組合せ化合物ライブラリーの製法
本発明は、固体又は半固体担体(ビーズ)、合成オリゴマー(リガンド)及び
これらのリガンドのモノマーをコードする同定構造(tag)から成る複数の異なる
ユニットの製法、並びに新規クラスの化合物及び個々の化合物の検索のためのそ
のライブラリーの使用に関する。さらに本発明は、上記新規プロセスにより検出
された化合物及びトロンビン・インヒビターとしてのその使用に関する。
近年、特定のレセプターにだけ選択的に結合する化学物質についての断えず増
大する要求が在る。これらの化合物は、そのとき、とりわけ、インヒビター、ア
ゴニスト、アンタゴニストとして、又はマーキングのために使用されることがで
きる。これに関して、その可能性は、多数(ライブラリー)の異なる化合物を同
時に合成し、そしてそれらを、特定のレセプターへのそれらの結合能力について
検定することから開発されてきた。これらのライブラリー(組合せ化合物ライブ
ラリー(combinatorial compound libraries=CCL))は通常、天然アミノ酸又は
ヌクレオチドから、そしてさらによりしばしば、修飾アミノ酸、修飾ヌクレオチ
ド及び他の化学物質であってモノマー構築ブロックとして使用されるものから成
る。
これらのライブラリーの個々の異なるリガンドは、通常、並行して、混合物又
は物理的に分離された個々のものとして合成される。これらの並行合成は、短時
間の間にひじょうに多数の異なるリガンドを含むライブラリーを製造することを
許容する。
文献は、複数のテスト候補から成るライブラリーの化学合成のための2つの内
因的な異なる主な戦略について記載している。
1.リガンドを溶液中一緒にそれらの合成後に得る(Houghten et al.Nature(
1991)354,84-86)。
2.リガンドを、固体担体上で合成し、その合成を、リガンドの1の種だけがそ
の担体に結合されるように制御する(1ビーズ1配列;Lam et al.Nature(1991)
,354,82-84; Furka et al.,14thIntern.Congr.Biochem.FR 013,1988)。
第1の戦略においては、問題は、全てのリガンドがその溶液中に同時に存在す
るとき、所望のリガンドをはっきりと同定することに在る。多数の可変位置をも
つリガンドを同定するために、その各々が20アミノ酸の中の1により占有される
ことができる4可変位置をもつリガンドについて、4×20=80プールが合成され
、そして検定されなければならないように、可変位置当り各可能性のあるモノマ
ーについて1のプールを合成するとが必要である。
第2の戦略は、1のプール内に全ての所望の可能性を作り出し、そしてその所
望のリガンドを含む固体担体の簡単な分離、そしてそれ故のそのリガンドの同定
を許容する。しかしながら、それらの組成物がその後同定されなければならない
ので、使用されるリガンドのタイプにおける制限が、ここに存在する。従って、
(天然アミノ酸からのペプチド、DNA及びRNA)を配列決定することによりはっきり
と同定されることができるリガンドだけが好適である。さらなる欠点は、固体担
体へのリガンドの付着が偽陽性結果を導くことができる、すなわち、多くの検定
が、固体担体に付着されたリガンドを用いて行われることができないということ
である。
これらの知られた可能性とは反対に、本発明は、各固体担体に、たった、1の
タイプのリガンド(1ビーズ、1配列)及びこのリガンドをコードする1の同定
構造(tag)が結合されているライブラリーを提供する。1のタグ(tag)への1のリ
ガンドの明確な指定は、
結合検定において、それらが担体上に存在する量において、公知の方法及び/又
は自動化された手順により明確に(unequirocally)同定されることができないよ
うなリガンドをも同定することを可能にする。さらに驚ろくべき利点は、その検
定の結果かいずれの方法においてもその固体担体により影響されないように、そ
の全体又は部分における同定能力の損失を伴わずに、そのリガンドがその担体か
ら分離されることができるということである。従って、均質溶液中に存在する同
定されたリガンドを用いて、担体に結合されたリガンドを用いては不可能である
検定、例えば、IR,NMR又はMSによるさらなる酵素検定又は純度検定を行うこと
ができる。
従って、本発明は、各々が、固体又は半固体担体(ビーズ)、合成オリゴマー
(リガンド)及びそれによりそのリガンドのモノマーが同定されることができる
同定構造から成る、複数の異なるユニットを含んで成るライブラリーであって:
a)各担体が、たった1のタイプのリガンドだけを担持し、
b)リガンドとタグが、異なる位置において同一担体に付着され、
c)リガンドが、上記タグに拘らず、そしてその情報内容を変更せずに、上記
