Description
(1)多角体病ウイルスの多角体の結晶構造に標的分子が封入された、多角体−標的分子複合体の製造方法であって、
配列番号1のアミノ酸配列からなるタンパク質をコードする核酸、又は、配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつ多角体形成能を有するタンパク質をコードする核酸を、細胞に導入する第1工程と、
第1工程の後、細胞に導入した核酸にコードされたタンパク質が発現し、発現したタンパク質が多角体の結晶の形成を完了する前までの所定時間、細胞をインキュベートする第2工程と、
第2工程後の細胞の培地に標的分子を添加して、更に細胞をインキュベートする第3工程と、を含み、
前記標的分子が、有機物質である、製造方法。

(3)前記配列番号1のアミノ酸配列からなるタンパク質、又は、前記配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつ多角体形成能を有するタンパク質が、金属原子に配位性を有するアミノ酸残基を有するタンパク質であり、
前記有機物質が、金属原子を含有する物質である、(1)又は(2)に記載の製造方法。
(4)前記金属原子を含有する物質が、金属錯体、金属原子含有タンパク質又は金属原子含有有機化合物(但し、金属錯体を除く。)である、(3)に記載の製造方法。
(5)多角体病ウイルスの多角体の結晶構造に標的分子が封入された、多角体−標的分子複合体であって、
配列番号1のアミノ酸配列からなるタンパク質、又は、配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつ多角体形成能を有するタンパク質が結晶化してなる多角体の結晶構造に標的分子が封入されており、

1実施形態において、本発明は、多角体の構成物質であるポリヘドリンタンパク質をコードする核酸を細胞に導入する第1工程と、第1工程の後、所定時間細胞をインキュベートする第2工程と、第2工程後の細胞の培地に標的分子を添加して、更に細胞をインキュベートする第3工程と、を含む、多角体−標的分子複合体の製造方法を提供する。以下、各工程について詳細に説明する。
第1工程では、多角体の構成物質であるポリヘドリンタンパク質をコードする核酸を細胞に導入する。ここで、ポリヘドリンタンパク質は、製造する多角体−標的分子複合体に封入する、標的分子に応じて選択する。ポリヘドリンタンパク質として、標的分子を結合可能な部位を有し、かつ結晶化により多角体を形成することが可能なものを使用する。ポリヘドリンタンパク質と標的分子との結合様式としては、配位結合、共有結合、水素結合等が挙げられる。
金属原子に配位性を有するアミノ酸としては、ヒスチジン、グルタミン酸、アスパラギン酸、システイン、セリン等が挙げられる。金属原子に配位性を有するアミノ酸を含む部位としては、上述したアミノ酸から選択される1種以上のアミノ酸を含む部位が挙げられ、例えば、ヒスチジンが約6個連続した部位、多角体の結晶内部の空隙に金属原子に配位性を有するアミノ酸が露出した部位、多角体の結晶表面に金属原子に配位性を有するアミノ酸が露出した部位等が挙げられる。
金属原子を含有する物質としては、金属錯体、金属原子含有タンパク質、金属原子含有有機化合物(但し、金属錯体を除く。)が挙げられる。
本実施形態の第1工程で細胞に導入する、ポリヘドリンタンパク質をコードする核酸としては、例えば、ポリヘドリンタンパク質のC末端にヒスチジン残基が6個連続したアミノ酸配列が付加されたタンパク質をコードする核酸が使用できる。この場合の標的分子としては、金属原子として、銅、ニッケル、亜鉛、コバルト、マンガン等から選択される1種以上を含有する物質が使用できる。核酸としては、ポリヘドリンタンパク質を発現させることができる限り、使用する発現系に応じてDNAであってもRNAであってもよい。ポリヘドリンタンパク質のC末端にヒスチジン残基が6個連続したアミノ酸配列が付加されたタンパク質のアミノ酸配列を配列番号1に示す。
ポリヘドリンタンパク質をコードする核酸を導入する細胞としては、昆虫細胞、甲殻類細胞、クモ類細胞等が挙げられる。より具体的には、昆虫細胞株であるSf9細胞株、Sf21細胞株等が挙げられる。
