発明の名称二非A非B型肝炎ウィルスに対する抗体に免疫化学反応性を示すペプ
チド
本発明は、C型肝炎(HCV)に対する抗体と免疫化学的に反応するペプチド及
び該ペプチドをコードする核酸配列に係わる。本発明は更に、検査液中のHCV
または抗HC■抗体の検出方法並びにかかる検出方法に使用し得る免疫化学試薬
及び検査キットに係わる。
HCVウィルスによって惹起される可能性が最も高い非A非B型肝炎は、ウィル
ス誘発性であることが示されている伝播性疾患または疾患系である。非A非B型
肝炎は、既知の肝炎ウィルス、即ちA型肝炎ウィルス(HAV) 、δ型肝炎ウ
ィルス(HBV)、δ型肝炎ウィルス(HDV)によって惹起されるもの、及び
サイトメガロウィルス(CMV)またはエプスタイン−バールウィルス(EBV
)によって誘発される肝炎を含む他の形態のウィルス関連肝疾患とは区別され得
る。NANBHは最初は輸血された個体において同定された。人からチンパンジ
ーへの伝播及びチンパンジーにおける連続継代は、NANBHが伝播性感染物質
に起因することを立証するものである。疫学的証拠は、水系感染流行型;血液ま
たは注射針関与型;及び散発(集団獲得)型の3つの型のNANBHがあり得る
ことを示している。しかしながら、NANBHを惹起し得る因子の数は未知であ
る。
NANBHによる感染の種々の段階において信頼性のある診断を行ない得る特異
的且つ高感度の方法を開発するためには、この型の免疫優勢ウィルスエピトープ
を同定することが極めて重要である。本発明は、抗HCV抗体に驚くほど予想外
の免疫化学反応性を示す新規のペプチドに係わる。
本発明の新規のペプチドは、抗HCV抗体と免疫化学反応性を示し、アミノ酸配
列(1文字コードで示す):A−X、−X2−X3−X4−L−X5−X、−E
−F−X7−X8−XI−B (I)〔ここで、Aは水素、アミノ酸またはポリ
ペプチドを表わし;XI、X2、X8、X4、X5、X6、X7、X、及びX、
は任意のアミノ酸であり得;Bはヒドロキシ、アミノ酸またはポリペプチドであ
る〕
または、抗HCV抗体と免疫学的反応性を示すそのフラグメントであるが、但し
、式A−D−R−E−V−L−Y−R−E−F −D −E −M−B (ここ
でA及びBは前記定義の意味を有する〕を有するペプチドは除くことを特徴とす
る。
配列D−R−E−V−L−Y−R−E−F−D−E−M (If)はHCv配列
の一部テアリ、SOD/HCV C100−3り0−ンのORF領域内に位置す
る。配列D−R−E−V−L−Y−R−E −F −D−E−Mを含む特異的免
疫反応性ペプチドは優先日1991年12月6日の同出願人名義の同時係属出願
に記載されている。
上述の配列(II)において、抗HCV抗体との免疫反応性に関してはただ3つ
のアミノ酸しか重要でない。驚くべきことに、他の全てのアミノ酸は、抗HCV
抗体に対するペプチドの高反応性を維持しながら多数の他のアミノ酸で置換し得
る。従って本発明は、当業者によって容易に製造され得る、抗HCV抗体に対し
て高免疫反応性を有するペプチドを提供する。上述の配列(I)においてアミノ
酸L1E及びFのみが重要なアミノ酸である。これら3つのアミノ酸は、図1か
ら判るように、ペプチドの抗HCV抗体に対する免疫反応性に影響することなく
他のアミノ酸で置換することはできない。
残りのアミノ酸X I−X eは、上記例外はあるが、任意のアミノ酸とし得る
。好ましい式Iのペプチドは、選択された1つのアミノ酸が別のアミノ酸で置換
されている、式A−D −R−E −V−L −Y −R−E −F −D −
E−M−Bを有するペプチドとは異なるペプチドである。かかるペプチドは、図
1に示したように、抗HCV抗体に対する高免疫反応性を有する。
上記ペプチドのほかに、かかるペプチドの機能的誘導体もまた本発明のペプチド
に属すると考えられる。ペプチドの機能的誘導体とは以下のものを含むとする:
a)ペプチドの酸付加塩;
b)ペプチドのアミド、特にC末端アミド;C)エステル、特にC末端エステル