担体から分離されることができ、
d)上記タグが、配列決定可能なポリペプチドであり、そして
e)上記タグ又はリガンドを合成するために、温和な酸条件下で除去されるこ
とができる保護基が使用される、ようなライブラリーに関する。
従って、上記の進歩性のあるプロセスにより、2つの異なる検定を実施するこ
とができる:
a)そのアクセプター(acceptor)に結合するリガンドを前選択するための第
1検定であって、そのリガンドが上記固体又は半固体
担体に結合されているもの、
b)そのリガンドのさらなる特徴付けのための、その担体及びその情報構造物
からのそのリガンドの分離後の、第2検定。この第2検定は、同様に、そのアク
セプターへのそのリガンドの付着を調べることができ、又はその付着されたリガ
ンドを用いては行われることができない完全に異なる検定であることができる。
温和な酸性条件下で除去されることができる保護基は、例えば、酢酸の、10%
までの、好ましくは5%までの濃度において完全に除去されることができる。こ
れらの好ましい保護基は、ジメチル−ジメトキシベンジルオキシカルボニル保護
基(Ddz)よりも少なくとも10倍より不安定であろう。特に好ましい基は、トリチ
ル・タイプのもの、例えば、非置換又はアルコキシ−置換トリチルである。
用語リガンドは、特別に、多数のモノマーを含んで成る全ての化合物を包含す
る。典型的には、これらのモノマーは、少なくとも2つの反応性基を含む。この
ような反応性基の例示的な例は、アミノ、アジド、イソシアニド、イソシアナト
、ヒドラジノ、カルボニル、カルボキシル、アシルハロゲノ、ヒドロキシル、ス
ルフヒドリル、塩化スルホニル、ホスフェート基、及びハロゲノ基である。この
合成を容易にするために、これらの基の反応性は、保護又は活性化基により修飾
されることができる。2以上の反応性基を含むモノマーの典型的な例は、天然及
び非天然アミノ酸、ω−アミノカルボン酸、糖類、ヌクレオチド及びヌクレオチ
ド・アナログである。2以上の反応性基を含むモノマーを使用するとき、そのリ
ガンドの任意的な分枝及び環化を挿入することもできる。たった1の反応性基を
含むモノマーを末端基として使用することができる。
以下、詳細に特徴付けする進歩性のあるプロセスは、個々の構築ブロックが、
連続するだけではなく、特定の組合せにおいて互いに
基本化合物(a basic compound)に結合されるようなリガンドのライブラリー
を作り出すことをも可能にする。これは、多数の反応性基を含む基本構築ブロッ
クを上記担体に直接的に又はリンカーを介して付着させ、そして次にこの基本構
築ブロックに個々のモノマーであって今度は1又は1以上の反応性基を含むもの
を付着させることにより、行われることができる。本発明の文脈中、基本構築ブ
ロック(Basic building blocks)は、典型的には、ステロイド核、ペニシリン
又はペネム(penem)核、ソラフェン(soraphens)、ベンゾジアジピン(benzodiazip
ines)、糖類又はデスフェロキシアミン(desferoxiamine)である。次に、異な
る反応性基を、標準的な化学的方法によりこれらの核に固定することができる。
好ましいリガンドは、さらなる実験的努力を伴わずに、例えば、公知の自動化
された手順により、配列決定することによっては、明確に同定されることができ
ないモノマーを含む。
配列決定により明確に同定されることができないモノマーは、天然の20アミノ
酸及び DNA及び RNAにおいて天然にあるヌクレオチド以外の全ての化学物質であ
る。典型的な例は、修飾されたアミノ酸又はヌクレオチド、ω−アミノカルボン
酸、D−アミノ酸、糖類、糖類側鎖をもつアミノ酸及びたった1の反応性基、例
えばアセチル又はベンジルを担持する末端基である。
リガンドは、通常、リンカーを通じて上記固体又は半固体担体に付着される。
このリガンドのためのリンカーは、典型的には、少なくとも2の反応性基を含む
化学物質である。好ましいリンカーは、リンカー及びタグが担体上で合成される
だけでなく検定もされることができるように、温和な塩基性及び温和な酸性条件
に耐えるものである。それ故、好ましいリンカーは、特定の反応により解裂され
ることができ、例えば、臭化シアンにより解裂されることができる
メチオニン、又は強塩基又は強酸性条件下でのみ、光分解により、又は還元的又
は酸化的条件下で解裂されることができるリンカーである。特に好ましいリンカ
ーは、リガンドが担体から選択的に除去されることができるように、塩基性条件
下で解裂性の結合を、酸性条件下で安定性の結合を作るものである。好適なリン
カーの例示的な例は、p−ヒドロキシメチル−安息香酸、4−ヒドロキシメチル
フェニル酢酸、ベンズヒドリルアミノ、アルキル、ヒドロキシクロトニルアミノ
メチル、3−ニトロ−4−ヒドロキシメチル安息香酸、p−ニトロベンズヒドリ
