親水性制御放出デバイス 技術分野
本発明は、予定された時間間隔後に活性物質を放出するように計画された、活
性物質(特に、薬剤学的活性物質)を含有するための制御放出デバイスに関する
。背景
国際特許WO90/09168明細書は、雌型体に係合する水膨潤性雄型プラ
グを含む、この種類の制御放出デバイスを開示する。カプセル形状でありうるデ
バイス内に薬剤学的活性物質が含まれる。カプセルが水に暴露されると、典型的
にヒドロゲル物質から形成される雄型プラグが膨潤して、結局は雌型体から外れ
る;それによって、カプセルに含まれる薬剤学的活性物質が放出される。ヒドロ
ゲルプラグを外して、それによってデバイスを開放するために要する時間が予測
可能かつ再現可能であるので、典型的に0.5〜12時間の範囲内である所定時
間間隔後に患者の体内で薬剤学的活性物質を放出するためにデバイスが使用可能
であることが判明している。これは例えば、時には患者が眠っている夜間を通し
て患者に薬剤学的活性物質を投与することが望ましいような医学的状態の治療に
おいて、夜間のある一定時間に又は患者の覚醒時に患者に所望のレベルの活性物
質を供給するために有用であると考えられる。カプセルが胃腸管を通過するとき
に所定の箇所における、例えば結腸における物質の投与を可能にするためにも有
用である。
特許WO92/13521明細書(Alza Corporation)は、
周囲環境から流体を吸収する膨張手段を包含する、活性物質の持続放出のための
流体吸収性投与デバイス(dispensing device)を述べている。この投与デバイス
は入れ子式に相互に係合する第1壁区分と第2壁区分とを有するハウジング、特
に中空カップと中空ボディとを有するカプセルを含む。カップ又はボディの一方
は他方の雌型部分の開放端部内に嵌合する雄型部分の形状である。このデバイス
内には膨張手段が含有され、流体を吸収すると膨張して、デバイスを開放するよ
うに2部分を分離させる。膨張手段は膨潤性ポリマー又は、流体を吸収したとき
に膨潤する浸透性製剤でありうる。デバイス内に含有される膨張手段に流体を接
触させるために、膨張手段に隣接する壁区分の1つは流体透過性である。区分が
分離した後に、流体がデバイスに入り、デバイス内の活性物質と接触し、それに
よって流体中に活性剤が投与される。
制御放出デバイスから薬剤学的活性物質を効果的に放出させるためには、2つ
の状況(thing)が起こらなければならない。第一に、液体の侵入を可能にするた
めに、デバイスは雄型部分と雌型部分との分離によって開放されなければならな
い。第二に、薬剤学的活性物質がデバイスから周囲液体中へ流入することを可能
にするためには、液体が実際にデバイスに侵入しなければならない。患者が特定
の時間に薬剤学的活性物質を投与されうることが目的であるので、これらの両方
が起こらなければならない。本発明は特に第二段階に関する、即ち、制御放出デ
バイスの開放後に物質の効果的な放出が起こることを保証することに関する。
制御放出デバイスの特定の用途は、薬剤学的活性物質の結腸中への放出である
。胃腸管の結腸部分では、そこに含まれる廃棄物が特に高い固形物含量と特に低
い水分含量とを有する。これは制御放出デバイスの含量を周囲流体中に、患者の
循環系中に吸収させるために有効になるように、効果的に放出させるという問題
を悪化させる。比較的低い水分含量は、制御放出デバイス内からの薬剤学的活性
物質を流出させる又は溶出させるために最少量の水が利用可能であるに過ぎない
ことを意味する。さらに、高い固形物含量は、結腸内での撹拌度が比較的低いの
で、胃腸管内での制御放出デバイスの運動によって制御放出デバイスの内容物が
放出される見込みが低いことを意味する。
制御放出デバイスのボディから物質を効果的に放出するという問題を扱うこと
が、本発明の目的である。発明の概要
本発明は、ボディが水性液体中に浸漬するときに放出される物質を含有するた
