The present invention is a neurotoxicity evaluation model system of a drug using nerve cells derived from artificial pluripotent stem cells (iPS cells) derived from a Charcot-Marie-Tooth disease (CMT) patient, and a drug using the model system. Regarding the neurotoxicity evaluation method.
Predicting the toxicity of candidate drugs is an essential step in drug development. Actually, the safety and efficacy of the candidate drug will be verified in clinical trials targeting humans through animal experiments, etc., but the action of the drug may differ between animals and humans, so candidates who have advanced to clinical research The reality is that about 40% of drugs have been forced to discontinue development due to unexpected side effects.
Therefore, in recent years, research has been conducted at an early stage of development to predict toxicity by investigating candidate drugs using human cells prepared from iPS cells so that drug discovery development can be carried out more efficiently and at low cost. It has been attracting attention, and toxicity evaluation systems using iPS cell-derived cardiomyocytes and hepatocytes have been reported so far.
Neurotoxicity, along with cardiotoxicity and hepatotoxicity, is an important item to be evaluated in drug discovery. For example, axonal degeneration, which is the predominant type of drug-induced neurotoxicity, is caused by certain anti-cancer drugs, but an in vitro model was used to test anti-cancer drug-induced neurotoxicity. It has been reported (Non-Patent Document 1). However, it has not yet been clarified what kind of evaluation system using iPS cell-derived neurons reflects actual clinical side effects. Establishing a neurotoxicity evaluation model system by identifying the phenotype of nerve cells that well reflects clinical symptoms from among many cell assay systems and phenotypes by cell assay using human iPS cell-derived neurons. Longed for to do.
Therefore, the subject of the present invention is to identify a cell assay system using human iPS cell-derived neurons that well reflects the clinical side effects of drug-induced neurotoxicity and cell phenotypes that are indicators of evaluation, and to identify in vitro nerves. To establish a toxicity evaluation model system, and to provide a means for predicting human neurotoxicity of a drug candidate drug with high accuracy using the model system.
In order to achieve the above-mentioned problems, the present inventors used iPS cells prepared from CMT patients who are highly sensitive to anticancer drugs and are prone to show neuropathy as a side effect. CMT is the most common hereditary peripheral neuropathy, affecting about 1 in 2,500 people worldwide. CMT causes progressive muscle atrophy, weakness, sensory impairment and foot deformity. The present inventors prepared iPS cells from CMT patients having a mutation in the gene encoding the mitochondrial protein Mitofusin-2 (MFN2) and differentiated them into nerve cells including motor nerves to prepare a neurotoxicity evaluation model. ..
Next, the present inventors comparatively analyzed the phenotypes between CMT patient iPS cell-derived neurons (CMT-N) and healthy human iPS cell-derived neurons (control), and in CMT-N, compared with controls. We found that mitochondria in neurites are morphologically short and have low mobility. Also, in the neurosphere assay, administration of anticancer drugs known to cause neurotoxicity resulted in mitochondrial aggregation with ATP deficiency in the neurites in both CMT-N and controls. , CMT-N was found to be more prominent.
Based on these findings, the present inventors conclude that this cell model is useful as a novel neurotoxicity evaluation system, and have completed the present invention.

Then, the cells are re-sown on feeder cells (for example, mitomycin C-treated STO cells, SNL cells, etc.), and about 10 days after the contact between the somatic cells and the reprogramming factor, the culture medium for bFGF-containing primate ES cell culture is used. It can be cultured to give rise to iPS-like colonies about 30-about 45 days or more after contact.

Desirably, instead of feeder cells, the reprogrammed somatic cells themselves are used (Takahashi K, et al. (2009), PLoS One. 4: e8067 or WO2010 / 137746), or extracellular matrix (eg, Laminin-). 5 (WO2009 / 123349) and Matrigel (BD)) are exemplified.