ルアミン、4−〔4,4′−ビス(メチルスルフィニル)−2−オキシベンズヒ
ドリルアミノ〕酪酸及びジスルフィド・リンカーであって、その後、固相合成に
おいて“ハンドル(handle)”といわれる基を通じて連結されることができるも
のである。本進歩性のあるプロセスにおける使用に好適なこれらのリンカーは、
上記の解裂性構築ブロックに加えて、その解裂反応に影響し又は影響しないさら
なる構築ブロックであって、その解裂性基から空間的に、そのリガンドの可変部
分を隔離するのに好適なものを含むことができる。
プロテアーゼ、例えば、トリプシン、yscα,yscF又はV8−プロテアーゼに
特異的な部位をもつ短いペプチドをリンカーとして使用することもできる。
特に好ましいリンカーは、好適な条件下、そのリンカーの特定の部分だけが解
裂され、それにより、例えば、さらなる検定のために、いくつかの部分において
固体担体からそのリガンドを分離する可能性を許容するように、その解裂がコン
トロールされることができるものでもある。これらのリンカーは、輝発性又はガ
ス状の剤、例えば、アンモニアにより解裂されることができるようなリンカーが
きわめて特に好ましい。なぜなら、これらのリンカーは、そのリガ
ンドの検定前に、残渣を完全に又はほとんど含まないように除去されることがで
きるからである。
正確に規定され、そしてそのリガンドの不均一性にも拘らず、等しい部分にお
いて、異なるリガンドを解裂させることができるために、その担体とリガンドの
間のリンカーが、上記のような解裂性部分、及び全リガンドについて同一である
部分から成ることができる。定常部分の好適な構築ブロックは、一般的に、その
リンカーの解裂性部分だけでなく、他の化学的構築ブロック又はリガンドへの付
着を許容する化学物質であることができる。例示的な例は、2官能価の化学物質
、例えば、アミノ酸、核酸又はそのリガンドの不変部分である。
情報構造(tag)は、そのコーディング単位が便利には、天然のアミノ酸(Ala,A
rg,Asn,Asp,Cys,Gln,Glu,Gly,His,Ile,Leu,Lys,Met,Phe,Pro,Ser
,Thr,Trp,Tyr及びVal)から成る配列決定可能なポリペプチドである。反応性
の側基を全く担持せず、そして/又は配列決定の間に容易に同定できるアミノ酸
、典型的には、Ala,Asn,Asp,Gly,Ile,Leu,Phe,Pro,Trp 及び Valを使用
することが好ましい。特に好ましいアミノ酸は、Ala,Asn,Asp,Gly,Leu,Phe
及び Proである。このタグは、次に、特定の条件下で解裂可能である上記リンカ
ーの中の1を通じて上記担体に結合されることができる。これに関して重要なこ
とは、リンカーとタグの分離が他から独立していることができ、そして担体から
のリガンドの分離がそのタグの情報を変えないということである。
全20の天然アミノ酸が使用される場合、可変部位当り20の異なるモノマーをコ
ードすることができる。可変部位当り20以上の異なるモノマーをコードすること
が望ましい場合、1以上のアミノ酸が各コーディング単位のために使用される。
従って、ジペプチドを使用
することにより、20の異なるアミノ酸を用いて202の異なるモノマーまでをコー
ドすることができる。
タグを配列決定するときの翻訳の誤りを回避するために、先に列記した好まし
いアミノ酸だけを使用することができる。これらのアミノ酸は、コード性モノマ
ーの数を増加させるためにジ−又はトリペプチドとして使用されることができる
。すなわち、リガンドの各モノマーは、タグ内で1以上のアミノ酸によりコード
される。
タグができるだけ小さくそのテスト結果に影響を及ぼし、そしてそのリガンド
の量ができるだけ多くあることを確保するため、固体又は半固体担体上のリガン
ド対タグの比は、望ましくは1より大きく、好ましくは、2〜100、そして、好
ましくは、4〜10である。
実施を考慮するために、タグは、そのタグを配列決定するとき、そのリガンド
をコードする部分の始め及び終りを正確に同定することができるように、そのリ
ガンドのモノマーをコードしない開始及び/又は終了配列を提供されることがで
きる。このような出発及び終了配列の例示的な例は、そのリガンドのモノマーを
コードしないアミノ酸並びに同一アミノ酸のジ−又はトリペプチド、例えば Val
-Val又は Leu-Leu-Leuである。
用語“固体又は半固体担体”は、反応媒質中で不溶性であり、そしてそれにリ
ガンドとタグの両方が十分な量で結合されることができる肉眼観察可能な粒子を
意味すると理解されるであろう。担体に結合されることができるタグの量は、そ
のタグを配列決定するとき明確に同定することができるシグナル(例えば>1pm
ol)を作り出すために十分に大きなものでなければならない。そのタグが放射性