めの制御放出デバイスの不透性ボディであって、物質がボディから放出されるた
めの開口を有し、ボディの内面が少なくとも開口に隣接するその領域において親
水性物質を含むボディを提供する。
したがって、一般的に言うと、本発明は開口に隣接するボディの内面の少なく
とも一部に、環境からの水性液体の侵入を促進する親水性物質をコーティングす
る又は他のやり方で備えることにある。一般的に言うと、制御放出デバイスから
放出されるべき物質はボディ中に必然的にある量の空気と共に充填される粉状物
質又は他の固体物質である。このことは、液体充填物が制御放出デバイス内に入
れられるときにも同様に該当すると考えられる。本発明者はデバイスの雄型部分
と雌型部分との放出によってボディが開放されるときに、開口部分内に気泡が形
成される傾向があり、この気泡がカプセルボディ中への水性液体の侵入を阻止す
ることを観察している。本発明者は、気泡(単数又は複数)が通常集中する部分
である、ボディの開口に隣接する内面に親水性物質をコーティングすることによ
ってこの問題に取り組んで好結果を得ている。親水性物質は通常被膜として存在
するが、必要な場合には、ボディが形成される物質から固有部分として形成され
ることもできる。親水性物質の供給は気泡をボディへの開口を塞ぐその位置から
放出させるのを助けると考えられる。気泡の放出はボディの内面の空気をけん引
する表面エネルギーの変化によって、又はより可能性があることには、水性液体
によるボディの内面の湿潤を促進し、ボディ中への水性液体の流動を促進し、そ
れによって気泡を押し出すことによって気泡の放出を促進することができると考
えられる。しかし、本発明者は特定の作用理論によって束縛されるのを望まない
。本明細書で実証されるように、このようにして、親水性表面の提供は制御放出
デバイスのボディの開口から気泡を除去することに特に有効である。
好ましくは、ボディは通常のカプセル形成方法によって製造されるカプセルボ
ディの形状であり、この方法は一般にカプセルを形成するための物質(例えばゼ
ラチン)の溶液に型ピン(mould pin)を浸漬することを含む。浸漬の代わりに噴
霧を用いることもできる。次に、ゼラチンを乾燥させ、ピンから剥離する。次に
、このカプセルボディを必要に応じてトリミングすることができる。或いは、例
えば熱可塑性物質の射出成形のような他の既知製造方法によって、カプセルボデ
ィを製造することができる。
活性物質を含有するように予定されるボディの水不透性壁は非常に多様な物質
から製造することができる。ボディの壁は水に対して不透性である(及び一般に
水中に不溶性でもある)ので、ボディが所定時間間隔後に開放するまで水性液体
はボディに侵入することができない。“水不透性”とは、ボディが制御放出デバ
イスの開放まで水性液体と接触する通常の期間中に水が被膜を実質的に通過しな
いことを意味する。ボディの壁は均質な構造でも積層状でもよい。ボディの構造
に用いるために適した物質の例はポリエチレン、ポリプロピレン、ポリ(メチル
メタクリレート)、ポリ塩化ビニル、ポリスチレン、ポリウレタン、ポリテトラ
フルオロエチレン、ナイロン、ポリホルムアミド、ポリエステル、酢酸セルロー
ス及びニトロセルロースを包含する。
しかし、好ましい構造は水不透性被膜を用いて、水溶性物質から形成されたボ
ディの外側を被覆する、この理由はこれが慣用的方法を用いて製造されることが
できるからである。水に対して不透性である層を形成する物質の溶液中にボディ
を浸漬することによって、被膜を形成することができる。或いは、ボディを噴霧
塗装することができる。カプセルボディの好ましい種類は慣用的な硬質ゼラチン
又は澱粉のカプセルボディであり、これにポリ塩化ビニル、ポリ酢酸ビニルコポ
リマー又はエチルセルロース溶液をコーティングして、乾燥させて、水不透性被
膜を形成する。
本発明の特に好ましい実施態様では、ボディは開口内に係合した雄型要素を含
む雌型ボディである。特に、雄型要素はボディの開口に隣接したボディのネック
