本開示の実施形態は、医学的状態を治療するか、又は老化の自然なプロセスの影響を低減する方法を含む。医学的状態は、老化関連障害又は加齢関連障害又はプロセスであり得る。個体は、より若い個体と比較して、少なくともいくつかの細胞及び構造の喪失を含む、損傷及び他の有害な変化の細胞蓄積を有し得る。特定のケースでは、個体は、成体の体細胞が、個体のために部分的に再プログラムされた細胞又は誘導された多能性幹細胞に変換される能力を失っている。
以下の実施例は、本開示の好ましい実施形態をより完全に例示するために提示される。しかしながら、それらは、本開示の広い範囲を限定するものとして決して解釈されるべきではない。

包皮線維芽細胞をAmerican Type Culture Collection(ATCC)から入手し、製造業者の説明書に従って培養した。線維芽細胞条件培地を作製するために、線維芽細胞をCD73の発現に基づいて単離し、10%ウシ胎仔血清を含む10mlのRPMI培地あたり100万細胞の濃度で24時間培養し、培地を条件培地として使用した。

Claims (33)
- 対象を治療するためのシステムであって:
(a)細胞を含む少なくとも1つのバイオリアクター、
(b)少なくとも1つの選択的透過性膜、及び
(c)少なくとも1つの前記バイオリアクターと、少なくとも1つの前記選択的透過性膜と、前記対象とを接続し、前記対象から前記バイオリアクターに流体を循環させるためのチュービング(但し、前記チュービングが中空糸を含む場合、前記選択的透過性膜は任意で含む)を含み、
前記細胞が、多能性羊水幹細胞(MAFSC)又は線維芽細胞であり、
前記MAFSCが線維芽細胞由来であり、前記線維芽細胞が、CD73を発現するものである、システム。 - さらに前記対象からの血液又は血漿を含む、又は臓器保存液を含む、請求項1に記載のシステム。
- 前記細胞が、1つ以上の再生因子を基底速度又は誘導可能な速度で分泌する、請求項1に記載のシステム。
- 前記再生因子が、AKT、BAMBI、BCL-2、BCL-2XL、BDNF、BIRC5 CDA、CXCR4、ドミナントネガティブCCL2、EGF、エキソソーム、FGF-2、GATA-4 GDF-11、GDNF、hCG、HGF、HIF-1α、HLA-G、HO-1、hTERT、IFN-b、IGF-1、IFT-1、LIGHT、miR-126、NK4、NUR77、OCT-4、PGE-1、SDF-1、STC-1、TERT、TRAIL、VEGF、WNT11、XIAP及びそれらの組み合わせからなる群より選ばれる1つ以上の因子である、請求項3に記載のシステム。
- 前記細胞が、前記対象の血液又は血漿からの変性因子に応答して、1つ以上の再生因子を産生する、請求項1~4のいずれか一項に記載のシステム。
- 前記細胞が、変性因子の検出によって設定された速度で、再生因子を分泌する、請求項1~5のいずれか一項に記載のシステム。
- 前記再生因子の分泌速度と前記変性因子の検出速度の比が、50:1、10:1、5:1、1:1、1:5、1:10、1:50、及びこれらの間の任意の比からなる群より選択される、請求項6に記載のシステム。
- 少なくとも1つの前記選択的透過性膜が、前記バイオリアクターと前記対象との間の前記チュービング内に配置される、
前記チュービングが、選択的透過性膜から構成される、及び/又は
前記バイオリアクターが、少なくとも部分的に、前記選択的透過性膜から構成される、 請求項1~7のいずれか一項に記載のシステム。 - 少なくとも1つの前記選択的透過性膜が、前記対象と前記バイオリアクターとの間の細胞物質の通過を阻害又は減少させる、請求項8に記載のシステム。
- 少なくとも1つの前記選択的透過性膜が、再生因子及び変性因子の通過を可能にする、請求項1~9のいずれか一項に記載のシステム。
- 前記対象が動物である、請求項1~10のいずれか一項に記載のシステム。
- 前記対象が動物由来の臓器又は組織である、請求項1~10のいずれか一項に記載のシステム。
- 前記対象がドナー由来の臓器又は組織である、請求項1~12のいずれか一項に記載のシステム。
- 前記臓器が、肝臓、膵臓、胆嚢、胃、小腸、大腸、肺、腎臓、心臓、脾臓、脳、眼球及びそれらの組み合わせからなる群より選択される、請求項12又は13記載のシステム。
- 前記対象が、前記臓器の任意の部分又は臓器系の任意の部分である、請求項12~14のいずれか1項に記載のシステム。
- 前記対象が臓器又は組織であり、
前記システムが、前記バイオリアクターの少なくとも1つと、前記対象を封入するための少なくとも1つの前記選択的透過性膜とに接続された、容器をさらに含む、
