c-fos プロモーター活性化タンパク質をコードする核酸を同定する方法
本発明は、ヒトc-fosガン原遺伝子プロモーターを活性化するシグナル伝達分
子およびこのような分子のアンタゴニストを同定する物質および方法に関する。発明の背景
ガン原遺伝子Ha-ras、c-fos、c-myc、およびc-junのようなシグナリング分子
の変異または過剰発現の結果としての細胞活性化は、新生物の基礎を形成する細
胞の異常増殖に関連している。DeFeoら、Proc .Natl.Acad.Sci.,78,3328-33
32(1981); Millerら、Cell,36,51-60(1984); Kelekarら、Mol .Cell.Biol.,6
,7-14(1986); およびVogtら、Adv .Cancer Res.,55,1-35(1990)を参照のこ
と。
c-fosの誘導は、マウス3T3細胞の血清刺激後の増殖関連シグナリング経路の活
性化に応答して、またはHa-rasの正常およびトランスフォーミング形態の過剰発
現に応答してそれぞれ起こる。c-fosの構成的発現はまた、特定のヒト腫瘍株で
起こることが示されている。これらの知見は、新生物表現型の異常な増殖特性が
、c-fosガン原誘導に関係するシグナル伝達経路の構成的活性化を含み得ること

c-fosプロモーターで駆動されるリポーター遺伝子を用いることにより、c-fos
ガン原遺伝子プロモーター中の特異的エンハンサーが同定されており、それは活
性化シグナル伝達経路に応答する。これらのエンハンサーには、チロシンキナー
ゼ応答性SCM、raf-応答性直接反復、プロテインキナーゼC-応答性AP-1部位、お

SV40 T抗原遺伝子の転写活性化を用いてエンハンサーおよび安定に相互作用す
る転写因子を同定する随伴複製系が知られている。Vasavadaら、Ind .J.Bioche m.Biophys.

細胞増殖の制御におけるシグナリング分子の重要性のため、増殖関連シグナリ
ング系に含まれる分子(それは次に抗腫瘍薬剤を発見するための生物学的標的を
同定するために用いられ得る)を同定する方法が必要である。異常細胞増殖が起
こったときこのような増殖関連シグナリング系を妨害して正常増殖を回復し得る
因子を同定する方法もまた必要である。発明の要旨
本発明は、シグナル伝達分子およびそのアンタゴニストを同定するための物質
および方法を提供することにより前述の必要性を満たす。より詳しく述べると、
本発明は哺乳類細胞株を提供し、この細胞は:(a)ポリオーマウイルスラージT抗
原をコードする核酸に作動可能に連結された誘導可能なまたは組織特異的プロモ
ーターを含む組換えベクター;および(b)ポリオーマウイルスの複製開始点およ
び上記プロモーターの活性化タンパク質をコードすると考えられる核酸を含む組
換え発現ベクターを含む。
好ましくは、プロモーターはヒトc-fosプロモーターであり、そして活性化タ
ンパク質はヒトc-fosプロモーター活性化タンパク質である。
本発明は、プロモーター活性化タンパク質をコードする核酸を同定する方法を
さらに提供し、この方法は:(a)哺乳類細胞株であって:(i)ポリオーマウイルス
ラージT抗原遺伝子のコード領域に作動可能に連結された誘導可能なまたは組織
特異的プロモーターを含む組換えベクター;および(ii)ポリオーマウイルスの複
製開始点および上記プロモーターの活性化タンパク質をコードすると考えられる
核酸を含む組換え発現ベクターを含む細胞を、このような核酸が発現する条件下
で培養する工程;および(b)ベクター複製を許容するに十分なインキュベーショ
ン期間の後細胞中の複製したベクターのレベルを測定する工程;それによってヒ
トプロモーター活性化タンパク質をコードする核酸が、細胞中のベクターの増加
したレベルの測定により同定される工程を包含する。
好ましくは、プロモーターはヒトc-fosプロモーターであり、そして活性化タ
ンパク質はヒトc-fosプロモーター活性化タンパク質である。
本発明において使用されるヒトc-fosプロモーターを含む好適な組換えベクタ
ーは、プラスミドPfLAG-8である。
ポリオーマウイルス複製開始点を含む好適な組換え発現ベクターは、プラスミ
ドLα2である。
本発明はまた、配列表の配列番号1および配列番号3で定義されるアミノ酸配
列を有するヒトc-fosプロモーター活性化タンパク質、またはその抗原性フラグ
メント、およびこのようなタンパク質またはフラグメントをコードする核酸を提
供する。
別の実施態様では、本発明は哺乳類細胞株を提供し、この細胞は:(a)ヒトc-f
osプロモーターに作動可能に連結されたリポーター遺伝子を含む第1の組換え発
現ベクター;および(b)ヒトc-fosプロモーター活性化タンパク質をコードする核
酸を含む第2の組換え発現ベクターを含む。
本発明はまた、ヒトc-fosプロモーター活性化タンパク質のアンタゴニストを
同定する方法を提供し、この方法は:(a)哺乳類細胞株を提供する工程であって
、この細胞が(i)ヒトc-fosプロモーターに作動可能に連結されたリポーター遺伝
子を含む第1の組換え発現ベクター;および(ii)ヒトc-fosプロモーター活性化
タンパク質をコードする核酸を含む第2の組換え発現ベクターを含み;(b)工程(
a)の細胞株と、ヒトc-fosプロモーター活性化タンパク質のアンタゴニストを含
むと考えられる試料とを接触させる工程;および(c)リポーター遺伝子の発現の
レベルを測定する工程;それによって試料中のヒトc-fosプロモーター活性化タ

好ましくは、第2の組換え発現ベクターは、CROC-1タンパク質、CROC-4タンパ

本明細書で用いられる用語「リポーター遺伝子」は、イントロンを含み得るま
