This invention relates to an analog peptide in which the N-terminal amino acid of YY (hereinafter referred to as PYY) is substituted.

It shows high homology (more than 50%) with peptides (PP) and is thought to belong to the same family.

Since peptides derived from the gastrointestinal tract, such as stokinin (CCK), were also found in the brain, the existence of PP-like peptides in the central nervous system was investigated immunochemically using APR antiserum, and many neural tissues were investigated. A positive reaction was observed.

2 pages (1987)). However, the affinity decreased from 1/20 to 1/1000 fold, and all of the reported C-terminal fragments were immovable even in VitrO.
It shows no physiological activity in vivo and does not act as antabonis. In particular, it has been shown that the entire sequence is essential for activity on blood vessels. On the other hand, regarding the vas deferens, there is a report that the fragment from position 13 to position 36 of PYY (hereinafter referred to as (13-36) PYY) exhibits approximately one-fourth the activity of PYY (C, W
ahlestedt et al., Regul, Pept., 13
Vol., pp. 307-318 (1986)). This indicates that this fragment adopts a conformation that is active for receptors in the vas deferens.

, 6-T, 7-Y) P Y Y can be synthesized as follows. Since the C-terminus is Tyr amide, p-methylbenzhydrylamine resin is used as a carrier in the in-phase method. Protected amino acids are sequentially coupled to this resin from the C-terminus. Subsequently, the peptide is excised from the resin using the HF method, and all protecting groups are removed at the same time. The resulting crude peptide is subjected to molecular sieve chromatography such as 5el) hadeX G-15 and eluted with about 0.2N acetic acid to remove small fragments and m1 products. Furthermore, the target peptide can be isolated by performing fractionation using high speed liquid chromatography using a column such as ODS, C4, (butyl), etc. Peptide purity can be confirmed by high performance liquid chromatography and amino acid analysis.

The conformation of each peptide is examined from the circular dichroism spectrum of P Y Y. Water, water-methanol, or water-trifluoroethanol is used as a solvent. Because methanol and trifluoroethanol have a lower dielectric constant than water, they promote the formation of intramolecular hydrogen bonds in peptide molecules rather than the interaction (hydrogen bonds) between peptide molecules and solvent, resulting in formation of intramolecular hydrogen bonds such as α-helices and β-structures. Stabilizes secondary structure.

The CD of P Y Y was measured using a JASCO J-20C self-recording optical rotation dispersion meter. Each peptide was dissolved in water, water-methanol, or water-1 to refluoroethanol (ratios were varied from 2 to 5) to a concentration of approximately 100 μM. The cell used is one with an optical path length of 05 mm.
Measurements were made from 0 to 260 nm.

It is fully conceivable that the active conformation of Y is an α-wax formed from the 14th position to the 32nd position and a disordered structure. Fragment (13-36) P Y Y
The fact that the activity of PYY is about one-fourth that of PYY can be explained as a difference in the degree of hydrophobicity of the peptide as a whole or the degree of stability of the α-helix.

。(Effects of the Invention) It has been revealed that PYY is widely distributed in vertebrates and exhibits remarkable pharmacological activity on the endocrine system as a gastrointestinal hormone and on the central and peripheral nervous systems as an agonist of NPY. In the present invention, we synthesized an analogue of the N-terminus of PYY and examined its functional structure and physiological activity.
It was revealed that it has an activity equal to or greater than that of natural PYY. By using an approach such as the present invention in which amino acids are substituted with consideration to the peptide conformation and its stabilization, it is possible to conduct research toward improving pharmacological activity, receptor selectivity, etc. for drug development.