;及びd)N−アシル誘導体、特にN末端アシル誘導体、とりわけN−アセチル
誘導体。
本発明のペプチドは、検査液中のHCV抗原または抗HCv抗体の存在を判定す
る診断方法に使用するのに特に適している。天然HCVと比較し、本発明のペプ
チドは、安全な非感染性起源のものであるという利点を有する。更に、本発明ペ
プチドは抗HCV抗体に対して特に高い親和性を有しており、このことで本発明
ペプチドは前記診断試験法に使用するのに極めて適当となる。
式Iのペプチドのフラグメントは、1つ以上のアミノ酸が他のアミノ酸で置換さ
れている、D −R−E −V −L −Y −R−E −F −D −E−M
のフラグメントとは異なるフラグメントである。
本発明のペプチドの製造は、公知の有機化学的ペプチド合成方法のいずれかまた
は組換えDNA技術によって行ない得る。後者は、1つ以上の問題のペプチドを
コードする核酸配列を含む組換え核酸配列を、宿主として適当な微生物中で発現
させることにより、所望のペプチドを製造することを含む。有機化学的ペプチド
合成方法は、均一相中または所謂固相を用いた縮合反応によって必要なアミノ酸
を結合することを含むと考えられる。
縮合反応は以下のように実施し得る:
a)遊離カルボキシル基を有すると共に他の反応基は保護されている化合物(ア
ミノ酸、ペプチド)を、遊離アミノ基を有すると共に他の反応基は保護されてい
る(ここで保護基の1つは(誘導体化)固体支持体であってもよい)化合物(ア
ミノ酸、ペプチド)と、縮合剤の存在下に縮合させる、
b)活性化カルボキシル基を有すると共に他の反応基は遊離または保護されてい
る化合物(アミノ酸、ペプチド)を、遊離アミノ基を有すると共に他の反応基は
遊離または保護されている(ここで保護基の1つは(誘導体化)固体支持体であ
ってもよい)化合物と縮合させる。
カルボキシル基の活性化は、特に、カルボキシル基を酸ハロゲン化物、アジ化物
、無水物、イミダゾリドまたは活性化エステル、例えば
N−ヒドロキシ−スクシンイミド、N−ヒドロキシベンゾトリアゾール、p−ニ
トロフェニル、3,4−ジヒドロ−3−ヒドロキシ−4−オキソ−1,2,3−
ベンゾトリアジン(ODhbt)またはペンタフルオロフェニル(OPfp)エ
ステルに変換することにより行ない得る。
上記縮合反応の最も一般的な方法は、カルボジイミド法、BOP法[ベンゾトリ

)に記載のごとき活性化エステルを使用する方法である。
縮合反応に関与し得ない反応基は、上述したように、酸、塩基または還元による
加水分解によって極めて容易に再び除去し得る基によって効果的に保護される。
カルボキシル基は、例えばメタノール、エタノール、第三級ブタノール、ベンジ
ルアルコールまたはp−ニトロベンジルアルコールを用いたエステル化、アミン
を用いたアミド化によって、或いは固体支持体に結合させたアルコール及びアミ
ンの誘導体を用いて効果的に保護し得る。
アミノ基を効果的に保護し得る基は、エトキシカルボニル、ベンジルオキシカル
ボニル、t−ブトキシ−カルボニル、9−フルオレニル−メトキシカルボニル(
Fmo c)もしくはp−メトキン−ベンジルオキシカルボニル基、またはスル
ホン酸から誘導される酸基、例えばp−トルエン−スルホニル、ペンタメチルベ
ンゼンスルホニル(Pms)、4−メトキシ−2,3,6−トリメチルーベンゼ
ンスルホニル(Mtr)もしくは1,2.5,7.8−ペンタメチルクロマン−
6−スルホニル(Pmc)基であるが、置換もしくは未置換アリールもしくはア
ラルキル基、例えばベンジル及びトリフェニルメチル、またはオルト−ニトロフ
ェニル−スルフェニル及び2−ベンゾイル−1−メチルビニルのごとき他の基を
使用することもできる。使用可能な保護基のより広範な記述は、The pep

“固相”法を使用する上記本発明ペプチドの製造は例えばJ、^m、 Chew
、 Soc、、 85.2149 (1963)及びInt、J、 Pepti
de Protein Res、、 35.161−214 (1990)に記
載されている。製造すべきペプチドのアミノ酸の結合は通常カルボキシル末端側