たは含み得ないゲノムDNAから単離されたDNA分子、またはメッセンジャーRNAを
テンプレートとして用いて調製された相補DNA(cDNA)のいずれかを意味する。い
ずれの場合にも、このDNAは、例えば、酵素活性、酵素連結イムノソルベントア
ッセイ(ELISA)またはラジオイムノアッセイ(RIA)により、容易に測定され得る発
現産物をコードする。本発明において使用される好適なリポーター遺伝子は、pC
H110(Stratagene #27-4508-01)由来のE.coli Lac-Z遺伝子を含む。この遺伝子の
発現レベルは、高感度の蛍光基質アッセイにより測定され得る。以下に記載する
CATリポーター遺伝子もまた好適であるが、当業者に周知の多くの他のリポータ
ー遺伝子がこの代わりに用いられ得る。
用語「組換え発現ベクター」は、組換え技術を用いて調製されたベクターを意
味し、このベクターは、タンパク質をコードする挿入された核酸を、このベクタ
ーが適切な宿主細胞中へのトランスフェクションまたは形質転換の際に上記タン
パク質を発現し得るように含む。好適なベクターは、プロモーター活性化タンパ
ク質をコードする核酸を含む。ヒトc-fosプロモーターに作動可能に連結された
リポーター遺伝子を含むベクターもまた好適である。
「安定に形質転換」された細胞は、それらのゲノムDNA中に組み込まれた組換
えDNAを有する。そのような安定に組み込まれたDNAは、形質転換された細胞に保
持される。何故なら、それは、G418耐性のような選択マーカーとともに細胞中に
導入され、選択培地で細胞が増殖するとき保持を強制するからである。本発明は

る。
本発明の誘導可能なまたは組織特異的プロモーターは、通常必要な(house-kee
ping)プロモーターではない。即ち、それらは通常の条件下で調節されるが構成
的発現の低い基礎レベルが生じ得る程度を除いて転写的に活性ではない。
本明細書で定義されるように、「誘導可能なプロモーター」は、その活性が、
ストレス、ホルモン刺激、または分化のような細胞応答を生じる細胞環境におけ
る変化に応答して活性化または増大されるプロモーターである。誘導は、シグナ
リングカスケードの活性化を経由して起こり、プロモーター部位で、転写因子の
増大した結合および活性を生じる。このような誘導に含まれる分子は、本明細書
で記載されるようなプロモーター活性化タンパク質を包含する。誘導可能なプロ

モーターである。
用語「組織特異的プロモーター」は、前立腺細胞でのみ活性であるプロモータ
ーのような、細胞タイプのサブセット内でのみ活性であるプロモーターを意味す
る。Youngら,Biochem.,31,818-824(1992);およびRiegmanら,Mol .Endocrin ol.
,5,(No.12)1921-1930(1991)を参照のこと。他の組織特異的プロモーター

本発明で用いられ得るプロモーターは、ガン原遺伝子c-fosおよびc-mycのプロ
モーターを含むがこれに限定されない。Millerら、前出;およびKelekarら、前
出を参照のこと。これらの両プロモーターは、過剰発現が異常な細胞増殖を導き
得る遺伝子のインビボ発現を調節する。c-fosプロモーターが最も好ましい。
用語「異常細胞増殖」は、本明細書では、新生物の異常なまたは制御されない
細胞増殖特性として定義される。
本明細書で用いられる用語「プロモーター活性化タンパク質」は、上記のプロ
モーターの1つの転写活性化を引き起こすタンパク質として定義される。好まし
くは、プロモーター活性化タンパク質は、ヒトc-fosプロモーター活性化タンパ
ク質である。最も好適には、α2-マクログロブリンレセプター結合タンパク質の
アミノ酸配列と実質的に同一のアミノ酸配列を有する活性化タンパク質である。
また、最も好適なのは、配列番号3および配列番号1でそれぞれ定義される配列
である、CROC-4タンパク質またはCROC-1タンパク質のアミノ酸配列と実質的に同
一であるアミノ酸配列を有する活性化タンパク質である。アミノ酸配列の実質的
同一性は、配列番号1または配列番号3のいずれかにより定義される配列と比較
した別のc-fosプロモーター活性化タンパク質の配列が、同一であるかまたは本
明細書で記載されるような転写活性化活性を実質的に損なわない1つまたはそれ
以上のアミノ酸改変(欠失、付加、置換)によって異なることを意味する。例えば
、配列番号1または配列番号3のいずれかにより定義される配列の対立遺伝子座
のまたは種間の改変体が存在し得る。
さらに、例えば、化学的合成によりまたは改変ポリメラーゼ連鎖反応(PCR)の
使用によりまたは部位特異的変異誘発の使用により、配列番号1または配列番号
3のいずれかにより定義される配列を有するc-fosプロモーター活性化タンパク
質をコードするDNAを改変して、単一または複数の塩基置換を生成しそれから活
性を実質的に損なわないc-fosプロモーター活性化タンパク質を生成することは
、十分に当業者の技術的範囲である。このような保存的に改変された改変体は、
本発明の範囲内にある。
配列同一性は、必要であれば、残基の一致を最適化することにより、そして必
要に応じてギャップを導入することにより測定され得る。これは、保存的置換を
一致として考慮する場合には異なる。保存的置換は、代表的には、以下のグルー
プ内の置換を包含する:グリシン、アラニン;バリン、イソロイシン、ロイシン
;アスパラギン酸、グルタミン酸;アスパラギン、グルタミン;セリン、スレオ
ニン;リジン、アルギニン;およびフェニルアラニン、チロシン。相同のアミ
ノ酸配列は、代表的には、各タンパク質配列の各々における天然の対立遺伝子座
および種間変動を含む。代表的な相同タンパク質またはペプチドは、CROC-1タン
