Description
The present invention relates to a cell culture apparatus and a cell culture method.
This application claims priority based on Japanese Patent Application No. 2017-176138 filed in Japan on September 13, 2017, the contents of which are incorporated herein by reference.
In order to evaluate the efficacy and toxicity of pharmaceuticals and chemical products, and the pharmacokinetics such as absorption, distribution, metabolism, and excretion using cultured cells, a cell culture device using a diaphragm is used (for example,
In the cell culture apparatus described in
In general, in order to evaluate the efficacy and toxicity of pharmaceuticals and chemical products, it is necessary to examine the effects of many candidate compounds in a wide concentration range, so the number of assay conditions (assay throughput) that can be evaluated at the same time is large. preferable. Therefore, 96-well plates and 384-well plates are often used in general two-dimensional culture. Therefore, even in the cell culture apparatus described in

By forming a hydrogel using a cross-linking agent having polyethylene glycol as a main chain, which is a hydrophilic polymer, the hydrogel can be easily swollen when the culture solution is added to the dried hydrogel.
Specifically, the
For example, as the
The molecular structure of the above DBCO-4armPEG is shown in FIG.
The DBCO-4armPEG shown in FIG. 22 has the following properties.
(1) It has polyfunctional polyethylene glycol (4armPEG; molecular weight around 10,000) as a basic skeleton. This makes it relatively easy to get used to water.
(2) It has four dibenzocyclooctyne (DBCO) groups capable of click reaction with an azide group at the end of the molecule.
When the DBCO-4armPEG is mixed with a polymer modified with an azido group, the DBCO group and the azido group undergo a click-crosslinking reaction, whereby a plurality of polymers are crosslinked via the DBCO-4armPEG and gelled (FIG. 23). ).
Therefore, a gel can be formed simply by mixing an "aqueous solution of a click-crosslinking agent" and an "aqueous solution of a polymer having an azido group".
The click-crosslinking agent (compound A constituting the hydrogel described later) used for preparing the
The click-crosslinking agent according to the present embodiment may have the above-mentioned two properties of (1) a water-soluble basic skeleton and (2) a plurality of "groups that react with an azide group by click-crosslinking".
Here, (1) the water solubility of the basic skeleton means that it can be dissolved in water or a buffer solution in the vicinity of neutrality in an amount of 10% by mass or more at a temperature from room temperature to 0 ° C.
For specific water solubility, a compound serving as a basic skeleton or a compound A containing a basic skeleton is dispersed in a buffer solution (pH 7.0-7.6) such as a HEPES buffer at a concentration of about 1-100 mg / mL and dissolved. It can be judged by visually examining whether or not it is possible.
Further, as the structure of the basic skeleton which is specifically water-soluble, a part of the basic skeleton may be replaced with a water-soluble group or the like.
(2) The click-crosslinking reaction with an azido group is a group capable of easily and specifically cross-linking with an azide group. In particular, the condensation of an azido group and an alkyne can be mentioned. In the present embodiment, specifically, cycloalkyne, azacycloalkyne, or the like has such a property, and in the present embodiment, a structure having a cyclooctyne ring or an azacyclooctyne ring is preferably mentioned.
For example, a cross-linking agent containing a main chain composed of linear or branched or branched polyethylene glycol and a group such as a dibenzocyclooctyne group arranged at both ends or branched ends of the main chain. , Can be used as a click-crosslinking type cross-linking agent according to the present embodiment.
The number of branches in the main chain may be 4 branches or 8 branches. Further, the main chain may have a neopentyl skeleton.
Specific examples of such a compound include the compound of the following general formula (1).

In equations (1) and (2), ΔP is the pressure difference between the inlet and outlet of the microchannel, R is the channel resistance, μ is the viscosity of the fluid, l is the length of the microchannel, and w is the microchannel. Width, h is the depth of the microchannel. The equations (1) and (2) are established under the condition of w> h.
Consider a case where the lengths of the resistance flow path portion and the portion other than the resistance flow path portion are the same in the first culture solution
When a resistance flow path portion having a flow path cross-sectional area of 1/10 or less is formed in a part of the flow path, the flow path is designed by considering only the pressure loss of the resistance flow path. It has the advantage of facilitating the design of the channel network.

