チエニルチアゾール誘導体
技術分野
この発明は、医薬として有用な新規チエニルチアゾール誘導体およびそれらの
医薬として許容される塩に関する。
背景技術
たとえば欧州特許出願公開第0183191号および日本特許出願公開昭59
−225186号において、若干のチエニルチアゾール誘導体が抗潰瘍剤として
既知である。
発明の開示
この発明は、新規チエニルチアゾール誘導体およびそれらの医薬として許容さ
れる塩に関する。
より詳しくは、この発明は、抗潰瘍作用、H2−レセプタ拮抗作用および抗菌
活性を有する新規チエニルチアゾール誘導体およびそれらの医薬として許容され
る塩、それらの製造法、それらを含有する医薬組成物ならびにヒトまたは動物に
おける胃炎、潰瘍(たとえば胃潰瘍、十二指腸潰瘍、吻合部潰瘍など)、ゾリン
ジャー ・エリソン症候群、逆流性食道炎、上部胃腸管出血、感染症などを治療
および/または予防する方法に関するものである。
従って、この発明の一目的は、抗潰瘍作用、H2−レセプタ拮抗作用および抗
菌活性を有する新規チエニルチアゾール誘導体およびそれらの医薬として許容さ
れる塩
を提供することである。
この発明の他の一目的は、該チエニルチアゾール誘導体およびそれらの塩の製
造法を提供することである。
この発明のさらなる目的は、該チエニルチアゾール誘導体またはそれらの医薬
として許容される塩を活性成分として含有する医薬組成物を提供することである
。
この発明のなお一つの目的は、該チエニルチアゾール誘導体またはそれらの医
薬として許容される塩を用いて、ヒトまたは動物における前記の諸疾患を治療お
よび/または予防するための医療法を提供することである。
この発明が目的とするチエニルチアゾール誘導体は、新規であり、次の一般式
(I)によって表わすことができる:
式中、R1は水素またはアルコキシで置換されていてもよいアルキルを、
R2は水素またはアルコキシで置換されていてもよいアルキルを、
R3は水素または低級アルキルを、
R4はアシルで置換されていてもよいアミノを、
Aは単結合または低級アルキレンを、
Qは水素または低級アルキルを、それぞれ表わす。
ただし、R1およびR2がともに水素である場合には、1)R4がエステル化され
たカルボキシで置換されたアミノであるか、または2)Qが低級アルキルである
ものとする。
ならびにそれらの医薬として許容される塩である。
目的化合物(I)またはその塩は、次の諸方法により製造できる。
上記反応式中、R1、R2、R3、R4、AおよびQは各々上に定義した通りであり
、
R5は低級アルキルを、
X1は酸残基を、
R4 aはアシルで置換されたアミノを、それぞれ表わす。
本明細書の上記および後記の説明における式に関して、次の点を注記しておか
なければならない。すなわち、当該化合物が式
(式中、R1およびR2は各々上に定義した通りである)の基を有している場合、
該化合物は互変異性体を包含しうる。それゆえ、該基は式
(式中、R1およびR2は各々上に定義した通りである)で表わすことができ、該
式(A)の基は、また、その互変異性体の式
(式中、R1およびR2は各々上に定義した通りである)によっても表わされうる
。
すなわち、式(A)および(A’)の該両基が次の平衡式で表わしうる互変異
性平衡状態にある:

上述の式(A)および(A’)間のこれらのタイプの互変異性は当該技術分野
で周知であり、当業者にとっては、両互変異性体が平衡して、相互に変換しうる
状態にあることは自明であり、従って、かかる異性体は当該化合物自体の同一範
疇に包含されると理解すべきである。従って、両互変異性形態は明らかに本発明
の範囲に包含される。本明細書においては、かかる互変異性体の基を含む目的化
合物および出発化合物を、単なる便宜上の目的で、それらの表現法の一方、すな
わち式
によって表わすこととする。
本明細書の上記および後記の説明において、本発明の範囲に包含される種々の
定義の好適な例を、以下に詳細に説明する。
「低級」なる語は、とくに断わらない限り、1〜6個の炭素原子を有する基を
意味するものとする。
「シクロ低級アルキル」なる表現における低級部分は3〜6個の炭素原子を有
する基を意味するものとする。
「アルキル」なる語は、低級アルキルおよび高級アルキルを包含する。
「アルコキシ」なる語は、低級アルコキシおよび高級アルコキシを包含する。
好適な「低級アルキル」ならびに「アル低級アルキル」および「低級アルコキ
シ低級アルキル」なる表現における好適な低級アルキル部分としては、メチル、