物質によりマークされたモノマーを含む場合、担体当り1pmol未満のタグも、放
射性化合物についてのより低い検出限界のために、同定されることができる。
リガンドとタグの結合は、その担体の表面における反応性基、例えば、アミノ
、カルボキシル、ヒドロキシル又はハロゲン基により行われる。これらの反応性
基は、通常、既にその担体の構成成分であるが、それらはその後に適用され又は
修飾されることもできる。固相合成において慣用される樹脂が、使用されること
ができ、例えば、それらは、Merrifieldペプチド合成において使用される。それ
らは、ほとんどジビニル・ベンゼンとの共重合により架橋されるポリスチレン分
子から成る。これらの分子は、固相合成においてそれらの反応体に付着させるた
めにさらに誘導体化される。
本発明は、直交(orthogoral)及び他の合成によるリガンド及びタグの合成を
含んで成る、上記ライブラリーの製法にも関する。
本発明のライブラリーの合成は、以下の段階;
a)タグの第1ユニット(好ましくは、等量未満、すなわち、担体上の反応性
基に基づき 50mol%未満)及びリガンドの第1モノマー又はその構築ブロック又
は各々についてのリンカーを、固体又は半固体担体に付着させ;
b)場合によりさらなる非−可変性モノマーを、上記リガンドに、又はさらな
る非−可変性コーディング・ユニットを、上記タグに付着させ;
c)上記固体又は半固体担体を、上記リガンドの可変性モノマーのための部分
に分割し;
d)各部分において別々に、コード性配列内で、上記リガンドにおいてさらな
る修飾を行い、又はそのリガンドのこの部位において可能性のある他の可変性モ
ノマー及びこの段階をコードするタグのユニットを付着させ;
e)これらの部分を混合し;
f)上記リガンドの可変性部分が完全に合成されるまで、段階b
)〜e)を繰り返し;そして
g)場合により、1又は1以上のさらなる不変モノマーを、上記リガンドに、
又はさらなる非−コーディング・ユニットを、上記タグに付着させ(反応スキー
ム1及び2参照);
を含んで成り、温和な酸性条件下除去されることができる保護基をタグ又はリガ
ンドの合成のために使用する。
上記タグの第1ユニット及び上記リガンドの第1モノマーと上記第1モノマー
のためのリンカーとの結合のタイプは、選ばれた反応性基のタイプに依存し、そ
してこのような基のための標準的な方法により行われる。
さらなる合成は、固相合成のための標準的な公知の方法(Fields et al.,Int
ern.J.Pept.Prot.Res.(1990),35,161-214)により行われる。リガンド及
びタグの合成のために使用される過渡的な保護基(各合成段階の前に除去される
ことができる保護末端基)は、直交保護基である。すなわち、リガンドとタグの
合成は、互いに独立して行われることができる。例えば、2グループの保護基が
使用されることができる。第1グループは温和な酸性条件下で除去され、そして
他のグループは塩基性条件下、光の働きにより、酸化又は還元条件下で除去され
る。温和な酸性条件下で除去されることができる保護基と塩基性条件下で除去さ
れることができる保護基との組合せを使用することが特に好ましい。
リガンド合成の間に無傷で残るであろう多数の保護基との適合性を確保するた
めには、酸除去性の過渡的保護基は、特に著しい酸不安定性をもつものである。
好適な酸不安定性基は、ジメチルジメトキシベンジルオキシカルボニル保護基(D
dz)よりも少なくとも10倍以上不安定なものである。
十分な直交性(orthogonality)を達成するために、塩基−除去性
のフルオレニルメトキシカルボニル(Fmoc)が、好ましくは、その1の構造の合
成のために使用され、そしてその他の構造の合成のために、酸−除去性トリチル
(Trt)又は置換トリチル保護基、例えば、アルコキシ−置換トリチルが使用され
る。
保護基のタイプの組合せの例示的な例は以下のものである:
β−脱離タイプの基は、典型的には、フルオレニルメチル・タイプの保護基で
あり;上記 NPSは、ニトロフェニルスルフェニル・タイプの基を示す。適格な基
についての一般的な記載は、Fields et al.(Intern.J.Pept.Prot.Res.(1
990),35,161-214)中に見られる。
また、光分解により除去されることができる保護基、例えば、ニトロベンジル
又はニトロベラトリルオキシカルボニル基とのリガンド又はタグのための保護基
の上記グループの中の1の組合せも好適である。
例えば、アミドの合成のために必要とされる反応段階は、本分野において広く
知られており、そして通常、その反応に参加するカルボン酸基の活性化のタイプ
に依存する。この反応は、通常、縮合剤の存在中で、又は、無水物の形態でその
カルボン酸を活性化すると
