【発明の詳細な説明】
大腸特異的還元に感受性のあるポリマーの製造方法
本発明は、(i)アゾ含有ポリマー、(ii)ジスルフィド含有ポリマー及び
(III)アゾ及びジスルフィド含有ポリマーよりなる群から選ばれた特定的還
元に感受性のあるポリマーを製造する方法に関し、この方法は、アゾ及び/又は
ジスルフィド含有α,ω−2官能性反応剤を適当なα,ω−2官能性コモノマー
と共重合させる段階を含む。
本発明に従う方法は、バクテリアにより人の大腸内の還元的媒質の中で短いイ
ンキュベーション時間の後に完全に各モノマー単位に減成又は分解され得る、ア
ゾ及びジスルフィド含有ポリマーを作ることを目的とする。上記ポリマーは薬剤
を胃腸管下部に位置特異的に送達するためのタブレットやカプセルを作るのに有
用である。
国際特許WO91/11175から、
(i) アゾ含有ポリマー、
(ii) ジスルフィド含有ポリマー及び
(iii)アゾ及びジスルフィド含有ポリマー
よりなる群から選ばれた大腸特異的還元に感受性のあるポリマーを製造する方法
の1つが既に公知である。
上記方法は、アゾ及び/又はジスルフィド含有α,ω−2官能性コモノマーを
下に示す反応式に従って共重合させる段階を含む:

Halはハロゲン基、例えばCl、Br であり、

Z は C=O、CH2-CH-OH、SO2であり、
そしてそれにより、
X と Y とは上記各式において相互交換可能である。
上記方法はそのα,ω−2官能性反応剤の変化により多くのアゾ及びジスルフ
ィド含有ポリマーの製造を許容する。最終的な物理化学的及び物理的諸性質(親
水性、熱的諸性質、レオロジー特性)は広く変化させることができる。
得られるポリマーの分子量はゲル透過クロマトグラフィーにより有利に決定す
ることができる。これは用いた反応混合物の中のモノマーのタイプ及び度合いを
調節することにより変化させることができる。
その発明の方法により作られるポリマーの分子量は、互いに反応する官能基の
モル比を修飾することにより調節される。高い分子量のポリマーのためには反応
性官能基の等量を使用すべきである。
胃腸管中での吸収を避けるにはポリマーの分子量は約1000以上でなければ
ならないこと、及び胃腸管中で使用するためのポリマーが10,000,000
までの平均分子量を有し得ることは当業者によく知られている(米国特許第4,
298,595号、第25欄14−33行参照)。
これらのポリマーは特に特異的薬剤送達系を作るのに好適化されている。この
後者はマトリックス型系又は保留型系であることができる。前者の場合にはアゾ
及び/又はジスルフィド含有ポリマーがその薬剤包含室の主要部である。後者の
部分においてそのアゾ及び/又はジスルフィド含有ポリマーは薬剤の充填された
芯を封入するのに用いられる。アゾ及び/又はジスルフィド含有ポリマーは口、
胃袋及び上部腸の各液体野中で安定であるために、この発明に従うポリマーは分
解することなく口、胃袋及び上部腸を通過することができる。胃腸管の下部にお
いてこの還元感受性ポリマーは場合により酵素により媒介されてその還元的媒質
によって非常にゆっくりと解裂され、分裂されてそれにより封入されている活性
薬剤はその分解の部位においては完全には放出されない。
セグメントに分かれたいくつかのジスルフィド含有ボリアミド及び還元的解裂
に適した親水性基が3,3’−ジチオビススクシニミジルプロピオネートとα,
ω−アミノ末端基のポリ(テトラメチレンオキシド)又はテトラエチレングリコ
ールジアミン(ジェファミンEDR−192)との縮重合によって調製された。
SHIME反応器、すなわち人の大腸内微生物性内容物をシミュレートするSimu
lating Human Intestinal Microbial Ecosystem 反応器から採取した15mlの
試料の中でいくつかのポリマーフィルムをインキュベートした。
また、特定の乾燥投与のカプセルを製造するための、ジスルフィドと親水性基
とを含む、セグメントに分かれたいくつかのポリウレタンを、4,4’−ジフェ