エチル、プロピル、イソプロピル、ブチル、イソブチル、第三級ブチル、ペンチ
ル、イソペンチル、ヘキシル、メチルブチル、エチルブチルなどの直鎖状または
分枝鎖状のものが挙げられる。
好適な「シクロ低級アルキル」としては、シクロプロピル、シクロブチル、シ
クロペンチル、シクロヘキシルが挙げられる。
好適な「アリール」としては、フェニル、ナフチル、低級アルキルで置換され
たフェニル[たとえばトリル、メシチル、クメニル、キシリル、ジエチルフェニ
ル、ジイソプロピルフェニル、ジ第三級ブチルフェニルなど]などが挙げられる
。
好適な「アル低級アルキル」としては、ベンジル、フェネチル、ジフェニルメ
チル、トリフェニルメチル、ナフチルメチルなどが挙げられる。
好適な「低級アルコキシ」ならびに「低級アルコキシ低級アルキル」なる表現
における好適な低級アルコキシ部分としては、メトキシ、エトキシ、プロポキシ
、イソプロポキシ、ブトキシ、イソブトキシ、第三級ブトキシ、ペンチルオキシ
、ヘキシルオキシなどが挙げられ、
なかでも好ましいのはC1〜C4のものである。
「高級」なる語は、とくに断わらない限り、7〜20個の炭素原子を意味する
ものとする。
好適な「高級アルキル」としては、ヘプチル、オクチル、ノニル、デシル、ウ
ンデシル、ドデシル、トリデシル、テトラデシル、ペンタデシル、ヘキサデシル
、ヘプタデシル、オクタデシル、ノナデシル、エイコシル、エチルヘキシル、メ
チルヘプチル、メチルオクチル、メチルノニル、メチルデシル、エチルヘプチル
、エチルオクチル、エチルノニル、エチルデシルなどの直鎖状または分枝鎖状の
ものが挙げられ、なかでも好ましいのは7〜10個の炭素原子を有するものであ
り、とくに好ましいのはヘプチル、オクチル、エチルヘキシルである。
好適な「高級アルコキシ」としては、ヘプチルオキシ、オクチルオキシ、ノニ
ルオキシ、デシルオキシ、ウンデシルオキシ、ドデシルオキシ、トリデシルオキ
シ、テトラデシルオキシ、ペンタデシルオキシ、ヘキサデシルオキシ、ヘプタデ
シルオキシ、オクタデシルオキシ、ノナデシルオキシ、エイコシルオキシ、エチ
ルヘキシルオキシ、メチルヘプチルオキシ、メチルオクチルオキシ、メチルノニ
ルオキシ、メチルデシルオキシ、エチルヘプチルオキシ、エチルオクチルオキシ
、エチルノニルオキシ、エチルデシルオキシなどの直鎖状または分枝鎖状のもの
が挙げられ、なかでも好ましいのはエチルヘキシルオキシである。
好適な「低級アルキレン」としては、メチレン、エチレン、トリメチレン、プ
ロピレン、テトラメチレン、ペンタメチレン、ヘキサメチレン、エチルエチレン
などの直鎖状または分枝鎖状のものが挙げられ、なかでも好ましいのはC1〜C4
のものであり、とくに好ましいのはメチレン、エチレンである。
好適な「アシル」ならびに「アシルアミノ」なる表現における好適なアシル部
分としては、カルボキシ;エステル化されたカルボキシ;低級アルキル、シクロ
低級アルキル、アリール、アル低級アルキル、低級アルコキシ低級アルキルおよ
び複素環基からなる群から選ばれた置換基で置換されていてもよいカルバモイル
;低級アルカノイル;アロイル;複素環カルボニル;低級アルキルスルホニルな
どが挙げられる。
エステル化されたカルボキシとしては、置換または無置換アルコキシカルボニ
ル[たとえばメトキシカルボニル、エトキシカルボニル、プロポキシカルボニル
、ブトキシカルボニル、第三級ブトキシカルボニル、ヘキシルオキシカルボニル
、2−ヨードエトキシカルボニル、2,2,2−トリクロロエトキシカルボニル
など]、置換または無置換アリールオキシカルボニル[たとえばフェノキシカル
ボニル、4−ニトロフェノキシカルボニル、2−ナフチルオキシカルボニルなど
]、置換または無置換アル低級アルコキシカルボニル[たとえばベンジルオキシ
カルボニル、フェネチルオキシカルボニル、ベ
ンズヒドリルオキシカルボニル、4−ニトロベンジルオキシカルボニルなど]な
どが挙げられ、なかでも好ましいのは無置換低級アルコキシカルボニルおよび無
置換アル低級アルコキシカルボニルであり、とくに好ましいのはメトキシカルボ
ニル、エトキシカルボニル、ベンジルオキシカルボニルである。
低級アルカノイルとしては、ホルミル、アセチル、プロピオニル、ブチリル、
イソブチリル、バレリル、イソバレリル、ピバロイル、ヘキサノイルなどが挙げ
られ、なかでも好ましいのはC1〜C4のものであり、とくに好ましいのはアセチ
ルである。
アロイルとしては、ベンゾイル、ナフトイル、低級アルキルで置換されたベン
