Description
本出願は、参照により本明細書に全体として組み込まれる、2018年9月5日に出願された米国仮特許出願第62/727,519号明細書の利益を主張するものである。
P450モノオキシゲナーゼ酵素は、不安定でバイオ触媒反応に使用するのが困難であることは有名である。しかしそれらは、薬物及び生体異物変換の代謝経路にとって重要な成分であるため、薬物代謝産物の生成及び化学物質の解毒において重要な役割を果たす。P450を含む代謝酵素によって化学的代謝産物の多様性を生じさせるための新規な方法に対するニーズが高まっている。それらは、新薬開発において化学物質の薬物動態研究及び生分解研究のために使用されている。組み換えシトクロムP450 BM3(BM3)は、その活性が高いため、及び例えば、B.メガテリウム(B.megaterium)又はE.コリ(E.coli)などの一般的宿主内での組み換えベクターから発現させるのが容易であるために、生体工学的及び化学的用途のための最も前途有望なモノオキシゲナーゼの1つであると考えられている。BM3は、それらが酸化活性及び補因子還元活性を有するので、一体型触媒である。構造的に、P450ドメインは、レダクターゼドメインと融合しているので電子の直接移動を容易にする。さらに、分子は可溶性であり、膜結合される必要はない。これは、バイオ触媒反応における製造及び使用のための利点を提供する。そこで、バイオ触媒反応においてP450を使用するための新規な方法を開発することには、商業的関心が有意に高い。
P450及び一般に大多数の代謝性酸化酵素は、それらの標的化合物を変換させるためには補因子を必要とする。陽子(H+)は、通常はCYP酵素内の特異アミノ酸を通して補因子NADH又はNADPHから送達される。それらは陽子を活性部位に中継し、そこでそれらは単一原子を基質に添加できるように酸素分子を還元的に分割する。CYP酵素は、グルコースデヒドロゲナーゼ(GDH)及びギ酸デヒドロゲナーゼ(FDH)を含むがそれらに限定されないある範囲の様々な酸化還元パートナー酵素からの電子を受け入れる。
ピンの製造に使用する材料は、Formlabsから購入した高温樹脂であった(0.45MPaで289℃、https://formlabs.com/store/us/form-2/materials/high-temp-resin/)。ピンはコンピュータ支援設計を用いてモデル化し、そのモデルの方向付け及びサポートを行うPreForm(Formlabs https://formlabs.com/tools/preform/)に送った。ピンは、Form 2(Formlabs https://formlabs.com/3d-printers/form-2/)プリンター及び高温樹脂を使用し、3D印刷した。印刷後、ピンを2つのイソプロピルアルコール槽で各々5分間洗浄した。ピンを槽から取り出し、室温で風乾した。次に、ピンをポスト硬化室内に配置し、60℃で405nmの光に60分間曝露した。最後に、支持体を除去し、ピンを水と800及び1200グリットの紙やすりで滑らかになるまでウェットサンドした。印刷されたピンを図1に示す。マイクロプレートピンアレイの図面を図2に示す。
機能化されたピンコーティングの製作に使用する材料は、鉄(II、III)酸化物(マグネタイト、Sigma-Aldrich<5μmの粒子、カタログ番号310069-500G)、カルボキシメチルセルロース(CMC、Aqualon TM、CMC 7H3SXF PH、Ashland、カタログ番号426352)、ポリビニルアルコール(PVA、89-98K 99%、Aldrich、カタログ番号341584-500G)、ナノフィブリル化セルロース(NFC、3%w/wの水分散液、Process Development Center,The University of Maine)、無水クエン酸(VWR)、キサンタンガム(Judeeのグルテンフリー、100%純粋)、及びミリQ水であった。
40%振幅で1分間、パルスの30秒オン及び10秒オフでの超音波処理を通じて、1gの3%w/w NFC分散液を6.3mLの水に添加することにより、NFCの分散液を生成した。次に、80mgのクエン酸を、500RPMで溶解するまで分散液に添加した。これに続いて、700RPMで撹拌中、2.7mLのCMC 2%w/w及び1.37mLのPVA 10%w/w水溶液を添加した。次に、823mgのマグネタイトを先の分散液に添加し、800RPMで2分間撹拌し、次に氷槽内、35%で1分間、パルスの6秒オン及び2秒オフで超音波処理した。