ニルメタンジイソシアネート(MDI)と、α,ω−ヒドロキシ末端基のプレポ
リマーと、及びジスルフィド含有の連鎖延長剤との反応によって合成した。ヒド
ロキシ末端のプレポリマーとしてはポリ(テトラメチレンオキシド)、ポリ(ブ
ロピレンオキシド)、ポリ(カプロラクトン)、ポリ(エチレンオキシド)、ポ
リ(エチレンオキシド/ポリプロピレンオキシド)を用いた。
同様に、アゾ−芳香族性の、及び親水性の各基を含む、セグメントに分かれた
ポリウレタンがm−キシリレンジイソシアネートと、m,m’−ジヒドロキシア
ゾベンゼン、ポリ(エチレングリコール)及び1,2−プロパンジオールの混合
物との反応により合成された。親水性の薬剤(FOY−305)をこれらの重合
物で作られたカプセルの中に封入し、或いはこれらの重合物で被覆した。これら
のカプセル又は被覆ペレットを、人腸管植物相(Flora)の培養を再現するSH
IME反応器の中でインキュベートした。還元感受性のポリマーのいくつかの試
料をいくつかの測定された時間的間隔で採取する。分子量の減成を、それら試料
についての透過クロマトグラフィーを実施することにより測定する。その薬剤は
これらのペレットから放出されたけれども、分子量は実質的に減少しなかった。
従って主鎖のアゾ基のアミン基への分解は存在しなかった。
1連のアゾ含有ポリアミド類をアゾ含有ジアシッドクロリドとオリゴマー性α
,ω−ジアミンとの縮重合により調製した。これらのポリマーを試験管内の還元
的媒質の中でインキュベートした。いくつかの試験によって、多くのアゾ含有ポ
リマーが人腸管植物相をシミュレートするSHIME反応器の中でのインキュベ
ーションの間に粘稠化して無色になることが示された。構造の変化を13C、ラマ
ン、UV及びGPCでの分析によってモニタした。これらのポリマーの観測され
た脱色は、必ずしもデグラデーションによる必要はなくて、そのアゾのヒドラゾ
型への部分的還元によることも可能である。
このアゾポリマーの還元的解裂のための熱力学的な諸条件を電位測定によって
決定した。
親水性のアゾ含有ポリアミドについてその還元的媒質の中でのインキュベーシ
ョンはそのアゾ結合の、アミンの形成を伴う還元に導くことが示された。
これらアゾポリマーの分子量は実質的に低下しない。アゾ基のヒドラゾ基への
部分的還元はゆっくりと起こり、そしてその主鎖のアゾ基のアミン基への分解は
存在しない。
インキュベーションの間にそれらポリマーは粘稠になり、そして無色になる。
再酸化の間にもとの色が再び現れ、そしてその分子量はもとの値に再び上昇する
。
これらの実験はこの型のポリアミドについてはアゾ結合のアミノ基へのデグラ
デーションが起こらないことを示している。色及び機械的諸性質の変化はそのア
ゾ基のヒドラゾ基への転化によるものである。このような発見はヨーロッパ特許
EP−A−0398472のKimura等のそれとよく一致する。かれらはP
olymer 33(1992)5294−5299に発表された報文の中で人
の腸内植物相の培養におけるアゾ含有ボリウレタンのインキュベーションによっ
ては分子量の減少が起こらないと言うことを示している。これらの結果からかれ
らはアゾ基のアミンへのデグラデーションは存在せず、ヒドラゾ基へのデクラデ
ーションしか存在しないと結論した。
本発明は、上記のポリマーの類似の還元的媒質の中でのインキュベーションに
よるアゾ又はジスルフィド含有ポリマーの還元的解裂のための熱力学的条件を改
善しようとするものである。
この改善を達成するために本発明は、請求の範囲第1項の上位概念部に記載し
た型の方法の1つを提案するものである。この方法は、
HX-R3-XH が親水性のコモノマーであり、その際
-XH は -NH2、-OH であり、
-XX- は -N=N-、-S-S-であり、
そして
R1、R2 は、場合によりメトキシ基により置換されている、アルキル、アリ
ール、アルキルアリール基であり、
R3 は全て置換されているか、又は置換されていないアルキリデン、アリー
リデン、アルキルアリーリデン、ポリエーテル、ポリエステルであり、
