皮膚等の生体組織に貼り付けられる膜の材料としてポリ乳酸が知られている。しかし、ポリ乳酸は、疎水性を示すので、蒸れ等の問題が生じる可能性があり、ポリ乳酸の膜を生体組織に長期間貼り付けることは必ずしも適切でないと考えられる。一方、セルロースの膜は、高い水蒸気透過度を有しやすく、汗などの水分を通しやすく、セルロースの膜を皮膚に貼りつける場合、蒸れ等の問題に起因する不快感を低減できる。(Findings on which this disclosure is based)
Polylactic acid is known as a film material that is attached to living tissue such as skin. However, since polylactic acid exhibits hydrophobicity, problems such as stuffiness may occur, and it is not necessarily appropriate to attach a polylactic acid film to living tissue for a long period of time. 2000~4000の重量平均分子量を有する、蛍光標識化したヒアルロン酸(PGリサーチ社製、Fluoresceinamine labeled Sodium Hyaluronate(3K2))と、5000~10000の重量平均分子量を有する、蛍光標識化したヒアルロン酸(PGリサーチ社製、Fluoresceinamine labeled Sodium Hyaluronate(U2))とを準備した。各ヒアルロン酸の水溶液(ヒアルロン酸の濃度:0.1重量%)を調製した。10μLの各ヒアルロン酸水溶液を、ろ紙上に設置した実施例1~3並びに比較例1及び2に係る生体貼付用膜のそれぞれに滴下した。ヒアルロン酸水溶液の滴下から30秒間経過したときに、実施例1~3並びに比較例1及び2に係る生体貼付用膜のそれぞれを取り外した。ろ紙を蛍光顕微鏡で観察することによって、ろ紙にヒアルロン酸が漏れ出しているか否かを確認した。蛍光顕微鏡は、キーエンス社製の蛍光顕微鏡を用いた。励起光波長:470nm、吸収波長:525nm、及び露光時間:1/200秒の条件で観察を行った。結果を表1に示す。表1において「OK」は、ろ紙におけるヒアルロン酸の検出レベルが検出限界以下であったことを示し、「NG」は、ろ紙においてヒアルロン酸が検出されたことを示す。いずれの場合も、ろ紙の変色を確認できたが、実施例1~3に係る生体貼付用膜がその上に置かれたろ紙では、蛍光標識したヒアルロン酸の検出レベルは検出限界以下であった。実施例1~3に係る生体貼付用膜は、高密度層の働きにより、ヒアルロン酸の溶出を抑制できることが示された。一方、比較例1及び2に係る生体貼付用膜がその上に置かれたろ紙では、蛍光標識したヒアルロン酸の検出レベルは検出限界を超えていた。<Dissolution evaluation of active ingredients>

90%以上の純度を有する、木材を原料とした漂白パルプ由来のセルロースを準備した。漂白パルプ由来のセルロースをイオン液体に溶解させることにより、セルロース溶液を調製した。イオン液体としては、1-エチル-3-メチルイミダゾリウムジエチルフォスフェイト(アルドリッチ社製、純度:98%)を用いた。セルロース溶液を、ジメチルスルホキシドで希釈した。ギャップコーティングによって基板の表面にセルロース溶液を塗布し、基板上にセルロース溶液の液膜を形成した。このとき、比較例3に係る生体貼付用膜の厚みが900nmとなることを狙ってギャップコーティングにおけるギャップの大きさを調整した。セルロース溶液の液膜を、70℃で1時間乾燥させ、さらに20℃及び40~60%RHの環境に置いてゲル化させ、高分子ゲルシートを得た。イオン液体が付着した高分子ゲルシートを所定のリンス液で洗浄し、高分子ゲルシートからイオン液体を除去した。その後、高分子ゲルシートを約0.1Nのテンションをかけながら乾燥させた後、アスコルビン酸ナトリウム水溶液(アスコルビン酸ナトリウムの濃度:3重量%)に浸漬させ、再度乾燥させた。これにより、約890nmの厚みを有する再生セルロース膜である、比較例3に係る生体貼付用膜を得た。比較例3に係る生体貼付用膜の厚みは、実施例1と同様にして測定した。(Comparative Example 3)

The liquid film of the cellulose solution was dried at 70° C. for 1 hour and then placed in an environment of 20° C. and 40 to 60% RH for gelation to obtain a polymer gel sheet. The polymer gel sheet to which the ionic liquid was attached was washed with a predetermined rinse liquid to remove the ionic liquid from the polymer gel sheet. Thereafter, the polymer gel sheet was dried while applying a tension of about 0.1 N, then immersed in an aqueous sodium ascorbate solution (concentration of sodium ascorbate: 3% by weight) and dried again. As a result, a bioadhesive membrane according to Comparative Example 3, which is a regenerated cellulose membrane having a thickness of about 890 nm, was obtained.