固定化CYP3A4は、7日にわたり貯蔵したとき、安定であった。遊離及び固定化CYP3A4の活性を評価するため、蛍光定量代替基質アッセイを開発した。CYP3A4ミクロソームは、Corningから購入した(カタログ456202:ヒトCYP3A4+P450レダクターゼ+チトクロムb5 SUPERSOMES(商標))。トリス緩衝液は、1Mトリス、pH7.5(KP BioMedical)から調製した。全ての水は、BarnStead Nanopure浄水器(Thermo Scientific、18.5Mオーム-cm)から入手した。トレハロース(D-(+)-トレハロース、二水和物)及びスクロースは、Fisher Scientificから購入した。マグネタイト粉末(鉄(II/III)酸化物粉末<5μm、95%)は、Sigma Aldrich(St.Louis,MO,USA)から購入した。
固定化CYP3A4活性は、無細胞アッセイにおいて、遊離CYP3A4と比較すると、有意に拡張された。CYP3A4ミクロソーム(カタログ456202:ヒトCYP3A4+P450レダクターゼ+チトクロムb5 SUPERSOMES(商標))は、Corningから購入した。ヘペス緩衝液は、Acros Organicsから購入した(ヘペスナトリウム塩、99%)。全ての水は、BarnStead Nanopure浄水器(Thermo Scientific、18.5Mオーム-cm)から入手した。スクロースは、Fisher Scientificから購入し、マグネタイト粉末(鉄(II/III)酸化物粉末<5μm、95%)は、Sigma Aldrich(St.Louis,MO,USA)から購入した。スーパーオキシドジスムターゼ(ウシ赤血球由来;MP Biomedicals);カタラーゼ(ウシ肝臓由来;Sigma Aldrichカタログ番号SRE0041)、G6DH(ロイコノストック・メセンテロイデス(Leuconostoc mesenteroides)由来のグルコース-6-リン酸デヒドロゲナーゼ;Alfa Aesarカタログ番号J60117-4I)、NADP(β-ニコチンアミドアデニンジヌクレオチド2’-リン酸還元テトラナトリウム塩水和物、>97%;Sigma Aldrich)、及びヘペス(ヘペスナトリウム塩、99%、ACROS Organics)を配合し、-20℃で貯蔵した。各固定化に対して、カタラーゼを新規に調製した。スーパーオキシドジスムターゼの保存液(水中、500U/ml)、G6DHの保存液(pH8の水中、500U/ml)を調製し、-20℃で貯蔵した。NADPの保存液(pH8の水中、26mM)を調製し、-80℃で貯蔵した。26mMのMgCl2の保存液を塩化マグネシウム六水和物(Macron Chemicals)から調製した。
ヒト肝ミクロソーム(Corning(著作権)UltraPool(商標)HLM150カタログ番号452117)は、Corningから購入した。トリス緩衝液を1Mトリス(pH7.5)(KP BioMedical)から調製した。全ての水は、BarnStead Nanopure浄水器(Thermo Scientific、18.5Mオーム-cm)から入手した。スクロースは、Fisher Scientificから購入し、マグネタイト粉末(鉄(II/III)酸化物粉末<5μm、95%)は、Sigma Aldrich(St.Louis,MO,USA)から購入した。
実施例3に記載の通り、PVA、CMC及びクエン酸の懸濁液(7%w/v)を円形ピンの形状で薄壁鋳型内に投じ、それを-80℃で凍結させることにより、酵素ピンデバイスを構築した。ピンを-12℃及び<100mTorrで3日間凍結乾燥させ、160℃の乾燥器内で1時間硬化させることにより直ぐに架橋させた。
CYP2B6は、酵素のチトクロムP450ファミリーのメンバーである。このファミリー内の酵素は、薬剤代謝に関与するモノオキシゲナーゼである。固定化CYP2B6の活性は、同じ濃度のタンパク質において遊離酵素の活性以上であった。
