【発明の詳細な説明】
抗関節炎剤および抗炎症剤としてのピリミジノン発明の背景
本発明は、抗炎症剤および抗関節炎剤として有用な、(式I)によって特徴付け
られるピリミジノンおよびその医薬上許容される塩に向けられる。
本発明の化合物は、急性の炎症疾患の安全かつ有効な治療として、ヒトおよび
より下等な動物に有用である。これらの疾病には、歯周病、慢性関節リウマチ、
骨関節炎、神経炎、滑液胞炎、塵肺症、クローン(Crohn)病、慢性の炎症性腸疾
患、慢性喘息、アテローム性動脈硬化症、多発性硬化症および類肉腫症が包含さ
れる。関連技術の説明
技術水準陳述の目的として、米国特許第4,746,954号は、抗炎症剤とし
て有用なビスホスホネートを開示している;オーストラリア国特許第A-515
34/85号は、異常なカルシウムおよびリン代謝の治療に有用であり、かつ関
節炎の治療に有用なビスホスホネートを開示している;および、米国特許第3,
683,080号は、動物組織におけるリン酸カルシウムの異常な沈着および動
員を阻害するのに有用なポリホスホネート;特にジホスホネートを開示している
。
欧州特許出願第0 168 262号は、対応するR2が水素または低級アルキ
ルのみである以外は主題の化合物に類似する式VIIaで示される中間体ピリミ
ジン化合物を開示している。該化合物は、高血圧および脳血管疾患の治療に有用
であると記載されている。発明の概要
1つの態様において、本発明は、式I:

ペリジニル、ピロリジニル、フェノキシ、フェニルチオ、フェニルスルホニル、
フェニルスルフィニル、-NHC(O)-C1-C6カルボン酸、N3、NH2、ジエチ
ルアミノ、水素(但し、R4はベンジルオキシ)、ハロゲン(但し、R3はC1-C6ア
ルキル)またはCH(「EWG」)2であり、ここに、「EWG」とは、各々独立し
て、CO2R6およびPO(OR7)2よりなる群から選択される電子吸引性基;また
は
ii)nが2である場合、YはCH(「EWG」)2、あるいは、
b) i)アリールまたはヘテロアリール、
ii)ヒドロキシルおよび、C1-C6アルキル、フェニルまたはmが
1〜3の(CH2)m-CO2R6
で置換された末端オレフィン、あるいは、
c)(所望により、ハロゲン、(PO(OC2H5)2)2またはCNで置換されて

R3は水素またはC1-C6アルキル;
R4は水素、ヒドロキシル、C1-C6アルキル、アルコキシ、ベンジルオキシま

R5は水素、ハロゲン、C1-C6アルキル、C1-C6アルコキシ、フェノキシ、
C1-C6アルキルチオ、チオフェニル、NH2、アリール(但し、R4およびYが共
に水素である場合に、R5がフェニル以外となる場合を除く)またはヘテロアリー

R6はH、C1-C6アルキル、ベンジル、フェニル、1〜5個のF、Cl、Br
、I、NO2、OCH3またはC1-C4アルキルで置換されたフェニル;および
R7はH、C1-C6アルキル、ベンジル、フェニル、1〜5個のF、Cl、Br
、I、NO2、OCH3またはC1-C4アルキルで置換されたフェニル、あるいは
双方のR7が一緒になった場合、CH2-CH2、CH2-CH2-CH2またはCH2-
C(CH3)2-CH2を形成し、それによって、結合したP原子および2個のO原子
を含有する複素環を形成する]
によって構造的に表されるピリミジノンまたはその医薬上許容される塩である。
もう1つの態様において、本発明は、慢性の炎症疾患の安全かつ有効な治療と
して、ヒトおよびより下等な動物におけるこれらの化合物の使用よりなる。これ
らの疾病には、歯周病、慢性関節リウマチ、骨関節炎、塵肺症、クローン(Croh
n)病、慢性の炎症性腸疾患、慢性喘息、アテローム性動脈硬化症、多発性硬化症
および類肉腫症が包含される。
さらにもう1つの態様において、本発明は、炎症の治療を要する患者(ヒトを