部分内に嵌合したプラグでありうる。このプラグは通常、実質的に円筒形の形状
であり、水膨潤性物質、好ましくはヒドロゲルから形成されることができる。ヒ
ドロゲルプラグは水を吸収するので、膨潤して、雌型ボディから分離する。ヒド
ロゲルは特許WO90/09168明細書に開示されるような、水膨潤性物質で
あることが好ましい。通常は、ネック部分は円筒形状の雌型プラグと緊密な嵌合
を形成するように、実質的に円筒形である。
他の実施態様では、雄型要素は一端が閉じた中空要素であり、その反対の開放
端部は雌型ボディのネック内に係合する。水膨潤性物質は制御放出デバイス内に
入れられ、所定時間後に水の存在中でこの物質が膨潤するときに雄型要素と雌型
ボディとを押し離すことによって、雌型ボディを分離することに役立つ。制御放
出デバイス内の膨潤性物質はオスマゲント(osmogent)又はオスモポリマー(osmop
olymer)であることができる。このような配置は特許WO92/13521明細
書に開示されている。水を制御放出デバイスに侵入させ、水膨潤性物質と接触さ
せるために、水膨潤性物質に隣接するデバイスの壁の一部が半透性であることが
好ましい、即ち、この壁の一部がデバイス中への水の通過に対しては透過性であ
るが、デバイス内からの他の物質の放出に対しては不透性であることが好ましい
。
本発明によるボディへの開口からの気泡(単数又は複数)の効果的な除去は、
開口に隣接する領域におけるボディの内面が親水性物質であることを必要とする
。ボディ自体が適当な親水性物質から形成されることができるが、一般的に言う
と、親水性物質は水溶性である傾向があり、デバイスの開放前にカプセルボディ
の壁を通して水を侵入させる危険性に陥る傾向があるので、適当な物質の選択は
限定される。これより通常は、ボディは水不溶性である物質又は水不溶性被膜で
被覆された物質から形成される。このような水不溶性物質はそれらの本質から親
水性でない傾向がある。それ故、本発明の好ましい実施態様では、親水性物質は
被膜として備えられる。被膜は浸漬被覆又は噴霧塗装によるような、任意の適当
な方法で与えることができる。噴霧塗装を用いる場合には、充分な親水性コーテ
ィング物質をボディの開口から噴霧して、気泡が沈着する可能性のある、開口に
直接隣接するボディの内面領域を効果的に被覆することができることが判明する
。一般的に言うと、ボディの内面全体を親水性物質で被覆する必要はない。
一般に、適当な水性又は有機の液体ビヒクル中に親水性物質の溶液又は懸濁液
の適用によって、親水性物質を塗布することができる。
一般的に言うと、制御放出デバイス(例えばカプセル)の正常な流動特性が損
なわれず、凝集が生じないように、被膜は非粘着性であり、平滑な連続した(coh
erent)被膜であるべきである。この物質は一般に乾燥して、固体被膜になるべき
である。
親水性物質は、上述した基準を満たす限り、適当な界面活性剤物質を含むこと
ができる。また、親水性のフィルム形成物質を用いることができ、多くのこのよ
うな物質が技術上周知である。このフィルム形成物質は水に不溶性であることも
水溶性を有することもできる。他の状況も同じであるが、水溶性被膜は比較的良
好な完全排出(emptying)特性を示す傾向がある。最後に、フィルム形成物質と界
面活性剤との混合物を用いることができ、これらの混合物はフィルム形成剤(fil
m former)の親水性の強化に特に有用であることが判明している。 種々な親水
性度の適当なフィルム形成物質はセルロース、ポリアクリレート、ポリメタクリ
レート、ポリアクリルアミド、ポリビニルアルコール及びポリビニルピロリドン
を包含する。特定のフィルム形成物質はヒドロキシプロピルメチルセルロース、
メチルセルロース、ヒドロキシプロピルセルロース、エチルセルロース、酢酸フ
タル酸セルロース、ポリビニルアセテートフタレート、例えばEudragit
