Description
本発明は非環式アミンを用いるエポキシステロ
イドの製法に関する。
発明の背景
Δ9(11)−ステロイドを対応するブロモヒドリン
(9α−ブロモ−11β−ヒドロキシステロイド)、さ
らに、対応する9β,11β−エポキシステロイド、
ついで最後にフツ化水素と反応させて所望の9α
−フルオロ−11β−ヒドロキシステロイドに変換
させることはよく知られている。メタノール中、
メトキシド、t−ブトキシドまたは水酸化ナトリ
ウムのような強塩基(米国特許第2838493号およ
び第3725392号)、アセトン中酢酸カリウム〔米国
特許第3980778号およびJ.Am.Chem.Soc.,
Vol.77,p4438(1955)〕、特定の溶媒中のフツ化
カリウム(米国特許第3255185号)およびDBUの
ような環式アミジン塩基(米国特許第4207235号)
を含み、種々の試薬が該エポキシ化反応に用いら
れてきた。
該9α−ブロモ−11β−ヒドロキシブロモヒドリ
ンの対応する9β,11β−エポキシドへの閉環は脱
ハロゲン化水素反応である。エイチ・エデイガー
ら〔H.Oediger,et al,Synthesis 591(1972)〕
は、種々の有機塩基による塩化水素の脱離につい
て比較している。その592頁の第1表は、環式お
よび非環式アミンならびに非環式アミジンが脱ハ
ロゲン化水素反応においてDBNと同等には作用
しないことを示している。そのため、非環式アミ
ジンは、DBNまたはDBUと同等には、9α−ブロ
モ−11β−ヒドロキシブロモヒドリンを対応する
9β,11β−エポキシドに変換しないと考えられて
いる。
デイ・エイチ・アール・バートン〔D.H.R.
Barton,J.C.S.Chem.Comm.1136,(1981)〕は強
有機塩基である多くのグアニジンの製法について
報告している。しかし、バートンはこれらの塩基
がブロモヒドリンの対応するエポキシドへの変換
のような脱ハロゲン化水素反応において有用であ
ることを報告していない。
本発明は、9α−置換−11β−ヒドロキシステロ
イド()(後記チヤート参照、以下同じ)の対
応する9β,11β−エポキシド()への変換に、
驚くべきことに、また、予期せぬことに、効率よ
く効果を及ぼす非環式のグアニジンおよびアミジ
ンを包含するものである。該グアニジン()お
よびアミジン()はDBUおよびDBNのような
多の公知の脱ハロゲン化水素試薬よりも安く、製
造も容易であり、かつ、大部分の他の塩基のよう
に21−アシルオキシ基を開裂しない。
本発明の概要
本発明は、式()の9α−置換−11β−ヒドロ
キシステロイドを式()のグアニジンおよび式
()のアミジンからなる群より選ばれる非環式
アミンと接触させ、エポキシド()を単離する
ことからなる式()のエポキシドの製法を提供
するものである。
本発明の詳細な説明
該9α−置換−11β−ヒドロキシステロイド
()出発物質は当該分野においてよく知られて
いるか、当該分野に公知の化合物から公知の方法
で容易に製造することができる。
9α−置換−11β−ヒドロキシステロイド()
をエポキシド()へ変換させる本発明方法は、
広範なステロイドA−環に対して実施することが
できる。Δ4−3−ケト(A)、Δ1,4−3−ケト(B)お
よび3β−ヒドロキシ−Δ5(C)のステロイドA環が
好ましい(チヤートB参照)。
C−6位上のフツ素原子またはメチル基は、本
発明の方法において何ら妨げになるものではな
い。脱離基R9は、塩素、臭素またはヨウ素、あ
るいはトシレート、メシレートまたはトリフラー
トエステルとしてのヒドロキシである。好ましく
は、R9は臭素である。R16は水素、ヒドロキシ、
炭素数2〜6のアシルオキシまたはメチルであ
る。R16がヒドロキシル基で、C−17位にヒドロ
キシが存在するとき、これらのヒドロキシはアセ
トニドの形態で存在し得る。R17は水素または−
COR17αで、R17αは炭素数1〜6のアルキルまた
はフエニル基である。R17が水素およびR16がヒ
ドロキシの場合、該ヒドロキシはアセトニドの形
態で存在し得る。R21は炭素数1〜6のアルキ
ル、フエニルあるいは、p−ニトロまたはm−メ
トキシ基で置換されたフエニルである。本発明方
法は、Δ4−3−ケトステロイドおよびΔ1,4−3
−ケトステロイドに等しく適用できる。好ましい
ステロイドD環をチヤートCに式(D)として示す。
非環式アミジン()は当該分野において公知
であるか、当該分野に公知の方法によつて公知の
化合物から容易に製造することができる。〔J.
Am.Chem.Soc.Vol.102,p7125(1980)およびM.
Bredereck et al,Chem.Ber.,96,1350(1963)
参照〕Raは炭素数1〜7のアルキルである。好
ましくは、Raはt−ブチルである。また、R′3お
よびR′4は、同一または異なつて、炭素数1〜7
のアルキルである。R′3およびR′4はいずれもメチ
ルであるのが好ましい。
式()のグアニジンは当該分野によく知られ
