Description
分注デバイスによって発生させられ、バルクサンプル流体の中に伝達されるエネルギーを制御する分注パラメータは、バルクサンプル流体から抜け出すために十分にエネルギーを与えられた標的化液滴体積を送達するために、そのバルクサンプル流体の流体性質に具体的に合わせられる必要がある。
本発明は、例えば、以下の項目を提供する。
(項目1)
液体サンプルからサンプル液滴を分注する液滴分注器を較正する方法であって、前記方法は、
既知の較正物濃度の較正物を含む液体サンプルを提供することと、
前記液体サンプルから仮定された液滴サイズの液滴を分注することと、
前記仮定された液滴サイズの液滴をイオン化することと、
質量分析計を使用して、前記イオン化液滴中の較正物の較正物質量を測定することと、
前記仮定された液滴サイズに基づく予期される較正物質量を前記測定された較正物質量と比較することによって、実際の液滴サイズを決定することと
を含む、方法。
(項目2)
前記実際の液滴サイズが、ある閾値サイズを上回って前記仮定された液滴サイズから変動していることを決定することと、
前記液滴サイズ相違の大きさおよび符号のうちの少なくとも1つに基づいて、前記液滴分注器の少なくとも1つの液滴分注器パラメータを調節することと
をさらに含む、項目1に記載の方法。
(項目3)
前記以前に調節された少なくとも1つの液滴分注器パラメータを使用して、前記液体サンプルから後続の仮定された液滴サイズの後続液滴を分注することと、前記後続液滴をイオン化することと、前記イオン化された後続液滴中の較正物の後続較正物質量を測定することと、前記仮定された後続液滴サイズに基づく予期される後続較正物質量を前記測定された後続較正物質量と比較することによって、実際の後続液滴サイズを決定することと、前記実際の後続液滴サイズが前記閾値サイズを下回って前記仮定された液滴サイズから変動するまで、前記液滴分注器の前記少なくとも1つの液滴分注器パラメータを調節することとを繰り返し行うことをさらに含む、項目2に記載の方法。
(項目4)
捕捉プローブからの輸送流体を用いて前記分注された液滴を捕捉することと、
前記分注された液滴を前記輸送流体とともに前記質量分析計のイオン化チャンバに輸送することと
をさらに含む、項目1-3のいずれか1項に記載の方法。
(項目5)
前記流体分注器は、音響液滴分注器を備えている、項目1-4のいずれか1項に記載の方法。
(項目6)
前記捕捉プローブは、サンプルを捕捉するために輸送流体を外側導管を通してサンプル処理領域に供給し、輸送流体および捕捉されたサンプルを受け取り、エレクトロスプレーイオン化源に輸送する同軸開放ポートプローブ(OPP)を備えている、項目1-5のいずれか1項に記載の方法。
(項目7)
基準化合物を前記輸送流体に導入することをさらに含む、項目1-6のいずれか1項に記載の方法。
(項目8)
流体分注器によって分注される流体の体積を測定するためのシステムであって、前記システムは、
前記流体分注器から流体の分注された体積を受け取り、捕捉するための捕捉プローブであって、前記流体の分注された体積は、較正標準を含む、捕捉プローブと、
前記流体の捕捉された体積をイオン化するためのイオン化源と、
前記流体のイオン化された体積内に含まれる前記較正物の質量を測定するための質量分析計と
を備えている、システム。
(項目9)
前記捕捉流体の輸送流体の中に導入された既知の濃度の基準化合物をさらに含む、項目8に記載のシステム。
(項目10)
前記較正物の質量に基づいて、前記流体の測定された体積を決定するようにさらに動作可能である、項目8または項目10に記載のシステム。
(項目11)
前記較正物の前記測定された質量と前記基準化合物の測定された量との比率に基づいて、前記流体の前記測定された体積を決定するようにさらに動作可能である、項目10に記載のシステム。
Claims (11)
- 液体サンプルからサンプル液滴を分注する液滴分注器を較正する方法であって、前記方法は、
既知の較正物濃度の較正物を含む液体サンプルを提供することと、
前記液体サンプルから仮定された液滴サイズの液滴を分注することと、
捕捉プローブの進入口において輸送流体液体を用いて前記分注された液滴を捕捉することと、
前記仮定された液滴サイズの液滴をイオン化することと、
質量分析計を使用して、前記イオン化液滴中の較正物の較正物質量を測定することと、
前記仮定された液滴サイズに基づく予期される較正物質量を前記測定された較正物質量と比較することによって、実際の液滴サイズを決定することと
を含む、方法。 - 前記実際の液滴サイズが、ある閾値サイズを上回って前記仮定された液滴サイズから変動していることを決定することと、
前記液滴サイズ相違の大きさおよび符号のうちの少なくとも1つに基づいて、前記液滴分注器の少なくとも1つの液滴分注器パラメータを調節することと
をさらに含む、請求項1に記載の方法。 - 前記以前に調節された少なくとも1つの液滴分注器パラメータを使用して、前記液体サンプルから後続の仮定された液滴サイズの後続液滴を分注することと、前記後続液滴をイオン化することと、前記イオン化された後続液滴中の較正物の後続較正物質量を測定することと、前記仮定された後続液滴サイズに基づく予期される後続較正物質量を前記測定された後続較正物質量と比較することによって、実際の後続液滴サイズを決定することと、前記実際の後続液滴サイズが前記閾値サイズを下回って前記仮定された液滴サイズから変動するまで、前記液滴分注器の前記少なくとも1つの液滴分注器パラメータを調節することとを繰り返し行うことをさらに含む、請求項2に記載の方法。
- 前記分注された液滴を前記輸送流体液体とともに前記質量分析計のイオン化チャンバに輸送すること
をさらに含む、請求項1-3のいずれか1項に記載の方法。 - 前記液滴分注器は、音響液滴分注器を備えている、請求項1-4のいずれか1項に記載の方法。
- 前記捕捉プローブは、サンプルを捕捉するために輸送流体液体を外側導管を通してサンプル処理領域に供給し、輸送流体液体および捕捉されたサンプルを受け取り、エレクトロスプレーイオン化源に輸送する同軸開放ポートプローブ(OPP)を備えている、請求項1-5のいずれか1項に記載の方法。
- 基準化合物を前記輸送流体液体に導入することをさらに含む、請求項1-6のいずれか1項に記載の方法。
- 流体分注器によって分注される流体の体積を測定するためのシステムであって、前記システムは、
前記流体分注器から流体の分注された体積を受け取り、輸送流体液体の中に捕捉するための捕捉プローブであって、前記捕捉プローブは、進入口を有し、前記流体の分注された体積は、較正標準を含む、捕捉プローブと、
前記流体の捕捉された体積をイオン化するためのイオン化源と、
前記流体のイオン化された体積内に含まれる較正物の質量を測定するための質量分析計と
を備えている、システム。 - 前記捕捉プローブの輸送流体液体の中に導入された既知の濃度の基準化合物をさらに含む、請求項8に記載のシステム。
- 前記較正物の質量に基づいて、前記流体の測定された体積を決定するようにさらに動作可能である、請求項8または請求項9に記載のシステム。
- 前記較正物の前記測定された質量と前記基準化合物の測定された量との比率に基づいて、前記流体の前記測定された体積を決定するようにさらに動作可能である、請求項9に記載のシステム。
