール安息香酸およびチオール酢酸を混合する
ことにより、3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサイド
の前駆体である、3H−1,2−ベンゾジチオール−3−オンが生成されること
が教示される。例えば、Beaucageら、'097。この方法を使用して、最終生成物で35
Sを適切な位置に有するためには、35Sをチオール酢酸中に組み込
むことが必要である。我々は、報告された手段で、チオール酢酸および35S元素
の間の高温(125℃)交換反応により、35Sチオール酢酸の製造を試みた。Ka
wamuraら、Chem.Lett.1231-1234(1975)。しかし、高い比活性を有する35S
(32mCi/μモル)を使用しても、チオール酢酸の揮発性(沸点81℃)お
よび蒸気圧が高いことが問題となって、高い比活性有する35S標識チオール酢酸
は単離できなかった。
チオール酢酸を35S標識しようとした際に遭遇した困難を解消するために、沸
点がより高く、蒸気圧がより低いチオール酸を探求したところ、市販のチオ安息
香酸(4)が理想的候補であるように思えた。35S−3H−1,2−ベンゾジチ
オール−3−オン−1,1ジオキサイド(1)の製造に35S−チオ安息香酸(4
)を使用する前に、3H−1,2−ベンゾジチオール−3−オン−1,1ジオキ
サイド(1)の前駆体である35S−3H−1,2−ベンゾジチオール−3−オン
(2)を製造することにより、35S−3H−1,2−ベンゾジチオール−3−オ
ン−1,1ジオキサイド(1)の製造におけるチオ安息香酸(4)の使用の有用
性を確認した(図2)。いかなる理論にも縛られることは望まないし、実際、こ
の合成方法は、いかなる理論にも依存するものではないが、35S−3H−1,2
−ベンゾジチオール−3−オン(2)は、中間体(3):
を経て形成されることが推測される(McKibbenおよびMaClelland,J.Chem.Soc
.170-173(1923))。収率は、チオール酢酸を使用する(約80%)より幾分
低く(約60%)、反応の完了にはより長い時間(4時間)が必要とされた。こ
のように合成された結晶化した35S−3H−1,2−ベンゾジチオール−3−オ
ン(2)のサンプルは、チオール酢酸を使用する報告された方法(Iyerら、J.A
m.Chem.Soc.112,1253-1254(1990)およびIyerら、J.Org.Chem.55,4693
-4698(1990))により得られるものと全ての点で同一であった(融点、1H−N
MRおよび13C−NMR)。35S−3H−1,2−ベンゾジチオール−3−オン
(2)の製造に35S−チオ安息香酸(4)を使用することの実行可能性を示し、35
S−チオ安息香酸(4)は高い放射化学的収率(78%)で都合よく製造され
た。こうして得られた35S−チオ安息香酸(4)は、35S−3H−1,2−ベン
ゾジチオ−ル−3−オン(2)に変換され、トリフルオロ酢酸中の過酸化水素を
使用して、注意深く制御して酸化された(図2)。Iyerら、J.Am.Chem.Soc.
112,1253-1254(1990)およびIyerら、J.Org.Chem.55,4693-4698(1990)
。過剰なH2O2の使用を避けることが重要であ
る。所望の生成物35S−3H−1,2−ベンゾジチオール−3−オン−1,1ジ
オキサイド(1)は、30%化学収率(使用された5の量に対し)で白色結晶固
体として得られ、90μCi/μモルの比活性を示した。反応混合物は、35S−
3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサイド(1)の分解
を避けるために、完了後ただちに使用されるべきである。
このように、本発明のこの態様による合成方法は、第1に35Sチオ安息香酸(
4)をチオサリチル酸(5)と接触させて、縮合物35S−3H−1,2−ベンゾ
ジチオール−3−オン(2)を得ることよりなる。この反応は酸で触媒される。
任意の適切な強酸が使用されうるが、好ましい実施例では、硫酸が使用される。
その後、35S−3H−1,2−ベンゾジチオール−3−オン(2)を酸化して、
所望の生成物である35S−3H−1,2−ベンゾジチオール−3−オン−1,1
ジオキサイド(1)を得る。任意の適切に強い酸化剤、例えば過酸化水素および
トリフルオロ酢酸、トリフルオロペルオキシ酢酸または、オキソン(oxone)、
