Abstract

PURPOSE:To provide the title inoculation method so designed that fungi are applied and inoculated on the surface of a culture medium using an inoculating tool with the tip of a substrate fitted with fiber, thereby markedly reducing the damage of the surface and enabling fungus inoculation of numerous generating colonies to be accomplished. CONSTITUTION:Natural or synthetic fiber such as cotton is wound, fixed with an adhesive or detachably fitted on the tip of a rod-like substrate made of e.g., wood, bamboo, synthetic resin, metal to obtain a tool for fungus inoculation. Using this tool, fungi are applied and inoculated on the surface of a culture medium in a similar way to the conventional method, pref. with the surface of the culture medium in a laboratory dish oriented downward.

(Conventional technology and its issues) Traditionally, in medical and pharmaceutical research and development research in the fermentation industry, bacteria have been planted and cultured in culture media. Bacteria have been applied to the surface of an agar medium using a platinum loop or a wooden toothpick.

However, in this conventional inoculation method using platinum loops and wooden toothpicks, these inoculation tools made of platinum loops and wooden toothpicks tend to damage the surface of the agar medium, making it impossible to separate colonies and forming colonies. The disadvantage was that there were only a few.

For example, as shown in Figure 2, the platinum loop for inoculation consists of a grip made of ebonite handle (A) and a platinum wire at the end (Currently, nichrome wire for electric heating is used) (B) The tip of this platinum wire (A) is a loop-shaped part (
It is known what forms the This platinum loop is essential when applying and inoculating bacteria onto the surface of an agar medium. However, this loop-shaped part
Even in the case of using a platinum loop having the same properties, it is unavoidable to damage the surface of the culture medium when applying it to the surface.

This invention was made in consideration of the above-mentioned circumstances, and it overcomes the drawbacks of conventional inoculation tools such as platinum loops and wooden toothpicks, and the inoculation methods using these, and damages the surface of the culture medium. The purpose of the present invention is to provide a new inoculation tool and an inoculation method using the same, which enable inoculation with a large number of colonies without the need for inoculation.

(Function) The inoculation tool of the present invention has M fibers attached to the tip of the support, so it does not have a hard tip like conventional platinum loops or wooden toothpicks, so it can be used to inoculate the culture medium surface with a soft touch. Can be bacterium. Moreover, when the culture medium surface is directed downward, unnecessary force is not applied to the contact between the culture medium surface and the fibers, and therefore the culture medium surface is not damaged.
Colony separation is good and large numbers of colonies can be produced.