パク質またはCROC-4タンパク質と、25-100%の相同性(ギャップが導入され得る
場合)、50-100%の相同性(保存的置換が含まれる場合)を有する。相同性の大き
さは、少なくとも約50%、および代表的には少なくとも60%またはそれ以上であ
る。
本発明はまた、ヒトc-fosプロモーター活性化タンパク質の「抗原性フラグメ
ント」を含む。抗原決定基(エピトープ)が一般に少なくとも約5のアミノ酸残基
を含むことは当業者に周知である。Ohnoら、Proc .Natl.Acad.Sci.USA,82,
2945(1985)。本発明の抗原性フラグメントは、約5から約100の、そして好まし
くは約5から約50のアミノ酸残基を含む。所定のポリペプチドが本発明の範囲に
入るか否かは、以下に記載する方法を用いるルーチンの実験により容易に決定さ
れ得る。
このような抗原性フラグメントは、全長のヒトc-fosプロモーター活性化タン
パク質のタンパク質分解により、または化学的もしくは組換DNA合成により作成
され得る。この抗原性フラグメントは、標準的方法により、好ましくは哺乳類で
、抗体産生を惹起するために用いられ得る。このように産生された抗体は、標準
の免疫アッセイまたは免疫吸着法を用いて活性化タンパク質をアッセイまたは精
製するために用いられ得る。
本発明は、ポリオーマウイルスT抗原遺伝子を含む組換えベクターを利用し、
そして随伴複製系を拡張して、その産生が遺伝子プロモーターの転写活性化に導
くタンパク質を同定する。Vasavadaら、前出、により用いられるSV40 T抗原遺伝
子とは対照的に、ポリオーマT抗原遺伝子の複製および形質転換性質は分離され
得る。
複製および形質転換性質の分離は、ミドルT抗原の中央コード配列と重複する
領域中のラージTイントロンに停止コドンを挿入することにより達成される。こ
の機能の分離は、c-fosプロモーターの場合重要であり、ここでミドルT発現の防
止は、プロモーターからの低レベルの、基礎転写に起因する、ミドルTで活性化
されるc-src-およびホスファチジルイノシトール3-キナーゼ関連シグナリング系
(Talmageら,Cell,59,55-65(1989)により同定された)を経由する転写活性化の
可能性を除外する。
ポリオーマウイルス系の使用は、いくつかの良く特徴付けられたマウスの系へ
の、随伴複製の拡張を可能にする。対照的に、Vasavadaら、前出、により用いら
れるSV40 T系は、主としてシミアン(サル)の系に限られる。さらに、本発明の系
は、SV40 T抗原をベースにした系で先に報告された高頻度の短縮型のまたは再配
列された挿入物(約25%)を生じないようてある。挿入物の改変は、本発明の系で
は2%未満の頻度で生じる。
好適な実施態様は、ポリオーマウイルスラージT抗原遺伝子のプロモーター上
流からの複数のエンハンサーの組み込みを含み、cDNAでコードされるシグナリン
グ分子の低レベル発現に応答するラージT誘導を許容するに十分なプロモーター
の感受性を達成する。ラージT誘導は、次に、プラスミド複製をもたらす。以下
に記載のようなcDNAライブラリーとの同時トランスフェクションは、このような
ラージTで誘導されるプラスミド複製により、シグナリングタンパク質をコード
するcDNAのパーセントが、ライブラリー集団内で濃縮されることを可能にする。
得られる濃縮は、cDNAライブラリー内のライブラリープラスミドのますます小さ
くなるグループの連続的スクリーニングを可能にし、プロモーターを活性化する
生物学的に活性な分子をコードする単一のライブラリープラスミドの同定をもた
らす。
本発明の自己増幅プロセスは、シグナリング分子をコードするcDNAの検出に対
するさらなる感度を提供する。誘導に応答して初期プラスミド複製は、より大き
な遺伝子コピー数に起因して活性なシグナリング分子の増大した発現をもたらす
。シグナリング分子におけるこの増加は、ラージT抗原発現のより大きな増幅を
もたらし、それは次に、より大きなプラスミド増幅をもたらす。
本発明の好ましいベクターは、以下に記載するように、PfLAG-8およびLα2と
名付けられる新規プラスミドを含む。
本発明は、プロモーター、好ましくはヒトプロモーター、そしてより好ましく
は、ヒトc-fosプロモーターを活性化し得るタンパク質をコードするcDNAを同定
する方法をさらに提供する。より好ましいのは、c-fosプロモーター活性化タン
パク質をコードするCROC-1およびCROC-4と名付けられたcDNAである。例えば、CR
OC-1は、配列番号1で示されるアミノ酸配列を有するCROC-1タンパク質と名付け
られる特異的c-fosプロモーター活性化タンパク質をコードする。同様に、CROC-
4は、配列番号3で示されるアミノ酸配列を有するCROC-4タンパク質と名付けら
れる特異的c-fosプロモーター活性化タンパク質をコードする。最も好ましいの
は、配列番号1および配列番号3で示されるヌクレオチド配列である。
本発明はまた、CROC-1またはCROC-4によりコードされるタンパク質の保存的変
異体であるc-fosプロモーター活性化タンパク質をコードするcDNAを提供する。
このような変異体は、それぞれCROC-1およびCROC-4によりコードされるタンパク
質の結合およびc-fosプロモーター活性化機能を有する。
さらに、本発明は、CROC-1またはCROC-4によりコードされるタンパク質のアン
タゴニストである化合物を提供する。これらのアンタゴニストは、CROC-1タンパ
ク質またはCROC-4タンパク質の欠失、置換または付加変異体であるタンパク質を
包含し、そしてそれは、ヒトc-fosプロモーターに結合するが活性化しない。
遺伝コードの縮重のため、本明細書で定義されるようなc-fosプロモーター活
