【発明の詳細な説明】[Detailed description of the invention]
本発明はL−セリンの製造法に関し、さらに詳
しくは、グリシンとホルムアルデヒドから微生物
の生産するセリンヒドロキシメチルトランスフエ
ラーゼを利用して、反応液に添加したグリシンに
対し高収率にL−セリンを製造する方法に関する
ものである。
L−セリンは医薬品、化粧品等に利用されるア
ミノ酸であり、現在は化学合成法またはグリシン
を前駆体とする発酵法により製造されている。化
学合成法の場合はDL体が合成されるためL体の
みを得るのには光学分割をしなければならないと
いう欠点があり、又、グリシンを前駆体とする発
酵法でも蓄積量、収率、精製、廃水処理等に欠点
があり、実用上有利な方法とは言い難い。これら
の方法に代つて最近は、セリンヒドロキシメチル
トランスフエラーゼを利用し、酵素法でL−セリ
ンを製造する方法が注目されている。セリンヒド
ロキシメチルトランスフエラーゼ(E.C.2.1.2.1)
はセリントランスヒドロキシメチラーゼまたはセ
リンアルドラーゼとも呼ばれ、哺乳動物、鳥類、
高等植物、エシエリヒア属を含む微生物等に広く
分布しており、テトラヒドロ葉酸とピリドキサル
リン酸を補酵素とし、グリシンとホルムアルデヒ
ドからL−セリンを生成せしめる反応を触媒する
ことも既に知られている。しかし従来の、微生物
のセリンヒドロキシメチルトランスフエラーゼを
利用し酵素法によりL−セリンを製造する方法と
して知られていることはプロテウス属(特公昭58
−2677)、サルシナ属、フラボバクテリウム属、
シユードモナス属、ミクロバクテリウム属(特開
昭53−81691)などの微生物を用い、グリシン単
独でまたはグリシンと微量のホルムアルデヒドよ
りL−セリンを製造する方法が知られているので
ある。そしてこれらの方法において使用された微
生物は、いづれもグリシンそのものからL−セリ
ンを生成せしめる強い活性を持つものであり、反
応液にホルムアルデヒドを添加した場合でも生成
したL−セリンのモル数と反応に添加したホルム
アルデヒドのモル数の比を見ると凡そ10対1にす
ぎず、反応にはセリンヒドロキシメチルトランス
フエラーゼの関与もさることながらこれに加えて
グリシンを開裂する反応を触媒する酵素が強く関
与しているものと理解され、そのため生成したL
−セリンの対グリシン収率は10%モル/モル以下
と著しく低い。なおこの場合L−セリンの蓄積濃
度は高々5g/程度でしかなく到底実用的な方
法とは言い難い。
本発明は、微生物の生産するセリンヒドロキシ
メチルトランスフエラーゼを利用し、グリシンの
存在下特にホルムアルデヒドを有効に利用してL
−セリンをグリシンに対し収率よく、しかも高い
反応液中の濃度で製造する方法について鋭意研究
の結果得られたものである。その結果我々はエシ
エリヒア属に属し、セリンヒドロキシメチルトラ
ンスフエラーゼ生産能を有し、かつグリシン単独
からはL−セリンを生成する能力が低いか又は能
力を有しない微生物の培養液、菌体もしくはそれ
らの処理物の存在下グリシンとホルムアルデヒド
を、反応系中のホルムアルデヒド濃度を20mM以
下に保つようにホルムアルデヒドを連続または間
けつ的に添加して反応させると添加したグリシン
に対し80%モル/モル以上の著しく高い収率で、
しかも50g/以上の著しく高い濃度でL−セリ
ンが生成することを見出し、本発明に完成するに
至つた。
本発明の方法により得られるこのような高収
率・高濃度でL−セリンを生成せしめる方法は全
く知られていない。更に、本発明の方法によれ
