Description
【発明の詳細な説明】
本発明は式()
で表わされるラセミまたは光学活性−スレオ−N
−カルボベンゾキシ−3−(3,4−メチレンジ
オキシフエニル)セリンに関する。
本発明によれば式()
で表わされるラセミ−スレオ−3−(3,4−メ
チレンジオキシフエニル)セリンをカルボベンゾ
キシクロリドと反応させることにより式()で
表わされるラセミ−N−カルボベンゾキシ−スレ
オ−3−(3,4メチレンジオキシフエニル)セ
リンを得ることができ、必要に応じ、続いてエフ
エドリン、キニジン、キニン、2−アミノ−1,
1−ジフエニルプロパノールから選ばれる光学活
性アミンの1つを作用させてDまたはL−スレオ
−N−カルボベンゾキシ−3−(3,4−メチレ
ンジオキシフエニル)セリンのアミン塩とし、溶
解度の差を利用してD−スレオ−N−カルボベン
ゾキシ−3−(3,4−メチレンジオキシフエニ
ル)セリンのアミン塩とL−スレオ−N−カルボ
ベンゾキシ−3−(3,4−メチレンジオキシフ
エニル)セリンのアミン塩とに分別し、しかる後
にそれぞれの塩に酸を作用させることにより、光
学活性(DまたはL)−スレオ−N−カルボベン
ゾキシ−3−(3,4−メチレンジオキシフエニ
ル)セリンを得ることができる。
本発明化合物である式()で表わされるラセ
ミまたは光学活性−スレオ−N−カルボベンゾキ
シ−3−(3,4−メチレンジオキシフエニル)
セリンは文献未記載の新規化合物であり、抗菌活
性を有し、それ自体医薬品として有用であると共
に末梢起立性低血圧症の治療剤(特開昭56−
104815号公報)あるいは抗うつ剤(特開昭55−
20747号公報)として有用であるラセミまたは光
学活性−スレオ−3−(3,4−ジヒドロキシフ
エニル)セリン(以下DOPSと略称する)の製造
上の中間体として極めて有用な化合物である。
従来、ラセミまたは光学活性−DOPSの製造方
法としては、式()
で表わされるプロトカテキユアルデヒドのカテコ
ール部分が2つのメチル基またはメチレン基で保
護されたバニリンまたはピペロナールを出発原料
として用い、メチル基またはメチレン基を除去す
ることによつて上記式()で表わされるプロト
カテキユアルデヒドを得た後、改めてカテコール
部分をエトキシカルボニル基またはベンジル基で
保護して式()
〔式中、R1はエトキシカルボニル基またはベ
ンジル基を意味する。〕
で表わされるベンズアルデヒド誘導体とし、この
ベンズアルデヒド誘導体をグリシンまたはグリシ
ン誘導体と縮合させて、スレオおよびエリスロ−
3−(3,4−ジヒドロキシフエニル)セリン誘
導体の混合物を得、必要に応じた光学分割操作を
経て保護基を除去することにより、ラセミまたは
光学活性−DOPSを製造するという方法が用いら

