Description
技術分野
本発明はヒトアポリポ蛋白質B48に対する単ク
ローン性抗体に関するものである。
従来技術
アポリポ蛋白質Bは、ヒト血漿トリグリセリド
リツチリポ蛋白質の合成および分泌に重要な役割
をもつている蛋白質である。このアポリポ蛋白質
Bには、肝で産生されVLDL(超低比重リポ蛋白
質)の合成に関与するアポリポ蛋白質B100(以
下、アポB100と略称する)と、小腸で産生され
カイロミクロン合成に関与するアポリポ蛋白質
B48(以下、アポB48と略称する)がある。アポ
リポ蛋白質Bの遺伝的欠損による疾患である無β
リポ蛋白血症には、アポB100とアポB48が共に
欠損する無βリポ蛋白血症古典型と、アポB100
のみの欠損するトリグリセリド正常型無βリポ蛋
白血症がある。そのいずれもが、赤血球異常、精
神神経症状など重篤な症状を呈するが、トリグリ
セリド正常型無βリポ蛋白血症ではカイロミクロ
ンが合成されうる。またアポリポ蛋白質Bを含む
リポ蛋白質は動脈硬化を惹起する性質を有すると
考えられ、アポB100とアポB48が異なる代謝経
路を有するため、動脈硬化の発症機序を知る上で
も両者の構造を知ることは重要である。
上記の如く、アポリポ蛋白質Bは、様々な機能
を有するにもかかわらず、難溶性であり、分解し
やすい高分子蛋白質から成るため、その研究は今
だ詳細になされていない。この研究のためには、
アポリポ蛋白質Bの各構成部分を特異的に認識す
る必要がある。
発明の目的
このために、本発明は、アポB48の精製手段を
確立し、精製したアポB48に対する単クローン性
抗体(以下、抗アポB48−と略称する)を得る
ことを目的とする。
発明の構成
本発明の抗アポB48−を産生するハイブリド
ーマは、アポB48で予め免疫されたマウスの脾細
胞と、マウスの骨髄腫細胞ラインからの細胞の融
合によつて形成される。このような本発明の抗ア
ポB48−およびこの抗体を産生するハイブリド
ーマは、例えば、次の4行程によつて作成するこ
とができる。1.アポB48の精製、2.免疫、3.細胞
融合、4.ハイブリドーマの選択および単クローン
化。
このようにして作成された抗アポB48−1は、
2元拡散法の結果、IgG1aサブクラスに属するこ
とがわかつた。トランス・ブロツテイング法によ
り検討すると、抗アポB48−は、アポB100お
よびアポB48に結合するが、アルブミン、アポリ
ポ蛋白質Eその他のアポリポ蛋白質とは結合しえ
ない。
更にこの抗体を用いて免疫酵素抗体の測定法を
開発した。この方法として、測定したいサンプル
をポリスチレン96穴プレートへ付着させる間接法
と、プレートにあらかじめウサギ抗アポBポリク
ローナル抗体を付着させておき、これにサンプル
を結合させるサンドイツチ法を用いた。本発明の
アポB48−を用いると、比較的よい精度で、ヒ
ト血中アポリポ蛋白質Bの定量が可能である。
前に述べた無βリポ蛋白血症の患者には、本邦
ではアポB100の単独欠損であるトリグリセリド
正常型(名古屋家系)と、アポB100とアポB48
が共に欠損する古典型(神奈川家系)とが知られ
ている。トランスブロツテイング法および免疫酵
素抗体法を用いると、名古屋家系患者のリポ蛋白
質およびアポリポ蛋白質Bは本発明の抗アポB48
−と結合しえたが、神奈川家系患者のリポ蛋白
質およびアポリポ蛋白質は結合を示さなかつた。
このことは抗アポB48−が、アポリポ蛋白質B
以外のいかなるアポリポ蛋白質とも結合できない
ことを支持している。
以上まとめると、本発明の抗アポB48−は、
(1)正常者人血中、アポB48と反応する、(2)アポリ
ポ蛋白質B以外のアポリポ蛋白質とは反応しえな
い、(3)TG正常型無βリポ蛋白血症患者の血中リ
ポ蛋白質とは反応しうる各性質を有することがわ
かつた。
次に、本発明の抗アポB48−およびこの抗体
を産成するハイブリドーマを作成するための好適
例を説明する。
まず、アポB48を精製する。このためには、カ
イロミクロンを多量必要とするため、乳び腹水患
者の乳び腹水、V型高脂血症患者の血漿または正
常人の脂肪食後血漿を用いる。サンプルが得られ
たならば、EDTA、NaN3、カナマイシンを加え
て処理した後、超遠心法にて分離してVLDL+カ
イロミクロン分画を得る。さらに、このVLDL+
カイロミクロン分画から、超遠心法でカイロミク
ロン分画を得る。こうして得られたカイロミクロ
ン分画を有機溶媒(例えば、エーテルとエタノー
ル、テトラメチルウレアまたはクロロホルムとメ
タノール)にて脱脂し、SDS(ドデシル硫酸ナト
リウム)を含む溶液に溶解する。この溶液をウオ
ータージヤケツトで加温したセフアロース4B・
CLカラムを用いてSDS溶液でゲル過する。こ