During the above culture, the fresh culture solution and the culture solution are exchanged once a day from the second day after the start of the culture. On the other hand, when a drug resistance gene expressed in conjunction with a gene expressed when somatic cells are reprogrammed (for example, Oct3 / 4, Nanog) is introduced as a marker gene, a culture medium containing the corresponding drug (selection). Established iPS cells can be selected by culturing with a culture solution). In addition, iPS cells are selected by observing with a fluorescence microscope when the marker gene is a fluorescent protein gene, by adding a luminescent substrate when it is a luciferase gene, and by adding a chromogenic substrate when it is a luciferase gene. can do.

No. 5,223,409), Spores (US Pat. No. 5,571,698, No. 5,403,484, and No. 5,223,409), plasmids (Cull et al. (1992) Proc. Natl. Acad. Sci. USA 89) : 1865-9) or Phage (Scott and Smith (1990) Science 249: 386-90; Devlin (1990) Science 249: 404-6; Cwirla et al. (1990) Proc. Natl. Acad. Sci. USA 87: 6378-82; Felici (1991) J. Mol. Biol. 222: 301-10; US Patent Application Publication No. 2002/0103360).
The concentration of the test compound added can be appropriately selected depending on the type of the compound, but in the case of a drug candidate compound, the concentration can be selected based on the concentration at which the desired medicinal effect can be sufficiently exhibited.
The time for contacting CMT-N with the test compound is not particularly limited, but preferably, various phenotypes that are indicators of neurotoxicity in step (2) are more prominent than those of healthy human iPS cell-derived neurons (control). The time to become is mentioned. For example, when the test compound is vincristine, paclitaxel, etc., it takes about 10 minutes to about 6 hours, preferably about 20 minutes to about 3 hours, more preferably about 30 minutes to about 2 hours (for example, about 1 hour). It can be exemplified.
In this sense, in a preferred embodiment of the evaluation method of the present invention, the above steps (1) and (2) are similarly performed on a healthy person iPS cell-derived nerve cell (control), and the control is used in CMT-N. In comparison, whether or not the phenotype showing neurotoxicity is more prominent can be used as a further evaluation index of neurotoxicity.
In step (2), the phenotypes of neurons that are indicators of neurotoxicity include (a) mitochondrial state in neurites, (b) ATP levels in neurites, and (c) MFN2 protein levels. Be done. Preferably, the phenotype of (a) or (b) is used as an index.
Here, examples of the "state of mitochondria in neurites" include mitochondrial length, mitochondrial morphology, mitochondrial aggregation, mitochondrial mobility, and mitochondrial number. The length and morphology of mitochondria can be verified by electron microscopy. For mitochondrial aggregation and mitochondrial mobility, for example, a construct in which a mitochondrial translocation signal sequence is linked to a fluorescent protein gene under the control of a motor nerve cell-specific promoter (eg, HB9 promoter) is introduced into nerve cells. It can be carried out by visualizing and observing mitochondria. The number of mitochondria can be counted by staining mitochondria with Mitotracker or the like.
Measurement of ATP level in neurites and MFN2 protein level in nerve cells can be performed by a conventional method.
When measuring the state of mitochondria in neurites and ATP levels, neurospheres are made from differentiated neurons to rule out mitochondria in the nerve body, which are then used in extracellular matrices (eg, laminin, matrigel, fibronectin). , Retronectin, etc., preferably laminin), inoculate the cells until neurite outgrowth is observed from the adherent neurites, and if necessary, remove the neurite core by pipetting or the like. , It is preferable to carry out.
In step (3), the phenotype of CMT-N when contacted with the test compound assayed in step (2) is compared to the phenotype when the compound is not contacted. As a result of comparison, the length of mitochondria in neurites is shorter when contacted with the test compound, the morphology of mitochondria in neurites is abnormal, and aggregation of mitochondria in neurites. High degree, decreased mitochondrial mobility in neurites, decreased number of mitochondria in neurites, decreased ATP levels in neurites, MFN2 protein levels in nerve cells If high, the test compound can be selected as a compound that is or is likely to cause neurotoxicity to humans. When the test compound is a drug candidate compound, the selected compound can be excluded from the development candidates. In other words, the test compound is unlikely to cause neurotoxicity to humans if there is no significant difference in the above phenotypes when contacted with the test compound compared to when not contacted. Or, it can be judged that the possibility is low, and drug discovery research can be continued as a development candidate.