s(商標)のようなアクリル樹脂、ヒドロキシプロピルメチルセルロースフタレ
ート、ヒドロキシプロピルメチルセルロースアセテートスクシネート、ゼイン(z
ein)、シェラク(shellac)、酢酸セルロース及び三酢酸セルロースを包含する。
また、例えば糖、ゼラチン、アラビアゴム及び澱粉のような、幾つかの適当な糖
ベースドコーティング物質も存在する。
親水性被膜に含めるために適当な界面活性剤はアニオン界面活性剤、非イオン
界面活性剤及びカチオン界面活性剤を包含する。典型的な例はラウリル硫酸ナト
リウム、ナトリウムドクセート(sodium docusate)、オレイン酸ナトリウム、ひ
まし油、水素化ひまし油、脂肪酸エステル及び脂肪酸アルコール(例えば、セチ
ル及びラウリル)を包含する。界面活性剤はポリオキシエチレン又はその誘導体
、例えばポリオキシエチレンソルビタンモノラウレート(例えば、Tween)
、ポリオキシエチレンソルビタンモノオレエート(例えば、Span)、脂肪酸
エトキシレート 例えばポリオキシエチレンステアレート、アルコールエトキシ
レート 例えばオレイルエーテル、ポリオキシエチレンーポリオキシプロピレン
コポリマー及びポリエチレングリコールであることもできる。SpanとTwe
enは商標である。
本発明の特に好ましい実施態様は、水不溶性にするためにエチルセルロースで
被覆されている、慣用的な硬質ゼラチンカプセルボディを用いる。本発明による
親水性被膜を与えるために、このカプセルボディにヒドロキシルプロピルメチル
セルロース(HPMC)のさらなる被膜を噴霧塗装する。或いは、患者に親しみ
やすい外観と感触とを与えるために、ゼラチンを水不溶性カプセルボディ上に親
水性被膜として用いることもできる。
親水性被膜は一般に制御放出デバイスのボディの0.1〜50重量%を占め、
特に1〜20重量%、とりわけ2〜10重量%を占める。
親水性物質の親水性の性質は物質の前進接触角度(標準Wilhelmyプレ
ート方法を用いて測定可能)として定量化することができ、これは物質の表面活
性を表す。前進接触角は、好ましくは80°未満、より好ましくは75°未満、
有利には70°未満である。これは50〜75°の範囲内、特に60〜70°の
範囲内であることができる。界面活性剤の親水性物質のHLB(親水性親油性平
衡)値は一般に10より大きく、通常は20より大きく、好ましくは30より大
きい。HLB値は25〜55の範囲内、好ましくは30〜50の範囲内であるこ
とができる。
本発明を薬剤学的活性物質の放出に関して考察してきたが、本発明は原則とし
て他の非薬剤学的物質の時間放出にも同様に適用されることができる。好ましい実施態様の詳細な説明
次には、本発明の実施態様を実施例のみととして説明する。
図面において、図1はボディ、ヒドロゲルプラグ及びキャップから成るカプセ
ルの形状の制御放出デバイスの断面図である。
このカプセルは我々の公開特許WO94/09745明細書にさらに詳しく述
べられており、雌型ボディ6のネック4中に挿入された、ヒドロゲル物質製のプ
ラグ2の形状の雄型要素を含む。カプセルはキャップ8で閉鎖される。ボディ6
は例えば全体長さ約18mmであり、円筒形の主要部分10と閉鎖端部12とを
含む。主要ボディはショルダー領域5においてネック部分4に狭くなる。ネック
部分は、密接な許容範囲で雄型プラグ2を受容するように実質的に円筒形(被覆
時の内径は約6.7mmである)である。ネック部分は次に広口(flared mouth)
部分14まで広がり、広口部分は主要ボディ部分10の直径と実質的に同じ直径
の開口15を有する。
雄型プラグ2はヒドロゲル物質(例えばWO90/09168に開示されたよ
うな)から形成され、通常、プラグの上端部がカプセルボディの上端部の上又は
この上端部と同じ高さ又はこの上端部の下になるように挿入される。
カプセルを次にキャップ8によってシールする、キャップ8はキャップを適所
で固定するように広口部分のリムの下で切り取られる戻り止め18を備える。
