Description
本発明はβ−ラクタム環を有する化合物の直腸
投与用組成物に関する。
β−ラクタム環を有する化合物、例えばセフア
ロスポリン、セフアマイシン系抗生物質は広い抗
菌スペクトルを有し、かつ毒性が小さいことか
ら、細菌感染症の予防および治療薬として広く使
用され、あるいは試験されている。
しかし、これらの抗生物質の中には、経口投与
では吸収され難く、生物学的利用能が小さいため
に、本来の抗菌活性を期待し得ない化合物例が多
い。これらの化合物は従来静脈内あるいは筋肉内
注射が行なわれているが、注射投与の場合、疼
痛、筋肉障害、便宜性、作用持続性などの欠点を
有している。かかる状況から最近では注射剤に代
つて種々の添加剤による吸収促進効果を利用した
直腸投与剤形が注目され、提案されている(例え
ば特開昭55−149209、同56−100712、同56−
104812、同56−104813)。しかしながら、従来の
直腸投与剤では実用上の問題点として、安定性、
刺激性、組織障害性等の欠点がある。
本発明者らは、β−ラクタム環を有する化合物
にある種の植物油、非イオン性界面活性剤および
縮合リン酸塩を配合することにより、上記問題点
を解決し、かつ非常に良好な直腸吸収性を示すこ
とを見出し、本発明を完成した。
本発明は、β−ラクタム環を有する化合物に、
オリーブ油、ゴマ油、ヤシ油、アマニ油およびナ
タネ油から選ばれた一種または二種以上の植物
油、非イオン性界面活性剤および縮合リン酸塩を
配合したことよりなる直腸投与用組成物に関す
る。
本発明の直腸投与用組成物は、例えば次のよう
な方法で製造することができる。
即ち、基剤は前記植物油の一種または二種以上
を混合したものに、非イオン性界面活性剤および
必要に応じて下記に述べる吸収促進助剤の一種ま
たは二種以上を混合し、必要ならば60℃以上に加
熱した後、混合攪拌下で室温まで冷却し、次いで
縮合リン酸塩を加え均一にすることによつて得ら
れる。直腸投与用組成物は、このようにして得ら
れた基剤にβ−ラクタム環を有する化合物を混合
し、ホモゲナイザーを用いて微細分散または溶解
することによつて得られる。分散物または溶解物
は、固形、軟膏、溶液の形態で0.5〜3gの適量
をそのまま一般坐薬、直腸投与用ソフトカプセル
または適当な直腸投与用容器に封入することによ
り使用に供される。
本発明のβ−ラクタム環を有する化合物として
は、例えば以下に示す化合物およびそれらの各種
塩(アルカリ金属塩、アルカリ土類金属塩含窒素
有機塩、有機酸塩など)があげられる。β−ラク
タム環を有する化合物の使用量は通常は基剤に対
して1〜50%、好ましくは3〜30W/W%であ
る。

This invention relates to a rectal administration composition containing one or more vegetable oils selected from olive oil, sesame oil, coconut oil, linseed oil, and rapeseed oil, a nonionic surfactant, and a condensed phosphate. The composition for rectal administration of the present invention can be produced, for example, by the following method. That is, the base is a mixture of one or more of the vegetable oils mentioned above, a nonionic surfactant and, if necessary, one or more of the absorption promoting aids described below. It is obtained by heating to 60° C. or higher, cooling to room temperature while stirring, and then adding condensed phosphate to make it homogeneous. エステル、例えばポリオキシエチレン(以下
POEと略称)ソルビタンモノラウレート、POE
ソルビタンモノパルミテート、POEソルビタン
モノステアレート、POEソルビタントリステア
レート、POEソルビタンモノオレート、POEソ
ルビタントリオレートなど;ソルビタン脂肪酸エ
ステル、例えばソルビタンモノラウレート、ソル
ビタンモノパルミテート、ソルビタンモノステア
レート、ソルビタンモノオレート、ソルビタント
リオレート、ソルビタンセスキオレートなど;ポ
リオキシエチレン脂肪酸エステル、例えばPOE
ラウレート、POEステアレート、POEオレート、
POEパルミテートなど;ポリオキシエチレンア
ルキルエーテル、例えばPOEラウリルエーテル、
POEセチルエーテル、POEオレイルエーテルな
ど;ポリオキシエチレン・ポリオキシプロピレン