Halはハロゲン基、例えばCl、Br であり、
その際
であり、そして
Z は C=O、CH2-CH-OH、SO2であり、
そしてそれにより、
X と Y とは上記各式において相互交換可能である
ことを特徴とするものである。
本発明の特徴の1つのよれば、この方法はジチオビス(スクシニミジルプロピ
オナート)と、親水性のα,ω−2官能性コモノマーとの共重合を含む。
この還元感受性ポリマーは以下に記述する方法に従って作られる。出発物質の説明
ジェファミンED−600(Mn=600)(7)はTexaco Chem
ical Company(米国オースチン)から購入される市販製品である。
このポリマーはトルエン溶液からの共沸的な水の除去によって乾燥される。
アミン含有量は滴定により決定される:ジェファミンED600:2.96m
eq/g。
文献[23]に記述されているようにして対応するアゾベンゼン−4,4’−
ジカルボン酸(4)からアゾベンゼン−4,4’−ジアシッドタロリド(5)を
調製した。他の全ての反応剤はJanssen Chimica(ベルギー国、
Beerse)から入手した。
クロロホルムを硫酸及び水で洗浄し、次いで水素化カルシウムの上で乾燥させ
た。
トリエチルアミンをそれぞれトシルアミンとニンヒドリンとの反応により精製
した。
これを最終的に水素化カルシウムの上で乾燥した。方法
Beckmann IR 4230装置により赤外線スペクトルを記録した。
Kontron Uvikon 810ダブルビーム分光光度計により紫外線ス
ペクトルを測定した。
1H−NMRスペクトルを360MHzのBruker WH−360装置を
用いて記録した。
13C−NMRスペクトルを、Varian Unity−400分光光度計に
より100MHzにおいて5mmの広幅プローブを用いて求めた。
各ポリマーの分子量はゲル透過クロマトグラフィーにより決定した。PLゲル
混合D(5μm)カラムを、溶離液としてのクロロホルムとともに用いた。較正
のためにポリスチレン標準物を用いた。
FT−ラマンスペクトルは、FRT 106 FT ラマンモジュールを備え
たBruher IFS 66FT赤外分光光度計により記録した。
電位的測定は作動電極としてPd電極を用い、そして照合電極として飽和カロ
メル電極(SCE)を用いてEG&G Princeton Applied
Research Potentiostat/Galvanostat モデ
ル372により行なった。
粘度は、ウベローデ粘度計(Haake)を用いて測定した。
ジェファミンED−600とアゾベンゼン−4,4’−ジアシッドクロリドと
から出発するポリアミドの調製は、Macromol. Chem 180(1
979)2517−1519にM.BALASUBRAMANIAN 等により
記述された方法に従って達成される。アルコールを含まない乾燥したクロロホル
ム100mlの中に溶解した10g(14.80mmol)のジェファミンED
−600を3口フラスコの中に入れて窒素雰囲気のもとに撹拌した。この溶液に
7ml(50mmol)のトリエチルアミンを加えた。この混合物を−10℃に
おいて15分間撹拌した。次にメタノールを含まない乾燥したクロロホルム10
0mlの中に溶解させた4.4g(13.03mmol)のアゾベンゼン−4,
4’−ジアシッドクロリドをその反応混合物にただちに加えた。この混合物を−
10℃において15分間撹拌した後、室温にした。撹拌を24時間続けた。この
反応混合物を0.1MHCl(3×150ml)、水、0.1MNaOH(3×
150ml)で抽出した。そのクロロホルム層をMgSO4の上で乾燥させ、濾
過し、そして蒸発乾固させた。得られたポリマーは橙色のワックス滋養の固体で
あった。
1HNMRスペクトロスコピー(CDCl3、360MHz):δ=1.1pp
m(d,PPGエーテルのCH3γ)、δ=1.35ppm(d,PPGエーテ
ルのCH3β)、δ=3.65ppm(m,PEGエーテルのCH2α)、δ=3
.8ppm(m,PPGエーテルのCHβ)、δ=4.4ppm(広幅化した,