含む動物)に、抗炎症に有効な量の式Iで示される化合物を投与するにより、か
かる治療を行う方法である。投与経路には、経口、筋肉内、静脈内、経皮、関節
内、皮下または腹腔内が包含される。有効な量とは、それによって痛みおよび不
快のごとき炎症または関節炎の症状が緩和または減じられ、あるいは患部の可動
性が向上する量である。典型的な投与量は、特定の投与様式、投与の使用および
頻度によって決定される用量により約0.001mg〜1.0gである。発明の詳細な説明
本発明は、抗炎症剤および抗関節炎剤として有用な、(前記の式I)によって特
徴付けられる、ピリミジノンおよびその医薬上許容される塩よりなる。これらの
化合物は、特に、関節炎ならびに炎症および過剰骨増殖または再構成のごときそ
れに関連する症状の治療に有用である。式Iにおいて、変数設計は以下のように
さらに定義される。
種々の炭化水素含有基の炭素含量は、基中の炭素原子の最小数および最大数を
示す接頭辞によって示され、すなわち、接頭辞Ci-Cjは整数「i」〜整数「j
」個存在する炭素原子の数を定義している。従って、C1-C3アルキルは、1-3
個の炭素原子のアルキルを示し、メチル、エチル、プロピルおよびイソプロピル
が
包含される。
前記と関連して、C1-C6アルキルとは、メチル、エチル、プロピル、ブチル
、ペンチル、ヘキシルおよびそれらの異性体である。「アリール」とは、フェニ
ルまたはナフチルである。「ヘテロアリール」とは、モルホリニル、ピペラジニ
ル、ピペリジニル、イミダゾリジニル、ピラゾリジニル、イソキサゾリル、ピロ
リル、イミダゾリル、ピラゾリル、ピリジル、ピラジニル、ピリミジニルまたは
ピリダジニルを意味する。「ハロゲン」なる語には、フルオロ、クロロ、ブロモ
およびヨードが包含される。「末端オレフィン」とは、-CH=CH2を意味し、
これは、末端水素の片側または両側を示したように置換できる。
医薬上許容される塩とは、本発明の化合物を投与するのに有用な塩か、または
イン・ビトロ(in vitro)またはイン・ビボ(in vivo)で化合物が取り得る有用な
形態を意味し、カリウム塩、ナトリウム塩、塩酸塩、臭化水素酸塩、ヨウ化水素
酸塩、硫酸塩、リン酸塩、酢酸塩、プロピオン酸塩、乳酸塩、メシラート、マレ
イン酸塩、マロン酸塩、コハク酸塩、酒石酸塩、クエン酸塩他が包含される。こ
れらの塩は水和形態としても存在し得る。
抗-炎症剤および抗-関節炎剤として有用なピリミジノンおよびその誘導体(式
I)は、以下の実施例1-35および表1に示すごとく調製する。
本発明の式Iの化合物は、炎症の遅延型過敏症肉芽腫アッセイ(DTH GRA
)モデルで試験した。このアッセイは、ダン,シイ・ジェイ(Dunn,C.J.)ら「慢
性の免疫-介在炎症疾患の実験用のマウスにおける遅延型過敏症肉芽腫モデルの
開発(Development of a delayed-type hypersensitivity granuloma model in t
he mouse for the study of chronic immune-mediated inflammatory disease.)」
エージェンツ・アンド・アクションズ(Agents and Actions)第27巻、3/4(
1989年)および「ネズミの遅延型過敏症肉芽腫(Murine Delayed-Type Hy
persensitivity Granuloma)」インターナショナル・ジャーナル・オブ・イムノフ
ァーマコロジー(Int.J.Immunopharmc.)第12巻、8:899-904頁(19
90年)によって記載されている。
簡単には、9日後に切除するmBSA-浸漬フィルターを皮下的に移植するこ
とによって、DTH肉芽腫(DTH GRA)病斑を有するmBSA-感作マウス
を誘導した。化合物をマウスに投与して、病斑に対するそれらの効果を測定する