性化タンパク質およびc-fosプロモーター活性化タンパク質アンタゴニストをコ
ードし得る多くの機能的に等価な核酸配列が存在することが理解される。このよ
うな機能的に等価な配列は、化学的合成、改変プライマーを用いるPCR、および
部位特異的変異誘発のような公知の方法を用いて容易に調製され得、本発明の範
囲内である。
本明細書で用いられる用語「組換えベクター」は、本明細書で述べられるよう
な組換えプラスミド、およびGellerら,Proc .Natl.Acad.Sci.USA,87,1149
(1990)に記載のように加工され得る組換えレトロウイルスベクターの両者を含む
。
前述の組換えベクターは、本明細書で定義されるように、トランスフェクショ
ンされ、そしてベクター複製を許容し得る任意の呻乳類細胞をトランスフェクト
するために用いられ得る。新鮮な組織外植体(一次細胞)由来の細胞が原則として
用いられ得るが、確立された細胞株の使用が好ましい。多くのこのような細胞株
が利用可能であり、例えば、NIH 3T3マウス(ATCC番号CRL 1658)、L-M(TK-)マウ
ス(ATCC番号CCL 1.3)およびBALB/c 3T3クローンA31マウス(ATCC番号CCL 163)細
胞株を含む。
本発明の方法における細胞または細胞株の選択は、用いられるベクターの既知
のまたは決定され得る特異性により導かれる。例えば、マウス細胞株は、ヒトc-
fosプロモーターのような、調節プロモーターの制御下でポリオーマウイルスラ
ージT抗原遺伝子を含む組換えベクター;およびポリオーマウイルス複製開始点
、ならびにヒトプロモーター活性化タンパク質をコードすると考えられる核酸を
含む哺乳類組換え発現ベクターであって、例えば、この核酸を発現し得るレトロ
ウイルスLTRを含むレトロウイルス発現ベクターのようなベクターとの使用に好
ましい。
本発明において使用する細胞は一次的にトランスフェクトされるけれども、安
定に形質転換された細胞もまた用いられ得る。哺乳類細胞株の安定な形質転換は
、細胞を、上記の組換えベクターの1つおよび抗生物質のような選択薬剤に耐性
を付与する第2のベクターと同時トランスフェクトする標準的方法を用いること
により達成され得る。
本発明の方法を用いて、ヒトc-fosプロモーター活性化タンパク質をコードす
る核酸を同定するために、細胞を、ポリオーマウイルスラージT抗原遺伝子に作
動可能に連結されたヒトc-fosプロモーター、およびポリオーマウイルスの複製
開始点を含む哺乳類組換え発現ベクター中に組み込まれたcDNAライブラリーを含
む組換えベクターで同時トランスフェクトする。次いで細胞を、ヒトc-fosプロ
モーター活性化タンパク質をコードするcDNAを含むベクターがベクター複製の増
大を刺激する条件下でインキュベートする。次いで細胞を回収し、プラスミドを
抽出し、複製しなかったベクターはDpnIで選択的に消化する。複製したプラスミ
ドは、DpnI消化物てコンピテント細菌を形質転換することにより回収される。
代表的なインキュベーションは、湿潤CO2インキュベーター中、37℃で2日間
実施されるが、条件の選択は、当業者に明らかであり、そして、例えば、細胞の
性質、用いられる培地、および培養コンテナのタイプに依存する。インキュベー
ションは、強い複製応答の進展を許容するに十分な時間続けられる。最適時間は
ルーチンの実験により決定され得るが、代表的には、約24時間〜72時間の範囲で
ある。
実質的に増加したレベルのベクター複製およびDpnI消化後の回収は、ヒトc-fo
sプロモーター活性化タンパク質をコードする核酸を含むようなベクターについ
て、このようなベクターを欠いているベクターの複製から得られるバックグラウ
ンドと比較して検出される。
ベクター複製および回収における実質的な増加は、ヒトc-fosプロモーター活
性化タンパク質をコードする核酸を含むプラスミドが全く存在しないときに測定
されるレベルの、代表的には、少なくとも約5倍、好ましくは約8倍、そして最
も好ましくは約20倍のレベルの増加てある。増加の程度は、主に、バックグラウ
ンドの複製レベルに依存する。
実質的に同じ手順が、他のヒトプロモーター活性化タンパク質をコードする核
酸を同定するために用いられるが、それはポリオーマウイルスラージT抗原をコ
ードする核酸に作動可能に連結されたプロモーターを含むベクターを利用するこ
とによる。
本発明の方法を用いる、ヒトc-fosプロモーター活性化タンパク質アンタゴニ
ストのスクリーニングでは、ヒトc-fosプロモーターに作動可能に連結されたリ
ポーター遺伝子を含む第1の組換え発現ベクター、およびヒトc-fosプロモータ
ー活性化タンパク質をコードする核酸を含む第2のベクターで同時トランスフェ
クトされる細胞が提供される。好ましいリポーター遺伝子は、以下に記載するfo
s-CATリポーター遺伝子またはfos-lacZリポーター遺伝子である。細胞は、用い
られる細胞の種類に適切な培養培地に植えられる。
次いで、細胞は、推定されるアンタゴニストを含む変化する量の試料の存在下
または非存在下(コントロール)、ヒトc-fosプロモーター活性化タンパク質をコ
ードする遺伝子が発現する条件下でインキュベートされる。このような条件下、
およびアンタゴニストの非存在下では、ヒトc-fosプロモーターの剌激が起こり
、リポーター遺伝子が発現する。試料は、例えば、単離化合物が溶解された、ま

代表的なインキュベーションは、約37℃で湿潤CO2インキュベーター中で実施
されるが、条件の選択は当業者に明らかであり、そして例えば、細胞の性質、用
いられる培地、および培養コンテナのタイプに依存する。
インキュベーションは、顕著なリポーター遺伝子誘導を可能にするに十分な時
