Description
実施例1.方法
1.1 金コロイドの合成
金コロイド(金ナノ球体(Au nanospheres;NSs)、金ナノキューブ(Au nanocubes、NCs)、および金ナノ八面体(Au nanooctahedra、NOs))を頂点/角の選択的なエッチングと共にシード−媒介法(seed-mediated method)により合成した。まず、金ナノロッド(Au nanorods、NRs)を銀援助方法(silver-assisted methods)を用いて合成した(図1)。金ナノロッドのための金シードは、10mMソジウムボロハイドライド300μLを、10mM HAuCl4の125μLおよび100mM ヘキサデシルトリメチルアンモニウムブロミド(hexadecyltrimethylammonium bromide、CTAB)5mLを含有する溶液に注入して製造した。その後、100mM CTAB 200mL、10mM HAuCl410mL、10mM硝酸銀1.8mL、100mM L−アスコルビン酸1.14mL、および240μLのシード溶液1.14mLを順次添加し、金ナノロッドを成長させた。1分間穏やかに攪拌した後、2時間静置した。
金コロイドに抗凍結オリゴペプチドを融合させるために、C−末端システイン(Cys)の擬似共有結合(pseudocovalent bond)を用いた。精製水(DI water)に分散された前記方法で製造された金コロイドを過量のオリゴペプチドと混合し、30分間培養して完全に覆われるようにした。培養後、3回の遠心分離により過量のオリゴペプチドを除去し、水中0.01wt.%ドデシル硫酸ナトリウム(sodium dodecyl sulfate、SDS)に再分散した。
金コロイドの分散特性を理論的に予測するために、商業的に利用可能なソフトウェアパッケージであるFDTD(2014 CST Microwave Studio)を用いた。金の誘電率は、 Johnson, P. B.; Christy, R. W. Optical constants of the noble metals. Phys. Rev. B 1972, 6 (12), 4370-4379 に基づいており、オリゴペプチドの厚さは、単量体の数値によって0.60nm〜1.40nmとした。この厚さはオリゴペプチドの構造(conformation)に基づいて決定し、これは全原子(AA)分子動力学(MD)シミュレーションで計算した。暗視野顕微鏡のスペクトルの分析(図11)を反映するために、金コロイドの分散横断面(nm2)スペクトルを数値的に計算した。オリゴペプチドが付着されていない金コロイドおよびオリゴペプチドが付着されている金コロイドの吸収横断面(nm2)スペクトルを、図3b−dに示されるUV−Vis吸収スペクトルを説明するために計算した。
金コロイド上に付着したオリゴペプチドを実験的に定量するために、オリゴペプチドをNaBH4で還元させて分離し、チオール−選択的検出器媒介蛍光測定法(Lee, H. E.; Ahn, H. Y.; Mun, J.; Lee, Y. Y.; Kim, M.; Cho, N. H.; Chang, K.; Kim, W. S.; Rho, J.; Nam, K. T. Amino-acid-and peptide-directed synthesis of chiral plasmonic gold nanoparticles. Nature 2018, 556 (7701), 360-365.)を行った。まず、オリゴペプチドで改質された金コロイドを3回遠心分離し、0.01wt%SDSに再懸濁して、結合されていないオリゴペプチドを完全に除去した。金コロイドからオリゴペプチドを分離するために、25μLの100mM NaBH4を75μLのオリゴペプチドで改質された金コロイドに添加した。室温で5分間培養し、溶液を再び遠心分離して、金コロイドと放出されたオリゴペプチドを分離した。自由分散したオリゴペプチドを含有する上澄み液を一晩培養し、残りのNaBH4を非活性化した。
スプラット分析法(splat assay)及びスクロースサンドイッチ分析法を同時に行って氷再結晶化の程度を確認することにより、IRI活性を分析した(Mitchell, D.E.; Clarkson, G.; Fox, D.J.; Vipond, R.A.; Scott, P.; Gibson, M.I. Antifreeze protein mimetic metallohelices with potent ice recrystallization inhibition activity. J. Am. Chem. Soc. 2017, 139 (29), 9835-9838.; (9) Budke, C.; Heggemann, C.; Koch, M.; Sewald, N.; Koop, T. Ice recrystallization kinetics in the presence of synthetic antifreeze glycoprotein analogues using the framework of LSW theory. J. Phys. Chem. B 2009, 113 (9), 2865-2873.)。オリゴペプチドで改質された金コロイドを遠心分離し、精製水に再分散することにより、特性を把握する前に残存した有機リガンドによる作用を完全に除去した。