過ヨウ素酸ナトリウム、NaOCl、RuCl3のような他の酸化剤、およびス
ルフィドをスルホンに酸化する際に使用される試薬が使用できる。例えば、M.H
udlicky,Oxidation in Organic Chemistry,ACS Monograph 186,1990を参照さ
れたい。好ましい実施例では、過酸化水素およびトリフルオロ酢酸を用いて酸化
を完了する。この概略を図2に表す。
本発明の第3の態様は、オリゴヌクレオチドを35S標識する新規方法からなる
。この方法は、任意の所望のヌクレオシド間連結に35Sを選択的に配置するのに
使用できる。1つから全部のヌクレオシド間連結のどこにでも35Sで標識できる
。この方法は、35S−3H−1,2−ベンゾジチオール−3−オン−1,1ジオ
キサイド(1)を酸化的硫化されやすいオリゴヌクレオチドと接触させることか
らなる。好ましい実施例では、オリゴヌクレオチドは、ホスホロアミダイト法に
より合成され、35S−3H−1,2−ベンゾジチオール−3−オン−1,1ジオ
キサイド(1)は、当業界で知られる標準条件下で1つまたはそれ以上のβ−シ
アノエチルホスホロトリエステルのヌクレオシド間連結を有するオリゴヌクレオ
チドと接触される。別の好ましい実施例では、1つまたはそれ以上のアルキルお
よび/またはアリールホスファイトのヌクレオチド間連結を有するオリゴヌクレ
オチドは、35S−3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサ
イド(1)と接触させて、対応する35S標識アルキルおよび/またはアリールホ
スホノチオエートを得る。この反応は、非放射性標識酸化的硫化剤を用いるもの
であり、Padmapriyaら、上述の文献により教示される。
当業者であれば、35S−3H−1,2−ベンゾジチオール−3−オン−1,1
ジオキサイド(1)が、未標識類似体である3H−1,2−ベンゾジチオール−
3−オ
ン−1,1ジオキサイドにより硫化され得る任意の化合物を35S標識するのに使
用できることが理解されよう。例えば、本発明の方法は、酸化的チオール化によ
り35S標識を組み込むことができる炭水化物、タンパク質、および任意の巨大分
子を35S標識することができる。したがって、ホスホペプチドのように、RNA
は部位特異的に35Sで標識することができる。ホスホロチオエートおよびリン脂
質、グリセロリン脂質、およびホスホ炭水化物の硫黄類似体(例えば、ホスホロ
ミオイノシトール、または他の巨大分子とのこれらの結合体)も、35Sで標識さ
れうる。35Sは、チオホスフェートおよびチオトリホスフェート(例えば、AT
P)に挿入され、化学的または酵素的リン酸化反応を用いて、任意の分子に組み
込まれうる。
以下の実施例は、例示目的にのみ提供されるものであって、いかなる様にも本
発明を限定することを意図するものでなく、また解釈されるべきでもない。
実施例
実施例1
35S−3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサイド(1
)の合成 35 S−3H−1,2−ベンゾジチオール−3−オン(2)の合成
35S(100μlのトルエン中の5mCi)(Amersham,
英国)と6.5μl(55μモル)の未標識チオ安息香酸(Aldrich,ウイスコ
ンシン、ミルウォーキー)の溶液とを、1.5mlのエッペンドルフ試験管に配
置し、内容物を97℃で5時間加熱した。溶液をアルゴン下で濃縮乾固し、5m
gのチオサリチル酸(Aldrich,ウイスコンシン、ミルウォーキー)を添加した
。反応混合物を0℃に冷却し、硫酸(98%、50ml、J.T.Baker,ニュー
ジャージー、フィリップスバーグ)を添加した。混合物を50℃で3時間保持し
た。得られた茶色の反応混合物を−78℃に冷却し、600μlの水を添加した
。塩化メチレン(4×3ml)(VWR,フィラデルフィア、ウエストチェスター
)で、その溶液を抽出し、有機層をNa2CO3(5%、2×2ml)(EM Scien
ce,ニュージャージー、ギブスタウン)で洗浄した。有機層をアルゴン流下で濃
縮乾固して、黄色固体を得た。その後、その物質を3mlの温ヘキサン(J.T.B
aker,ニュージャージー、フィリップスバーグ)に溶解し、遠心分離した後、上
清溶液をアルゴン下で濃縮乾固し、淡黄色固体を得た(4mg、44%収率)。