ゾイル[たとえばトルオイル、キシロイルなど]が挙げられる。
「複素環カルボニル」なる表現における好適な複素環部分としては、1〜4個
の窒素原子を含有する3〜8員、好ましくは5または6員不飽和複素単環基、た
とえばピロリル、イミダゾリル、ピラゾリル、ピリジル、ピリジルN−オキシド
、ジヒドロピリジル、テトラヒドロピリジル、ピリミジル、ピラジニル、ピリダ
ジニル、トリアジニル、トリアゾリル、テトラジニル、テトラゾリルなど;
1〜4個の窒素原子を含有する3〜8員、好ましくは5または6員飽和複素単
環基、たとえばピロリジニル、イミダゾリジニル、ピペリジノ、ピペラジニルな
ど;
1〜5個の窒素原子を含有する不飽和縮合複素環基、たとえばインドリル、イ
ソインドリル、インドリジニル、ベンズイミダゾリル、キノリル、イソキノリル
、インダゾリル、ベンゾトリアゾリルなど;
1〜2個の酸素原子と1〜3個の窒素原子とを含有する3〜8員不飽和複素単
環基、たとえばオキサゾリル、イソオキサゾリル、オキサジアゾリルなど;
1〜2個の酸素原子と1〜3個の窒素原子とを含有する3〜8員飽和複素単環
基、たとえばモルホリノ、シドノニルなど;
1〜2個の酸素原子と1〜3個の窒素原子とを含有する不飽和縮合複素環基、
たとえばベンゾオキサゾリル、ベンゾオキサジアゾリルなど;
1〜2個の硫黄原子と1〜3個の窒素原子とを含有する3〜8員不飽和複素単
環基、たとえばチアゾリル、イソチアゾリル、チアジアゾリルなど;
1〜2個の硫黄原子を含有する3〜8員不飽和複素単環基、たとえばチエニル
など;
1〜2個の硫黄原子と1〜3個の窒素原子とを含有する不飽和縮合複素環基、
たとえばベンゾチアゾリル、ベンゾチアジアゾリルなど;
1個の酸素原子を含有する3〜8員不飽和複素単環基、たとえばフリルなど;
1〜2個の硫黄原子を含有する不飽和縮合複素環基、たとえばベンゾチエニル
など;
1〜2個の酸素原子を含有する不飽和縮合複素環基、たとえばベンゾフラニル
など;
などが挙げられる。
好適な「酸残基」としては、ハロゲン(たとえばフルオロ、クロロ、ブロモ、
ヨード)、アレーンスルホニルオキシ(たとえばベンゼンスルホニルオキシ、ト
シルオキシなど)、アルカンスルホニルオキシ(たとえばメシルオキシ、エタン
スルホニルオキシなど)などが挙げられ、なかでも好ましいのはハロゲンである
。
好ましい化合物(I)は、R1がアルコキシで置換されていてもよいアルキル
、R2が水素、R3が水素または低級アルキル、R4が、低級アルカノイル、エス
テル化されたカルボキシもしくはカルバモイルで置換されていてもよいアミノ、
Aが低級アルキレン、Qが水素または低級アルキルであるものである。
より好ましい化合物(I)は、R1が低級アルコキシで置換されていてもよい
低級アルキル、R2が水素、R3が水素、R4が低級アルカノイルアミノ、Aが低
級アルキレン、Qが水素であるものである。
目的化合物(I)の医薬として許容される好適な塩は慣用の無毒性塩であり、
それらとしては、無機酸付加塩(たとえば塩酸塩、沃化水素酸塩、臭化水素酸塩
、硫酸塩、燐酸塩など)、有機酸付加塩(たとえば蟻酸塩、酢酸塩、トリフルオ
ロ酢酸塩、マレイン酸塩、酒石酸塩、メタンスルホン酸塩、ベンゼンスルホン酸
塩、トルエン
スルホン酸塩など)などの酸付加塩が挙げられる。
目的化合物(I)の製造法を、以下に詳細に説明する。方法1
化合物(I)またはその塩は、化合物(II)またはその塩を化合物(III)ま
たはその塩と反応させることにより製造できる。
化合物(II)および(III)の好適な塩としては、化合物(I)について例示
したものと同じものが挙げられる。
この反応は、通常、アルコール(たとえばメタノル、エタノールなど)、ベン
ゼン、N,N−ジメチルホルムアミド、テトラヒドロフラン、塩化メチレン、塩
化エチレン、クロロホルム、ジエチルエーテル、その他反応に悪影響を及ぼさな
い任意の溶媒中で実施する。
反応温度はとくに限定されないが、通常は、加温ないし加熱下に反応を実施す
る。方法2
化合物(I)またはその塩は、化合物(IV)を化合物(V)またはその塩と反
応させることにより製造できる。
化合物(V)の好適な塩としては、化合物(I)について例示したものと同じ
ものが挙げられる。
反応は、通常、慣用の溶媒中で、たとえばアルコール(たとえばメタノール、
エタノールなど)、テトラヒドロフラン、N,N−ジメチルホルムアミド、ジク
ロロメ
タン、アセトン、酢酸、その他反応に悪影響を及ぼさない任意の溶媒中で実施す
る。