Neurotoxicity can be classified into axonal degeneration, neuronal cell injury, and myelin sheath disorder according to the site of injury, and the evaluation method of the present invention is an evaluation model system for axonal degeneration.
Although described with emphasis on its use as a toxicity evaluation system for drug candidate compounds, the CMT-N of the present invention evaluates the susceptibility of existing drug compounds to individual patients (or a group of patients having the same causative gene mutation). It can also be used to classify patient groups (eg, responders and non-responders). Furthermore, it can be applied to identify drug susceptibility marker genes by comparing the results of comprehensive gene expression analysis among classified patient groups.

キモグラフは、Meta Morph Software(Molecular Device、Sunnyvale、CA)を用いて作成した。 Live imaging of axonal mitochondrial mobility
Images of living cells were acquired on the 50th day. HB9 :: Turbo RFP-mito-positive neurons were selected and mitochondrial mobility in neurites was observed by time-lapse photography. Living cells were photographed using a Carl Zeiss LSM710 microscope equipped with a x60 lens and an incubation chamber. The cells were kept at 37 ° C and 5% CO 2 during imaging. Nucleic acid-introduced cells were selected and mitochondrial mobility in neurites was observed by time-lapse photography. Images were taken every 3 seconds for 5 minutes and kymograph analysis was performed. The kymograph was created using Meta Morph Software (Molecular Device, Sunnyvale, CA). Mitochondria that were migrating were classified as either migrating or quiescent based on whether there was a 2 μm migration. In each kymograph, mitochondria migrating were counted.
全ての選別手順は、FACS AriaII(BD Biosciences、Franklin Lakes、NJ)を用いて行った。 Electron microscopic observation According to the previously reported method, motor neurons were purified by flow cytometry. On day 50 after infection with HB9 :: GFP lentivirus, differentiated cells were dissociated into single cells by Accutase (Innovative Cell Technologies) for cell selection. HB9 :: GFP-positive cells (about 2% of all sorted cells) were sorted and seeded on Matrigel-coated 96-well plates to 20,000-30,000 cells per well. The next day, purified motor neurons were fixed with paraformalin (PFA) adjusted to 4% with PBS at room temperature and 2% glutaraldehyde and incubated overnight at 4 ° C. Ultrathin section analysis was performed with a transmission electron microscope (Hitachi, Tokyo, Japan, H7650) at 80 kV. Mitochondrial length in neurites was measured on 7 random images using ImageJ.
Development of neurite assay and analysis of mitochondria in neurites A neurosphere assay was introduced to exclude mitochondria in the nerve body and efficiently evaluate mitochondria in neurites.
ATP measurement in neurites The cells were seeded on a 24-well plate so that there were 10 neurospheres per hole, and cultured for 7 days. After removing the neurosphere core by pipette suction, the medium was removed from the wells and ATP levels in the neurites were measured using the CellTitier-Glo fluorescent cell survival assay (Promega, Fitchburg, WI). The protein concentration in the cytolyte used for the ATP measurement was measured by the BCA assay, thereby correcting the ATP level.

<Result>
IPS cells generated from CMT patients
IPS cells were generated from two CMT patients with a mutation in the MFN2 gene of H128Y or R94Q and two healthy subjects (Fig. 1a, Table 1). Next, motor neurons were prepared from iPS cells. The iPS cells derived from CMT patients and the motor neurons prepared from them had mutations in the MFN2 gene (Fig. 1b).
Abnormalities in mitochondrial morphology and mobility in neurites were observed in motor neurons derived from iPS cells prepared from CMT patients.