N−Hアミド)δ=7.95ppm(m,芳香族プロトン)。
赤外スペクトロスコピー(KBr円板の上のフィルム):ν=3300cm-1
N−H伸縮振動、ν=1650cm-1C=O伸縮振動、ν=1540cm-1N−
H曲げ振動、ν=1600cm-1C−H芳香族性伸縮振動、ν=1100cm-1
C−O伸縮振動。フィルムの成形
クロロホルムの中のアゾポリアミドの濃厚溶液(約30%)をシリコン被覆し
たガラス板の上に注流した。熱空気循環炉の中で溶媒を蒸発させた後でそのフィ
ルムをガラス板から取りはずした。試験管内還元性媒質の調製
燐酸ナトリウム緩衝液(0.25M、pH6.5)を煮沸し、そして更にアル
ゴン雰囲気のもとで冷却した。撹拌しながら硫化ナトリウム及びシステインを加
えてそれぞれについて50mMの濃度にした。
この媒質の酸化還元電位はPtレドックス電極で測定して−430mV(±1
0mV)であり、そして少なくとも4日にわたり一定に保たれた。この媒質の還
元性はスルフアラジンのインキュベーションによって評価した。生じた5−AS
Aを定性的及び定量的にHPLC分析によって求めた。化学的な還元性媒質の中でのフィルムのインキュベーション
約0.1g(厚さ±100μm)のアゾポリアミドを3時間から2日までに変
化する時間にわたり30mlの還元性媒質の溶液の中でシンキュベートした。規
則正しい時間間隔でその媒質からそれらのフィルムを取り出し、水で洗浄し、真
空において乾燥させ、そしてGPC、FT−ラマン及び13CNMR測定により分
析した。シミュレートした人腸管内媒質(SHIME)の中でのフィルムのインキュベー ション
約0.05g(厚さ±100μmのアゾポリアミドをSHIME−反応器の媒
質のバッチ15mlの中で12時間から3日間までに変化する時間にわたりイン
キュベートした。そのSHIME反応器は回腸(2容器)及び大腸(3容器)を
代表する5つの容器よりなる。その酸化還元電位−250mV(±50mV)を
Ptレドックス電極により毎日測定した。c;oインキュベーションフラスコを
37℃に保った。規則正しい時間間隔で各フィルムをその媒質から取り出し、水
で洗浄し、真空知友で乾燥し、そしてGPC、ラマン及び13CNMRの測定によ
り分析した。結果及び検討
I)ジスルフィド含有コポリアミドの合成
各種ジスルフィド含有ポリマーを3,3’−ジチオビススクシニミジルプロピ
オナートとα,ω−アミノ末端基のプレポリマーとの縮重合により調製した。
3,3’−ジチオビススクシニミジルプロピオナート(DDSP)は下記の反
応式に従って合成した:
125mlの乾燥ジオキサンの中の5g(23.778mmol)の3,3’
−ジチオビススタシニミジルプロピオナート(DDSP)の溶液に、250ml
の2口フラスコの中で窒素雰囲気のもとに6.25g(54.3mmol)のN
−ヒドロキシスクシニミド(NHS)の溶液を加えた。この反応混合物を氷−ア
セトンの浴の中で0℃に冷却し、次いで11.25g(54.52mmol)の
ジシクロヘキシルカルボナート(DCC)を加えた。この反応混合物を0℃にお
いて1時間にわたり撹拌し、そして撹拌を室温において1夜継続した。析出した
ジシクロヘキシル尿素(DCU)を濾過除去し、乾燥ジオキサンで洗浄し、そし
て濾液を1/3まで濃縮して冷蔵庫の中に1夜保った。
DCUの析出した痕跡を濾過除去し、そしてその濾液を回転蒸発機で蒸発させ
た。白色の粗生成物を乾燥アセトンとジエチルエーテルとの(1:1)混合物(
150ml)の中で1−2時間還流させることにより再結晶させた。そのDCU
不純物はその還流混合物の中により可溶性であり、そして純粋なDDSPは可溶
性がより低いことが見出されたが、その不溶性の生成物を濾過して真空の中で乾
燥させた。この最後の段階を、その生成物のNMRにおいてDCUが認められな
くなるまで2、3回繰り返した。
B)DDSPとジェファミンEDR−192及びPTHF−ジアミン750との 共重合
B−1)1:1のPTHF−750:EDR−192
2.