ば、反応終了時に残存するグリシンが著しく少な
く、このことは反応液からのL−セリンの単離精
製にかゝる負担を著しく軽減するものであり、本
発明の方法は酵素法によるL−セリンの工業的製
法として極めて優れている。
本発明において用いられる微生物は、エシエリ
ヒア属に属し、セリンヒドロキシメチルトランス
フエラーゼ生産能を有し、かつ、グリシン単独か
らはL−セリンを生成する能力を実質的に有しな
い菌株である。この性質を有する菌株であれば自
然界から分離されたものでも、突然変異、遺伝子
組み替え等の手段により創製されたものでも本発
明に使用できる訳であるが例えば、エシエリヒ
ア・コリ(Escherichia coli)MT−10350(微工
研菌寄第7437号)、エシエリヒア・コリMT−
10351(微工研菌寄第7438号)をあげることができ
る。
本発明に言う「実質的に有しない」とは、全く
有しない場合は勿論含むものであるが、本発明の
実施を妨げない程度の微弱な活性を有する場合は
格別本発明の効果をもたらすことに支障はない訳
であるからそのような場合も含む意味を示すもの
である。
又「酵素的処理物」とは、本発明の実施におい
て主役を果すものは、微生物に由来するセリンヒ
ドロキシメチルトランスフエラーゼであるから、
この酵素の活性を損うことのないように微生物そ
れ自体又はその培養液などを処理した一切の物を
示すものである。
本発明の実施につき微生物の培養に当つては、
培地およびその他の培養条件に特に制限はなく、
使用菌株の利用しうる炭素源、窒素源、無機塩
類、有機栄養物質などを含有するものであれば合
成培地、天然培地のいずれも使用できる。培養は
好気的条件下、PH5〜9、25〜40℃で行なうのが
好ましい。
そして得られる培養液はそのままでも、培養液
から遠心分離、過等により集菌した生菌体、そ
の乾燥菌体あるいは菌体処理物(例えば、生菌体
を磨砕、超音波、自己消化等により処理したも
の、菌体抽出液、該抽出物より得られる酵素区
分)などでも酵素源として用いることが出来る。
本発明の酵素反応は、PH6〜9、温度20〜60℃
で振とうもしくは撹拌条件下で行なうのが好まし
い。
セリンヒドロキシメチルトランスフエラーゼは
補酵素としてテトラヒドロ葉酸とピリドキサルリ
ン酸を要求するため、これらの物質を反応系に添
加することにより反応が高められることがある。
本発明の反応は、還元剤の添加又は窒素の通気
により高められることがある。この還元剤として
は、アスコルビン酸、ジチオスレイト−ル、2−
メルカプトエタノール、ジチオエリスリトール、
還元型グルタチオン、システイン、亜硫酸ナトリ
ウムなどがあり、使用濃度は通常0.1〜10mMが
好適である。
反応基質であるグリシンの使用濃度には特に制
限はなく、通常1〜40%程度であり、反応開始時
に全量添加しておいてもよく、反応の進行にとも
ない分割添加してもよい。
もう一つの反応基質であるホルムアルデヒドは
気体のまゝ、水溶液にして、アルコール溶液とし
て、更には固形重合物のパラホルムアルデヒドな
どで反応液に供給して使用できるが、37%程度の
水溶液であるホルマリンの使用が好適である。ホ
ルマリンは、そのまま反応液に添加しても、更に
希釈したのち添加してもよい。
反応液中のホルムアルデヒド濃度を20mM以下
に保つ手段としては、ホルムアルデヒドを連続的
にまたは間けつ的に添加する方法でよく、この場
合反応液中のホルムアルデヒド濃度を直接または
間接的な方法で随時測定して反応液へのホルムア
ルデヒド添加量を制御する。反応液中のホルムア
ルデヒド濃度の測定は、例えば、ガスクロマトグ
