Abstract

PURPOSE:To constantly keep the pH of a medium at 7.0, to remarkably reduce the labor and cost and to minimize the danger of contamination, by adding calcium carbonate which is a hardly soluble salt. CONSTITUTION:In a system which lowers the pH of medium according to the progress of culture, excess calcium carbonate 3 is put together with culture medium components into a culture vessel 1, the content is sterilized and the culture is started after the inoculation of microorganisms. The acid generated by the growth of the microorganism causes neutralization reaction with precipitated calcium carbonate 2 to equalize the hydrogen ion concentration to the hydroxyl ion concentration and to stabilize the pH of the medium constantly at a level near 7.0. If the calcium ion or carbonate ion inhibits the production of the objective substance of enzyme or causes irreversible reaction, the use of calcium sulfate or barium sulfate is effective. The pH of the medium can maintained at about 7.0 with simple means even in a system which lowers the pH of the medium according to the progress of the culture of microorganisms.

DETAILED DESCRIPTION OF THE INVENTION [Industrial Application Field] The present invention relates to microbial culture, and particularly to pH adjustment of a culture medium.

Conventionally, this rare medium 1) H adjustment has been carried out by placing a medium 2 containing bacterial cells in an Erlenmeyer flask 1 as shown in FIGS. 3 and 4, for example.
Using an aeration device from the aeration pump 4 through the aeration pipe 6, the neutralization reagent 7 is poured into the medium 2 by opening the cock 8.
The pH was adjusted to the optimum H value by adding the solution dropwise to the solution and performing a neutralization reaction while measuring the pH value with a pH meter 9.

The above-mentioned conventional pH adjustment of the growing medium is carried out by dropping the sterilized neutralizing reagent 7 shown in Figure 3, or by dropping the neutralizing reagent passed through a membrane filter, but this is not possible at a laboratory level. However, if you want to perform a simple culture in a culture medium, it will take too much time and money, and if you dilute the concentration of the neutralizing reagent to prevent sudden temporary changes in pH, the amount of reagent added will increase, especially if the culture time becomes longer. The concentration of other medium components becomes diluted, which affects the culture and causes problems in reproducibility. It takes a lot of effort and money to maintain a stable and constant pH at all times, and the probability of contamination increases. There are disadvantages such as being expensive.

First, an excess of calcium carbonate 3 is placed in a culture container 1 together with the medium components, and after sterilization, microorganisms are inoculated and culture is started. The acid generated by the growth of microorganisms causes a neutralization reaction with the precipitated calcium carbonate 2, and the hydrogen ion concentration becomes equal to the hydrogen ion concentration, and the pH of the medium is always 7.

However, in applying the present invention, it is necessary to ensure that the production of target substances and oxygen produced by microorganisms is not inhibited by excessive calcium ions, etc., and that irreversible reactions do not occur, and that the optimum pH is close to 7.0. The present invention should be applied after confirming the following.

In the above embodiments, calcium carbonate was used as the poorly soluble salt, but calcium sulfates and barium sulfate can also be applied to the present invention. Calcium sulfate and barium sulfate are more effective when calcium ions or carbonate ions inhibit the production of target substances or enzymes or cause irreversible reactions.

As explained above, the present invention maintains the pH of the medium at around 7.0 by a simple method of adding calcium carbonate, which is sparingly soluble in water, in a system where the pH of the medium decreases due to the cultivation of microorganisms. There is an effect that can be done.