キャップは、キャップの開放端部にまで達する円筒形状スカート部分9を有す
る。スカート部分の下端部はボディのショルダー5を過ぎて下方に伸びて、ごく
小さいクリアランスでボディの上部を閉鎖する。これはキャップを安定化するの
に役立ち、キャップが左右に傾斜するのを阻止する。これはまたキャップがボデ
ィとキャップとの間の接合部上を通る環状帯によってボディにシールされるのを
可能にする。実施例1
.ヒドロゲルプラグの製造
6,000gの、数分子量Mn8700とMw/Mn比=1.03とを有する
ポリエチレングリコールPEG8000(Pharma)を111.04gのヘ
キサントリオールと、506.8gのDesmodur W(ジシクロヘキシル
メタン−4,4−ジイソシアネート)と共に、0.6gの無水塩化第2鉄によっ
て触媒して重合させてヒドロゲルロッドを製造した。モル比はPEG8000(
1モル)、ヘキサントリオール(1.2モル)、Desmodur W(2.3
モル)及び塩化第2鉄(PEGの0.01重量%)であった。PEG8000を
溶融し、Buchi Rotavapor内で95℃、5ミリバール未満の圧力
において2時間の期間にわたって、0.05%w/w未満の水分含量になるまで
乾燥させた。次に、塩化第2鉄をヘキサントリオール中に75℃において溶解し
、混合物を乾燥PEG中に100rpmにおいて5分間にわたって撹拌して混入
した。混合物を85℃において次にDesmodur Wと1500回転/分で
回転するミキサーに供給することによって(by pumping)混合した。約80℃の溶
融ポリマーを次に、管状ポリテトラフルオロエチレン型(25cm長さ、約6.
7mm内径)に50ミリバール未満の真空下で分配した。ファン装備オーブン内
で硬化を95℃において4時間おこなった。このヒドロゲルポリマーを次に冷却
させた。
ヒドロゲルロッドを、安定剤としてブチル化ヒドロキシアニソール(BHA)
を含有する循環水流中に浸漬することによって洗浄した。
この洗浄はポリマーから水溶性の抽出可能物を除去し、BHA安定剤はポリマ
ー中に混入される。
膨潤率(swelling factor)は(Ws−Wd)/Wdx100[Wsは膨潤した
重量、Wdは乾燥重量である]として定義される。このヒドロゲルは270±2
5の膨潤率を有することが判明した。
次に、ヒドロゲルロッドを各々が一般に公称長さ4mmのプラグに切断した。実施例2
.エチルセルロース被膜
図1に示すようなゼラチンカプセルボディを水不溶性水不透性のエチルセルロ
ース被膜によって次にように被覆した。ゼラチンカプセルボディのバッチをAc
celacota噴霧塗装機(Manesty Machines社,英国)に
供給した。この噴霧塗装機は製造者の指示に従って操作した。イソプロピルアル
コールとアセトン(50:50重量比)の混合物中のエチルセルロース(E10
0)の溶液をカプセルボディに、80重量%増加を示すエチルセルロース被膜が
付着されるまで噴霧した。塗装機への入口空気温度は45℃であり、出口空気温
度は40℃であった。ドラム速度は9.5rpmであった。エチルセルロース被
覆ゼラチンボディを将来の使用のために貯蔵した。エチルセルロース被膜はカプ
セルボディの全外面を被覆し、カプセルボディの内面の一部にまで達するので、
ヒドロゲルプラグを取り付けたときに使用中に水性液体と接触するボディの全て
の領域が水不透性エチルセルロース被膜によって被覆される。実施例3
.エチルセルロース被覆ゼラチンボディの、親水性物質の追加トップコ ートによる被覆
エチルセルロース被覆ゼラチンボディからの完全排出が不良であると言う問題
が観察されている。ここでは、種々なバッチの親水性カプセルボディを得るため
に用いられる種々な加工条件とコーティング溶液組成物とについて述べる、次い
で、これらのカプセルボディを気泡形成と錠剤完全排出時間とに関して試験する
。方法
