Description
The present invention relates to an oral care composition. In particular, the present invention relates to a denture care composition containing liposomal cetylpyridinium chloride, an oral care composition used when an implant or denture is mounted, or a denture care composition outside the oral cavity.
In many countries as well as in Japan, the proportion of elderly people in the population structure is increasing, and accordingly, how to maintain a high QOL is being emphasized. Recent studies are revealing the importance of oral care as one of the means. Specifically, it is known that oral bacteria can cause systemic diseases such as bacterial endocarditis, subclinical pneumonia, glomerulonephritis, arthritis as well as infections in the oral cavity. is there. On the other hand, oral bacteria are known to be related to age, and are referred to as so-called opportunistic bacteria such as Staphylococcus aureus and Pseudomonas aeruginosa that are not normally present in the oral cavity in the elderly. There is a tendency for the percentage of bacteria that are resident to be high. In particular, Staphylococcus aureus is not only a disease of the oral region such as cellulitis such as cheek cellulitis and floor cellulitis, jaw osteomyelitis, periodontitis after implant placement, and root abscess, but also erroneous. Control of Staphylococcus aureus is considered to be an important factor in oral care in the elderly because it causes aspiration pneumonia.
Many bacteria in the oral cavity are present in bacterial aggregates floating in the oral cavity or called biofilms. Although it is easy to remove floating bacteria, biofilms are resistant to both physical and chemical effects, and removing biofilms uses a powerful method. If not, it will be difficult. For example, a special mechanical tooth surface cleaning (PMTC) + 3DS removal method utilizing a chemical action and a physical action has been implemented. This method usually eliminates bacteria remaining after removing the biofilm by cleaning the inside of the oral cavity with a strong mechanical force using a dedicated exfoliating / cleaning composition by using a disinfectant-containing composition. Is the method. However, this method is not preferable for so-called weak oral persons, such as elderly people and persons having abnormalities in oral tissues, because it may cause various problems because the physical and chemical stimuli are too strong. In addition, in the case of removable dentures, powerful sterilization and sterilization utilizing active oxygen etc. can be performed outside the oral cavity without affecting the human body, so it is possible to prevent bacterial adhesion after wearing these in the oral cavity It becomes an important point. In addition, since many bacteria in the oral cavity form a biofilm and proliferate, in order to reduce the bacterial abundance in the oral cavity even when it cannot be detached like an implant, it is necessary to prevent the attachment of oral bacteria. It is considered effective to use a method that does not form a biofilm.
The mechanism by which bacteria floating in the oral cavity adhere to teeth and oral mucosal surfaces has been studied for a long time and various findings have been obtained. In order to effectively inhibit the formation of biofilm, it adheres to clean tooth surfaces in a short time and adheres to the oral tissues, implants, denture bases, etc. It is considered important to prevent this. This early colonization bacterium has a receptor that binds to salivary proteins and is known to promote adhesion when bound to specific salivary proteins. This saliva protein is roughly classified into a high proline protein and a high proline glycoprotein. Representative bacteria with the former receptor, Streptococcus mutans, Streptococcus gordonii, are known, such as Porphyromonas gingivalis, Representative bacteria with the latter receptor, Staphylococcus aureus, Fusobacterium nucleatum, etc. Streptococcus oralis is known Therefore, it is necessary to develop a technique having an effect of preventing adhesion to these two fungal groups. Many techniques for preventing adhesion of oral bacteria to the oral tissue surface by preventing aggregation of oral bacteria have been proposed (Patent Documents 1 to 6), but sufficient effects cannot be obtained, No technique has yet been found that is highly safe and prevents adhesion to not only oral tissues but also artificial parts such as implants and denture bases.
Prevents oral bacteria such as Streptococcus mutans and Staphylococcus aureus from adhering to the oral cavity with implants or dentures, prevents biofilm formation, and is highly safe for so-called vulnerable oral persons who are sensitive to chemical stimulation An object of the present invention is to provide an oral care composition capable of realizing high oral hygiene care.