ているか、当該分野によく知られている方法によ
つて公知の化合物から容易に製造することができ
る(前記バートンら参照)。Rgは水素である。
R1、R2、R3およびR4は、同一または異なつて、
水素または炭素数1〜7のアルキルである。好ま
しくは、R1、R2、R3およびR4は全てメチルまた
は水素である。
後記の第1表および第2表には本発明の方法で
用いるグアニジン()およびアミジン()を
示してあり、そのうちのいくつかは、特に好まし
いものである。第1表および第2表中の評価の符
号については、−よりも+が優れ、+よりも++が
優れ、++よりも+++が優れ、+++よりも++
++が優れていることを意味する。従つて、第1
表の最初の2つのグアニジン()が最も優れて
いる。
9α−置換−11β−ヒドロキシステロイド()
を塩化メチレン、アセトン、酢酸エチル、
DMSO、DMF、THF、ジオキサン、アセトニト
リル、トルエンおよびクロロホルムからなる群よ
り選ばれる中性極性有機溶媒中で非環式グアニジ
ン()または非環式アミジン()のいずれか
と接触させる。好ましい中性極性有機溶媒は
DMFである。
ステロイド1当量に対して1〜2当量のグアニ
ジン()またはアミジン()を使用する。好
しくは、ステロイド1当量に対して約1.1当量の
グアニジン()またはアミジン()を用い
る。
該反応は、必要ではないが、窒素雰囲気下で行
なうのが好ましい。
該反応は、使用するグアニジン()またはア
ミジン()の性質に応じて0〜100℃の温度範
囲内で行なう。非環式アミンがグアニジン()
であるとき、該反応は、一般に、0〜40℃の温度
範囲内、好ましくは、25〜35℃で行なう。非環式
アミンがアミジン()であるとき、該反応は、
一般に、70〜100℃の温度範囲内、好ましくは、
80〜90℃で行なう。該反応は密封容器内で加熱し
て行なうことができ、これにより反応時間を短縮
させる。一般に、該反応は、用いる有機溶媒の還
流温度にて行なわれる。
反応をTLCによつてモニターする。一般的に、
グアニジン()では約1時間、また、アミジン
()では約4〜5時間で反応が完了することを
示す。
TLCが反応の完了を示したら、反応混合液を、
塩酸、酢酸、硫酸、リン酸のような酸またはp−
TSAを含有する水で希釈する。所望のエポキシ
ド()を、当該分野によく知られた方法によつ
て反応溶媒より抽出するか、または水析出および
過によつて単離する。
エポキシド()は、当該分野でよく知られて
いるように、該エポキシドをフツ化水素と反応さ
せ、対応する9α−フルオロ−11β−ヒドロキシス
テロイドへ変換するのに有用である。フリードら
〔Fried,et al.,J.Am.Chem.Soc.,Vol.76、
p1455(1954)〕;ヒルシユマンら〔Hirschmann
et al.,J.Am.Chem.Soc.,4957(1956)〕;米国特
許第2894007号、第3049556号、第3086032号、第
3007923号、第3211758号および3980778号参照。
かくして、本発明方法によれば、DBUおよび
DBNよりも安価で容易に製造される塩基を利用
して、9α−置換−11β−ヒドロキシ21−エステル
を対応する9β,11β−エポキシド21−エステル
()へ、該21エステル基を開裂させずに変換す
ることができる。
定 義
つぎに、本明細書で用いる略号、記号の定義を
示す。
THFとはテトラヒドロフランのことである。
DMSOとはジメチルスルホキシドのことであ
る。
DMFとは、ジメチルホルムアミドのことであ
る。
p−TSAとはp−トルエンスルホン酸の一水
塩のことである。
混合溶媒を用いる場合、その溶媒の比率は容
量/容量(V/V)である。
DBUとは1,8−ジアザビンクロ〔5,4,
0〕ウンデセン−7のことである。
DBNとは1,5−ジアザビシクロ〔5,4,
0〕ノネン−5のことである。
炭素数を示したアルキル、アシル等には異性体
も包含する。
Raは炭素数1〜7のアルキルである。
Rgは水素である。
R1、R2、R3およびR4は水素または炭素数1〜
7のアルキルである。
R′3およびR′4は炭素数1〜7のアルキルであ
る。
R6は水素、フツ素、またはメチルである。
R9は塩素、臭素、ヨウ素またはトシレート、
メシレート、またはトリフレートエステルとして
のヒドロキシである。
R16は水素、ヒドロキシ、アシルオキシ(該ア
シル基の炭素数は2〜6)またはメチル基で、た
だし、R16がヒドロキシおよびR17が水素のとき
はC−16位およびC−17位におけるヒドロキシは
アセトニドの形態で存在してもよい。
R17は水素または−COR17〓で、ただしR17が水
素およびR16がヒドロキシのときはC−16位およ
びC−17位におけるヒドロキシはアセトニドの形
態で存在してもよい。
R17〓は炭素数1〜6のアルキルまたはフエニル
である。
R21は炭素数1〜6のアルキル、フエニルある
いはp−ニトロまたはm−メトキシ基で置換され
たフエニルである。
「〜」は結合した原子または基がαまたはβ配