き、形成されたカルボン酸に結合する剤の存在中で、行われる。ある場合には、
カオトロピック(chaotropic)試薬、例えば、NB−メチルピロリドン中の LiFを
添加することもできる。これらの反応は、−30℃〜+ 150℃の、好ましくは+10
℃〜+70℃の、そして最も好ましくは+20℃〜+50℃の温度レンジ内で、そして
適宜、不活性ガス雰囲気中でも行われる。
有用な縮合剤の例示的な例は、カルボジイミド、例えば、N,N′−ジエチル
−、N,N′−ジイソプロピル−、N,N′−ジシクロヘキシル−又はN−エチ
ル−N′−(3−ジメチルアミノプロピル)−カルボジイミド;カルボニル化合
物、例えば、カルボニル・ジイミダゾール;1,2−オキサゾリウム化合物、例
えば、2−エチル−5−フェニル−1,2−オキサゾリウム−3′−スルホネー
ト及び2−tert−ブチル−5−メチルイソキサゾリウム・パークロレート;アシ
ルアミノ化合物、例えば、2−エトキシ−1−エトキシカルボニル−1、2−ジ
−ヒドロキノリン;及びウロニウム化合物、例えば、2−(1H−ベンゾトリア
ゾール−1−イル)−1,1,3,3−テトラ−メチルウロニウム・テトラフル
オロボレート(TBTU);又はホスホニウム化合物、例えば、ベンゾトリアゾール
−1−イルオキシ−トリス(ジメチルアミノ)ホスホニウム・ヘキサフルオロホ
スフェート(BOP)又はベンゾトリアゾール−1−イル−オキシ−ピロリジノホス
ホニウム・ヘキサフルオロホスフェート(PyBOP)である。
有用な酸アクセプターは、典型的には、アルカリ金属、カーボネート又はビカ
ーボネート、例えば、ナトリウム又はカリウム・カーボネート又はビカーボネー
ト(通常スルフェートと共にある)、又は有機塩基、例えば、立体的に妨害され
たトリ−低級アルキルアミン、例えば、N−ジイソプロピル−N−エチルアミン
である。
反応に参加しないリガンドとタグのモノマーの反応性側鎖は、第3グループの
保護基により保護されることができる。それらの導入と除去のために有用な保護
基とプロセスは、とりわけ、“Protective Groups in Organic Chemistry”,Pl

ヒドロキシ保護基の典型的な例は、アシル基、例えば、tert−ブトキシカルボ
ニル基、エーテル化基、例えば、tert−ブチル基、及びシリル又はスズ基、例え
ば、トリ−n−ブチル・スズ基又はtert−ブチルジメチルシリルである。
カルボキシル基は、上記tert−ブチル・タイプ、ベンジル、トリメチルシリル
エチル又は2−トリフェニルシリル基とのエステル形成により保護されることが
できる。
アミノ基は、便利には、除去性アシルアミノ、アリールメチルアミノ、エステ
ル化メルカプトアミノ、2−アシルアルク−1−エニルアミノ、シリルアミノ、
スズ・アミノ又はアジド基であって、tert−ブトキシカルボニル、アリルオキシ
カルボニル、ベンジルオキシカルボニル、4−ニトロベンジルオキシカルボニル
、ジフェニルメトキシカルボニル、ニトロフェニルスルフェニル、2,2,2−
トリクロロエトキシカルボニル、ペンタメチルクロマノスルホニル
(PMC)又はメトキシトリメチルベンジルスルホニル(Mtr)保護基を含むものにより
保護されることができる。
チオールは、アセトアミドメチル基により保護されることができる。
これらの保護基は、通常、ペプチド化学の常法により、便利には95%トリフル
オロ酢酸により、リガンドとタグの完全な合成後、一緒に除去される。ある場合
には、強い求核基、例えば、1,2−エタンジチオールがさらに、その生成され
た保護基を捕獲するために、添加されることができる。
これらの保護基の除去のさらなる方法は公知であり、そして、とりわけ、β−
脱離、ソルボリシス、加水分解、アルコリシス、アシドリシス、塩基による処理
又は還元を含んで成る。
d)に記載した反応のための部分の数は、好ましくは、各ケースにおいて、リ
ガンドの次のモノマーが付着される可能性の数に対応する。上述のように、タグ
の1ユニットは、1又は1以上のアミノ酸から成ることができる。1ユニットが
数個のアミノ酸から成る場合、これらの各々は順番に、その成長タグ付着され、
又はユニットとして別々に合成され、そして次に1反応においてそのタグに付着
されることができる。タグの配列決定は、その開始及び終了に、その情報を担持
する部分の出発又は終了(開始配列/終結配列)を示す1以上のアミノ酸をさら
に付着させることによりより容易に行われることができる。
所望のアクセプターに結合するリガンドを同定するために、上記のように調製
したライブラリーを、調査すべきアクセプターにより処理し、そしてアクセプタ
ーが付着した担体を洗浄し、そして単離する。
本発明の範囲内で可能性のあるアクセプターは、1又は1以上の
リガンドに結合するアフィニティーをもつ巨大分子ユニットである。例示的な例