から開始する。この方法には、反応基がその上に存在するまたはかかる基を導入
し得る固相が必要とされる。これは例えば反応性クロロメチル基を有するベンゼ
ン及びジビニルベンゼンのコポリマー、またはヒドロキシメチルもしくはアミン
官能基に対して反応性にされたポリマー固相とし得る。
特に適した固相は、例えば、fang (1974) J、^m、 CheIl
。
Soc、、 95.1328によって記載されているp−アルコキシベンジルア
ルコール樹脂(4−ヒドロキシメチル−フェノキシ−メチル−コポリスチレン−
1%ジビニルベンゼン樹脂)である。合成後、ペプチドをこの固相から緩和条件
下で分離し得る。他の適当な支持体としては、Geysen、 Proc、 N
atl、 Acad、 Sci、、 81.3998 (1984)及びPro
c、 Natl、 Acad。
導体化ポリエチレンまたはポリプロピレンロッドが挙げられる。
溶液中または固体支持体上に所望のアミノ酸配列を合成した後、例えばトリフル
オロ酢酸を用いたり、水素及び触媒、例えばパラジウムによる緩和還元によった
り、ピペラジンもしくは水酸化物イオンのごとき塩基または氷酢酸中のHBrを
用いて処理するなど、特定の基の特性に従って種々の慣用方法によって保護基を
分離し得る。
ペプチドを固体支持体上に合成し、そこから引き離し得るとすれば、それは、リ
ンカ−のタイプに従って例えばトリフルオロ酢酸、トリフルオロメタンスルホン
酸、または、トリフルオロ酢酸中に溶解したメタンスルホン酸を用いたり、低級
アルコール、好ましくはメタノールまたはエタノールを用いてエステル変換する
ことにより行ない得る。エステル変換の場合、ペプチドの低級アルキルエステル
が直接形成される。同様に、アンモニアによって分離すれば本発明のペプチドの
アミドが得られる。
上述したように、本発明のペプチドは組換えDNA技術によって製造することも
できる。この可能性は、ペプチドを(縦列)反復配列中に取込む場合や、ペプチ
ドを(はるかに大きい)タンパク質またはポリペプチドの必須成分として製造す
る場合に特に重要である。従ってこの種のペプチド製造も本発明の範囲内に含ま
れる。このためには、組換えDNAの構成成分として、本発明のペプチドはコー
ドするが、天然HCVゲノムにおいて上述の核酸配列に隣接するポリヌクレオチ
ドセグメントを実質的に含まない核酸配列を使用する。
本発明のペプチドをコードする上記のごとき核酸配列、及びこの核酸配列が取込
まれた組換えDNAも本発明の範囲内に含まれる。
本明細書において使用される“核酸配列”なる用語は、任意の長さのポリマー形
態のヌクレオチド、即ちリボ核酸配列及びデオキシリボ核酸配列の両方を指す。
原則としてこの用語は分子の一次構造を指す。従ってこの用語は、二本鎖及び−
重鎖DNA、並びに二本鎖及び−重鎖RNA。
更にこれらの変性体を含む。本発明の核酸配列は、実際には関連もないし連結も
していない種々の複製可能DNA配列に連結し、適当な宿主の形質転換に使用し
得る所謂組換えベクター分子となり得る。有効な組換えベクター分子は、例えば
プラスミド、バクテリオファージ、コスミドまたはウィルスから誘導されるのが
好ましい。本発明の組換えベクター分子の構築に使用される方法は当業者には公
知であり、特に、Maniatis、 T、ら(Molecular Clon
ing A Laboratory Manual、第二板; Co1d Sp
ring Harbor Laboratory。
1989)に記載されている。例えば、本発明の核酸配列をクロ−ニグベクター
中に挿入することは、遺伝子及び所望のクローニングビヒクルの両方を、相補的
DNA末端が生成されるのと同じ制限酵素で切断したならば容易に行ない得る。
発現ベクター中の本発明の核酸配列を発現させるためには、該配列を発現制御配
列に作動可能なように連結する。かかる制御配列は、プロモーター、エンハンサ
−、オペレーター、インデューサー、リポソーム結合部位などを含み得る。