ラフイーによつても良いし、クロモトロプ酸、ア
セチルアセトンまたは4−アミノ−3−ヒドラジ
ノ−5−メルカプト−1,2,4−トリアゾール
等とホルムアルデヒドによる発色をそれぞれ固有
の波長の吸光度で測定してもよい。
反応液中に生成したL−セリンを単離するに
は、濃縮、イオン交換樹脂や活性炭による吸脱着
処理など常法が適用できる。
生成したL−セリンの確認および定量は、ペー
パークロマトグラフイーによるニンヒドリン発色
位置、液体クロマトグラフイーおよびロイコノス
トツク・メゼンテロイデスによるバイオアツセイ
法により行なうことができる。
以下、実施例により本発明を具体的に説明す
る。
実施例 1
エシエリヒア・コリMT−10350(微工研菌寄第
7437号)をブイヨンスラント上にて37℃、40時間
生育させたのち、第1表に示す栄養素を含む液体
培地(PH7.2)100mlに1白金耳づつ接種し、37℃
で40時間振とう培養した(5本)。この培養液を
20ジヤーフアメンタ−中の上記組成の培地10
に接種し、PH7.2、37℃で30時間通気撹拌培養し
た。培養液を遠心分離して菌体を集め、更にこの
菌体を生理食塩水にて洗浄し、湿菌体約50gを得
た。この湿菌体を−15℃で2日間凍結し反応直前
に解凍したものを酵素源として使用した。第2表
に示す栄養素を含む組成の反応液500mlに凍結菌
体40gを加え、反応液中のホルムアルデヒド濃度
をガスクロマトグラフイーで分析しその濃度が7
〜12mMの範囲であるようにホルマリンをペリス
タポンプで連続的に供給した。反応は50℃、PH
7.0、反応液中に1ml/minで窒素ガスを通じ、
ゆるやかな撹拌条件下で実施した。反応液へのホ
ルマリンの供給は37時間継続し、反応液に供給し
たホルマリン(37%w/wホルムアルデヒド溶
液)の総量は、24mlであつた。反応終了時には反
応液中に、29gのL−セリンが蓄積し、L−セリ
ンの蓄積濃度は55g/、生成したL−セリンの
反応液に添加したグリシンに対する収率はモル比
で83%、ホルムアルデヒドに対する収率はモル比
で86%であつた。

実施例 2
実施例1と同様の方法で、使用した微生物を代
えてエシエリヒア・コリMT−10350(微工研菌寄
第7438号)を用い反応を行なつた。反応液へのホ
ルマリン供給は57時間継続し、反応液に供給した
ホルマリンの総量は25mlであつた。反応液中には
28gのL−セリンが蓄積し、L−セリンの蓄積濃
度は53g/、生成したL−セリンの反応液に添
加したグリシンに対する収率はモル比で80%、反
応液に添加したホルムアルデヒドに対する収率は
モル比で80%であつた。
本発明に用いる微生物がグリシンよりL−セリ
ンを産生する能力につき、ホルムアルデヒドの存
在下と不存在下で比較した結果は次の実験例に示
す通りである。
実験例
エシエリヒア・コリMT−10350およびMT−
10351をそれぞれ肉エキス10g/、ペプトン10
g/、NaCl5g/を含む液体培地(PH7.0)
100mlに接種し、30℃で24時間振とう培養を行な
つた。培養終了菌体を遠心分離し、生理食塩水で
2度洗浄し湿菌体約0.4gを得た。この湿菌体0.2
gをグリシン5000μmoleホルムアルデヒド
150μmole、テトラヒドロ葉酸10μmole、ピリド
キサルリン酸0.1μmoleを含む50mMリン酸緩衝液
(PH7.0)10mlに懸濁し、37℃で2時間振とうしな
がら反応させた。
第3表に示すようにL−セリンの蓄積がみられ
た。
一方上記と全く同様の操作でそのうちホルムア
ルデヒドの添加のみを行わずに行なつた実験の結
果も第3表のとうりであつた。