は、セロトニン・レセプター、GABAレセプター及びベンゾジヤゼピン・レセプタ
ーを含むレセプター、輸送タンパク質、例えば、Na及びKチャンネル、抗体及び
酵素、例えば、プロテアーゼ、トロンビン、レニン(renin),ACE、アロマターゼ
及び逆転写酵素;又はリガンドについて同一の結合特性をもつそれらの断片であ
る。これらのアクセプターに結合するリガンドは、アンタゴニスト、インヒビタ
ー、アゴニストとして、又はアクセプターのためのマーカーとして働く。
既に天然には存在しない場合、これらのアクセプターは、同定可能な基、例え
ば、蛍光性、化学発光性又は放射性基、アビジン、ビオチン、リポーター酵素、
免疫学的に検出可能な基(ELISA)その他を提供される。上記のマーカーによりマ
ーキングする方法及びアフィニティー・クロマトグラフィーの方法は、一般に、
本分野において公知である。
アクセプターが結合する担体の単離は、例えば、UV又は蛍光性アクセプターの
場合には青色光下で、又は細胞をソーティングのために典型的に使用される自動
化装置の助けを借りて、担体をソーティングすることにより手動で行われる。
このやり方で単離された各担体のタグは、慣用のペプチド分析法、例えばEdma
n分解により配列決定されることができ、そしてその担体に付着されたリガンド
ははっきりと同定される。配列決定は、リガンドの分離前又は後に、その担体に
付着されたタグにおいて、例えば、自動化配列決定装置により行われることがで
きる。リガンドの分離後の配列決定が好ましい。この場合においては、開放され
たリガンドのモノマーは、その配列決定を妨害しない。他の可能性は、それらが
タグの分析により分解されないように、好適なやり方
で(例えば、アセチル化)によりそのリガンドを末端で誘導体化することにある。
さらなる可能性は、担体からタグを分離し、そして次にそのタグを配列決定す
ることにある。
上記結合テストの偽陽性結果を除外するために、これらのリガンドは、個々の
担体から全体的又は部分的に分離されることができ、そして均質液液中で、さら
なる検定、例えば、アクセプターへのそれらの付着のための第2ラウドの検定に
供されることができる。リガンドの分離は、例えば、そのリンカーに特異的な反
応によりその担体とリガンドとの間のリンカーにおける解裂により行われる。リ
ンカーがp−ヒドロキシ安息香酸である場合、この解裂は、気体アンモニア、ア
ンモニア/THF の飽和溶液又は液体アンモニアにより(例えばマイクロタイトレ
ーションプレート内で)個々の担体を処理することにより行われることができる
。次に分離されたリガンドは、担体から濯がれ、そしてもう一度、そのアクセプ
ターへのその結合1についてテストされることができ、又は、さらなる検定であ
って、担体に結合されたリガンドにより行われることができないもの、典型的に
は、結合性試験、光吸収が計測されるテスト、又はMS,IR又は NMR調査が行われ
る。
リンカーの好適な選択は、第一検定について、気体アンモニアによる処理によ
る単離担体からのリガンドの第1部分、とアンモニア/THF 又は液体アンモニア
の溶液による処理によるさらなる検定についてのリガンドの第2部分とを分離す
ることを可能にする。この点で好適なリンカーは典型的には、4−ヒドロキシメ
チル安息香酸である。
典型的な例は、非天然アミノ酸及び他の構築ブロックを含む潜在的なトロンビ
ン・インヒビターのライブラリーの合成であり、そし
てそれ故、よりプロテアーゼ−耐性である。
ライブラリーは、例えば、2の定常及び3の可変部位をもち、そして以下の組

このライブラリーをスクリーニングした後、ペンタマーD-Phe-D-Pro-Arg-Pro-
GABA及びD-Phe-Pro-D-Arg-Pro-GABAが全て有効なトロンビン・インヒビターであ
ることが発明される。
従って、本発明のさらなる目的は、トロンビンを阻害するための、式D-Phe-D-
Pro-Arg-Pro-GABA,D-Phe-Pro-D-Arg-Pro-GABAの化合物、又は医薬として許容さ
れるそれらの塩;及び単独で又はさらなる任意的賦形剤との組合せにおいてその
トロンビン阻害剤を含む医薬組成物である。
本発明は、さらに、ヒト又は動物体の治療及び予防処置の方法における新規の
使用方法により検出される各化合物に関する。好ましい使用分野は、トロンビン
の働き、例えば塞栓症及び血栓症に関する治療的及び予防的処置の分野である。
本発明の医薬組成物は、温血動物への、経腸、例えば経口、そしてまた直腸及
び非経口投与、例えば皮下、静脈内、又は腹腔内投与のためのものであり、そし
てそれらは、薬理学的に活性な化合物を単独で又は医薬として許容される担体と
一緒に含む。その日用量は、その患者の年齢及び個体の状態並びに投与モードに
依存するであろう。