As a result of intensive studies in view of such circumstances, the present inventors surprisingly have a composition containing liposomes containing cetylpyridinium chloride and non-hydrogenated phospholipids or hydrogenated phospholipids. By using the product, it has been found that bacteria such as Streptococcus mutans and Staphylococcus aureus can be prevented from adhering to the surface of oral tissues and dentures and proliferating, and the present invention has been completed.

According to the present invention, for oral bacteria having different properties such as Streptococcus mutans and Staphylococcus aureus , it effectively prevents adhesion to the surface of oral tissues and the like, particularly artificial surfaces such as implants and denture bases. Nevertheless, oral care compositions that are extremely safe and can be used every day with peace of mind even for oral tissues that are vulnerable to irritation called so-called weak oral cavity, especially oral care compositions for denture and implant wearers Can provide things. The oral care composition referred to in the present application is not only a composition directly applied to the oral cavity, but also a composition applied externally to the artificial object (for example, a denture) attached to the oral cavity (for example, denture). , Denture mounting agents, denture cleaning agents, denture preservatives, etc.).
The cetylpyridinium chloride used in the present invention is present in a state of being included in the liposome with respect to the total amount of the composition. Here, “included state” means a state in a liposome prepared by using phospholipid, cetylpyridinium chloride and other liposome components, and for example, cetylpyridinium chloride is the surface of the liposome. Adsorbed to the liposome, present as one of the membrane components of the liposome, or present in the continuous layer in the liposome formed by the lipid membrane of the liposome, ie other liposomes such as phospholipids It means a state in which a component and cetylpyridinium chloride form a liposome. In addition, as a preparation method of a liposome, the normal method shown below can be used and it is not specifically limited. Although the compounding quantity with respect to the oral care composition whole quantity of this invention is not specifically limited, It is preferable that it is a compounding quantity which will be 0.0004 mass% or more in a density | concentration at the time of use. That is, in the case of a product form that is used after being dissolved in a solvent or a product form that is diluted and used after being diluted with a solvent such as water, it means the concentration when actually used. In addition, about an upper limit, it is preferable from an economical viewpoint that it is 0.5 mass% or less, Preferably it is 0.3 mass% or less.
In the present invention, the degree of effect varies depending on the type of phospholipid constituting the liposome. For example, when the main phospholipids constituting the liposome are phosphatidylinositol, phosphatidylserine, and phosphatidylcholine (“main” means 50% by mass or more, preferably 70% by mass or more based on the total amount of phospholipids) The same applies to the following description.), The concentration of cetylpyridinium chloride with respect to the total amount of the oral care composition of the present invention when used in the oral cavity is preferably 0.0008% by mass or more, 0.0125 mass% is further more preferable, and 0.025 mass% or more is most preferable. Here, the reason for describing “the concentration of cetylpyridinium chloride relative to the total amount of the oral care composition of the present invention when used in the oral cavity” is one of the methods for improving the content stability of cetylpyridinium chloride. This is because a composition having a concentrated specification may be used. In this case, it means the concentration when diluted based on the product description, that is, the concentration when actually used in the oral cavity. Further, when the main phospholipid constituting the liposome is hydrogenated phosphatidylcholine, the concentration of cetylpyridinium chloride with respect to the total amount of the oral care composition of the present invention at the time of use in the oral cavity is preferably 0.0004% by mass or more. 0008% by mass or more is more preferable, 0.0016% by mass or more is more preferable, and 0.003% by mass or more is most preferable.

That is, cetylpyridinium chloride present in the continuous layer in which the liposome is suspended but not included in the liposome is removed using a conventional method, and then the liposome is destroyed using a conventional method. By checking the presence, cetylpyridinium chloride contained in the liposome can be confirmed. It is also possible to quantify using a similar method. The method for confirming and quantifying cetylpyridinium chloride is not limited. For example, the Japanese Pharmacopoeia Standard 2002 (ISBN4-8407-3054-7) pages 151-152 and the Quasi-drug Raw Material Standard “Confirmation test and quantification method of cetylpyridinium chloride” described in the second volume 371 of the 2006 integrated version (ISBN978-4-8408-1227-6) can be used.