間継続し、その時点でリポーター遺伝子の発現レベルが適切なアッセイにより測
定される。測定を行う最適時間はルーチンの実験により決定され得るが、代表的
には、約24〜72時間の範囲、好ましくは約48時間である。
リポーター遺伝子発現の最も高いレベルは、コントロール(アンタゴニストが
ない)培養で測定され得る。培養がヒトc-fosプロモーター活性化タンパク質アン
タゴニストを含む場合、リポーター遺伝子発現レベルの低下が測定され、その程
度は、培地に添加されたアンタゴニストの量の直接の関数である。培養のいくつ
かに添加された試料中に存在するアンタゴニストは、リポーター遺伝子発現の実
質的に低減したレベルを、コントロール培養で測定されたレベルと比較して測定
することにより同定される。
リポーター遺伝子発現の実質的に低下したレベルは、ヒトc-fosプロモーター
活性化タンパク質のアンタゴニストの全くない条件で測定されたレベルの少なく
とも50%、そして好ましくは少なくとも約70%の減少として定義される。勿論、
減少の程度は、用いたヒトc-fosプロモーター活性化タンパク質およびアンタゴ
ニストの効果と比較した試料中に存在するアンタゴニストの質により影響される
。
試料の一般毒性に起因するリポーター遺伝子発現の低減したレベルは、β-ア
クチンプロモーターにより駆動されるlac-Zのような構成的に発現する第2のリ
ポーター遺伝子をトランスフェクトし、そしてlac-Z発現に対してc-fosリポータ
ー遺伝子活性を補正することにより説明され得る。
以下の限定されない実施例は、本発明の例示のために供される。
実施例 材料および一般的方法
特に示さない限り、以下の、固体混合物中の固体、液体混合物中の液体および
液体中の固体に対するパーセントは、それぞれ、wt/wt、vol/vol、およびwt/vol
ベースである。細胞の培養中は無菌状態が保たれる。
Sambrookら、「Molecular Cloning.A Laboratory Manual,第2版」、Cold S
pring Harbor Laboratory Press(1989)に記載されているような標準的な組換え
方法を使用した。
DpnIは、Diplococcus pneumoniaeから単離された既知の制限エンドヌクレアー
ゼであり、ICN Biomedicals,Sigma Chemical CompanyまたはNew England BioLa
bs,Inc.から購入し得る。
制限エンドヌクレアーゼAsel、BamHI、BglII、BstXI、ClaI、FspI、HincII、N
arI、NotI、SacII、SalI、ScaI、XbaIおよびXhoIは既知であり、例えば、Sigma
Chemical Companyから購入し得る。
制限エンドヌクレアーゼBamHI、BssHII、BstXI、HincII、SalI、ScaIおよびXb

ベクターLα2を調製するために用いられる合成ポリリンカーはNew England B
ioLabs,Inc.から得、配列番号2に示される配列を有している。NcoIリンカーd
(pAGCCATGGCT)は既知てあり、New England BioLabs,Inc.(カタログ番号1150)
から購入し得る。
ベクターpUC19(ATCC 37254、GenBank Accession受託番号X02514)は、New Engl
and BioLabs,Inc.またはICN Biomedicalsから購入し得る。pUC19のヌクレオチ
ド配列および制限部位は、Yanisch-Perronら、Gene,33,103-119(1985)に記載
されている。
以下のDNAは、本発明のプラスミドの調製に使用され、公衆に入手可能である
:ポリオーマウイルスDNA A2株(ATCC番号45017);およびヒトゲノムc-fos(ATCC
番号41042)。付言すると、ポリオーマウイルスA2株のDNA配列は、DNA Tumor Vir uses
、Tooze,J編、(1980)(Cold Spring Harbor Press)、834-838頁に報告さ
れている。
レトロウイスルベクターpMV7の構築は、Kirschmeierら、DNA,7,219-225(198
8)に記載されており、プラスミドpPyoriおよびpMV(ATCC番号37190)から出発する
。ベクターpMV7は当業者に周知であり、多くの研究所に、無料で広く配布されて
いる。さらに、pMV7に類似し、本願発明において代わりに用いられ得るレトロウ
イスルは容易に入手可能であり、例えば、pV-mos(ATCC番号41037)がある。
以下に記載されるfos-CATリポーター遺伝子構築物は、購入可能なpCAT-ベーシ
ックベクター(Promegaカタログ番号E1041)を用いて調製された。
赤血球凝集素エピトープおよび蛍光結合ウサギ抗-マウスIgGに対するマウスモ
ノクローナル抗体は、Boehringer Mannheimから購入し得る。
cDNAライブラリーのスクリーニングのために、一方向性cDNAライブラリーを作
製したが、これは、ヒト脳ポリA RNA(Clontech,Palo Alto,CA)からGIBCO(Gran
d Island,NY)のSuperscriptクローニングキットを用い、プラスミドLα2のSalI
/NotI部位に挿入することにより行った。
核酸の分離と視覚化は、Sambrookら、前出、に記載のように行い、アガロース
ゲルで電気泳動し、エチジウムブロマイドを用いて視覚化した。すべてのヌクレ
オチド配列決定は、ジデオキシを介するチェーンターミネーション法を用いて実
施した。この方法は、Sangerら、Proc .Natl.Acad.Sci.USA,74,5463-5467(
