イソフラボン類は、たとえば大豆内では、糖と共有結合した配糖体の形、ダイジン(daidzin)、グリシチン(glycitin)、ゲニスチン(genistin)として存在しており、アグリコンの形ではごく少量存在しているのみである。これら配糖体はさらにマロニル化、アセチル化されているものも存在している。これらの配糖体は、ヒトや動物の体内に入ると消化酵素又は腸内細菌の産生する酵素であるβグルコシダーゼ等の働きにより、それぞれダイゼイン(daidzein)、グリシテイン(glycitein)、ゲニステイン(genistein)となる。さらに、ダイゼインは腸内細菌の働きにより、ジヒドロダイゼイン(dihydrodaidzein)を経て、O-デスメチルアンゴレンシン(O-desmethylangolensin:O-DMA)又はエクオール(equol)へと酵素的に変換されることが知られている。

When these glycosides enter the human or animal body, they are converted to daidzein, glycitein, and genistein, respectively, by the action of digestive enzymes or β-glucosidase, an enzyme produced by intestinal bacteria. Furthermore, it is known that daidzein is enzymatically converted by the action of intestinal bacteria to dihydrodaidzein, and then to O-desmethylangolensin (O-DMA) or equol.

これらの課題を克服するために、乳酸菌等の嫌気性微生物を用いて体外的にエクオールを産生させる試みがなされている(特許文献1~4)。

なお、サポニンの分析は、次のように行った。
検体(サポニン含有量1mg相当)から水1mL+メタノール18mLの混液で抽出した。不溶物に対し、さらにメタノール20mLで抽出し、抽出液を混合した。この混合液から減圧濃縮によりメタノールを留去した。10%塩酸メタノール10mLを添加し、80℃、2時間加熱還流後、エタノールを加え、これをHPLC分析用サンプルとした。
ソヤサポゲノールA(分子量:474.72)およびソヤサポゲノールB(分子量:458.73)の標準品それぞれ10mgを100mLのメスフラスコに精秤し、エタノールに溶解、定容して標準溶液とした。
大豆サポニンAb(分子量:1437.52)、大豆サポニンBb(分子量:943.12)の含有量を求めるには、ソヤサポゲノールA、ソヤサポゲノールBの分子量から換算した。

[実施例1]
製造例1で得られたエクオール含有大豆胚芽発酵物に、アセスルファムK、アスパルテーム、スクラロース、ソーマチン、ラカンカエキス(モグロシドV 72%含有)、及びステビアエキス(ステビオール配糖体を80.0%以上含む)のうち1種を添加し、よく混合しエクオール含有組成物を得た。該エクオール含有組成物について、パネラー5名による試食を行った。なお、添加量は、砂糖換算で同等の甘味となるように算出した。
試食は、甘み又は苦みが口に残るため、適宜、水道水で口をゆすぎながら行った。無添加のものの苦みを5、甘みを1、大豆胚芽発酵物と当重量の砂糖を添加したものの甘みを5として、5段階で数値化を行った。その結果を表1に示す。
表1から、アセスルファムK、アスパルテーム、スクラロースの添加では、甘みはあるものの苦みも感じられ、効果としては十分ではないことがわかった。一方、ソーマチン、ラカンカエキスでは、甘みはあまり感じられないものの、苦みが軽減されることがわかり、マスキング剤として有用であることがわかった。

The taste test was conducted while rinsing the mouth with tap water as necessary, since sweetness or bitterness remained in the mouth. The bitterness of the additive-free product was rated as 5, the sweetness as 1, and the sweetness of the product with the same weight of sugar added as the soybean germ fermented product was rated as 5 on a 5-point scale. The results are shown in Table 1.
From Table 1, it was found that the addition of acesulfame K, aspartame, and sucralose produced a sweet taste but also a bitter taste, and was not effective enough. On the other hand, it was found that the sweet taste was not felt much with thaumatin and swingle extract, but the bitter taste was reduced, and it was found that they are useful as masking agents.

添加物をソーマチンとしその量を表2に示す量に変えた以外、実施例1と同様にエクオール含有組成物を得た。その組成物について、実施例1と同様に、パネラー5名による試食を行った。その結果を表2に示す。
表2から、ソーマチンの添加量を増やすことによって、甘みは弱めであるが、苦みが低減されることがわかった。試食を行ったパネラーによると、ソーマチンの添加量を増やすことによりきな粉様の味に近づくことがわかった。

表3から、ラカンカエキスの添加量を増やすことによって、甘みは徐々に増し、苦みが低減されるが、量を増やすと、エクオール含有大豆胚芽発酵物とは異なる苦みがでてくることがわかった。