すなわちDOPSを製造するにあたり、原料化合
物となるベンズアルデヒド誘導体のカテコール部
分の保護基の変更という煩雑な操作を必要とする
欠点がある。
かかる情況下、本発明者らはカテコール部分の
保護基の変更を必要としないラセミ及び光学活性
−DOPSの製造方法を鋭意検討した。その結果ピ
ペロナールから保護基を変更することなく製造す
ることができる、本発明化合物である文献未記載
なラセミ及び光学活性−スレオ−N−カルボベン
ゾキシ−3−(3,4−メチレンジオキシフエニ
ル)セリン()を中間体として用いることによ
り、容易にラセミ及び光学活性−DOPSを製造で
きるという知見を得、本発明を完全した。
即ち本発明化合物であるラセミ及び光学活性−
スレオ−N−カルボベンゾキシ−3−(3,4−
メチレンジオキシフエニル)セリン()を塩化
アルミニウム等のルイス酸で処理することにより
ラセミ及び光学活性−スレオ−N−カルボベンゾ
キシ−3−(3,4−ジヒドロキシフエニル)セ
リンとした後、パラジウム・カーボン等を触媒と
する接触還元反応によりラセミまたは光学活性−
DOPSを得ることができる。
従つて、本発明化合物であるラセミ及び光学活
性−スレオ−N−カルボベンゾキシ−3−(3,
4−メチレンジオキシフエニル)セリン()
は、ラセミ及び光学活性−DOPSを経済的に製造
する上で重要な中間体となるものである。
以下に本発明を具体的に説明する。
ラセミ−スレオ−N−カルボベンゾキシ−3−
(3,4−メチレンジオキシフエニル)セリン
()は、ラセミ−スレオ−3−(3,4−メチレ
ンジオキシフエニル)セリン()とカルボベン
ゾキシクロリドとのいわゆるSchotten Baumann
反応により得ることができる。
即ち、ラセミ−スレオ−3−(3,4−メチレ
ンジオキシフエニル)セリン()をPH7以上の
アルカリ水溶液に溶解し、カルボベンゾキシクロ
リドを加えることにより実施することができる。
カルボベンゾキシクロリドはラセミ−スレオ−
3−(3,4−メチレンジオキシフエニル)セリ
ン()に対し1〜2倍モル使用し、反応温度は
0〜30℃にて数分〜20時間反応させれば良く、反
応中、反応液のPHを7〜10に保つ事が好ましい。
反応液のPHを7〜10に保つには、PHの変化に応
じアルカリ水を滴下するかあるいは反応液にあら
かじめ炭酸水素ナトリウム、ホウ砂等、水溶液と
した場合にPH7〜10である塩を加えておく。
アルカリ水として用いるアルカリとしては、炭
酸水素ナトリウム、炭酸ナトリウム、炭酸カリウ
ム、水酸化ナトリウム、水酸化カリウム等を挙げ
ることができる。
反応は水溶液で充分進行するが、トルエン、ベ
ンゼン等の芳香族炭化水素系溶媒、ヘキサン、ヘ
プタン等の脂肪族炭化水素系溶媒、ジエチルエー
テル、テトラヒドロフラン、ジオキサン等のエー
テル系溶媒、アセトン、メチルエチルケトン等の
ケトン系溶媒、クロロホルム、ジクロロメタン、
ジクロロエタン等のハロゲン化アルキル系溶媒、
酢酸エチル、酢酸ブチル等のエステル系溶媒及び
これらの混合物を添加する事もできる。
光学活性(DあるいはL)−スレオ−N−カル
ボベンゾキシ−3−(3,4−メチレンジオキシ
フエニル)セリン()を得るには、ラセミ−ス
レオ−N−カルボベンゾキシフエニル)セリン
を、エフエドリン、キニジン、キニン、2−アミ
ノ−1,1−ジフエニルプロパノールから選ばれ
る光学活性アミンの1つを用いて適当な溶媒中で
反応させてDおよびL−スレオ−N−カルボベン
ゾキシ−3−(3,4−メチレンジオキシフエニ
ル)セリンと光学活性アミンとの塩とし、溶解度
の差を利用してD−スレオ−N−カルボベンゾキ
シ−3−(3,4−メチレンジオキシフエニル)
セリンの光学活性アミン塩と、L−スレオ−N−
カルボベンゾキシ−3−(3,4−メチレンジオ
キシフエニル)セリンの光学活性アミン塩とに分
別し、しかる後にそれぞれの塩に酸を作用させて
塩を分解する方法が行われる。
塩を形成し分別する温度としては0〜80℃で行
う事ができるが、溶媒の沸点付近まで加温した後
0〜30℃まで冷却する事もできる。塩を形成する
時間は数分間で充分であるが、数時間をかけても
良く特に制限はない。
光学活性アミンはラセミ−スレオ−N−カルボ
ベンゾキシ−3−(3,4−メチレンジオキシフ
エニル)セリンに対し0.5〜1倍モル用いて実施
する事ができる。
上記塩の形成、分別に用いる溶媒としては、メ
タノール、エタノール、2−プロパノール等のア
ルコール系溶媒、テトラヒドロフラン、ジオキサ
ン等のエーテル系溶媒、アセトニトリル、水及び
これらの混合溶媒を好ましい例として挙げること
ができる。
得られた光学活性(DあるいはL)−スレオ−
N−カルボベンゾキシ−3−(3,4−メチレン
ジオキシフエニル)セリンの光学活性アミンとの
塩に酸性水溶液を加えることにより、塩を分解
し、有機溶媒により抽出することにより光学活性
(DあるいはL)−スレオ−N−カルボベンゾキシ
−3−(3,4−メチレンジオキシフエニル)セ
リンを得ることができる。
この酸性水溶液の酸としては塩酸、硫酸、リン
酸等の鉱酸が挙げられ、その使用量は塩に対し1
〜10倍モル用いることができる。
抽出に用いる有機溶媒としては、酢酸エチル、
クロロホルム、ジクロロエタン、ジクロロメタ
ン、ジエチルエーテル等を挙げることができる。
本発明において原料となるラセミ−スレオ−3
−(3,4−メチレンジオキシフエニル)セリン
()は立体構造(スレオ、エリスロ)の不明な
ものとしては既に知られているが〔薬学雑誌,
67、218(1947),Can.J.Chem.,42、1901
(1964)〕、スレオ体としては本発明者等により初
めて単離、同定されたものである。すなわちグリ
シンとグリシンに対し2倍モルのピペロナールと
2倍モルの水酸化ナトリウム、水酸化カリウム等
の塩基とを反応させた後、反応液に酢酸水を加え
る事によりラセミ−スレオ/エリスロー3−(3,
4−メチレンジオキシフエニル)セリン・酢酸塩
を得、この塩を水から再結晶するかあるいは水に
懸濁後取することにより、ラセミ−スレオ−3
−(3,4−メチレンジオキシフエニル)セリン
()を得ることができる。
次に実施例により本発明方法を説明する。本発
明はもとよりこれに限定されるものではない。
参考例
水酸化カリウム61.1g、メタノール1055mlを
撹拌下溶解し、それにグリシン33.05g加え溶
解した。さらにピペロナール145gを加え30分
間62℃〜65℃で撹拌した。反応後減圧下濃縮し
た。残渣にメタノール315ml、次いで酢酸308g
を加え30分間40℃〜45℃で撹拌した。さらに水
135g、トルエン1000gを加え2時間40℃〜45
℃で撹拌後、更に1時間0〜5℃で撹拌した。
析出した結晶を取し、乾燥するとスレオ/エ
リスロ−3−(3,4−メチレンジオキシフエ
ニル)セリン・酢酸塩87.0gが得られた(収率
69.3%)。
融点は162℃(分解点)であり、下記条件で
のHPLC分析結果はスレオ:エリスロ=85:15
の比率であつた。