、Staphylococcus aureus IFO13276(以下「S.a.」

(About test bacterial solution)
The test bacterial solution was prepared by the following method. Streptococcus mutans ATCC25715 (hereinafter abbreviated as “Sm”) and Staphylococcus aureus IFO13276 (hereinafter abbreviated as “Sa”) previously subcultured were inoculated into 10 ml of BHI liquid medium (Becton Dickinson). Pre-cultured at 37 ° C. for 18 hours. 100 μl of the preculture was added to each 10 ml of fresh BHI medium and cultured for 18 hours. After that, each BHI liquid medium was added to adjust to OD660 = 1.0, and the test bacterial solution was used for the test. did.
(Evaluation test procedure)
In the case of the liposome suspensions A and B, each of the specimens had a CPC culture concentration of 2, 4, 8, 16, 31, 63, 125, 250 (unit: μg / ml). Was diluted with a BHI liquid medium concentrated twice. As for liposome suspensions C and D, the liposome content in the subject at each CPC concentration of the liposome suspension A for the liposome suspension C and the liposome suspension B for the liposome suspension D In order to be the same, a solution prepared by diluting each liposome suspension with BHI liquid medium concentrated twice was used.
100 μl of the specimen prepared as described above was added to each well of a resin-made 96-well plate, and then 10 μl of the test bacterial solution prepared by the above method was added to each well, followed by incubation at 37 ° C. for 24 hours. After culturing, the supernatant in each well was removed, and each well after removal was washed with PBS (manufactured by Dainippon Sumitomo Pharma Co., Ltd.). The washing was performed by adding and removing 200 μl of PBS to each well three times.
As a control, a BHI liquid medium added and cultured instead of the specimen, that is, a culture using only the BHI liquid medium was used.
Each sample was evaluated using 3 wells and the control using 6 wells to obtain measured values.
The number of bacteria remaining (in the well) in the well after washing was measured according to the following procedure. That is, 100 μl of AlamarBlue (manufactured by Invitrogen) diluted 10-fold with PBS is added to each well after washing, and left in a dark place at 37 ° C. for 120 minutes to leave bacteria remaining in the plate. And reacted. After the reaction, using a Gemini XPS fluorescent plate reader (Molecular Devices), the fluorescence intensity (RFUxi) having a wavelength of 590 nm emitted upon excitation with light having a wavelength of 560 nm was measured. The survival rate of bacteria was calculated according to the following formula. That is, when each fluorescence intensity measurement value obtained above is (RFUxi) and the average value of the RFU values of the 6 wells of the control is (RFUca),
Bacterial residual rate (%) = {(RFUxi) / (RFUca)} × 100
According to the above, the bacterial residual rate in each well of each subject was calculated, and the average value and standard deviation value of the bacterial residual rate in 3 wells of each subject were calculated.
The obtained results are shown in Table 1 and FIG. 1 (result in Sm) and Table 2 and FIG. 2 (result in Sa).
Liposomes C and D do not contain CPC, but in Table 1 and FIG. 1 above, in order to facilitate comparison of the results of liposome A and liposome C, and liposome B and liposome D, the corresponding data are displayed side by side. did. The liposome amount (in terms of mass) at each CPC concentration is the same for liposome A and liposome C, and liposome B and liposome D. Table 2 and FIG. 2, Table 3 and FIG. 3 were processed similarly.
As shown in Table 1 and FIG. 1, liposomes containing phospholipids and CPC (Liposome B) subjected to hydrogenation treatment with Streptococcus mutans showed excellent effects at all concentrations. On the other hand, liposomes containing phospholipid and CPC not treated with hydrogenation (liposome A) showed an effect at a CPC concentration of 8 to 250 μg / ml, but weak at a low concentration (CPC: 2 to 4 μg / ml). It turns out that it can only be obtained. In addition, for liposomes not containing CPC (liposomes C and D), liposome C containing phospholipid without addition of hydrogen showed no effect at all liposome concentrations (phospholipid concentration), but was hydrogenated. Liposome D containing phospholipid was found to be effective at a liposome concentration (phospholipid concentration) corresponding to a CPC concentration in liposome C of 63 to 250 μg / ml.