実施例

ジオン21−酢酸エステル2.406g〔()、
JACS77,4438、(1955)〕を12℃の水浴中、窒素
雰囲気下で撹拌する。テトラメチルグアニジン
0.656mlを該混合液に加え、一夜撹拌する。1N塩
酸1.0mlを含有する水20mlを撹拌しながら加える。
この水/塩酸添加の間、該混合液を氷水浴中で冷
却する。水13mlを該混合液に−10℃にて撹拌しな
がら徐々に加える。混合液を6℃で20分間撹拌
し、固体を取し、水で3回洗滌し、ついで、50
〜60℃にて3.5時間減圧乾燥して標記化合物を得
る。
実施例 2
9α,11β−エポキシ−17α,21−ジヒドロキシ
プレグナ−4−エン−3,20−ジオン21−酢酸エ
ステル()
9β−ブロモ−11β,17α,21−トリヒドロキシ
プレグナ−4−エン−3,20−ジオン21−酢酸エ
ステル2.00g〔()、JACS77,4438(1955)〕を
DMF5mlに溶解する。テトラメチルグアニジン
0.475gを加え、該混合液を20〜25℃で撹拌する。
TLCが反応の完了を示したとき、10%塩酸を含
有する水を加え、該混合液を撹拌し、固体を取
し、洗浄し、ついで減圧乾燥して標記化合物を得
る。
実施例 3
9α,11β−エポキシ−17α,21−ジヒドロキシ
−16β−メチルプレグナ−1,4−ジエン−3,
20−ジオン−安息香酸エステル()
9β−ブロモ−11β,17α,21−トリヒドロキシ
−16β−メチルプレグナ−1,4−ジエン−3,
20−ジオン21−安息香酸エステル2.0g〔()、
米国特許第3164618号、実施例30〕を塩化メチレ
ン10ml中でスラリー化する。t−ブチルジメチル
アミジン0.86gを加え、該混合液を加熱還流す
る。6時間後、TLCは反応が90%完了したこと
を示す。該有機溶媒を酢酸エチルで置き換え、該
混合液を過し、固体を減圧下で乾燥させて標記
化合物を得る。

5ml中でスラリー化する。t−ブチルジメチルア
ミジン0.43mlを加え、該混合液を一夜撹拌する。

液を過し、ついで固体を減圧下で乾燥して標記
化合物を得る。

ステル()4.0gおよびDMF20mlを5℃で混合
する。DBU1.49ml(9.94m mole)を加える。
15分後、10%塩酸水溶液を含む水50mlを加え、該
混合液を撹拌し、過し、固体を水洗し、ついで
乾燥して9β,11β−エポキシ−17α,21−ジヒド
ロキシ−プレグナ−4−エン−3,20−酢酸エス
テル()を収率93.2%で得る。