スプラット分析法によりIRI活性を試験するために、10μLの試料を1.5mの高さから−70℃に既に冷却されたカバーガラスの表面に水滴を落として薄い氷膜を形成させた。薄い氷膜が形成された硝子カバーガラスを−20℃のペルチェ冷却器(Peltier cooler)に移し、硝子カバーガラスの温度を−6℃まで5℃/minの速度でゆっくりと昇温した後、この温度で試料をアニールした。氷が再結晶化する30分間、暗視野光学顕微鏡(Dark-field optical microscopy、DFOM)画像を撮影した。これらのDFOM画像を再結晶化した氷の粒子サイズを確認するために開発したコードで定量した。見える範囲で最も大きな10個の氷結晶を選定し、これらの平均直径を求めてIRI活性を測定した。3回の独立した実験の平均値から平均最大粒子径(mean largest grain size、MLGS)を算出した。
まず、スライドガラス上に両面テープを貼着し、その上にカバーガラスを位置させることにより、ガラスマイクロ流体チャネルを製造した。その後、このチャネルに精製水を注入し、透明なマニキュア液で密封した。空間的に温度(T)勾配を誘導するために、チャネルの一端は、−20℃のペルチェ冷却器上に位置させ、他端は室温(15〜20℃)に位置させた。これらの条件の下で、単結晶氷−水界面がペルチェ冷却器と室温エリアとの間に形成された。
DIS活性を確認するために、氷結晶の形状をスクロース水溶液で観察した。まず、45wt%スクロース溶液に溶解された試料を二つのカバーガラスの間に位置させ、50℃に冷却した。その後、試料を0.5℃/minの速度で、氷がほとんど残らなくなるまで昇温した後、試料を−0.1℃/minの速度で冷却させながら氷結晶の成長を誘導した。DFOM画像を冷却10分後に撮影した。
試料のTH活性を確認するために、単一の氷結晶の成長速度を様々な過冷却温度でモニタリングした。試料を、注文製作したナノリットル浸透圧計を用いて−50℃まで急速に冷却し、多結晶質の氷結晶を製造した。その後、単一の氷結晶が残るまでゆっくり昇温した。単一の氷結晶の大きさが4分以上維持された場合、その温度を融点に設定した。その後、温度を標的温度まで下げて、氷結晶の成長速度を4分間モニタリングした。THは、融点と氷結晶の成長が観察される温度との差で測定した(成長速度>0μm/min)。
AA MDシミュレーションは、CHARMM36力場を用いて、GROMACS v5.1.4で行った(Abraham, M.J.; Murtola, T.; Schulz, R.; Pall, S.; Smith, J.C.; Hess, B.; Lindahl, E. GROMACS: high performance molecular simulations through multi-level parallelism from laptops to supercomputers. SoftwareX2015, 1, 19-25.; Huang, J.; MacKerell Jr, A. D. CHARMM36 all-atom additive protein force field: validation based on comparison to NMR data. J. Comput. Chem. 2013, 34 (25), 2135-2145.)。オリゴペプチドの構造は、マービンスケッチ(MarvinSketch)を用いて製作し、パラメータはCGenFFを用いて誘導した(Vanommeslaeghe, K.; MacKerell, A. D. Automation of the CHARMM general force field (CGenFF) I: bond perception and atom typing. J. Chem. Inf. Model. 2012, 52 (12), 3144-3154.)。氷の実際の溶融点(Tm)(272.2K)を考慮して、TIP4P/ICE水モデルを使用した(Berendsen, H. J. C.; Postma, J. P. M.; van Gunsteren, W. F.; DiNola, A.; Haak, J. R. Molecular dynamics with coupling to an external bath. J. Chem. Phys. 1984, 81 (8), 3684-3690.)。金ナノキューブは、665金原子で構成されるように設計した(すなわち、2nmの一辺の長さ)。CHARMM−Metal力場は、無機界面での生体分子間の相互作用を説明する(Abascal, J. L.; Sanz, E.; Garcia Fernandez, R.; Vega, C. A potential model for the study of ices and amorphous water: TIP4P/Ice. J. Chem. Phys. 2005, 122 (23), 234511.)。様々なサイズと形状の金コロイドをMaterials Studio(Accelrys Inc.,San Diego,CA,2011)でナノクラスター製造機を用いて製造した。