。結果は、%阻害で記録する。阻害が大きい程、化合物の効果も高い。10〜2
0%阻害で、抗-肉芽腫活性を示すと考える。また、30%より大きな阻害は、
良好な活性である。式Iで示される化合物から得たDTH GRAデータを以下
の表2に示す。化合物は、10-20%阻害で抗-炎症活性を有すると、30%よ
り大きな阻害で良好な活性を有すると記録する。
「化合物の名称」は、実施例の表示に対応する。示した特定の化合物は以下の
通りである:
実施例1:4(3H)-ピリミジノン,2-(クロロメチル)-3-メチル-6-フェニル-工程A:
THF(55ml)中の4(3H)-ピリミジノン,2,3-ジメチル-6-フェニル(1
1.0g、54.9ミリモル)を、LiHMDS(60ミリモル、THF中の1M、
60ml)の-78℃溶液に徐々に添加した。45分間撹拌した後に、次いで、こ
れを、THF(50ml)中のヘキサクロロエタン(13.64g、57.6
ミリモル)の-40℃溶液に45分間にわたってカニューレ処理した。この反応物
を室温に1時間加温し、次いで、NH4Clでクエンチし、酢酸エチルで3回抽出
し、飽和NaHCO3および食塩水で洗浄し、MgSO4で乾燥し、真空下にて濃
縮した。生成物は、メチルt-ブチルエーテルから再結晶化させた(6.555g
、27.9ミリモル、51%)。
融点121-122℃
実施例2:4(3H)-ピリミジノン,3-メチル-2-(フェノキシメチル)-6-フェ
ニル-工程B:
トルエン(9ml)中の水素化ナトリウム(0.191g、3.98ミリモル)の0℃
懸濁液に、フェノール(0.34g、3.61ミリモル)を添加した。15分後に、
2-(クロロメチル)-3-メチル-6-フェニル-4(3H)-ピリミジノン(0.775
g、3.2ミリモル)を添加し、その反応物を2.5時間加熱還流した。冷却した
後に、その反応混合物を塩化メチレンで希釈し、次いで飽和NaHCO3で3回
洗浄し、MgSO4で乾燥し、真空下にて濃縮した(57%)。
融点104-105℃
実施例2記載と同じ方法によって実施例3-6を行い、それらの化合物は以下
のごとく同定された。

THF(12ml)中の2-(クロロメチル)-3-メチル-6-フェニル-4(3H)-ピ
リミジノン(0.507g、2.2ミリモル)をモルホリン(0.40ml、4.5ミリ
モル)で処理し、次いで1.5時間還流加熱した。その反応混合物を冷却し、セラ
イト・パッドを通して濾過し、真空下にて濃縮した。その残渣を塩化メチレンに
溶解し、2回濾過した。その有機層を真空下にて濃縮して明黄色固形物を得た。
粗製物質は酢酸エチルから再結晶化することによって精製した(27%)。
融点135-136℃
実施例8:4(3H)-ピリミジノン,3-メチル-6-フェニル-2-ピペリジニル-
実施例7と同一の方法を行って、標題化合物を調製した。

下にて濃縮した。その残渣を塩化メチレンに溶解し、飽和NaHCO3、3×1M
のNaHSO3、飽和NaClで洗浄し、MgSO4で乾燥し、真空下にて濃縮した(
40%)。
融点111-112℃
実施例13:4(3H)-ピリミジノン,2-(ヨードメチル)-3-メチル-6-フェニ
ル-
2-(クロロメチル)-3-メチル-6-フェニル-4(3H)-ピリミジノン(0.98
g、4.16ミリモル)およびヨウ化ナトリウム(0.654g、4.37ミリモル
)をアセトン(10ml)中で合わせ、30分間加熱還流した。その反応混合物を
冷却し、濾過し、真空下にて濃縮した。得られた固形物を高真空下にて乾燥し、
明黄色固形物を回収した。
実施例14:4(3H)-ピリミジノン,2-(アミノメチル)-3-メチル-6-フェ
ニル-
2-(クロロメチル)-3-メチル-6-フェニル-4(3H)-ピリミジノン(3.98
g、17.0ミリモル)およびアジ化ナトリウム(2.21g、34ミリモル)をア