The biochemical properties of MFN2 were investigated by Western blot analysis in a group of neurons including motor neurons derived from iPS cells of CMT patients. An increase in MFN2 was observed in the group of neurons derived from iPS cells of CMT patients (Fig. 2a, b). This indicates that mutations in the gene encoding MFN2 alter the biochemical properties of the MFN2 protein.
MFN2 is known to function in mitochondrial fusion. It has already been reported that biopsies of the calf nerve of CMT patients showed abnormalities such as small, round, and aggregated mitochondria in the axons. Therefore, since it was known that CMT patients with a mutation in the MFN2 gene showed damage in motor neurons, the mitochondrial length in the neurites of purified CMT motor neurons was investigated. The mitochondrial length in the neurites of CMT neurons was shorter than that of healthy subjects, and was clearly aggregated in the neurites with abnormal mitochondria lacking raised parts (Fig. 2c, d). These data suggest that mitochondrial fusion does not occur in CMT neurons, recreating the pathological features seen in biopsy samples of CMT patients.
Next, we investigated whether mitochondrial migration was impaired in the neurites of CMT neurons by time-lapse photography. To visualize mitochondria in live motor neurons, lentivirus expressing a red fluorescent protein (turbo RFP) was introduced into the motor neurons on day 36 of differentiation. The turbo RFP was fused to the mitochondrial targeting sequence under the control of the HB9 promoter (HB9 :: turbo RFP). And on the 50th day, time-lapse photography was performed. Images were taken every 3 seconds for 5 minutes. Typical chymographs of motor neuron mitochondria showed that the number of mitochondria migrating in CMT neurons was significantly reduced compared to controls (Fig. 2e). Quantitative analysis showed that in CMT neurons, the proportion of mitochondria migrating was significantly lower than in controls. In control neurons, migrating mitochondria accounted for 40-50%, whereas in CMT neurons, it was about 20% (Fig. 2f).
Furthermore, a neurosphere assay system was established and the number of mitochondria in the neurites was analyzed. This assay system made it possible to exclude mitochondria in the nerve body and efficiently evaluate mitochondria in neurites. In order to separate neurites from the nerve body, neurospheres were prepared from differentiated neurons on the 60th day, seeded in a laminin-coated culture vessel, and cultured for 3 days. This was followed by neurite outgrowth from the neurosphere with slight movement of the nerve body (Fig. 3b). Within the neurite, the number of mitochondria stained with Mitotracker was analyzed by a multifunctional analyzer. It was found that the number of stained neurite spots was reduced in CMT neurons compared to the control nerve (Fig. 3c, d). We also assessed ATP levels in neurites collected after removal of the neurosphere core. ATP levels were reduced in CMT neurons compared to control neurons (Fig. 3e). These results suggest that CMT neurons exhibit mitochondrial dysfunction in neurites.
Mitochondrial aggregate formation in neurites by drugs known to cause neuropathy
Patients with CMT develop severe drug-induced neuropathy after administration of anticancer drugs. The effect of the anticancer drug vincristine on CMT neurons was investigated. Neurites from neurospheres were exposed to 1 μM vincristine and stained mitochondria to assess their effects. Prolonged exposure to vincristine resulted in mitochondrial aggregates in neurites in both CMT and control neurons. However, at short exposures, CMT neurons were more sensitive to their toxicity than control neurons (Fig. 4a, b). 1 μM paclitaxel also increased mitochondrial aggregates in CMT neurons compared to control neurons (Figs. 5a, 5b). The multifunctional analysis system can recognize the objects of interest and automatically analyze their sizes. The system was also used to evaluate aggregated mitochondria.
These results suggest that genetic susceptibility promotes mitochondrial abnormalities in neurites in the form of drug-induced neuropathy. And the difference in susceptibility to drug toxicity between CMT patients and healthy individuals supports this idea. Among the many phenotypes of cells, mitochondrial abnormalities found in neurites are an appropriate cell assay system and represent the phenotype for predicting the neurotoxicity of pharmaceuticals using human iPS cells.