勿論、クローニングベクターの選択部位に挿入されたヌクレオチド配列は、形質
転換宿主が少なくとも1つ以上の免疫原決定基を有するポリペプチドを産生ずる
限りは、本発明のペプチドをコードする全核酸配列のフラグメントしか含まなく
てよいことが理解される。
組換えベクター分子は更に、例えばpBR322におけるアンピシリン及びテト
ラサイクリン耐性、pUC8におけるアンピシリン耐性及びβ−ガラクトシダー
ゼのα−ペプチドのごとき所望の形質転換体を選択するのに使用し得る1つ以上
のマーカー活性を含み得る。
上述のごとく製造されるペプチドまたはそのフラグメントを使用し、ポリクロー
ナル及びモノクローナルいずれの抗体をも生産し得る。本発明のペプチドに対す
るモノクローナル抗体は、当業者には容易に生産し得る。ハイブリドーマにより
モノクローナル抗体を生産することはよ(知られている。細胞融合により不死化
抗体産生細胞系を生成することもできるし、また、腫瘍原性DNAによるBリン
パ球の直接的形質転換またはエプスタイン−バールウィルスによるトランスフェ
クションのごとき他の方法も使用し得る。本発明のペプチドに対するモノクロー
ナル及びポリクローナルの両抗体は診断に極めて適しており、中和性の抗体は受
動免疫療法に極めて有効である。特にモノクローナル抗体は、抗イデイオタイプ
抗体を誘導するために使用し得る。抗イデイオタイプ抗体を誘導する方法は当分
野において知られている。抗イデイオタイプ抗体は、非A非B型肝炎の予防及び
/または治療、並びにHCV抗原の重要なエピトープ領域の解明にも有効である
。
本発明は更に、1種以上の本発明ペプチドを含む免疫化学試薬を提供する。本発
明の“免疫化学試薬“は通常1種以上の本発明ペプチドと、適当な支持体または
標識物質とを含む。この点で使用し得る支持体としては、例えば、BSAのごと
き担体タンパク質、微量試験ウェルまたはキュベツト、試験管もしくは毛細管の
内壁、膜、フィルター、試験ストリップまたは粒子(例えばラテックス粒子、赤
血球、染料ゾル、金属ゾル)、または金属化合物(例えばゾル粒子)が挙げられ
る。
使用し得る標識物質は、放射性同位元素、蛍光化合物、標識用タンパク質(例え
ば酵素)、酵素、染料ゾル、金属ゾル、または金属化合物(例えばゾル粒子)で
あり得る。
免疫化学試薬は、検査液中のHCVに対する抗体の検出方法に必須の成分である
。免疫化学試薬を前記検査液と接触させると免疫化学反応が起こり、(本発明の
)ペプチドと抗HCV抗体とで免疫複合体が形成される結果となる。免疫化学試
薬の特性及び更なる特徴に従って、起こる免疫化学反応は所謂サンドイッチ反応
、凝集反応、競合反応または阻害反応である。上記反応の結果形成される免疫複
合体は、検査液中の抗体の存在についての(直接または間接的な)尺度である。
本発明の免疫化学試薬は、検査液中のHCV抗原を検出するために使用すること
もできる。
本発明は更に、1種以上の本発明ペプチドを使用する、検査液中のHCVに対す
る抗体を検出する方法をも提供する。本発明は更に、1種以上の本発明ペプチド
を使用する、検査液中のHCVを検出する方法にも係わる。この検出方法におい
ては、検査液を抗HCV及び本発明の免疫化学試薬と順次及び同時に接触させ得
る。
検査液中のHCVを検出するのに特に適した方法は、標識物質が与えられた本発
明ペプチドと(検査液中に存在する)HCV抗原との競合反応に基づいており、
本発明ペプチドと抗原とが、固体支持体に固定されたHCVに対する抗体に関し
て競合する。
本発明は更に、少なくとも1種の本発明の免疫化学試薬を含む、イムノアッセイ
を実施するために使用される検査キットをも提供する。本発明の検査キットは必
須成分として上述の免疫化学試薬を含む。抗IイCV抗体検出のためにサンドイ
ッチ反応を実施する際には、検査キットは、例えば、
1)固体支持体(例えば微量試験ウェルの内壁)に固定された本発明のペプチド
、及び
2)本発明の標識ペプチドまたは標識抗抗体のいずれかを含み得る。
HCV抗体検出のために競合反応を実施する際には、検査キットは、