セトニトリル(25ml)中で合わせ、1時間穏やかに加熱還流した。室温に冷却
した後に、真空下にて溶媒を除去した。その残渣を塩化メチレンで希釈し、6%
NaHCO3、H2Oおよび食塩水で洗浄し、チャコールで処理し、MgSO4のパ
ッ
ドを通して濾過した。真空下にて濃縮した後に、粗製生成物を5%炭素パラジウ
ム(0.4g)を入れたエタノール混合液(80ml)に再懸濁し、水素大気中にて2
時間水素添加した。その反応混合物をセライトを通して濾過し、真空下にて濃縮
した。生成物は、酢酸エチル/ヘキサンから再結晶化した(0.80g、3.7ミリ
モル、22%)。
融点90-92℃
実施例15:酪酸,4-[[(1,6-ジヒドロ-1-メチル-6-オキソ-4-フェニル-2
-ピリミジニル)メチル]アミノ]-4-オキソ-
クロロホルム(5ml)中に溶解した4(3H)-ピリミジノン,2-(アミノメチル)-
3-メチル-6-フェニル-(304mg、1.41ミリモル)を、無水スクシン酸(1
41mg、1.41ミリモル)で処理し、次いで1時間加熱還流した。得られた固
形物を濾過し、クロロホルムで洗浄し、乾燥した(0.377g、1.20ミリモ
ル、85%)。
融点200-201℃
実施例16:ペンタン酸,5-[[(1,6-ジヒドロ-1-メチル-6-オキソ-4-フェ
ニル-2-ピリミジニル)メチル]アミノ]-5-オキソ-
実施例15の方法と同様にして、標題化合物を無水グルタル酸から調製した(
62%)。
融点181-182℃
実施例17:4(3H)-ピリミジノン,3-メチル-6-フェニル-2-[(フェニルス
ルホニル)-メチル]-
ベンゼンスルフィン酸のナトリウム塩(0.346g、2.1ミリモル)をトルエ
ン(5ml)中でスラリー化し、次いで、4(3H)-ピリミジノン,2-(ヨードメチ
ル)-3-メチル-6-フェニルー(0.517g、1.6ミリモル)で処理し、70℃
にて20時間加熱した。その反応物を室温に冷却し、真空下にて大部分の溶媒を
除去した。その残渣を塩化メチレンに溶解し、飽和NaHCO3および1MのNa
HSO3で3回洗浄し、MgSO4で乾燥し、真空下にて濃縮した。生成物は、シ
リカゲル上のクロマトグラフィーによって精製した(226mg、0.66ミリ
モル、41%)。
融点174-175℃
実施例18:4(3H)-ピリミジノン,3-メチル-6-フェニル-2-[(フェニルス
ルフィニル)-メチル]-
0℃のメタノール(10ml)中に溶解した4(3H)-ピリミジノン,3-メチル-
6-フェニル-2-[(フェニルチオ)メチル]-(0.589g、1.91ミリモル)を、
水(21ml)中のオキソン(7.23g、5.7ミリモル)の水性溶液で処理した。
その反応混合物を0℃にて5時間撹拌し、次いで、試料を40℃に加熱しつつ1
MのNaHSO3で処理し、真空下にて濃縮した。塩化メチレンを添加して分離
させた後に、その有機層を1MのNaHSO3で3回、飽和NaHCO3で1回洗
浄し、MgSO4で乾燥し、真空下にて濃縮した。試料は、シリカゲル上のクロ
マトグラフィーによって精製した(217mg、0.67ミリモル、35%)。
融点158-159℃
実施例19:4-ペンテン酸,5-(1,6-ジヒドロ-1-メチル-6-オキソ-4-フェ
ニル-2-ピリミジニル)-4-ヒドロキシ-,(Z)-
4(3H)-ピリミジノン,2,3-ジメチル-6-フェニル(5.45g、27.2ミ
リモル)をTHF(25ml)に溶解し、-20℃に冷却し、LiHMDS(60ミリ
モル、THF中の1M、60ml)で処理した。30分間撹拌した後に、無水スク
シン酸(3.0g、30.0ミリモル)を添加し、その反応物を2時間室温に加温し
た。それを水でクエンチし、酢酸エチルで2回抽出し、その水性溶液を1NのH
Clで酸性化し、次いで濾過した。生成物は、iPrOHから再結晶化した(4.