実施例1によって得られた50gのエチルセルロース被覆ボディを以下に述べ
る種類の塗装機に加え、コーティング溶液を噴霧塗装する前に10分間加熱した
。必要な重量負荷が得られるまで噴霧を続けた。
下記表は、完全排出速度の測定と気泡形成試験とで試験した被覆ボディの各バ
ッチに関して、使用装置を記録されたプロセス条件と共に示す。
最後のバッチD195155BをHicoater装置(Freund製,ド
イツ)を用いて塗装した以外は、全てのバッチをStrea 1塗装機(Nir
o/Atomoser,スイス)を用いて噴霧塗装した。塗装機は製造者の指示
に従って操作した。IPAはイソプロピルアルコールの略号である。HPMCは
ヒドロキシプロピルメチルセルロースの略号である。
塗装後の各バッチのボディを適当にラベルした容器に入れて、次の実験まで貯
蔵した。実施例4
.気泡形成に対する種々な親水性被膜と湿潤剤との効果
エチルセルロース(E100)被覆ゼラチンカプセルボディに対するトップコ
ートとしての、湿潤剤及び/又は親水性フィルム層の組込みを研究した。プラグ
放出時の気泡発生を、その後の内容物完全排出と同様に、記録した。実験はさら
に完全な分析を可能にするために時間経過ビデオに記録した。方法

各カプセルボディに低置換(low substituted)ヒドロキシプロピルセルロース
(LH21)粉末の200mgスラグを充填した。この物質は水と接触すると膨
張する。次に、各カプセルに実施例1によって製造した3.00mm長さのヒド
ロゲルプラグを嵌合させた。一度に最大10カプセルをビデオ録画することがで
きたにすぎなかった。それ故、全体で6種類の被膜が試験されるまで実験を繰り
返した。組み立てたならば、デバイスをプラスチックスタンドに入れて、ガラス
の水タンクの底部に取り付けた。水(室温)をタンクに加え、ビデオカメラを記
録するようにセットした。実験のビデオ再生を観察することによって結果を得た
。結果
これらの結果は、エチルセルロース被膜のみで被覆したボディでは気泡が僅か
に残留したことを示唆する。実験5
.親水性被覆カプセルボディの完全排出時間の測定
実施例4の結果は、慣用的な噴霧塗装方法による親水性物質のトップ層の組込
みがプラグ放出時の気泡付着(air bubble retention)の発生を顕著に減じたこと
を実証した。この実験はカプセルボディからの充填物質の完全排出に見られる改
良を定量化することを目的とした。方法

各カプセルボディに100mgのLH21粉末を充填した。これを圧縮した。
プラセボ錠剤を頂部においた。LH21粉末は水との接触時に膨張して、ボディ
から錠剤を放出するように意図される。このデバイスに実施例1で製造した2.
5mm長さのヒドロゲルプラグを嵌合させた、このプラグはボディのネックから
1.5mm突出し、直接観察するために好都合な短いプラグ放出時間を生じた。
これらを次にプラスチックホルダーに載せて、ガラスタンクの底部に固定した。
水(室温)を加えた。実験をビデオカメラによってリアルタイムで記録し、全体
で4種類の被膜が試験されるまで繰り返した。ビデオを再生して、各デバイスに
関してプラグ放出から錠剤完全排出までの時間を知ることができた。次に、平均
時間を算出することができた。結果は以下に示す。
ラウリル硫酸ナトリウムとナトリウムドクセートとを含むフィルムでトップコ
ートされた(topcoated)カプセルは非常に迅速に完全排出した。気泡がボディの
内側に観察されたが、これらはプラグ放出時に瞬時に分散した。
対照的に、HPMCでトップコートされたボディは、プラグ放出時に形成され
た気泡があまり迅速に分散しないので、やや緩慢な速度で完全排出した。
エチルセルロース被覆された対照ボディは全く排出しなかった。放出されたプ
ラグがカプセルボディ上に位置して残留し、ボディのネックに気泡が存在した。
プラグを物理的に除去しても、錠剤の完全排出は見られなかった。
この証拠から、トップフィルム層として、例えばラウリル硫酸ナトリウム又は