固定化CYP2B6は、14日間にわたり貯蔵したとき、安定であった。CYP2B6酵素(Corning(登録商標)Supersomes(商標)ヒトCYP2B6+オキシドレダクターゼ、カタログ番号456210)は、Corningから購入した。トリス緩衝液を1Mトリス(pH7.5)(KP BioMedical)から調製した。スクロースは、Fisher Scientificから購入した。全ての水は、BarnStead Nanopure浄水器(Thermo Scientific、18.5Mオーム-cm)から入手した。マグネタイト粉末(鉄(II/III)酸化物粉末<10μm)は、Reade Advanced Materialsから購入した。
UGT1A6は、小さい親油性分子を水溶性の排泄可能な代謝産物に変換するグルクロン酸抱合経路の酵素のUDP-グルクロノシルトランスフェラーゼである。固定化UGT1A6活性は、同じタンパク質濃度で遊離UGT1A6の活性以上であった。
UGT1A6酵素(Corning(登録商標)Supersomes(商標)ヒトUGT1A6、カタログ番号456416)は、Corningから購入した。トリス緩衝液を1Mトリス(pH7.5)(KP BioMedical)から調製した。全ての水は、BarnStead Nanopure浄水器(Thermo Scientific、18.5Mオーム-cm)から入手した。マグネタイト粉末(鉄(II/III)酸化物粉末<10μm)は、Reade Advanced Materialsから購入した。
マグネタイト粉末上でのCYP3A4の固定化及び活性。固定化反応緩衝液pHは、固定化反応における重要なパラメータであった。

- ミクロソーム及び磁気ナノ粒子の自己集合メソ多孔性凝集体を含み、ここで第1酵素活性を有する拡散性補因子を必要とする第1酵素は前記ミクロソーム内に含有され、補因子再生活性を含む第2酵素は前記メソ細孔内に磁気的に封入され、
前記補因子は前記第1酵素活性において利用される、
デバイスであって、
前記第1及び第2酵素は、拡散性基質を拡散性生成物に変換することにより機能し;
前記磁気ナノ粒子は、マクロ多孔性骨格と磁気的に会合され;
前記ミクロソームは、前記マクロ多孔性骨格と会合され;且つ
前記磁気ナノ粒子を含む前記マクロ多孔性骨格は、前記マクロ多孔性骨格を反応溶液に入れるか又はそれから除去するための非反応性ハンドルと会合される、
デバイス。 - 前記ミクロソーム、前記磁気ナノ粒子、前記第1酵素、前記第2酵素、及び前記マクロ多孔性骨格を安定に維持するための緩衝液を含む機能部分をさらに含む、請求項1に記載のデバイス。

- 前記非反応性ハンドルは、金属、プラスチック、セラミック、コンポジット材、又はそれらの組み合わせを含む、請求項1~4のいずれか一項に記載のデバイス。
- 磁気ナノ粒子の前記メソ多孔性凝集体は、酸化鉄組成物を有する、請求項1~4のいずれか一項に記載のデバイス。
- 磁気ナノ粒子の前記メソ多孔性凝集体は、前記磁気ナノ粒子の少なくとも90%が少なくとも3nm~30nmまでのサイズを有する磁気ナノ粒子サイズ分布、及び磁気ナノ粒子の前記メソ多孔性凝集体の少なくとも90%が少なくとも10nm~500nmまでのサイズを有する凝集粒径分布を有する、請求項1~6のいずれか一項に記載のデバイス。
- 磁気ナノ粒子の前記メソ多孔性凝集体は、少なくとも10emu/gの飽和磁化を有する、又は
磁気ナノ粒子の前記メソ多孔性凝集体は、5emu/gまでの残留磁化を有する、

- 前記ミクロソーム及び前記磁気ナノ粒子を含む前記マクロ多孔性骨格を前記溶液から除去する工程をさらに含む、請求項26に記載の方法。
- 複数の化合物の毒性をスクリーニングするためのハイスループットスクリーニング法に組み込まれる、請求項26に記載の方法。
- 代謝酵素の混合物からの代謝産物をスクリーニングするためのハイスループットスクリーニング法に組み込まれる、請求項26に記載の方法。
- 請求項1~25のいずれか一項に記載のデバイスを製造する方法であって、前記第2酵素を磁気ナノ粒子の前記メソ多孔性凝集体内に磁気的に封入する工程と、前記第2酵素を有する前記凝集体を前記第1酵素を含む前記ミクロソームと組み合わせる工程と、前記凝集体及びミクロソームを前記マクロ多孔性骨格上でテンプレート化する工程と、前記骨格を前記非反応性ハンドル上でテンプレート化する工程と、を含む方法。
- 前記凝集体は、補因子レダクターゼ活性を有する第3酵素をさらに含む、請求項30に記載の方法。
- 前記凝集体は、カタラーゼ、スーパーオキシドジスムターゼ(SOD)、又はグルタチオンペルオキシダーゼ/グルタチオン-ジスルフィドレダクターゼである第4酵素をさらに含む、請求項31に記載の方法。
- 前記凝集体は、第II相代謝に関与する第5酵素をさらに含む、請求項32に記載の方法。