The composition was subjected to tasting by five panelists in the same manner as in Examples 1 and 2. The results are shown in Table 3.
As can be seen from Table 3, by increasing the amount of Monk Fruit Extract added, the sweetness gradually increases and the bitterness decreases, but increasing the amount produces a bitterness different from that of the equol-containing soybean germ fermented product.
[実施例4]
添加物を表4に示すアスパルテーム及びソーマチンとし且つその量を表4に示す量に変えた以外、実施例1~実施例3と同様に、エクオール含有組成物を得た。その組成物について、実施例1~実施例3と同様に、パネラー5名による試食を行った。その結果を表4に示す。
[Example 4]
An equol-containing composition was obtained in the same manner as in Examples 1 to 3, except that the additives were changed to aspartame and thaumatin shown in Table 4 and their amounts were changed to those shown in Table 4. The composition was subjected to tasting by five panelists in the same manner as in Examples 1 to 3. The results are shown in Table 4.
表4から、アスパルテーム量が多くなると甘みが強くなり、ソーマチン添加による苦み抑制効果を軽減することがわかった。また、表4から、特に実施例4-3~4-7から、ソーマチン量が多くなりすぎるとと、苦みを感じることがわかった。なお、パネラーによると、アスパルテーム量が多くなると後に残る甘さを感じることがわかった。また、パネラーによると、総じて、きな粉砂糖のような風味を感じることがわかった。
以上のことから、アスパルテーム量が5~10重量部において、適度な甘みを感じ、ソーマチン量2~7重量部において、後に残る苦みをマスキングできることがわかった。
It was found from Table 4 that increasing the amount of aspartame increases the sweetness, and reduces the bitterness suppressing effect of adding thaumatin. From the above, it was found that when the amount of aspartame was 5 to 10 parts by weight, a moderate sweetness was felt, and when the amount of thaumatin was 2 to 7 parts by weight, the lingering bitterness was masked.
[参考例2]
実施例4-4の苦みを改善したエクオール含有組成物を、1)もち、2)わらび餅、3)トースト、4)白玉団子、5)アイスクリームに用いて、食品として摂取を試みた。
1)もち
市販の切り餅を、水に漬けた後電子レンジで温め、やわらかくした。このもちに実施例4-4の苦みを改善したエクオール含有組成物をまぶし、パネラーによる試食を行った。パネラーによると、きな粉餅様であり、おいしく食することができたとのことであった。
2)わらび餅
市販のわらび餅に、実施例4-4の苦みを改善したエクオール含有組成物をまぶし、パネラーによる試食を行った。パネラーによると、わらび餅に添付されたきな粉砂糖と食べ比べてもそん色のない味であるとのことであった。
[Reference Example 2]
The bitterness-reduced equol-containing composition of Example 4-4 was used in 1) mochi, 2) warabimochi, 3) toast, 4) shiratama dango, and 5) ice cream, and was tried to be ingested as a food.
1) Mochi Commercially available cut mochi was soaked in water and then heated in a microwave oven to soften it. The mochi was sprinkled with the equol-containing composition with reduced bitterness of Example 4-4, and a panel of people tasted it. The panelists said it tasted like kinako mochi and was delicious.
2) Warabimochi Commercially available warabimochi was sprinkled with the equol-containing composition with reduced bitterness of Example 4-4 and a panel of people tasted it. The panelists said that the taste was comparable to the soybean powder sugar that came with the warabimochi.
3)トースト
市販の食パン(六枚切りのうちの1枚)をトースターで焼きマーガリンを塗った。これに実施例4-4の苦みを改善したエクオール含有組成物をまぶし、パネラーによる試食を行った。パネラーによると、きな粉をかけたトーストと同様の味であるとのことであった。
4)白玉団子
市販の白玉粉に水を加え、よく混ぜた後、3cmほどの大きさに丸めた。これを沸騰したお湯に入れ、浮き上がってきたところを掬い上げた。これに実施例4-4の苦みを改善したエクオール含有組成物をまぶし、パネラーによる試食を行った。パネラーによると、苦みを感じることなく、きな粉白玉団子様であり、食することができたとのことであった。
5)アイスクリーム
市販のバニラアイスに実施例4-4の苦みを改善したエクオール含有組成物をまぶし、パネラーによる試食を行った。パネラーによると、アイスクリームのバニラ味が和風テイストとなり、苦みを感じることなく食することができたとのことであった。
3) Toast A piece of commercially available bread (one of six slices) was toasted in a toaster and spread with margarine. The toast was then sprinkled with the equol-containing composition with reduced bitterness of Example 4-4, and a panel of people tasted it. The panel of people said it tasted the same as toast sprinkled with soybean flour.
4) Shiratama Dango Commercially available shiratama flour was mixed with water and then rolled into balls of about 3 cm in size. These were placed in boiling water, and the balls that rose to the top were scooped up. The equol-containing composition of Example 4-4 with reduced bitterness was sprinkled on top, and a panel of people tasted it. According to the panel, it tasted like soybean flour shiratama dango without any bitterness, and was easy to eat.
5) Ice cream Commercially available vanilla ice cream was sprinkled with the equol-containing composition with reduced bitterness of Example 4-4, and a panel of people tasted it. According to the panelists, the vanilla flavor of the ice cream had a Japanese taste, and they were able to eat it without feeling any bitterness.
Claims (16)
(B)エクオール含有物に、ソーマチン及びラカンカエキスからなる群から選ばれる少なくとも1種のマスキング剤を加えて混合する工程;