操作
15mlの乾燥クロロホルムの中の2.51g(24.75meq)のEDR
−192、15mlの乾燥CHCl3の中の10.55g(24.75meq)
のPTHF−750及び13.69mlのトリエチルアミンの、冷却された溶液
(0℃)に、65mlのCHCl3の中のDDSP(49.5meq)の溶液を
加えた。この反応混合物を0℃において1時間撹拌し、そしてその撹拌を室温に
おいて36時間にわたり継続した。
そのクロロホルム層を0.1MのHCl(3×)、H2O(3×)、0.1M
のNaOH(3×)及び最後にH2O(3×)により抽出した。そのCHCl3の
層をMgSO4の上で1夜乾燥させ、次いで濾過し、そしてその濾液を回転蒸発
機で蒸発させた。この生成物を更に真空の中で乾燥させた。結果
1)良好な膜形成
2)GPC(NMP):平均分子量=21791、分散度=2.3
3)大腸内デグラデーション(第1表参照)
II)分離した大腸媒質の中でのジスルフィド含有コポリアミドの分解
SHIME反応器から採取した15mlの試料の中でポリマーフイルムをイン
キュベートした。このSHIME反応器(人腸管内微生物生態系をシミュレート
した)は人の大腸の微生物性内容物をトミュレートしたものである。各フラスコ
を窒素でフラッシングすることにより脱ガスし、次いで37℃においてインキュ
ベートした。規則的な時間においていくつかの試料を採取し、そしてGPC(溶
離剤=NMP)により分析した。溶解しなかったポリマーの試料を洗浄し、真空
のもとに乾燥し、そしてGPCにより分析した。可溶性のポリマーはその媒質を
凍結乾燥することにより除去し、そして残渣をNMPの中に溶解し、無機塩類を
除くために濾過し、そしてGPCにより分析した。
III)機械的試験 引張試験
各ポリマー試料の引張強度をHounsfieldの type H10KM
引張試験機を用いて測定した。各試料を25℃においてクロスヘッド速度(引張
速度)20mm/分で100Nセルを用いて試験した。フィルムの調製
クロロホルムの中の10%(w/w)のポリマーを含むポリマー溶液をテフロ
ンの板の上に注流した。その板を時計ガラスで覆って溶媒の蒸発を遅延させ、大
気の空気乾燥により固化させた。各フィルムを注意深くその板から取りはずして
30℃において真空中で一定重量になるまで乾燥させた。そのフィルムの厚さは
顕微鏡Forster−Isometer S 2.320(ベルギー)を用い
て測定した。このフィルムを引張強度測定のためにBerg & Schmid
モデルHK500 からの手動のプレスカットナイフを用いて切り出した。
1.3 ジスルフィド含有ポリウレタン
いくつかのジスルフィド含有ボリウレタンを4,4’−ジフェニルメタンジイ
ソシアネート(MDI)と、α,ω−ヒドロキシ末端基のプレポリマーと、及び
ジスルフィド含有連鎮延長剤との下記反応式に従う縮重合により調製する。
ヒドロキシ末端基のプレポリマーとしてはポリ(テトラメチレンオキシド)、
ポリ(プロピレンオキシド)、ポリ(カプロラクトン)、ポリ(エチレンオキシ
ド)、ポリ(エチレンオキシド/プロピレンオキシド)を用いた。アゾ含有ポリアミドの合成
各種アゾ含有ポリマーを、アゾベンゼン−4,4’−ジアシッドクロリドとオ
リゴマーのα,ω−ジアミンとの、溶媒としてのクロロホルム及びプロトン受容
体としてのトリエチルアミンの中での縮重合により合成した。アゾベンゼン−4
,4’−ジアシッドクロリドは対応するアゾベンゼン−4,4’−ジカルボン酸
から、溶媒としてのトルエンの中でのチオニルクロリドとの反応により調製した
。選んだオリゴマーのジアミン類は親水性の(ジェファミンED−600) (
7)であったが、これは水溶性の(9)のポリマーをもたらした:
それらポリマーの特性決定は1H−NMR及びIRスペクトロスコピーにより
行なった。第1表に、標準としてポリスチレンを用いたGPCにより求めた両方
のアゾ重合物の分子量をあげる。
化学的還元性媒質の中でのアゾ含有ポリアミドの還元
各アゾポリマーのデグラデーションを試験管内で還元性媒質の中で調べた。イ
ンキュベーションの間にそれらフィルムは粘着性となり無色になった。そのイン