The structure and function of the
The design of the Laplace valve will be described below.
The pressure at which air flows into the Laplace valve (Laplace pressure, critical pressure) (ΔP Lap ) depends on the interfacial tension (γ) and the width (w L ) and depth (h L ) of the microchannels constituting the Laplace valve. It can be calculated by the following equation (3).
The realistic pressure range for driving the cell culture device is considered to be determined by the pressure range adjustable by a commercially available pressure control device and the pressure resistance of the cells.
Assuming that the pressure resistance of the cells is about the upper limit of blood pressure in the living body (30 kPa = 225 mmHg), the realistic pressure range for driving the cell culture apparatus according to the embodiment is about 1 kPa to 30 kPa. The interfacial tension of the culture solution is about 60 mN / m, and when the cross section of the microchannel constituting the Laplace valve is square, that is, when w L = h L , the dimension (length) of the microchannel in which air flows in at 30 kPa. From the above equation (3), it is estimated that the dimension (length) of the microchannel in which air flows in at 1 kPa is about w L = h L = about 8 μm and about w L = h L = 240 μm.
Assumed by making the dimension (length) of the microchannel constituting the Laplace valve smaller than the above dimension (w L = h L = 8 μm at 30 kPa) and w L = h L = 240 μm at 1 kPa). It is possible to prevent air from flowing into the Laplace valve when it is operated at the same pressure.
That is, the microchannels constituting the Laplace valve should be formed so that the Laplace pressure ΔP Lap , which is the limit pressure for the Laplace valve to function, is larger than the pressure range used in the cell culture apparatus according to the embodiment. For this reason, it is possible to prevent air from flowing into the Laplace valve.
Even when the ratio of w L to h L is not 1: 1, it is possible to design the dimensions of the flow path based on the equation (3) in the same manner.

In the

Therefore, highly efficient tests can be performed in drug discovery screening and the like.
As in the present embodiment, at least two of the plurality of culture solution storage chambers in the other embodiments may be communicated with each other by a gas flow path.
In addition, in the lungs in the living body, the alveoli are constantly expanding and contracting to breathe.
When measuring vital capacity, volume contraction occurs about 5 times as much as the volume of gas in the lungs, and in normal respiration, about 500 to 1,000 mL of the functional residual capacity (2,400 mL) is ventilated. (Standard Physiology 6th Edition, Medical School, p633-634).
Since the change in the volume of the alveoli is considered to be about the same as the change in the gas volume in the lung, the expansion of the alveoli surface area is estimated to be about 2/3 of the volume change, and is about 15 to 30% during normal breathing. Area change is occurring.
In
In principle, if the operation channel is formed deeper than the membrane area of the diaphragm, the area change of the diaphragm can be made large, but for this purpose, it is necessary to prepare a structure larger in the depth direction than in the plane direction. be. With general microfabrication technology, it tends to be difficult to fabricate a structure with a large aspect ratio, that is, a ratio between the plane direction and the depth direction, and it is very difficult to fabricate a structure with a large aspect ratio from a stretchable material. ..
Therefore, it is difficult to form a deep operation channel.
On the other hand, in the cell culture apparatus and the cell culture method according to the above-described embodiment, it is possible to cause expansion of the membrane based on the pressure difference between the top and bottom of the membrane surface.
According to the cell culture apparatus and cell culture method according to the above embodiment, the membrane can be stretched to about 100% (for example, the stretch ratio of FIG. 21 corresponds to 200%) as compared with the non-stretchable state of the membrane. It is possible, and it is possible to impart a larger stretching stimulus as compared with the method as shown in
Considering the direction of expansion and contraction of the membrane, some membrane-type organs and membrane-type tissues in the living body of an animal have an anisotropic expansion and contraction stimulus, while others are isotropic.
For example, in the dilation of blood vessels and the expansion and contraction of muscles, anisotropic expansion and contraction is imparted to tissues.
On the other hand, in the expansion / expansion / contraction of the alveolar tissue of the lung, an isotropic expansion / contraction stimulus is applied to the tissue.
According to the above embodiment of the present invention, for example, in FIGS. 12 to 14, since the elastic membrane is formed in the rectangular slit, as shown in FIGS. 16 to 20, in each of the drawings, the vertical direction is formed. It has been confirmed that the membrane expands and contracts.
On the other hand, the shape of the slit forming the film is arbitrary, and for example, as shown in FIGS. 3 and 10, it is possible to add an isotropic expansion / contraction stimulus by forming the film in a circular shape.
It is not feasible to apply such an isotropic expansion / contraction stimulus to the membrane in a configuration in which the diaphragm is expanded / contracted by pressurizing / depressurizing an operation channel adjacent to a flow path under the diaphragm as in

FIG. 21 is a graph showing the relationship between the focal length and the pressure, and the relationship between the expansion / contraction rate and the pressure.
As shown in FIG. 21, since the height of the microchannel was 600 μm, the displacement of the gel thin film was maximum at about 600 μm (2.6 kPa). On the other hand, regarding the expansion / contraction rate, even if the gel thin film came into contact with the wall of the flow path due to deformation, it continued to expand, and even if it doubled, it did not reach the expansion limit. From these results, it was confirmed that the gel thin film can cope with the expansion and contraction operation.