第2工程では、第1工程の後、所定時間細胞をインキュベートする。インキュベート中に、第1工程で細胞に導入した核酸が発現し、ポリヘドリンタンパク質が翻訳される。第2工程におけるインキュベートの時間を適切な範囲に設定することが、多角体への標的分子の封入効率を高めるうえで重要である。インキュベートの時間は、細胞への核酸の導入後、ポリヘドリンタンパク質が多角体の結晶の形成を完了する前までの時間であることが好ましく、細胞への核酸の導入後、ポリヘドリンタンパク質が発現し、多角体の結晶化が始まる前までの時間であることがより好ましい。
第3工程では、第2工程後の細胞の培地に標的分子を添加して、更に細胞をインキュベートする。標的分子の終濃度は0.001〜100mMであることが好ましく、0.01〜10.0mMであることがより好ましく、0.1〜5.0mMであることが更に好ましい。標的分子の終濃度が0.5〜2.0mMであると、標的分子の多角体への高い封入効率が得られる傾向にある。
製造された多角体−標的分子複合体は、遠心法、界面活性剤法等の方法により精製することができる。遠心法では、例えば多角体−標的分子複合体を産生した細胞に蒸留水を加えて破砕し、遠心分離で回収する操作を繰り返す手順により、多角体−標的分子複合体を精製することができる。界面活性剤法では、例えば多角体−標的分子複合体を産生した細胞に界面活性剤を加えて細胞を溶解し、遠心分離で回収をする手順により、多角体−標的分子複合体を精製することができる。
1実施形態において、本発明は、多角体病ウイルスの多角体の結晶構造の内部に標的分子が封入された(担持された)、多角体−標的分子複合体を提供する。ここで、多角体−標的分子複合体を構成するポリヘドリンタンパク質は、配列番号1のアミノ酸配列からなるタンパク質、又は、配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつ多角体形成能を有するタンパク質であってもよい。
標的分子として、金属錯体であるMn(CO)5Brを用いて、多角体−標的分子複合体を製造した。
ポリヘドリンタンパク質のC末端にヒスチジンが6個連続したアミノ酸配列が付加されたタンパク質(以下、「PH−His」という場合がある。)をコードするDNA(配列番号2)を組み込んだトランスファーベクターを作製した。具体的には、QUickChangeKit(アジレントテクノロジー製)と別途発注したプライマーを用いて変異を導入した。このDNAを制限酵素DpnIにて処理し、ヒートショック処理にて大腸菌に導入した後37℃で16時間寒天培地上培養、続いて37℃で16時間振盪培養を行った。大腸菌からDNAを精製し、エントリーベクター(pENT)として用いた。続いて、pENT 100μg及びpDEST 150μgを混合し、LR反応によりPH−His遺伝子を挿入したトランスファーベクターを作成した。LR反応物をヒートショック処理にて大腸菌に導入した後、37℃で1時間SOC培地中で培養し、続いて37℃で一昼夜寒天培地上で培養した。大腸菌からDNAを精製し、トランスファーベクターとして用いた。
PH−His発現用のバキュロウイルスを作製した。具体的には、上述のトランスファーベクター、BaculoGold linearized DNA(BDバイオサイエンス社製)、pTrans plasmid DNA 100ng/μL、Lipofectin reagent(Lifeテクノロジー社製)14μLを混合し、トランスフェクション試薬とした。Sf21細胞に27℃で24時間吸着させた。10%ウシ胎仔血清を含む培地2mLを加え、27℃で4日間培養した。この細胞培養上清を組換えバキュロウイルスとして使用した。
標的分子として、金属錯体であるMn(CO)5Br(シグマアルドリッチ社製)を用いた。Mn(CO)5Brは、アセトニトリル又は水に溶解し、波長300〜450nmの光を照射するとCOガスを放出する。
(第1工程:バキュロウイルスの細胞への導入)
上述したPH−His発現用バキュロウイルスを細胞株Sf21に導入した。具体的には、1×107個のSf21細胞を225cm2細胞培養フラスコに播種し、10%ウシ胎仔血清を含む培地40mLを加え、増殖させたウイルスを含む上清1mLを導入し27℃でインキュベートした。
PH−His発現用バキュロウイルスを感染させたSf21細胞を、ウイルス感染から36時間、27℃の環境下でインキュベートした。これにより、導入されたPH−Hisタンパク質が発現し、かつ多角体の結晶の形成が認められる前の段階になった。理論に拘泥するものではないが、この状態の細胞は、細胞膜の透過性が上昇しているため、通常は細胞膜を透過しにくい標的分子であっても、培地中に添加するだけで細胞膜を透過して、細胞内に入るものと考えられる。