30g、14.3ミリモル、53%)。
融点232-233℃
実施例20:5-ヘキセン酸,6-(1,6-ジヒドロ-1-メチル-6-オキソ-4-フ
ェニル-2-ピリミジニル)-5-ヒドロキシ-,(Z)-
実施例19と同様の方法で、無水グルタル酸から標題化合物を調製した(26
%)。融点170-171℃
実施例21:4-ペンテン酸,5-(1,6-ジヒドロ-1-メチル-6-オキソ-4-フェ
ニル-2-ピリミジニル)-4-ヒドロキシ-,メチルエステル,(Z)-
メタノール(25ml)およびH2SO4(1滴)中の4-ペンタン酸,5-(1,6-ジヒ
ドロ-1-メチル-6-オキソ-4-フェニル-2-ピリミジニル)-4−ヒドロキシ-,(Z)
-(693mg、2.31ミリモル)を3時間加熱還流した。真空下にて大部分のメ
タノールを除去した後に、結晶物を濾過し、エーテルで洗浄した(646mg、
2.06ミリモル、89%)。
融点153℃
実施例22:ホスホン酸,[(1,6-ジヒドロ-1-メチル-6-オキソ-4-フェニル-
2-ピリミジニル)シクロプロピリデン]ビス-,テトラエチルエステル
4(3H)-ピリミジノン,2-(クロロメチル)-3-メチル-6-フェニル-(705
mg、3.0ミリモル)、エテニリデンビス(ホスホン酸),テトラエチルエステル(
900mg、3.0ミリモル)およびDBU(0.5ml、3.3ミリモル)をTHF(
5ml)中、室温にて24時間撹拌した。その反応物をNH4Clでクエンチし、酢
酸エチルで3回抽出し、MgSO4で乾燥し、ストリッピングした。その粗製物
をシリカゲル・クロマトグラフィーによって精製した(471mg、0.95ミリ
モル、32%)。

実施例25:マロン酸,[2-(1,6-ジヒドロ-1-メチル-6-オキソ-4-フェニル-
2-ピリミジニル)エチル]-,ビス(1,1-ジメチルエチル)エステル
4(3H)-ピリミジノン,2,3-ジメチル-6-フェニル(0.744g、3.72
ミリモル)をTHF(37ml)に溶解し、-78℃に冷却し、次いで、リチウムビ
ス(トリメチルシリル)アミド(4.1ml、4.1ミリモル)で処理した。-78℃に
て30分間撹拌した後に、THF(10ml)中の2-t-ブチルカルボキシ-2-プ
ロペン酸t-ブチルエステル(0.424g、1.86ミリモル)の溶液を、シンリ
ンジ・ポンプを用いて約1時間にわたり徐々に添加した。その反応物を4.1M
のNH4Cl(1ml、4.1ミリモル)で直ちにクエンチし、次いで22℃に加温し
た。その反応混合物を真空下にて濃縮し、酢酸エチル中に溶解した。その有機層
を、1NのHClで3回、H2Oで1回、NaHCO3で3回および食塩水で1回
洗浄し、次いで、MgSO4で乾燥し、真空下にて濃縮した。その粗製混合物を
シリカゲル上のクロマトグラフィーに付した(0.317g、0.74ミリモル、
40%)。
融点94-95℃
実施例26:4(3H)-ピリミジノン,2,3-ジメチル-6-フェニル-5-(フェニ
ルメトキシ)-
水素化ナトリウム(19.2g、0.40モル、油中の50%)をトルエン(20
0ml)に懸濁し、次いで、ベンジルアルコール(43.2g、0.40モル)を滴下
した。45分間撹拌した後に、灰色泡状物をトルエン(100ml)中のクロロ酢
酸(18.8g、0.20モル)の溶液で徐々に処理し、次いで加熱還流した。2時
間還流した後に、その反応物を室温に冷却し、水(100ml)に注入した。その
有機層を分離し、トルエンを水で2回抽出した。その水性層をメチルt-ブチル
エーテルで洗浄し、有機物を捨てた。濃HClでpHを2に調整した後に、水性
画分を塩化メチレンで3回抽出し、MgSO4で乾燥し、真空下にて濃縮した(2
9.50g)。この粗製物質、エタノール(40ml)、トルエン(50ml)および濃
H2SO4(3ml)を一晩加熱還流した。冷却した後に、それを水、NaHCO3で
洗浄し、MgSO4で乾燥し、真空下にて濃縮した。生成物は、蒸留によって
単離した(24.38g、0.126モル、63%)。
Bp0.155-60℃
この合成は、オーストラリア・ジャーナル・オブ・ケミストリー(Aust.J.Che
m.)1976年、第29巻、1335-1339頁に記載されているものと実質的
に同一である。
そのエステル(22.10g、0.113モル)をTHF(5ml)に溶解し、LiH
MDSの-78℃溶液(0.230モル、THF中の1M、230ml)に徐々に添
加した。45分間撹拌した後に、塩化ベンゾイル(13.1ml、0.113モル)
を添加し、その反応物を45分間室温に加温した。それを1NのHClでクエン
チし、メチルt-ブチルエーテルで3回抽出し、MgSO4で乾燥し、ストリッピ
ングした(45.68g、0.153モル)。粗製物質、塩酸アセトアミド(28.9
5g、0.306モル)および25%ナトリウムメトキシド/メタノール(77ml