1977)に記載されている。CROC-1およびCROC-4の配列を得るために、両鎖のDNA配
列決定を実施した。
PfLAGと、生物学的に活性なシグナリング分子をコードするcDNAを含むLα2と
で細胞を同時トランスフェクトすることにより、c-fosプロモーターの活性化を
引き起こし、ラージT抗原が生産された。ラージT抗原の生産はポリオーマウイ
ルスの複製開始点を有するプラスミドの細胞内複製を刺激する。プラスミドを、
Hirt、J .Mol.Biol.,26,365-369(1967)に記載の方法を用いて「Hirt抽出」す
ることによりトランスフェクトされた細胞培養物から回収する。複製しなかった
プラスミドはDpnIによる制限切断で選択的に破壊される。複製されたプラスミド
は、次いで、コンピテントな細菌を形質転換することにより回収される。
初期の継代NIH 3T3マウス繊維芽細胞(ATCC番号CRL 1658)およびラット2繊維
芽細胞(ATCC番号CRL 1764)を10%の仔ウシ血清と50μg/mlの硫酸ゲンタマイシン
とを補足したDMEM培地で増殖させた。
本発明に使用するDH10B E.coliは、GIBCOから購入し得る。プラスミドの構築:
本発明に用いる2つの基本的なプラスミドを構築した。第一のプラスミド(PfL
AGと表される)は、プロモーターの活性化に際しラージT抗原の供給源として機
能し、そして、ヒトc-fosプロモーターに基づくヒトプロモーターで調節される
ポリオーマウイルスラージT抗原遺伝子を有していた。第二のプラスミド(Lα2
と表される)は、ポリオーマウイルスの複製開始点を有するレトロウイルスcDNA
発現ベクターであった。
レトロウイルスcDNAベクターLα2を以下のように構築した。ポリオーマウイル
スDNA A2株をBamHI/NarIで消化し、生じた750bpのフラグメントをpUC19のBamHI/
NarI部位に連結し、得られたプラスミドをpOriと名付けた。レトロウイルスベク
ターpMV7をFspI/AflIIIで消化し、生じた、2つのモロニーマウス(Molony murin
e)肉腫ウイルスLTRを含む4KbバンドをpOriのHincII/AflIIIフラグメントに連結
し、pMV7-2と名付けたプラスミドを得た。pMV7-2中の2つのモロニーマウス肉腫
ウイルスLTRの間に存在するネオマイシン耐性遺伝子をSauI/ClaI消化で除去し、
次に、合成ポリリンカー(上記)で置換し、プラスミドpMV7-3を得た。ブルー-
ホワイトスクリーニングを行うために、pUC19中のポリリンカーをNcoIリンカー
で置換し、次いで、360bpのlac Z領域をAseI/NarI消化で除去し、mung beanヌク
レアーゼで平滑末端化し、pMV7-3のポリリンカーに連結した。生じたプラスミド
(Lα2と名付けた)は4.5kbであり、lac Z遺伝子の5'末端および3'末端にそれぞ
れ、ただ一つのSalI部位およびNotI部位を有していた。翻訳開始コドン(これに
は、ヒスチジンのヘキサマーをコードするDNA配列が続いている)を、cDNA挿入
部位の5'側に挿入し、開始コドンを欠いている短縮型cDNA挿入物からのcDNAでコ
ードされた蛋白質の発現を確実にし、そして引き続く蛋白質の精製を助ける。
PfLAGプラスミドを以下の手順で調製した。ヒトc-fosプロモーターの制御下に
あるポリオーマウイルスラージT抗原を導入したが、これは、pcfos-1の5.9Kb B
amHIフラグメント(Curranら、Mol .Cell.Biol.,3, 914-921(1983)に記載)をN
aeIで消化し、c-fos遺伝子の全コード領域を取り除き、そしてポリオーマウイル
ス由来の2.8Kb BstXI/HincIIバンド(ポリオーマT抗原をコードしている)を挿
入することにより行った。ミドルTおよびスモールT発現を、ポリオーマウイル
スDNA配列(Toozeら、前出に報告)の605位にあるScaI部位に停止コドンを挿入
することにより排除した。生じた構築物をPfLAG-1(プロモーターfos/ラージT
抗原として)と命名した。
HELと名付けた第3のベクターを調製して、CROC-1の細胞内位置を同定するた
めに用いた。Lα2のヒスチジンヘキサマーをコードする配列は、BglII/SalI消化
により取り除き、そしてFieldら、Mol .Cell.Biol.,8,2159-2165(1988)に記
載されている9つのアミノ酸のインフルエンザウイルスHA1のエピトープをコー
ドする配列で置換した。次いで、SV40の複製開始点を、ポリオーマの複製開始点
と5'LTRとの間にある唯一のXbaI部位に挿入して、HELを作成した。
第4のベクターを調製して、推定のヒトc-fosプロモーターの活性化タンパク
質がc-fosプロモーターを刺激する能力を確認するために用いた。fos-CATリポー
ター遺伝子(Deschampsら、Science,233,1174-1177(1985)に記載)は、-735(B
amHI部位)から+42(NaeI部位)までのヒトc-fosプロモーターを、pCATベーシック
ベクター(Promega)中の細菌CAT遺伝子の前に挿入することにより調製した。PfLAG- 依存性随伴複製のためのエンハンサー要求性の決定:
本願発明の目的には、PfLAG中のヒトc-fosプロモーターは、静止期の細胞中で
転写的に沈黙していなければならないが、プラスミドの複製を引き起こすに十分
なT抗原を生産することによる、活性な、cDNAにコードされるシグナリング分子
の低レベルの発現に応答するために十分感受性でなければならない。プロモータ
ーからの遺伝子誘導の感受性とレベルは、プロモーターにさらにエンハンサー要