又、上記塩を取した母液からトルエン層を
分液し、濃縮することによりピペロナール88g
を回収することができた。
−1 水1105gにスレオ/エリスロ−3−(3,
4−メチレンジオキシフエニル)セリン・酢
酸塩85gを加え加熱撹拌下還流し、溶解させ
た後冷却し、1時間0〜5℃で撹拌した。析
出した結晶を取し、乾燥するとスレオ−3
−(3,4−メチレンジオキシフエニル)セ
リン48.37gが得られた。(収率72.1%)。融
点は193℃(分解点)であり、前記HPLC条
件での分析結果はスレオ体100%であつた。
−2 水20gにスレオ/エリスロ−3−(3,
4−メチレンジオキシフエニル)セリン・酢
酸塩1gを加え室温にて30分撹拌した。析出
した結晶を取し、乾燥するとスレオ−3−
(3,4−メチレンジオキシフエニル)セリ
ン0.45gが得られた(収率57・0%)。融点
は188℃(分解点)であり、前記HPLC条件
での分析結果はスレオ:エリスロ=98.7:
1.3であつた。
実施例 1
ラセミ−スレオ−3−(3,4−メチレンジオ
キシフエニル)セリン11.3gを5℃以下で水酸化
ナトリウム4.0gを含む水溶液160mlに加え、溶解
後5℃以下でカルボベンゾキシクロリド9.4gを
滴下した。同時に30%水酸化ナトリウムを滴下
し、PH8.5〜9.5になる様に調節した。2時間後、
濃塩酸水にてPH≒2として酢酸エチルにて抽出
し、有機層を飽和食塩水で洗浄後、硫酸マグネシ
ウムで乾燥し溶媒を減圧下留去した。残渣をトル
エンにて結晶化し、結晶を取することによりラ
セミ−スレオ−N−カルボベンゾキシ−3−(3,
4−メチレンジオキシフエニル)セリン17.3gを
得た。

実施例 2
実施例1におけるカルボベンゾキシクロリド
9.4gに代えて、カルボベンゾキシクロリド9.4g
とトルエン22gとの溶液を用い、実施例1と全く
同様に反応を行つた。反応後PH≒9.0に調整して
トルエン100mlを加え、トルエン抽出を行つた。
水層を濃塩酸にてPH≒2とし、実施例1と同様に
抽出、水洗、乾燥、濃縮を行い、トルエンにて結
晶化して、ラセミ−スレオ−N−カルボベンゾキ
シ−3−(3,4−メチレンジオキシフエニル)
セリン16.9gを得た。

ラセミ−スレオ−N−カルボベンゾキシ−3−
(3,4−メチレンジオキシフエニル)セリン
10.0gをアセトニトリル100mlに溶解し、キニジ
ン9.02gを加え均一な溶液とした。氷水にて5時
間冷却し、析出晶を取してL−スレオ−N−カ
ルボベンゾキシ−3−(3,4−メチレンジオキ
シフエニル)セリン・キニジン塩9.0gを得た。

上記塩の一部2.0gに3%塩酸水50mlを加えた
後、酢酸エチルにて抽出して、L−スレオ−N−
カルボベンゾキシ−3(3,4−メチレンジオキ
シフエニル)セリン0.98gを無定形粉末として得
た。

10.0gに2−プロパノール100mlとキニジン9.02
gを加え均一な溶液とした。
室温にて一夜放置後析出晶を取し、L−スレ
オ−N−カルボベンゾキシ−3−(3,4−メチ
レンジオキシフエニル)セリン・キニジン塩8.3
gを得た。

ラセミ−スレオ−N−カルボベンゾキシ−3−

gをアセトニトリル50mlに溶解後、キニン4.5g
を加え均一な溶液とした。
室温にて一夜放置後、析出晶を取してD−ス
レオ−N−カルボベンゾキシ−3−(3,4−メ
チレンジオキシフエニル)セリン・キニン塩5.1
gを得た。

−1,1−ジフエニルプロパノール塩7.8gを得
