発明の概要
出願人は、新規のアルカリ性ホスファターゼを好熱菌種サーマス・サーモフィ
ルス(Thermus thermophilus)から単離し且つ精製した。
この酵素は、極めて高い最適pH(pH13またはそれより大)を有し且つ耐熱
性であり、65℃で24時間のインキュベーション後でさえも、その活性の少な

サーマス・サーモフィルス菌株HB8を、この微生物中のアルカリ性ホスファ
ターゼを減少させる無機リン酸塩を制限量で含む規定の培地中(イェーおよびト
レラ(Trela);251 J.Biol.Chem.3134,1976年
から修飾された)において75℃の好気的条件下で増殖させた。培地は、1リッ
トルにつき以下の塩類を含んでいた。ニトリロトリ酢酸100mg、CaSO4
・2H2O 60mg、MgSO4・7H2O 100mg、NaCl 8mg、
KNO3105mg、ZnSO4・7H2O 5mg、H3BO35mg、CuSO4
・5H2O 0.16mg、Na2MoO4・2H2O 0.25mg、CoCl2
・6H2O 0.4mg、MnSO4・H2O 22mg、FeCl3・6H2O
0.28mg。ビタミン類を1リットルにつき以下のように加えた。ビオチン0

液は、特に断らない限り、6mM p−ニトロフェニルリン酸、100mM C
APS(pH11)および15%グリセロールを含んでいた。
凍結細胞を融解させ、10mMトリス−HCl(pH8)、1M MgCl2
および1mM CaCl2中に再懸濁させ、そして音波処理によって溶解させた

SDS−PAGEによる標品の予備的分析は、酵素活性のピークが、見掛けの

ある。最終生成物は、49kdポリペプチドに対して約80%均一であり且つ比

Tthアルカリ性ホスファターゼは、かなり広範囲のpH値にわたって測定可
能な活性を示すが、異常に高い最適pHを有するらしく、13.0が検定した最
高値である(図1)。このような極端なpHでの用途は希であるので、特性決定
の大部分はpH11.0で行われた。最適条件(100mM CAPS pH1
1.0、15%グリセロール)下において、酵素は37℃で≧250単位/mg
の比活性を示す。酵素活性は、緩衝液(図1および5)、イオン強度(図2およ
び3)、グリセロール(図4)および温度(図5)を含めた様々な他の因子によ
って影響される。酵素はまた、それが1mM EDTAによって阻害されるので
(データは示されていない)、二価の陽イオンを必要とすると考えられる。しか