反応温度はとくに限定されないが、通常は、加温ないし加熱下に反応を実施す
る。方法3
化合物(Ib)またはその塩は、化合物(Ia)またはその塩を脱アシル反応
に付すことにより製造できる。
化合物(Ia)の好適な塩としては、化合物(I)について例示したものと同
じものが挙げられる。
化合物(Ib)の好適な塩としては、化合物(I)について例示した酸付加塩
が挙げられる。
この反応は、加水分解、還元などの常法に従って実施する。
(i)加水分解の場合:
加水分解は、塩基もしくはルイス酸を含めての酸の存在下に実施するのが好ま
しい。
好適な塩基としては、アルカリ金属[たとえばナトリウム、カリウムなど]、
その水酸化物、炭酸塩もしくは重炭酸塩、アルカリ金属低級アルコキシド[たと
えばナトリウムメトキシド、ナトリウムエトキシドなど]、水素化物[たとえば
水素化リチウムアルミニウムなど]、トリアルキルアミン[たとえばトリメチル
アミン、トリエチルアミンなど]、ピコリン、1,5−ジアザビシクロ[4.3
.0]ノナ−5−エン、1,4−ジアザビシクロ[2.2.2]オクタン、1,
8−ジアザビシクロ
[5.4.0]ウンデカ−7−エンなどの無機塩基および有機塩基が挙げられる
。
好適な酸としては、有機酸[たとえば蟻酸、酢酸、プロピオン酸、トリクロロ
酢酸、トリフルオロ酢酸など]および無機酸[たとえば塩酸、臭化水素酸、硫酸
、塩化水素、臭化水素など]が挙げられる。トリハロ酢酸[たとえばトリクロロ
酢酸、トリフルオロ酢酸など]などを用いての脱離は、カチオン捕捉剤[たとえ
ばアニソール、フェノールなど]の存在下に実施するのが好ましい
反応は、通常、水、アルコール[たとえばメタノール、エタノールなど]、塩
化メチレン、テトラヒドロフラン、これらの混合物、その他反応に悪影響を及ぼ
さない任意の溶媒中で実施する。液状の塩基もしくは酸は、溶媒としても使用で
きる。反応温度はとくに限定されないが、通常は、冷却下ないし加温下に反応を
実施する。
(ii)還元の場合:
還元は、化学還元、接触還元を含めての常法に従って実施できる。
化学還元に使用すべき好適な還元剤は、金属(たとえば錫、亜鉛、鉄など)も
しくは金属化合物(たとえば塩化クロム、酢酸クロムなど)と有機酸もしくは無
機酸(たとえば蟻酸、酢酸、プロピオン酸、トリフルオロ酢酸、p−トルエンス
ルホン酸、塩酸、臭化水素酸など)との組合せである。
接触還元に使用すべき好適な触媒は、白金触媒(たとえば白金板、海綿状白金
、白金黒、コロイド状白金、酸化白金、白金線など)、パラジウム触媒(たとえ
ば海綿状パラジウム、パラジウム黒、酸化パラジウム、パラジウム炭、コロイド
状パラジウム、パラジウム/硫酸バリウム、パラジウム/炭酸バリウムなど)、
ニッケル触媒(たとえば還元ニッケル、酸化ニッケル、ラネーニッケルなど)、
コバルト触媒(たとえば還元コバルト、ラネーコバルトなど)、鉄触媒(たとえ
ば還元鉄、ラネー鉄など)、銅触媒(たとえば還元銅、ラネー銅、ウルマン銅な
ど)などの慣用のものである。還元は、通常、水、メタノール、エタノール、プ
ロパノール、N,N−ジメチルホルムアミド、テトラヒドロフラン、これらの混
合物などの、反応に悪影響を及ぼさない慣用の溶媒中で実施する。なお、化学還
元に使用すべき上記の酸が液状である場合には、それらを溶媒としても使用でき
る。
この還元の反応温度はとくに限定されないが、通常は、冷却下ないし加温下に
反応を実施する。方法4
化合物(Ia)またはその塩は、化合物(Ib)またはその塩をアシル化剤と
反応させることにより製造できる。
化合物(Ia)の好適な塩としては、化合物(I)について例示したものと同
じものが挙げられる。
化合物(Ib)の好適な塩としては、化合物(I)に
ついて例示した酸付加塩が挙げられる。
化合物(Ib)は、そのアミノ基における慣用の反応性誘導体の形で使用して
もよい。
アシル化剤は、式
(式中、R6は上に定義した通りのアシルを表わす)の化合物およびそのヒドロ
キシ基における反応性誘導体によって代表されうる。
好適な例としては、酸ハロゲン化物(たとえば酸塩化物など)、酸無水物、活
性アミド、活性エステルなどが挙げられる。
この反応において、化合物(VI)を遊離酸またはその塩で使用するときには、
1−(3−ジメチルアミノプロピル)−3−エチルカルボジイミドなどの慣用の
縮合剤の存在下で反応を実施するのが好ましい。
導入すべきアシル基がカルバモイル型アシルである場合には、アシル化剤を、
通常は、シアネート、イソシアネートもしくはそれらのアルカリ金属塩の形で使