この物質は、さらに精製することなく次の段階に使用することができ、使用する
まで−20℃で貯蔵した。 35 S−3H−1,2−ベンゾジチオール−3−オン(2)からの35S−3H−1 ,2−ベンゾジチオール−3−オン−1,1ジオキサイド(1)の合成
共に0℃に冷却された4mgの35S−3H−1,2−
ベンゾジチオール−3−オン(2)(上述のとおり製造された)を含む1.5m
lのエッペンドルフ試験管に、25mlのトリフルオロ酢酸(Aldrich,ウイス
コンシン、ミルウォーキー)および12mlの30%過酸化水素を添加した。反
応混合物を42℃に温めた。約2時間(シリカゲル、クロロホルムを用いたTL
Cによりモニター、Iyerら、J.Org.Chem.)後、反応混合物を0℃に冷却し、
300mlの水を加えた。35S−3H−1,2−ベンゾジチオール−3−オン−
1,1ジオキサイド(1)の白色沈殿がただちに形成された。スラリーを遠心分
離し、沈殿物を水(2×300μl)で洗浄し、真空中で乾燥させ、2mgの35
S−3H−1,2−ベンゾジチオール−3−オン−1,1ジオキサイド(1)を
得た(総活量350μCi、比活性90μCi/μモル)。
無水アセトニトリル中の35S−3H−1,2−ベンゾジチオール−3−オン−
1,1ジオキサイド(1)の溶液(20mg、200mlアセトニトリル中90
μCi/μモル)を以下に記載の酸化的硫化反応に使用した。溶液は、使用まで
は−20℃で貯蔵した。
実施例2
35S−標識オリゴヌクレオチドの合成
オリゴヌクレオチドの作製の際の35S−3H−1,2−ベンゾジチオール−3
−オン−1,1ジオキサイド(1)の使用を示すために、ホスホロアミダイト化
合物
を用いて自動化DNA合成機で0.1μモル規模で35S−d[TpsT](ここ
で、「ps」は、ホスホロチオエートヌクレオシド間連結を意味する)を作製し
た。BeaucageおよびCaruthers,Tetrahedron Lett.22,1859-1862(1981)およ
びBeaucageおよびIyer,Tetrahedron48,2223-2311(1992)。35S標識を組み込
むために、ヌクレオチド間のホスファイト連結の形成後、合成サイクルを中断し
た。CPGをカラムから取り出し、溶液(15ml、90μCi/μモル、30
分)で処理した後、「冷」(すなわち、非35S標識)3H−1,2−ベンゾジチ
オール−3−オン−1,1ジオキサイドの溶液(アセトニトリル中2%、100
ml、10分)で処理した。「非放射性」の35S−3H−1,2−ベンゾジチオ
ール−3−オン−1,1ジオキサイド(1)を使用し、厳格な条件下で作製され
た(TpsT)のサンプルは、TpsTの変換が>99%であることを示した。
酸化的硫化は、合成中行われた「トリチルアッセイ」(trityl assays)によ
り定めたとおり定量的であった。Agrawal,Protocols in Molecular Biology)
上述文献。水酸化アンモニウム水溶液でCPGからの開裂およびリン酸脱保護(
30%、2時間、35℃)後、ポリアクリルアミドゲル電気泳動(PAGE、2
0%)により2量体を調べた。オートラジオグラフ画像をそのUV造影バンドに
重ねることができた。放射性化学的検知器と接続したUV検知器を使用する逆相
HPLCにかけると、R
p−35S−d[TpsT](保持時間=22.7分)およびSp−35S−d[T
psT](保持時間=24.0分)の保持時間に対応するUV吸収ピークにその
活量プロフィールを重ねられた(図4、パネルA)。(「Rp」および「Sp」
は、リンのキラル中心での2つの立体配置を表す)。フローシンチレーション解
析による検出は図4、パネルBに表される。8NV C18 4μ Radial
Pak(Waters,マサチューセッツ、ミルフォード)カートリッジカラム、緩
衝液A(0.1M CH3CO2NH4)と緩衝液B(80:20、CH3CN:0
.1M CH3CO2NH4)とからなる勾配(60分かけて100%A〜60%
B)を設けた溶液、1.5ml/分の流量比を用い、Radiomatic(Me
riden Ct.,マサチューセッツ)500TRv3.00放射化学検知機と接続し
たフォトダイオードアレイUV検知機を具備したWatersカラム(マサチュ
ーセッツ、ミルフォード)でHPLC解析を行った。
その後、所定の35S標識組み込み部位を有する多数のオリゴヌクレオチド(配