請求項1~10又は12~15のいずれか一項に記載のシステム。 - 前記対象が臓器又は組織であり、
前記容器が、臓器保存液又は他の流体を前記チュービングへ移送するのを可能にするように、前記対象を保持するのに適している、
請求項16に記載のシステム。 - 再生因子を産生する体外的方法であって、
前記対象を、前記システム中の細胞からの1つ以上の再生因子の分泌を可能にする条件下で、請求項1~17のいずれか一項に記載のシステムに供するステップを含む、方法(但し、前記対象が、ヒトである場合、またはヒトから採取され治療目的のために前記採取されたヒトに戻されるものである場合を除く)。 - 前記細胞が、皮膚、心臓、血管、骨髄、骨格筋、肝臓、膵臓、脳、脂肪組織、胎盤、及び/又は包皮を含む組織に由来する線維芽細胞である、請求項18に記載の方法。
- 前記線維芽細胞が、CD73、CD90、CD56、SSEA3、SSEA4、Tra-1-60、Tra-1-81、Tra-2-54、HLAクラスI、CD13、CD44、CD49b、CD105、アミノペプチダーゼN、ヒアルロン酸結合受容体、コラーゲン/ラミニン結合インテグリンα2、OCT4、NANOG、SOX-2及びそれらの組み合わせからなる群より選択される1つ以上の表面マーカーを有する、請求項19に記載の方法。
- 前記線維芽細胞が、CD14、CD34、CD45、HLAクラスII及びそれらの組み合わせからなる群より選択される1つ以上の表面マーカーを欠く、請求項17~19のいずれか一項に記載の方法。
- (a)未分化状態にある線維芽細胞を、1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25日又はそれ以上の日数培養すること;
(b)ステップ(a)からの線維芽細胞を、神経成長因子、bFGF、ジブトリルcAMP、IBMX、レチノイン酸、エキセンジン-4及びそれらの組み合わせからなる群より選択される1つ以上の因子の存在下で培養すること;及び
(c)ステップ(b)からの前記線維芽細胞を活性化すること;
を含む、請求項19~21に記載の方法。 - ステップ(c)において前記線維芽細胞を活性化することが、ステップ(b)からの前記線維芽細胞を細胞培養培地中の1つ以上のサイトカインに曝露することを含む、請求項22に記載の方法。
- 前記サイトカインが、IL-1、IFNγ、及びそれらの組み合わせからなる群より選択される、請求項23に記載の方法。
- IL-1の濃度が、1~100ng/mL、5~100ng/mL、10~100ng/mL、又は20~40ng/mLである、請求項24に記載の方法。
- IL-1の濃度が、1ng/mL、5ng/mL、10ng/mL、20ng/mL、30ng/mL、40ng/mL、50ng/mL、60ng/mL、70ng/mL、80ng/mL、90ng/mL、又は100ng/mLである、請求項24又は25に記載の方法。
- IFNγの濃度が、1~1000IU、5~1000IU、10~1000IU、1~500IU、5~500IU、10~500IU、100~500IU、又は250IUである、請求項24に記載の方法。
- IFNγの濃度が、1IU、5IU、10IU、50IU、100IU、200IU、250IU、300IU、400IU、500IU、600IU、700IU、800IU、900IU、又は1000IUである、請求項24又は27に記載の方法。
- 前記線維芽細胞を1つ以上の前記サイトカインに曝露することにより、前記線維芽細胞からの1つ以上の補体阻害分子の発現の増加が誘導される、請求項23~28のいずれか一項に記載の方法。
- 前記補体阻害分子が、CD35、CD46、C4BP、CD55、H因子、及びそれらの組み合わせからなる群より選択される、請求項29に記載の方法。
- ステップ(c)において前記線維芽細胞を活性化することが、ステップ(b)からの前記線維芽細胞を1つ以上のウイルス及び/又は非ウイルス発現系でトランスフェクトして、1つ以上の再生因子の発現を誘導することを含む、請求項22に記載の方法。
- 前記再生因子が、AKT、BAMBI、BCL-2、BCL-2XL、BDN、BIRC5 CDA、CXCR4、ドミナントネガティブCCL2、EGF、エキソソーム、FGF-2、GATA-4 GDF-11、GDNF、hCG、HGF、HIF-1α、HLA-G、HO-1、hTERT、IFN-b、IFT-1、LIGHT、miR-126、NK4、NUR77、OCT-4、PGE-1、SDF-1、STC-1、TERT、TRAIL、VEGF、WNT11、XIAP、及びそれらの組み合わせからなる群より選択される、請求項31に記載の方法。
- 請求項1~17のいずれか一項に記載のシステムを含む、キット。