すべてのMDシステムは、平衡および生産稼動のために、それぞれBerendsen及びParrinello−Rahmanアルゴリズムを用いて、1barで維持した(Heinz, H.; Vaia, R. A.; Farmer, B. L.; Naik, R. R. Accurate simulation of surfaces and interfaces of face-centered cubic metals using 12-6 and 9-6 Lennard-Jones potentials. J. Phys. Chem. C 2008, 112 (44), 17281-17290.; Parrinello, M.; Rahman, A. Polymorphic transitions in single crystals: a new molecular dynamics method. J. Appl. Phys. 1981, 52 (12), 7182-7190.)。近接リスト(Neighbour list)は、カットオフ半径1.2nmでVerletカットオフ方式を用いて作成し、それぞれの段階ごとに更新した。結合の長さを制限するために、LINCSアルゴリズムを使用した(Hess, B.; Bekker, H.; Berendsen, H. J.; Fraaije, J. G. LINCS: a linear constraint solver for molecular simulations. J. Comput. Chem. 1997, 18 (12), 1463-1472.)。すべてのシミュレーションは、2fsの時間段階でリープ・フロッグ法(leap-frog integrator)を用いて行った。静電相互作用は、1.2nmのカットオフでPMEを用いて計算した(Darden, T.; York, D.; Pedersen, L. Particle mesh Ewald: An N log(N) method for Ewald sums in large systems. J. Chem. Phys. 1993, 98 (12), 10089-10092.)。
ペプチドが結合された金コロイドと氷結晶との間の結合エネルギー(即ち、自由エネルギー変化)を計算するために、本発明者は、プリングシステム(pulling system)(umbrella sampling simulation)を数値的にシミュレートした。このために、金粒子、氷表面および液体の水層を含有する合計200,000個の原子を有する10×10×12nm3のシステムボックスを製作した。Z−軸の反応配位を作るために、金粒子を1000kJ/mol nm2の力定数のDMPでプリングコードを使用して、氷の界面から500psの間、約4nm引っ張った。その後、1Aで試料ウィンドウの空間を定義し、40アンブレラサンプリング(umbrella sampling)ウィンドウを抽出した。それぞれのウィンドウを1nsの間、平衡を合わせ、10ナノ球体の生産実行を行った。PMF(potential of mean force)をGROMACSパッケージでWHAMツールを用いて取得した。PMF最大エネルギー状態と最低エネルギー状態との差として曲線から結合自由エネルギーを計算した。
2.1 抗凍結金コロイドの合成
異なる形態の、非常に均一な金ナノ粒子を十分に高い収率および拡張性を持って合成した。図2aは、本発明で使用される金ナノ粒子のSEM(scanning electron microscopy)およびTEM(transmission electron microscopy)画像を示す。氷の成長および再結晶化の抑制に二つの界面間の接触の効果を調査するために、金ナノ粒子が75nmと同じサイズでありながら、ナノ球体、ナノ八面体、およびナノキューブを有するように合成した。多角形の金コロイドの成長およびその後の頂点の選択的なエッチングにより、非常に均一な金コロイドを十分に多くの量で取得した(図2b)。金ナノ球体のLSPRは、合成バッチを20mLから200mLまで拡張した後でも維持した(図2bの右側図でのUV-Vis吸収スペクトル)。これは本発明での金コロイドの合成方法の収率および拡張性を裏付ける。
次に、オリゴペプチドが結合された金ナノ粒子の氷再結晶化の抑制(ice recrystallization inhibition、IRI)の効果を確認した(図6)。そのために、IRIの定量測定の標準であるスプラット分析法(すなわち、非平衡、急速冷却)を行った(Voets, I. K. Nat. Commun. 2017, 8, 15154.)。具体的には、精製水の水滴を冷却段階(−70℃)に位置させることにより、氷切片を作製した。その後、暗視野(dark field、DF)モードで光学顕微鏡(OM)を用いて、再結晶化された氷粒子サイズの空間的分布を確認した。DFOM画像に基づいて、再結晶化された氷ドメインの平均最大粒子サイズ(MLGS)を調査し、IRI特性を導出した。スプラット分析法で引き起こされる偽陽性を回避するために、スプラット方法から観察されるIRI特性を二重に確認するために、スクロースサンドイッチ分析を行った。
金コロイドの形状がIRI効果に及ぼす影響を評価した(図19)。異なる形状の金コロイドは、成長する氷とオリゴペプチドアセンブリとの界面接触を調節することができる(図19a)。金ナノ球体、ナノ八面体であるナノキューブのサイズは、75nmとすべて同一であり、(Thr)5−Cys−結合された金ナノ粒子の濃度は、0.2nMに固定した。図19bに示すように、IRI効果は、金ナノキューブ、金ナノ八面体および金ナノ球体の順に低くなった。この実験結果は、点接触に比べて面接触時に、成長する氷と(Thr)5−Cys−結合された金コロイドとの界面相互作用に有利であることを説明する。