本新規医薬組成物は、約10〜80%、好ましくは約20〜60%の活性化合物を含む
、経腸又は非経口投与のための医薬組成物は、典型的には、単位投与形態、例え
ば糖衣剤、錠剤、カプセル又は座剤、そしてまたアンプルにおけるものである。
これらの投薬形態は、それ自体公知のやり方で、典型的には、慣用の混合、粒状
化、糖剤調製(confectioning)、溶解又は凍結乾燥法により調製されることがで
きる。
経口投与のための医薬組成物が好ましい。好適な担体は、特に増量剤、例えば
、糖、便利にはラクトース、サッカロース、マンニトール又はソルビトール、セ
ルロース調製物、及び/又はリン酸カルシウム、典型的には、トリカルシウム・
ホスフェート又はリン酸水素カルシウム、そしてまたバインダー、例えば、デン
プン・ペーストであって、便利には、メイズ、コーン、米又はポテト・デンプン
、ゼラチン、トラガント、メチル・セルロース及び/又はポリビニル・ピロリド
ンを使用するもの、及び/又は所望により、崩壊剤、例えば、上述のデンプン、
またカルボキシメチル・デンプン、架橋ポリビニルピロリドン、寒天、アルギン
酸又はその塩、例えば、アルギン酸ナトリウムである。賦形剤は、特に、滑沢剤
、流動調節剤及び潤滑剤、便利には、シリカ、タルク、ステアリン酸又はそれら
の塩、典型的にはステアリン酸マグネシウム又はステアリン酸カルシウム、及び
/又はポリエチレン・グリコールである。糖衣剤のコ
アは、好適な非−経腸又は経腸コーティングにより、典型的には、アラビア・ガ
ム、タルク、ポリビニルピロリドン、ポリエチレン・グリコール及び/又は2酸
化チタン、を含むことができる濃縮糖溶液、好適な有機溶媒又は溶媒混合物中の
シェラック溶液、又は、経腸コーティングの調製のために、好適なセルロース調
製物、例えばアセチル・セルロース・フタレート又はヒドロキシプロピルメチル
・セルロース・フタレートの溶液を使用して、提供されることができる。染料又
は顔料が、便利には活性化合物の異なる投与を同定し又は示すために、錠剤又は
糖衣剤コーティングに添加されることができる。
経口投与のためのさらなる医薬組成物は、ゼラチンから作られた乾燥充填カプ
セルそしてまた、ゼラチン及び可塑剤、例えば、グリセロール又はソルビトール
から成るソフト−シール・カプセルである。この乾燥充填カプセルは、粒状物の
形態において、便利には、増量剤、例えば、ラクトース、バインダー、例えば、
デンプン、及び/又は滑沢剤(glidants)、例えば、タルク又はステアリン酸マ
グネシウムと混合されて、そして安定剤を伴って又は伴わずに、上記活性成分を
含むことができる。ソフト・カプセル内では、活性成分は、好ましくは、好適な
液体、典型的には、脂肪油、パラフィン油又は液体ポリエチレン・グリコール中
に溶解され又は懸濁され、これに安定剤が添加されることもできる。
直腸投与のために好適な医薬組成物は、典型的には、座剤基材と上記活性化合
物の組合せから成る座剤である。好適な座剤基材の例は、天然又は合成トリグリ
セリド、パラフィン炭化水素、ポリエチレン・グリコール及び高級アルカノール
である。基材物質と活性化合物の組合せを含む直腸投与のためのゼラチン・カプ
セルを使用することもできる。好適な基材物質は、典型的には、液体トリグリセ

非経口投与のための医薬組成物は、活性化合物の水溶液又は懸濁液、便利には
、好適な親油性溶媒又は媒質、例えば脂肪油、典型的にはゴマ油、又は合成脂肪
酸エステル、例えば、オレイン酸エチル又はトリグリセリド、を使用した油状注
射懸濁液、又は粘度増加物質、便利にはナトリウム・カルボキシメチル・セルロ
ース、ソルビトール及び/又はデキストランを含むことができる水性注射懸濁液
を、そしてまた、安定剤と伴って又は伴わずに、含む。
本発明は、好ましくは医薬組成物の形態における、上述のトロンビン阻害剤の
使用にも関する。活性化合物の投与量は、その温血動物の種、患者の年齢及び個
々の症状、そしてまた投与モードに依存するであろう。約75kg体重の患者への非
経口投与のために企図される日用量は、約 0.1mg〜500mg、好ましくは、約1mg
〜50mgであろう。

サルコシル NA実施例1:トリチル保護アミノ酸とジペプチドの調製
10mmolの各ジペプチド又はアミノ酸を、36mlのクロロホルム/アセトニトリル
(4:1)中に懸濁する。次に10mmolの塩化トリメチルシリルをゆっくりと滴下
し、そしてその反応混合物を約65℃において2時間還流させる。次に20mmolのト
リエチルアミンを滴下し、そして最後に10mlのクロロホルム中の10mmolの塩化ト
リチルの溶液を添加する。60分後、メタノールを過剰に添加し、そしてさらに5
分後、その反応混合物を蒸発により濃縮し、そしてその残渣を乾燥させる。この
粗材料を、氷冷硫酸水素カリウム(又はクエン酸)の2M溶液中に溶かし、そし