用する。
反応は、通常、慣用の溶媒、たとえば水、アルコール[たとえばメタノール、
エタノールなど]、アセトン、ジオキサン、アセトニトリル、クロロホルム、ジ
クロロメタン、塩化エチレン、テトラヒドロフラン、酢酸エチル、N,N−ジメ
チルホルムアミド、N,N−ジメチルアセトアミド、ピリジン、酢酸、その他反
応に悪影響を
及ぼさない任意の有機溶媒中で実施する。これらの慣用の溶媒は、水との混合物
として用いてもよい。
反応温度はとくに限定されないが、通常は、冷却下ないし加温下に反応を実施
する。
反応を、アルカリ金属重炭酸塩、トリ低級アルキルアミン、ピリジン、N−低
級アルキルモルホリン、N,N−ジ低級アルキルベンジルアミンなどの無機塩基
もしくは有機塩基の存在下に実施することもできる。
出発化合物(II)および(IV)もしくはそれらの塩は、次の方法で製造できる
。
上記反応式中、R1、R2、R3、R4、A、QおよびX1は、各々上に定義した通
りであり、
R7はアシルを、
X2は酸残基を、
X3は酸残基を、それぞれ表わす。
上記した出発化合物(II)および(IV)の製造法を以下に詳細に説明する。方法A
化合物(IXa)は、化合物(VII)またはその塩を化合物(VIII)と反応させ
ることにより製造できる。
化合物(VII)の好適な塩としては、化合物(I)について例示したものと同
じものが挙げられる。
反応は、通常、慣用の溶媒、たとえば水、アルコール(たとえばメタノール、
エタノール、イソプロピルアルコール、t−ブチルアルコールなど)、アセトン
、テトラヒドロフラン、ジオキサン、ジクロロメタン、クロロホルム、ジメチル
アセトアミド、N,N−ジメチルホルムアミド、その他反応に悪影響を及ぼさな
い任意の有機溶媒中で実施する。これらの溶媒のうち、親水性の溶媒は、水との
混合物として使用してもよい。
反応温度はとくに限定されないが、通常は、冷却下ないし加温下に反応を実施
する。方法B
化合物(X)またはその塩は、化合物(IX)またはその塩を脱アシル反応に付
すことにより製造できる。
化合物(IX)および(X)の好適な塩としては、化合物(I)について例示し
たものと同じものが挙げられる。
この反応は、方法3と実質的に同様にして実施する。従って、この反応の反応
態様および反応条件[たとえば溶媒、反応温度など]については、方法3のとこ
ろでの説明を参照されたい。方法C
化合物(II)またはその塩は、化合物(X)またはその塩を化合物(XI)と反
応させることにより製造できる。
化合物(II)および(X)の好適な塩としては、化合物(I)について例示し
たものと同じものが挙げられる。
反応は、通常、慣用の溶媒、たとえば水、アルコール(たとえばメタノール、
エタノール、イソプロピルアルコール、t−ブチルアルコールなど)、テトラヒ
ドロフラン、ジオキサン、ジクロロメタン、クロロホルム、ジメチルアセトアミ
ド、N,N−ジメチルホルムアミド、その他反応に悪影響を及ぼさない任意の有
機溶媒中で実施する。これらの溶媒のうち、親水性溶媒は、水との混合物として
使用してもよい。
反応温度はとくに限定されないが、通常は、冷却下ないし加温下に反応を実施
する。方法D
化合物(IV)は、化合物(XII)を化合物(XIII)と反応させることにより製
造できる。
反応は、ハロゲン化アルミニウム(たとえば塩化アルミニウム、臭化アルミニ
ウムなど)、ハロゲン化チタン(たとえば四塩化チタンなど)などのルイス酸の
存在下で実施するのが好ましい。
反応は、通常、二硫化炭素、ジクロロメタン、ジクロロエタン、ベンゼンなど
の慣用の溶媒、その他反応に悪影響を及ぼさない任意の有機溶媒中で実施する。
反応温度はとくに限定されず、通常、冷却下ないし加熱下に反応を実施する。
上記の各方法で得られた化合物は、粉末化、再結晶、カラムクロマトグラフィ
ー、再沈殿などの常法により、単離、精製できる。
目的化合物(I)の各々が、不斉炭素原子および二重結合にもとづく光学異性
体、幾何異性体などの1または2以上の立体異性体を包含しうるが、かかる異性
体およびそれらの混合物はすべて本発明の範囲に包含されるものである。
新規チエニルチアゾール誘導体(I)およびそれらの医薬として許容される塩
は、抗潰瘍作用およびH2−レセプター拮抗作用を有し、胃炎、潰瘍(たとえば
胃潰瘍、十二指腸潰瘍、吻合部潰瘍など)、ゾリンジャー ・エリソン症候群、
逆流性食道炎、上部胃腸管出血などの予防または治療に有用である。
さらに、本発明の化合物(I)およびその医薬として許容される塩は、最近ヒ