素を導入することにより増大させ得る。複数のエンハンサー要素を、PfLAG-1に
導入するが、それは、c-fosエンハンサー要素を含む約500bpまたはそれより大き
いXhoI/BssHII(平滑末端化されている)を単離し、このエンハンサー領域を前
のPfLAGのXhoI/SacII部位(平滑末端化されている)に連結することにより行う
。
随伴する複製を示すために必要とされるエンハンサーの数を決定するために、
1、2、4、および8個のエンハンサー領域を含む一連のPfLAGを構築した。次
いで以下の実験を行って、PfLAG依存性随伴複製のためのエンハンサー要求性を
規定した。
Muramatsuら、Mol .Cell.Biol.,9,831-836(1989)は、プロテインキナーゼC
の触媒ドメインの発現がc-fosプロモーターを誘導することを示した。ラットの
プロテインキナーゼC-β1のヌクレオチド配列および推定アミノ酸配列は、House
yら、Cell.52, 343-354(1988)に記載されている。ラットプロテインキナーゼC-
β1の触媒ドメインをLα2に導入し、pMVPkCΔβ1と名付けた構築物を作成した。
pMVPkCΔβ1/Lα2混合物と1、2、4、または8個のエンハンサー領域を含むPf
LAGとの同時トランスフェクションは、それ故、PfLAGの各々の感受性をテストす
る手段を提供し得る。
cDNAスクリーニングのために、40個から約1個のプラスミドを検出する閾値の
感受性を用いた。従って、NIH3T3細胞へのPfLAGの各々との同時トランスフェク
ションについて1:40(wt/wt)のpMVPkCΔβ1/Lα2の比を用いて、以下の手順を行
った。トランスフェクション後、細胞を48時間インキュベートした。プラスミド
を抽出し、そしてDpnI消化後、随伴複製を示す高められたプラスミドの回収を調
査した。これらの条件下で得られた結果を表1に示す。これらのデータは8個の
エンハンサー領域(PfLAG-8)が、プラスミドの複製の有意な活性化に必要であり
、そしてPfLAG-8をベクター単独で同時トランスフェクトした結果から生じるバ
ックグラウンドに対して、プラスミドの回収が8倍増大されたことを示す。PfLA
G-8の誘導は、プラスミド回収において6倍から20倍以上増加の範囲であり、主
にバックグラウンドのレベルに依存した。
プロモーター活性化タンパク質をコードしているプラスミドの濃度の、プラス
ミドの全集団におけるpMVPkCΔβ1の回収に対する影響は、PfLAG-8との同時トラ
ンスフェクションの前に、pMVPkCΔβ1/Lα2混合物中のpMVPkCΔβ1の濃度を変
えることにより測定した。Lα2はpUC19由来の改変されたlacZ遺伝子を有してい
るので、Lα2で形質転換された細菌は、アンピシリン、X-gal、およびIPTGを含
有する寒天平板上にプレートしたときに青色にかわるが、pMVPkCΔβ1で形質転
換された細菌は白いままである。pMVPkCΔβ1のパーセントは、DpnI消化Hirt抽
出物で細菌を形質転換して生じた総コロニー数のパーセントとして白色コロニー
の数を表すことにより測定した。
PfLAG-8をpMV7PkCΔβ1/Lα2混合物と同時トランスフェクトすることにより行
われた実験は、1:80の比(wt/wt)で開始し、次いで、希釈して1:400の比まで下げ
たが、以下に記載の方法で行った。DpnI消化Hirt抽出物でコンピテントDH10B E.
coliを形質転換し、そしてアンピシリン、X-gal、およびIPTGを含有する寒天上
にプレートした。回収されたコロニー中のpMV7PkCΔβ1のパーセントは、総コロ
ニーに対する白色コロニーの数により測定した。
pMVPkCΔβ1の形質転換から白色コロニーが生じたことを確認するために、プ
ラスミドを回収し制限酵素でマップした。全ての白色コロニーは、正しいpMVPkC
Δβ1の制限パターンを示した。表2に示した結果は、回収されたコロニーの数
が高いpMV7PkΔβ1希釈でバックグラウンドレベルに対して減少したが、pMV7Pk
Δβ1コロニーの実際のパーセントは増加すること;を示し、最低限400のライブ
ラリーコロニーが、pfLAG-8でトランスフェクトされ、シグナル伝達分子をコー
ドするcDNAについてcDNAライブラリー集団を濃縮し得ることを示す。従って、c-
fosプロモーターのアクチベーターをコードするcDNAが濃縮されたライブラリー
集団を得るために、最初のcDNAライブラリースクリーニングを、400またはそれ
以上のプラスミドからなるプラスミドプールを用いて行った。
細胞培養とトランスフェクション:
トランスフェクションのために、8×105の3T3細胞を100mmディッシュ中の増殖
培地に移植し、そして一夜付着させた。翌日、リン酸カルシウムを用いて、Wigl
erら、Cell,11, 223-232(1977)の方法によりトランスフェクションを行った。
リン酸カルシウム沈殿に4時間さらした後、細胞をリン酸緩衝化生理食塩水で2
回洗浄し、0.5%仔牛血清を補足したDMEMを再供給し、そして37℃で40-48時間イ
ンキュベートした。細胞を回収し、そしてプラスミドを、Hirt、前出の手順によ
り抽出した。抽出したプラスミドを、DpnIを用いて最低24時間消化した。DpnI消
化物をフェノール抽出し、そしてエタノール沈澱させた。DNAを20μl TE(1mM E
DTA+10mM Tris,pH8.0)に再縣濁し、そしてコンピテントDH10B細菌(GIBCO)中に
形質転換した。
同時トランスフェクションは上記の手順で行ったが、9:1(wt/wt)のcDNA/PfLAG
比で、皿あたり20μgのDNAを用いた。随伴複製を用いるcDNAライブラリースクリーニング:
ヒト脳cDNAライブラリーを、上記の方法でPfLAG-8とともにNIH 3T3細胞に同時
トランスフェクトした。約30-40プラスミドからなるプラスミドプールをPfLAG-8
とともに同時トランスフェクトし、そしてプラスミド回収を最低5倍増加させる
かについて検討した。活性なプールからのプラスミドを回収し、そして、それぞ
れ、4つのプラスミドの第2のプールに分け、随伴複製の活性化について同様に
調べた。次いで、それぞれの活性な第2プールからのプラスミドの個々について
、随伴複製を調べた。約1400のプラスミドから、最初に、2つのプラスミド(CRO
C-1およびCROC-2と名付けられた)を、(cDNAの随伴複製について)をスクリーニン
グし、PfLAG-8と同時トランスフェクトしたとき、一貫して増加したプラスミド
回収を与えた。CROC-1のヌクレオチド配列を配列番号1に示す。
第3のプラスミド(CROC-4と名付けた)は、さらなるプラスミドスクリーニング
により同定した。プラスミドCROC-4もまた、PfLAG-8と同時トランスフェクトし
たとき、一貫して増加したプラスミド回収を与えた。CROC-4のヌクレオチド配
列を配列番号3に示す。fos-CAT リポーター遺伝子を用いるc-fosプロモーター活性化の確認:
特定の外来性の因子がまた、随伴複製アッセイにおいて観察される増加したプ
ラスミド回収を引き起こし得た。例えば、DpnI部位の不完全な細菌のメチル化は
、DpnI耐性、またはトランスフェクションもしくは形質転換効率の差異を与える
。これらの可能性を除くために、CROC-1,CROC-2およびCROC-4のそれぞれをfos-
CATリポーター遺伝子と同時トランスフェクトし、そして以下のように、CAT活性
の増加についてテストした。ラット2細胞を、18μgのLα2-発現cDNA(即ち、CRO
C-1,CROC-2またはCROC-4)+2μgfos-CATで、4時間、同時トランスフェクトし
、次いで、DMEM+0.5%ウシ血清を再度供給した。トランスフェクション後、72時

CAT活性は、CROC-1、CROC-2およびCROC-4により、有意に誘導され、c-fosプロ
モーター活性化を示した。活性化の程度は、pMVPkCΔβ1との同時トランスフェ
クションにより引き起こされる活性の約50%であった。対照的に、ベクター単独
ては実質的なCAT活性を誘導しなかったし、CROC-1、CROC-2およびCROC-4とおな
じプラスミドプールから単離されたランダムに選択されたcDNAライブラリープラ
スミドであるが、随伴複製を活性化しなかったプラスミドも活性化しなかった。
これらの結果は、CROC-1、2または4をPfLAG-8と同時トランスフェクトした際に
、プラスミドの回収が増加することは、PfLAG-仲介随伴複製におけるc-fosプロ
モーターの活性化によることを確証した。c-fos 活性化タンパク質の解析:
配列決定の結果、CROC-2は、最近同定されたα2-マクログロブリンレセプター
結合タンパク質(AMRAP)(Stricklandら、J .Biol.Chem.,266,13364-13369(1991
)に記載されている)をコードしていることが示された。挿入物はほぼ全長を有
しており、そして開始コドン(これは、内部ベクターの開始コドンとインフレー
ムである)からポリAテールまで伸びていた。
CROC-4のヌクレオチド555-897に対応する347塩基対の配列は、発現配列tagと
してGenBank(受託番号Z40809)に提出されている。
CROC-1cDNAは、配列番号1に示すように、酸性のアミノ末端半分と塩基性のカ
ルボキシ末端とを有する19kdのタンパク質をコードしている。このタンパク質は
、キナーゼ標的ドメインを含んでおり、このドメインは、シグナル伝達に関与す

CROC-1タンパク質のキナーゼ標的ドメインは、塩基性ドメインの開始点に位置
する12アミノ酸の長さである。既知の酸性ドメインのトランスアクチベーター活
性は一般に、塩基性ドメインをDNAと結合する可能性と組み合わせると、CROC-1
が、その活性がキナーゼ標的ドメインのリン酸化により調節される転写アクチベ
ーターとして作用し得ることを示唆する。リン酸化は、この領域の酸性度をさら
に増大させ、それによって、転写活性化のその能力を増加させ、また、このタン

CROC-1 mRNAの長さと組織分布を、CROC-1の1.8kb SalI/NotI挿入物をプローブ
として用い、種々のヒト組織から単離したポリAを含むRNAのノーザン解析により
測定した。CROC-1 mRNAは、約2.3kbの長さであり、本発明者らのcDNA挿入物より
も約0.5kb長く、そして調べた全ての組織に存在し、脳、骨格筋、および腎臓で
最も高いレベルであった。これに比べて、1.5kb CROC-2 mRNAは、調べた全ての
組織に存在したが、心臓、胎盤、および腎臓で最も高いレベルで発現していた。
Strickland、前出、によってCROC-2について報告されているような二者択一のス
プライシングまたは転写-終結-ポリアデニル化シグナルの複数のセットの結果と
してのさらなる転写物に対する証拠は見い出されなかった。
CROC-1タンパク質の細胞内局在化を、HEL中にCROC-1をクローニングし、そし
て生じたプラスミドをCOS-7細胞(ATCC番号CRL 1651)にエレクトロポレーション
することにより測定した。CROC-1核酸のHELベクターへの導入により、赤血球凝
集素エピトープのCROC-1タンパク質へのインフレームでの融合を可能にする。次
いで、CROC-1タンパク質の細胞内位置を、赤血球凝集素エピトープに対するマウ
スモノクローナル抗体を用いる免疫蛍光顕微鏡により測定した。CROC-1のHELへ