キュベートしたポリマーの50℃における空気中での乾燥に際して各試料は無色
から橙色への急速な色回復を示した。
第3表に、上記親水性のアゾポリマー(9)の化学的な還元的媒質の中でのイ
ンキュベーションの前後における、ポリスチレンを標準として用いたGPCによ
り測定された分子量及び粘度測定の結果を示す。
異なったインキュベーション時間(第3表)において採取した試料のGPC分
析は分子量の著しい減少を示す。3時間のインキュベーションの後でその分子量
は見かけ上、最初の値から10のファクターで減少し、そして更に48時間のイ
ンキュベーションの間そのようにとどまる(第3表参照)。しかしながら再酸化
の間にもとの色が現れ、そしてGPCにより測定した分子量は最初の値に上昇す
る。それらのフィルムは6時間のインキュベーションの後でその還元性媒質の中
に完全に溶解する。アゾ結合の還元の間におけるヒドラゾ結合の形成の分光計による証拠
疎水性アゾポリマー(8)の還元の間におけるヒドラゾ基の形成の証拠は13C
NMR分析によって得られた。
第4表はこの疎水性アゾポリマーの酸化型及び還元型についての異なった化学
シフトをあげる。芳香族性炭素の共鳴は、その酸化されたポリマーについてY=
-N=N-Ph及びX=-CON(CH3)2としたとき、及びその還元されたものについてY=-
NH-NH2 及びX=-CON(CH3)2としたときのパラX−Ph−Y構造を用いて加成性法
則[25]に基づくものである。各計算値はその酸化型及び還元型につい
て実験的に求められたそれらとよく一致している。
結論として、若干の特性的炭素共鳴線(例えばC3及びC5)を有する両方のポ
リマー型が13CNMRによって容易に互いに判別し得ると言うことができる。
再酸化されたポリマーの13CNMRスペクトルは化学的な還元性媒質の中及び
SHIME反応器の中でのインキュベーションの後で明らかにその還元型及び酸
化型の混合物である。化学的還元性媒質の中でインキュベートしたポリマーを3
日間にわたり大気中で再還元したが、これはなお主として還元型(約90%)を
含んでいる。SHIME反応器の中でインキュベートしたポリマーを大気中で約
2週間にわたり再酸化したが、このものは主として酸化型(約55%)を含んで
いる。
FT−ラマンスペクトロスコピーはアゾ基の還元を調べるための魅力的な手段
の1つである。両方のスペタトルの中の芳香族環の振動は1613cm-1及び1
607cm-1のところに現れる。1460cm-1における典型的なラマンシフト
はそのN=N官能基に基づくものとすることができ、そしてその酸化型で存在し
ている。このラマン信号は酸化型化合物と還元型化合物とを区別するのに用いる
ことができる。FT−ラマンは従ってこの目的のための補充的な非破壊技術とし
て応用することができる。還元されたポリマーについては1460cm-1のとこ
ろのN=N伸縮バンドは消失し、そして1298cm-1のところの広幅のバンド
で置き換えられている。このバンドはNH−NHの対称的伸縮によるものであ
る。再酸化されたポリマーはN=N基に特性的な1460cm-1のところのシフ
トを含む。しかしながらSHIME反応器の中でインキュベートされたポリマー
の1460cm-1のところのラマン線の比較的高い強度は、その酸化された型の
ポリマーのより大きな量と関連させることができる。これらの結果は全体として13
CNMRのデータと一致している。電量計的実験
分限のデータ〔28、29〕はアゾ結合の電気化学的還元速度が2電子段階機
構によりヒドラゾ基へ進行することを示している。ヒドラゾ誘導体を超える還元
は一般にアゾ−ヒドラゾ反応よりも多いエネルギーを必要とする。もしエネルギ
ー差が充分に大きければその反応は2つの波により表わすことができよう(第5
図)。
しかしながら或る場合には、いくつかのアゾ化合物は4電子段階反応によって
アミンまで還元されることが知られている(第6図)。

125mlの乾燥ジオキサンの中の5g(47.62mmol)の3,3’−
ジチオプロピオン酸の溶液に、250mlの2口フラスコの中で窒素雰囲気のも