トの胃粘液ゲル下で発見されたグラム陰性桿菌のヘリコバクター・ピロリ(Heli
cobacterpylori)などの病原菌に対して高い抗菌作用を有する。現に、本発明の
化合物(I)は、ヘリコバクター・ピロリの成育を阻害した。
医療目的には、本発明の化合物(I)およびその医薬として許容される塩を、
経口投与、非経口投与もしくは外用(局所適用)に適した固体状もしくは液状の
有機もしくは無機賦形剤などの製薬上許容される担体との混合物の形で該化合物
の一種を活性成分として含有する医薬製剤の形態で使用できる。該医薬製剤とし
ては、カプセル剤、錠剤、糖衣錠、顆粒剤、坐剤、液剤、懸濁剤、乳剤などが挙
げられる。所望により、これらの製剤に、補助物質、佐剤、安定剤、湿潤剤ない
し乳化剤、緩衝剤、その他の常用添加剤を配合できる。
化合物(I)の用量は、患者の年令および状態によって異なるであろうが、化
合物(I)として約0.1mg、1mg、10mg、50mg、100mg、250mg、5
00mg、1000mgの平均一回量が、潰瘍の治療に有効でありうる。一般に、0
.1mg/個体/日〜約1000mg/個体/日の量を投与すればよい。
目的化合物(I)の有用性を例証するために、若干の代表的な化合物(I)の
薬理試験データを以下に示す。試験A
(抗菌活性)試験方法
寒天希釈法により、インビトロ抗菌活性を調べた。試験菌株を、3%のウマ血
清および2%のデンプンを加えたブルセラブロス寒天中37℃で3日間前培養し
、各薬物の2倍段階希釈液を含有するブルセラ寒天プラス7%ウマ血液平板に、
多点レプリケーターを用いて、105cfu/mlを接種し、37℃で3日間培養する
。インキュベー
ションは、10%CO2雰囲気中で行う。インキュベーション後、肉眼的コロニ
ー成育を阻止した最低薬物濃度として、MICを読取る。
試験B (HCI−アスピリン潰瘍の阻止)試験方法
6週令、体重約200gの雄性スプレイグ・ドーリーラット各群7頭を、24
時間絶食させたのち、HCI−アスピリン潰瘍試験に用いる。アスピリン投与3
0分前に、0.1%メチルセルロース溶液に懸濁させた試験化合物の各々(32
mg/kg)を経口投与する。0.2NHCl含有0.1%メチルセルロース溶液に
懸濁させたアスピリンを、用量200mg/kg/10mlで、経口投与する。1時間
後、動物を屠殺し、胃を摘出する。つぎに、胃を2%ホルマリンで固定する。各
動物について、潰瘍の長さを測定し、試験動物における潰瘍の平均長さ(mm)を
対照動物におけるそれと比較して、阻害率を計算する。
以下の製造例および実施例は、本発明をより詳細に説明するために挙げるもの
である。製造例1
チオシアン酸ナトリウム(5.1g)のアセトン(200ml)溶液に、還流下
、塩化ベンゾイル(8.2g)をゆっくり加える。混合物を10分間還流させる
。この混合物に、4−(5−アセチルアミノメチル−2−チエニル)−2−アミ
ノチアゾール(12.3g)を加える。混合物を7.5時間還流させる。生じた
沈殿を濾取する。沈殿を水、つぎにアセトンで洗って、4−(5−アセチルアミ
ノメチル−2−チエニル)−2−(3−ベンゾイルチオウレイド)チアゾール(
15.1g)を得る。
製造例2
4−(5−アセチルアミノメチル−2−チエニル)−2−(3−ベンゾイルチ
オウレイド)チアゾール(15.0g)のメタノール(200ml)中懸濁液に、
外界温度で、水酸化ナトリウム水溶液(30ml中に3.5g)を加える。混合物
を60℃に5時間加熱する。混合物を濃縮し、つぎに、水(100ml)を加える
。混合物を6N塩酸で中和する。生じた沈殿を濾取して、4−(5−アセチルア
ミノメチル−2−チエニル)−2−チオウレイドチアゾール(11.0g)を得
る。
製造例3
4−(5−アセチルアミノメチル−2−チエニル)−2−チオウレイドチアゾ
ール(11.9g)と沃化メチル(7.6g)とのメタノール(120ml)懸濁
液を5時間還流させる。溶媒を減圧下に除去する。残留物を酢酸エチルで洗って
、4−(5−アセチルアミノメチル−2−チエニル)−2−[(アミノ)(メチ
ルチオ)メチレンアミノ]チアゾール沃化水素酸塩(11.8g)を得る。
製造例4
N−[(3−メチル−2−チエニル)メチル]アセトアミド(1.4g)と塩
化クロロアセチル(1.4g)とのジクロロメタン(100ml)溶液に、氷浴上
で冷却しながら、塩化アルミニウム(2.65g)をゆっくり加える。混合物を
外界温度で2時間撹拌し、つぎに、氷水(100ml)中に注ぐ。有機層を集め、