糖エステル;アルキル(ポリオキシエチレン)リ
ン酸エステル、例えばジPOEアルキルエーテル
リン酸エステル、トリPOEアルキルエーテルリ
ン酸エステルなど;ポリオキシエチレンアルキル
アリールエーテル、例えばPOEノニルフエニル
エーテル、POEオクチルフエニルエーテルな
ど;ポリオキシエチレンコレステロール誘導体が
用いられる。これらの中ではPOEソルビタン脂
肪酸エステル、POEアルキルエーテル、POE脂
肪酸エステル、ポリオキシエチレン・ポリオキシ
プロピレン共重合体が良好であり、さらにこれら
の中でもPOEソルビタン中級脂肪酸エステル、
POE中級アルキルエーテル、POE中級脂肪酸エ
ステルが最適である。これらの非イオン性界面活
性剤は一種または二種以上を混合して使用するこ
ともできる。使用量としては通常は基剤全量の
0.05〜70W/W%、好ましくは0.5〜20W/W%
である。
次に本発明で使用される縮合リン酸塩としては
例えばピロリン酸、トリポリリン酸、トリメタリ
ン酸、ヘキサメタリン酸などのナトリウム塩、カ
リウム塩、リチウム塩のようなアルカリ金属塩、
アンモニウム塩が好ましい。これらの縮合リン酸
塩は一種または二種以上を混合して使用すること
もできる。使用量としては通常は基剤全量の0.05
〜50W/W%、好ましくは0.5〜10W/W%であ
る。
本発明においては、更に必要に応じて脂肪酸グ
リセライドを吸収促進助剤として使用することも
できる。使用される脂肪酸グリセライドとしては
脂肪酸のモノ、ジおよびトリグリセライドが限定
なく使用される。脂肪酸モノグリセライドとして
は、モノアセテート、モノプロピオネート、モノ
カプロエート、モノカプリレート、モノカプレー
ト、モノラウレート、モノミリステート、モノパ
ルミテート、モノステアレート、モノオレートな
ど;脂肪酸ジグリセライドとしては、ジカプリレ
ート、ジカプレート、ジカプロエート、ジラウレ
ート、ジミリステート、ジオレート、オクチルデ
シルグリセライドなど;脂肪酸トリグリセライド
としては、トリカプリレート、トリカプレート、
トリカプロエート、トリラウレート、トリミリス
テート、ジオクチルデシルグリセライドなどが用
いられるが、中級脂肪酸のトリグリセライドが好
適である。これらの脂肪酸グリセライドは、非イ
オン性界面活性剤と共に用いると、吸収を更に促
進する効果を有する。脂肪酸グリセライドは一種
または二種以上を混合して使用することもでき
る。使用量としては通常は基剤全量の0.01〜
30W/W%である。
本発明においては、常法により油脂安定化剤
(例えば抗酸化剤、保存剤)、主剤安定化剤(例え
ばキレート剤、錯化剤)、製剤安定化剤(沈降防
止剤、ゲル化剤、高分子物質)等も添加すること
ができる。
次に実施例を示す。なお部はいずれも重量部を
示す。
実施例 1
オリーブ油93部にポリオキシエチレンラウリル
エーテル(商品名Brij35)5部を加えて加温し溶
解後、室温に冷却した。次いでヘキサメタリン酸
ナトリウム2部を加えて均一な基剤とした。この
基剤175gに化合物1のナトリウム塩25gを加え
て均一に分散した。これを2gずつ直腸投与用ソ
フトカプセルに充填した。
実施例 2
ヤシ油95部にポリオキシエチレンオレイルエー
テル(商品名ニツコールBO−10TX)3部を加
えて加温し溶解後、室温に冷却した。次いでヘキ
サメタリン酸ナトリウム2部を加えて均一な基剤
とした。この基剤を用いて以下、実施例1と同様
にして調製した。
実施例 3
ゴマ油96.5部にポリオキシエチレンオレイルエ
ーテル(商品名ニツコールBO−10TX)3部を
加えて加温し溶解後、室温に冷却した。次いでヘ
キサメタリン酸ナトリウム0.5部を加えて均一な
基剤とした。この基剤を用いて以下、実施例1と
同様にして調製した。
実施例 4
ナタネ油94部にポリオキシエチレンオレイルエ
ーテル(商品名ニツコールBO−10TX)3部を
加えて加温し溶解後、室温に冷却した。次いでヘ
キサメタリン酸ナトリウム3部を加えて均一な基
剤とした。この基剤を用いて以下、実施例1と同
様にして調製した。
実施例 5
オリーブ油90部にポリオキシエチレンラウリル
エーテル(商品名Brij30)5部を加えて加温し溶
解後、室温に冷却した。次いでピロリン酸ナトリ
ウム5部を加えて均一な基剤とした。この基剤を
用いて以下、実施例1と同様にして調製した。
実施例 6
オリーブ油93部にポリオキシエチレンラウリル
エーテル(商品名Brij30)5部を加えて加温し溶
解後、室温に冷却した。次いでトリポリリン酸ナ
トリウム2部を加えて均一な基剤とした。この基

均一に分散した。これを以下、実施例1と同様に
して調製した。

代りに化合物9のナトリウム塩を用いて、以下同

解後、冷却して均一な基剤とした。この基剤175

次に直腸投与試験例を示す。
試験例 1
実験前24時間絶食させたビーグル犬(体重10〜
13Kg)の直腸に、各実施例および対照例により調
製した製剤を投与した。投与24時間後に集めた全
尿中のβ−ラクタム環を有する化合物の濃度をバ
イオアツセイ法または液体クロマトグラフ法によ
り求め、尿中回収率(%)を算出した。

基剤とした場合に比べて、本発明の植物油を使用
した場合は非常に良好な結果を示した(対照例
8,9との比較)。更に縮合リン酸塩を基剤に含
有する場合は、含有しない場合に比べて非常に良
好な結果を示した(実施例7と対照例10との比
較)。
試験例 2
はつかねずみ(体重約300g)の直腸に、固形
試料は分割して、液体試料は注入器を用いて、各
実施例および対照例により調製した製剤をβ−ラ
クタム環を有する化合物を50mg/Kg量投与した。
投与後、尾静脈より一定時間ごとに血液を採取
し、液体クロマトグラフ法により血液中のβ−ラ
クタム環を有する化合物の濃度を求めた。