てその溶液を酢酸エチルにより抽出する。(約5℃において)水性アルカリによ
り逆抽出した後、その水相を硫酸水素カリウム溶液によりpH6−7に調整する。
Trt−ジペプチド又は Trt−アミノ酸を沈澱させ、そして酢酸エチルにより抽出
する。10mmolのトリエチルアミンをその有機相に添加し、そしてそのバッチを蒸
発により濃縮する(Trt−ジペプチド又は Trt−アミノ
酸のトリエチルアンモニウム)。
Trt−ジペプチドDG,GD,GF,AA,AG,DA,GA,GG,NG,AN及びNA並びにA,
D,F,G及びNの Trt−アミノ酸のトリエチルアンモニウム塩をこのやり方で
調製する。実施例2:タグの定常ユニットの等量未満をもつ担体樹脂の部分的誘導体化
(Novabiochem)の反応性アルキルアミノ基 180μmol を、それぞれ、20% Pip/D
MA により5回40秒間、そしてその後室温においてDMAにより10回60秒間洗浄する
。
ローディング:約 480μmol /gは約 2000pmol /樹脂粒子(ビーズ)と等価
である。
前活性化のために、実施例1の、60μmol のTrt-Gly-OH・ NEt3を66μmol(HO
BT に基づく)のHOBT/DMA の 0.5M溶液中に溶解し、そしてこの溶液に66μmol
(DICD に基づく)のDICD/DMA の2M溶液を添加する。室温において40分後、
この混合物を2mlの DMAにより希釈し、そしてその溶液を樹脂上にピペットで添
加する。直後に、66μmol の DIPEAを添加する。このカップリング反応を60分間
続ける。この樹脂を DMAにより10×45秒間濯ぐ。実施例2.1 :遊離アミノ基の計測によるタグ・ローディングの測定
リポーター基Fmocを樹脂に結合させ、そして過剰量で洗い流した後、再び分裂

275μmol のFmoc-OSuを 2.1mlの DMAに溶解し、そして上記樹脂に添加する。
直後に、 413μmol(DIPEAに基づく)の DIPEA/DMA の 1.5M溶液を樹脂上にピ
ペットで添加する。室温において80分後、その樹脂を DMAにより10×18秒間洗浄
する。Fmoc基を除去するために、この樹脂を室温において20%の Pip/DMA によ
り15×40秒間
処理する。集めた濾液を所定のやり方で希釈し、そしてそれらの吸光度(extinc
tion)を 299.8nmにおいて計測する。遊離アミノ基の全数は約 154μmol、すな
わち、その技術の全ローディングの85%(リガンドのパーセンテージ)である。
従って、その樹脂のタグ・ローディングは約15%である。実施例3:リガンドのためのリンカーの、樹脂へのカップリング
前活性化のために、 769μmol の4−ヒドロキシメチル安息香酸を 845μmol
(HOBT に基づく)のHOBT/DMA の 0.5M溶液中に溶解し、そしてこの溶液に 84
5μmol(DICD に基づく)のDICD/DMA の2M溶液を添加する。室温において40
分後、この反応混合物を 0.3mlの DMAにより希釈し、そして樹脂上にピペットで

まず、Fmoc基を、実施例2に記載したようなリガンドにおいて除去する。次に
、第1可変位置(Z位)のために個々のカップリング反応を行う:
上に列記したFmoc構築ブロックの各々を、室温において40分間84μmol のHOBT

ギ酸により解裂させ(1分間2回)、そして濯ぐ:

を次に付着させる。実施例7:リガンドの脱保護
保護形態において得られたライブラリーの樹脂粒子の適当な数(但し、多数の
成分数)を、ペプチド化学において一般的に使用される脱保護反応に供してFmoc
及びtert−ブチル保護基を除去する。 Fmoc 保護基を除去するために、実施例6
.5の樹脂を再び20% Pip/DMA により繰り返して処理し(実施例2参照)、そ
して次に、 t−ブチル及び Pmc保護基を除去するために、室温において40分間
、95%トリフルオロ酢酸(5%水+2%エタンジチオール)により処理する。樹
脂へのリガンドの連結が無傷である場合、リガンド内及びタグの官能基を全て解
放する。次に濯ぎを以下のように行う:

イソチオシアネートを、ヒト・トロンビンの溶液(100mlのボレート・バッファ
ーpH8中0.53mg)に添加する(290μmol のDMSO中 2.9mgの溶液10μl)。この
反応を、室温において60分間続け、そしてその反応混合物を、ゲル・クロマトグ
ラフィー・カラム(Sephadex
20μlのフルオレセイン−トロンビン画分(約3%画分容量)を 500μlに希
釈し、そして 495nmにおいて光計測により測定する。これは、約2μmのフルオ

る。実施例9:不均質相におけるトロンビン結合性リガンドの同定
実施例7の樹脂の約 500ビーズを、室温において10分間実施例8のフルオレセ