硫酸マグネシウムで乾燥する。溶媒を減圧下に除去して、N−[(5−クロロア
セチル−3−メチル−2−チエニル)メチル]アセトアミド(1.99g)を得
る。
製造例5
塩化アルミニウム(18.6g)と塩化クロロアセチル(11.9g)との塩
化メチレン(80ml)中混合物を、外界温度で30分間撹拌する。生じた溶液に
、外界温度で撹拌下に、2−(エトキシカルボニルアミノメチル)チオ
フェン(13g)を加え、混合物を25〜35℃で20分間撹拌する。反応混合
物を氷水中に注ぎ、クロロホルムで抽出する。抽出液を飽和重炭酸ナトリウム水
溶液および食塩水で洗い、硫酸マグネシウムで乾燥する。溶媒を減圧下に濃縮し
、沈殿を濾取して、2−クロロアセチル−5−(エトキシカルボニルアミノメチ
ル)チオフェン(14.3g)を得る。
製造例6
製造例5と同様にして、次の化合物を得る。
1)2−クロロアセチル−5−(2−メトキシカルボニルアミノエチル)チオフ
ェン

製造例7
塩化アルミニウム(4.4g)、塩化クロロアセチル(3.39g)およびジ
クロロメタン(10ml)からなる混合物に、−5℃で、3−アセチルアミノメチ
ルチオフェン(4.66g)を加え、つぎに、同温度で20分間撹拌する。外界
温度で1時間撹拌後、生じた混合物を氷水で希釈し、酢酸エチルで抽出する。抽
出液を水および食塩水で洗い、無水硫酸マグネシウムで乾燥し、減圧下に蒸発さ
せる。残留物をシリカゲルクロマトグラフィー(クロロホルム中2%メタノール
)に付して、4−アセチルアミノメチル2−クロロアセチルチオフェン(1.5
2g)を得る。
製造例8
実施例18と同様にして、次の化合物を得る。

4−(5−アセチルアミノメチル−2−チエニル)−2−[(アミノ)(メチ
ルチオ)メチレンアミノ]チアゾール沃化水素酸塩(2.0g)と2−メトキシ
エチルアミン(5ml)とのエタノール(30ml)中懸濁液を、72時間還流させ
る。溶媒を減圧下に除去し、残留物を水(100ml)に懸濁させる。混合物を炭
酸カリウム水溶液でアルカリ性とし、つぎに、酢酸エチル(200ml)−テトラ
ヒドロフラン(50ml)混合物で抽出する。抽出液を硫酸マグネシウムで乾燥し
、つぎに、減圧下に蒸発させる。残留物をシリカゲルカラムクロマトグラフィー
に付し、クロロホルム−メタノール(20:1)混合物で溶出する。酢酸エチル
から再結晶して、4−(5−アセチルアミノメチル−2−チエニル)−2−[(
アミノ)(2−メトキシエチルアミノ)メチレンアミノ]チアゾール(0.77
g)を得る。

ミノメチレンアミノ)チアゾール塩酸塩(630mg)の水(20ml)中混合物を
、炭酸カリウム水溶液でpH11までアルカリ性にする。生じた沈殿を濾取する
。メタノール−テトラヒドロフラン−ジイソプロピルエーテル混合物から再結晶
して、4−(5−アセチルアミノメチル−4−メチル−2−チエニル)−2−(
ジアミノメチレンアミノ)チアゾール(430mg)を得る。
実施例5
3−メトキシ−3−メチルブチルフタルイミド(8.7g)とヒドラジン水和
物(1.76g)とのエタノール(100ml)溶液を、3時間還流させる。反応
混合物に、4−(5−アセチルアミノメチル−2−チエニル)−2−[(アミノ
)(メチルチオ)メチレンアミノ]チアゾール沃化水素酸塩(4.0g)を加え
る。混合物を45時間還
流させる。不溶物を濾去する。溶媒を減圧下に除去し、残留物を水(100ml)
−テトラヒドロフラン(30ml)混合物に溶解させる。混合物を30%炭酸カリ
ウム溶液でpH10に調整し、つぎに、酢酸エチル(150ml)で抽出する。抽
出液を硫酸マグネシウムで乾燥する。溶媒を減圧下に除去し、残留物をシリカゲ
ルカラムクロマトグラフィーに付し、クロロホルム−メタノール(50:1)混
合物で溶出する。溶媒を減圧下に除去する。メタノール−テトラヒドロフラン−
ジイソプロピルエーテル混合物から再結晶して、4−(5−アセチルアミノメチ
ル−2−チエニル)−2−[(アミノ)(3−メトキシ−3−メチルブチルアミ
ノ)メチレンアミノ]チアゾール(0.57g)を得る。
実施例6
2−アセチルアミノメチル−5−クロロアセチルチオフェン(1.5g)、N
−(2−エトキシエチル)アミジノチオ尿素酢酸塩(1.6g)および炭酸水素
ナトリウム(1.2g)のエタノール(30ml)中懸濁液を、55℃に4.5
時間加熱する。溶媒を減圧下に除去し、残留物を水(70ml)−テトラヒドロフ
ラン(20ml)混合物に溶解させる。混合物を30%炭酸カリウム溶液でpH1