の工程を2日以内に終える場合、アポB48の分解
が少なく、ほぼ純粋なアポB48を得ることができ
る。
このようにして精製したアポB48を用いると、
免疫が良好に行われた。すなわち、アポB48をマ
ウスに投与し、免疫されたマウスの脾細胞を取り
出す。このマウスの脾細胞とマウスの骨髄腫
(NS−1)からの細胞とを融合させて本発明のハ
イブリドーマを得る。このハイブリドーマの作成
は、例えばオーイらの方法を用いることができる
(SELECTED METHODS IN CELLULAR
IMMUNOLOGY、351−372、Freeman、1980)。
得られたハイブリドーマから、本発明の抗アポ
B48−を作成するには、例えば次の2方法を用
いることができる。
1の方法は、ハイブリドーマを適当な培養液中
(in vitro)で培養し、その上澄みに生成された
抗体を回収する方法である。他の方法は、ハイブ
リドーマをマウスに注射し(in vivo)生体内で
インキユベートし、マウスの腹水中に生成された
抗体を回収する方法である。前者の方法では、抗
体価が低いが純度の良いものが得られ、後者の方
法では純度はやや劣るが非常に抗体価の高いもの
が得られる。どちらの方法を選択するかは目的に
よつて使いわけられる。
以下、本発明を実施例に基づきさらに詳細に説
明する。
実施例 1
この実施例では、アポB48を精製する。
カイロミクロンの精製は種々のサンプルを用い
て行われるが、ここでは乳び腹水から精製する。
乳び腹水約2を採取後、直ちに1mM
EDTA、0.02%NaN3を加えて調製し、ベツクマ
ンSW・27ローターで27000rpm、20時間超遠心
し、カイロミクロン+VLDL分画を得る。更に、
この分画をベツクマン40.3ローターで20000rpm、
30分間超遠心し、浮上したカイロミクロン分画を
回収する。このカイロミクロン分画を、20倍量の
エーテルとエタノール(1:3vol/vol)溶液中
にて10時間脱脂する。沈澱物は、2000rpm、5分
間遠心して回収し、約10c.c.の10%SDSと1%2−
メルカプトエタノール溶液に溶解する。このうち
5c.c.を、50℃にウオータージヤケツトした2cm×
100cmのセフアロース4B・CLカラムに通してゲ
ル過する。その際に、流出液として0.5%SDS
および0.01MトリスHClを0.15M NaClでPH7.2に
調整した緩衝液を流速20ml/時で使用した。その
結果、ほぼ純粋なアポB48が得られる。脱脂はほ
ぼ完全である。
実施例 2
この実施例では、本発明の単クローン性抗体
(抗アポB48−)、およびそれを産生するための
ハイブリドーマを作成する。
1 免疫
精製したSDS溶液中のアポB48を100μg用い
て、フロインド完全アジユバンドと共に雄のバ
ルブ/cマウス(6週令)に皮下注射した。3
週間後に再度、前記アポB48の10μgを前記ア
ジユバンドと共に腹腔内へ注射した。
2 細胞融合
2回目の免疫から3日後に、マウスの脾臓を
取り出し、脾細胞の懸濁液とする。この脾細胞
1×108個を、3×107個の8−アザグアニン耐
性骨髄細胞腫NS−1とポリエチレングライコ
ール#4000を用いて融合した。細胞は96穴マイ
クロ培養プレート5枚に分配した。
24時間後、上澄みの半分をHAT培地(ヒポ
キサンチン1×10-4M、アミノプテリン4×
10-7M、チミジン1.6×10-3M)に置きかえた。
HAT耐性細胞(ハイブリドーマ)が2〜3週
間後に大半の培地に増殖するのが観察される。
3 ハイブリドーマの選択および単クローン化
マイクロプレート中の培養液上澄み10μ
を、脂肪食後血漿から得られたアポB48を多く
含むトリグリセリドリツチリポ蛋白をコートし
たアツセイ用プレートに入れ、室温で1時間放
置後、3回洗浄する。そこへ、1000倍に希釈し
たパーオキシデース標識抗マウスIgGを加え、
1時間室温で反応させ、さらに3回洗浄後、発
色液を加え、光度計で比色した。
トリグリセリドリツチリポ蛋白と強く反応す
る抗体を分泌するハイブリドーマを選択した。
単クローン化は、この選択したハイブリドー
マを、フイーダー細胞にマウス胸腺細胞を用
い、限界希釈法に2度かけることによつて行つ
た。
4 抗アポB48−の作成
(a) インビトロ法
前記工程で得られたハイブリドーマは、適
当な培養液(例えば、牛胎児血清10%を含む
RPMI1640培地)で培養し、その培養上澄み
を回収した。上澄み中に分泌された抗体は、
精製せずそのまま使用できる純度の高いもの
が得られた。
(b) インビボ法
ハイブリドーマを注射するマウスにあらか
じめ(4日前)、2,6,10,14−テトラメ
チルペンタデカンを腹腔内に注射しておく。
次に抗体を産生するハイブリドーマをマウ