ウイルス感染から36時間後のSf21細胞の培地にMn(CO)5Brを終濃度1mMとなるように添加し、更に6日間遮光下でインキュベートした。以上の手順により、多角体−標的分子複合体が製造された。
(多角体−標的分子複合体の精製)
製造した多角体−標的分子複合体を精製した。具体的には、多角体−標的分子複合体を産生した細胞に蒸留水を加えて破砕し、遠心分離で回収する操作を5回繰り返した。
精製した多角体−標的分子複合体を光学顕微鏡で観察した。具体的には、多角体−標的分子複合体を含む緩衝生理食塩水を一滴取り、カバーグラスで挟んで倒立実体顕微鏡で観察した。図1(c)に、実施例1の多角体−標的分子複合体の光学顕微鏡写真を示す。多角体−標的分子複合体の結晶が得られたことが確認された。また、図1(a)に実施例1の多角体−標的分子複合体と同様にして撮影した、野生型のポリヘドリンの結晶化により得られた多角体の光学顕微鏡写真を示す。また、図1(b)に実施例1の多角体−標的分子複合体と同様にして撮影した、PH−Hisのみの結晶化により得られた多角体の光学顕微鏡写真を示す。
精製した多角体−標的分子複合体の赤外線吸光分析を行い、多角体の結晶構造にMn(CO)5Brが封入されているか否かを確認した。具体的には、多角体−標的分子複合体を蒸留水に分散させた後、減圧乾燥により粉末状の多角体を回収し、KBr(臭化カリウム)錠剤法にて観測を行った。図2に赤外線吸光分析の結果を示す。多角体に配位したMn(CO)5Brに由来する赤外線の吸収ピークが検出され、多角体の結晶構造にMn(CO)5Brが封入されたことが確認された。

実施例1の多角体−標的分子複合体を用いて、COを制御放出し、細胞に刺激を与える検討を行った。
Claims (7)
- 多角体病ウイルスの多角体の結晶構造に標的分子が封入された、多角体−標的分子複合体の製造方法であって、
配列番号1のアミノ酸配列からなるタンパク質をコードする核酸、又は、配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加され、C末端に少なくとも6個連続したヒスチジン残基を有するアミノ酸配列からなり、かつ多角体形成能を有するタンパク質をコードする核酸を、細胞に導入する第1工程と、
第1工程の後、細胞に導入した核酸にコードされたタンパク質が発現し、発現したタンパク質が多角体の結晶の形成を完了する前までの所定時間、細胞をインキュベートする第2工程と、
第2工程後の細胞の培地に標的分子を添加して、更に細胞をインキュベートする第3工程と、を含み、
前記標的分子が、金属原子を含有する物質である、製造方法。 - 前記所定時間が12〜48時間である、請求項1に記載の製造方法。
- 前記金属原子を含有する物質が、金属錯体、金属原子含有タンパク質又は金属原子含有有機化合物(但し、金属錯体を除く。)である、請求項1又は2に記載の製造方法。
- 多角体病ウイルスの多角体の結晶構造に標的分子が封入された、多角体−標的分子複合体であって、
配列番号1のアミノ酸配列からなるタンパク質、又は、配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加され、C末端に少なくとも6個連続したヒスチジン残基を有するアミノ酸配列からなり、かつ多角体形成能を有するタンパク質が結晶化してなる多角体の結晶構造に標的分子が封入されており、
前記標的分子が、金属原子を含有する物質である、多角体−標的分子複合体。 - 前記金属原子を含有する物質が、金属錯体、金属原子含有タンパク質又は金属原子含有有機化合物(但し、金属錯体を除く。)である、請求項4に記載の多角体−標的分子複合体。
- 前記配列番号1のアミノ酸配列からなるタンパク質、又は、前記配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加され、C末端に少なくとも6個連続したヒスチジン残基を有するアミノ酸配列からなり、かつ多角体形成能を有するタンパク質。
- 配列番号1のアミノ酸配列からなるタンパク質をコードする核酸、又は、配列番号1のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加され、C末端に少なくとも6個連続したヒスチジン残基を有するアミノ酸配列からなり、かつ多角体形成能を有するタンパク質をコードする核酸。