0に調整し、つぎに、酢酸エチル(150ml)で抽出する。抽出液を硫酸マグネ
シウムで乾燥する。溶媒を減圧下に除去し、残留物を酢酸エチルから結晶化させ
る。メタノール−水混合物から再結晶して、4−(5−アセチルアミノメチル−
2−チエニル)−2−[(アミノ)(2−エトキシエチルアミノ)メチレンアミ
ノ]チアゾール(810mg)を得る。

2−クロロアセチル−5−(エトキシカルボニルアミノメチル)チオフェン(
2.62g)とグアニジノチオ尿素(1.8g)とのエタノール(50ml)中混
合物を、撹拌下に5時間還流する。反応混合物を水中に注ぎ、生じた混合物を2
0%炭酸カリウム水溶液でpH8.5に調整し、クロロホルム−メタノール混合
物で抽出する。抽出液を食塩水で洗い、硫酸マグネシウムで乾燥し、減圧下に溶
媒を蒸発させ、残留物を酢酸エチル−アセトン混合物から再結晶して、2−ジア

ム溶液でpH10に調整したのち、酢酸エチル(100ml)で抽出する。溶媒を
減圧下に除去し、残留物をシリカゲルカラムクロマトグラフィーに付す。クロロ
ホルム−メタノール(200:1)混合物で溶出する。所望の画分を集め、溶媒
を減圧下に除去する。残留物をメタノール(10ml)−
4N塩化水素ジオキサン溶液(10ml)混合物に溶解させる。混合物を外界温度
で10分間撹拌する。溶媒を減圧下に除去する。エタノール−ジイソプロピルエ
ーテル混合物から再結晶して、2−[(アミノ)(2−エトキシエチルアミノ)
メチレンアミノ]−4−[5−(2−エトキシカルボニルアミノエチル)−2−
チエニル]チアゾール塩酸塩(460mg)を得る。
実施例14
2−(2−アセチルアミノエチル)−5−クロロアセチルチオフェン(1.0
g)とN−メチルアミジノチオ尿素(0.54g)とのエタノール(20ml)溶
液を、4時間還流させる。混合物を減圧下に濃縮したのち、残留物を20%炭酸
カリウム水溶液で塩基性とし、酢酸エチルで抽出する。抽出液を食塩水で洗い、
無水硫酸マグネシウムで乾燥し、減圧下に濃縮する。残留物をシリカゲルクロマ
トグラフィー(クロロホルム中5%メタノール)に付して、油状物を得る。この
遊離塩基を常法により蓚酸塩に
転化させて、4−[5−(2−アセチルアミノエチル)−2−チエニル]−2−
[(アミノ)(メチルアミノ)メチレンアミノ]チアゾール蓚酸塩(1.5g)
を得る。
実施例15
2−アセチル−4−アセチルアミノチオフェン(916mg)と三臭化フェニル
トリメチルアンモニウム(2.63g)とのテトラヒドロフラン(15ml)溶液
を、外界温度で1時間撹拌する。混合物を食塩水(30ml)で希釈し、酢酸エチ
ル(20m1)で抽出する。有機層を無水硫酸マグネシウムで乾燥し、減圧下に蒸
発させる。得られた4−アセチルアミノ−2−ブロモアセチルチオフェンを含有
する残留物および3−[(アミノ)(n−ブチルアミノ)メチレン]チオ尿素(
870mg)をアセトニトリル(20ml)に溶解させ、2時間還流させる。溶媒を
除去後、残留物を水で希釈し、炭酸カリウム水溶液でアルカリ性とし、酢酸エチ
ルで抽出する。抽出液を食塩水で洗い、無水硫酸マグネシウムで乾燥し、濃縮す
る。残留物をシリカゲルクロマトグラフィーに付し、クロロホルム中5%メタノ
ールで溶出して、4−(4−アセチルアミノ−2−チ
エニル)−2−[(アミノ)(n−ブチルアミノ)メチレンアミノ]チアゾール
(0.30g)を得る。
実施例16
N−(5−クロロアセチル−3−メチル−2−チエニルメチル)アセトアミド
(1.0g)とN−メチルアミジノチオ尿素(540mg)とのエタノール(20
ml)中懸濁液を、55℃に3.5時間加熱する。生じた沈殿を濾取する。メタノ
ール−ジイソプロピルエーテル混合物から再結晶して、4−(5−アセチルアミ
ノメチル−4−メチル−2−チエニル)−2−[(アミノ)(メチルアミノ)メ
チレンアミノ]チアゾール塩酸塩(580mg)を得る。

実施例16と同様にして、次の化合物を得る。

4−(5−アミノメチル−2−チエニル)−2−[(アミノ)(3−メチルブ
チルアミノ)メチレンアミノ]チアゾール二塩酸塩(1.1g)とシアン酸カリ
ウム(270mg)との水(10ml)中懸濁液を、外界温度で4時間撹拌する。反
応混合物を、30%炭酸カリウム溶液でpH10に調整する。生じた沈殿を濾取
する。メタノールから再結晶して、2−[(アミノ)(3−メチルブチルアミノ
)メチレンアミノ]−4−(5−ウレイドメチル−2−チエニル)チアゾール(
0.5g)を得る。
