Description
〔産業上の利用分野〕
本発明は、イオントフオレーゼにおける、生体
電極用基剤組成物に関するものであつて、特に、
全身性薬物を投与できる前記組成物を提供できる
ものである。
〔従来技術〕
従来、局所あるいは全身性薬物を外皮より投与
する方法としては、軟膏、クリーム等、外皮に薬
物を基剤と共に塗擦する方法、あるいは、テープ
製剤を貼付する方法がとられている。しかしなが
ら、この方法は、単に皮膚表面にのみ薬物を塗布
又は貼付する性質のものであつた。そこで、外用
薬を経皮的に吸収させるための、経皮吸収促進剤
(特開昭58−210026号公報)が提案されている。

きた。これらの観点から、イオン滲透法(イオン
トフオレーゼ)が見直されている。本法は、直流
電流による電気治療で、イオン化した分子に対す
る極性効果を利用して、薬物を皮膚中に浸透させ
る方法であり、これまでに、殺菌、静脈瘤性潰
瘍、ケロイド、慢性関節炎等に利用されている。
一方、近年全身性薬物を経皮投与する新しい製
剤技術が開発されているが、イオントフオレーゼ
による全身性薬物の投与方法は試みらていない。
これは、イオントフオレーゼにより薬物を経皮的
に体内に浸透させるには、比較的高電圧、高電
流、短時間処理が必要であると考えられていたた
めである。
〔発明が解決しようとする問題点〕
本発明者らは、低電圧、低電流、長時間処理に
よる薬物の経皮的に制御された速度での投与を実
現すべく、鋭意研究を重ねた結果、次の知見を得
た。すなわち、イオントフオレーゼにおいて生体
電極として後記する特定の化合物および該化合物
に有機溶媒、必要に応じて水を配合してなる組成
物にイオン性薬物を配合することにより低電圧、
低電流にて薬物の経皮吸収性が容易に進行するこ
とができることを見い出した。
本発明は、上記の知見に基づいて完成されたも
のであり、その第1の目的は、イオントフオレー
ゼにおいて低電流密度にて薬物の経皮透過を可能
とする生体電極用基剤組成物を提供することにあ
る。
本発明の第2の目的は、イオントフオレーゼに
おいて低電流密度にて薬物の経皮浸透性を高める
方法を提供することにある。

本発明の生体電極用基剤組成物は、イオントフ
オレーゼによるイオン解離性薬物の皮膚浸透、経
皮吸収を促進するので、例えば0.1〜0.5mA/cm2
程度の低電流でも当該当薬物の皮膚吸収を促進す
ることができる。
本発明において用いられる基剤組成物は、無通
電下でもイオン性薬物の皮膚透過を促進するが、
通電を行うことによつて、相乗的に薬物の皮膚透
過、経皮吸収を促進するものであるから、低電流
でもイオン導入が可能になる。また、長期間にわ
たつて当該薬物投与が可能であるところから、薬
効の持続化が可能である。さらに、イオン解離性
薬物の皮膚透過、経皮吸収が改善されるので、小
型電池の使用が可能となり、携帯でき、かつ、長
時間通電による放出、投与、制御可能な装電化が
可能になるという利点がある。
〔実施例〕
以下、実施例、実験例などによって本発明をよ
り具体的に説明するが、本発明はこれらによつて
何ら限定されるものではない。なお、以下におい
て部とあるのはすべて重量部を示し、またPCA
はピロリドンカルボン酸を意味する。
実施例 1

上記(1)を(2)に溶解して薬物溶液を作成し、直径
20mm、厚さ2mmのポリエステル不織布に0.5mlの

のあるガラスセルの両端にラツト剥離皮膚を設置
し、片側に実施例1または比較例1の電極部を取
りつけ、反対側には実施例1における薬物溶液の
代わりに、0.9%NaCl溶液を含浸した実施例1と
同様の電極部を取りつけ、薬液側を陰極とし、
0.25mA/cm2の電流を10分間通電し、薬物の皮膚
透過量を高速液体クロマトグラフで測定した。
結果を第1図に示した。

実験例 7
ウイスター系雄性ラツト(体重約180g)の下
腹部を除毛し、右腹部に実施例20または比較例9
−aおよび9−bを粘着テープで貼着した。
左腹部には薬物含有液と同様の構成の電極部材
に0.9%NaCl溶液を含浸し、電極として粘着テー
プで貼着した。
薬物含有側を陽極とし、0.25mA/cm2の電流を
5分間通電し、ラツト尾動脈圧を測定した。