In addition, as shown in Table 2 and FIG. 2, the liposome containing lipophilic phospholipid and CPC (liposome B) with respect to Staphylococcus aureus shows an effect when the CPC concentration is 8 to 250 μg / ml. Was 31-250 μg / ml, showing an extremely excellent effect. On the other hand, liposomes containing phospholipids and CPC not treated with hydrogenation (liposome A) showed an effect only when the CPC concentration was 125 to 250 μg / ml. In addition, for liposomes not containing CPC (liposomes C and D), liposome C containing phospholipids without added hydrogen showed no effect on the liposome concentration (phospholipid concentration), but hydrogenated phospholipids. It was found that the liposome D containing sucrose exhibits an effect at a liposome concentration (phospholipid concentration) corresponding to a CPC concentration of 31 to 250 μg / ml in the liposome C.
Liposome D was found to have the same or better effect than liposomes containing CPC, even though it did not contain CPC as a fungicide.

Cytotoxicity evaluation for oral mucosal cells
(Method for preparing test cells)
As oral mucosal cells, human gingival epithelial cancer-derived cell line Ca9-22 (hereinafter abbreviated as “Ca9-22 strain”) was used. The Ca9-22 strain was cultured according to the evaluation test. The culture is carried out using a minimum essential medium (Sigma) containing 10% fetal bovine serum (Funakoshi) and 1% Antibiotic-Antimycotic, 100X (Gibco) in an incubator at 37 ° C, 5 ° C. % CO 2 went under. Cells in the logarithmic growth phase of the culture were adjusted to 5 × 10 5 cells / ml by diluting with the medium. 100 μl of the prepared cell suspension was seeded in each well of a 96-well culture flat plate (manufactured by Sumitomo Bakelite Co., Ltd.) and cultured for 1 day so as to be confluent.
(Sample preparation method)
In the case of the liposome suspensions A and B, each of the specimens had a CPC culture concentration of 2, 4, 8, 16, 31, 63, 125, 250 (unit: μg / ml). Was diluted with a BHI liquid medium concentrated twice. As for liposome suspensions C and D, the liposome content in the subject at each CPC concentration of the liposome suspension A for the liposome suspension C and the liposome suspension B for the liposome suspension D In order to be the same, a solution prepared by diluting each liposome suspension with BHI liquid medium concentrated twice was used. For CPC alone, samples were dissolved in PBS so that the concentration of CPC was accurately 500 μg / ml, and then serial dilution was performed using PBS to prepare specimens of CPC alone at each concentration.
(Method of cytotoxicity evaluation)
The medium present in each well of the 96-well culture flat plate in which the test cells confluent by the above method were cultured was removed, and 200 μl of PBS was added to each well for removal. 100 μl of the specimen prepared by the above method was added to each well after washing, and left in an incubator under conditions of 37 ° C. and 5% CO 2 for 3 minutes. The control was left for 3 minutes in an incubator under conditions of 37 ° C. and 5% CO 2 without removing the medium. After standing, the liquid in each well was removed, and each well after removal was washed with PBS. The washing was performed by adding and removing 200 μl of PBS to each well three times. To each well after washing, 100 μl of alamarBlue (manufactured by Invitrogen) diluted 10-fold with PBS was added and allowed to stand for 2 hours. After being allowed to stand, the fluorescence intensity (RFUxi) having a wavelength of 590 nm emitted during excitation with light having a wavelength of 560 nm was measured using a Gemini XPS fluorescent plate reader (manufactured by Molecular Devices).
The cell viability was calculated according to the following formula. That is, when each fluorescence intensity measurement value obtained above is (RFUxi) and the average value of the RFU values of the 6 wells of the control is (RFUca),
Cell viability (%) = {(RFUxi) / (RFUca)} × 100
Thus, the cell viability of each well of each subject was calculated, and the average value and standard deviation value of the cell viability of 3 wells of each subject were further calculated.
The obtained results are shown in Table 3 and FIG.
As shown in Table 3 and FIG. 3, in the case of CPC alone, the cell viability is greatly reduced in the concentration region where the CPC concentration is 63 μg / ml or more, and the cell viability is particularly low in the concentration region of 125 μg / ml or more The value is shown. On the other hand, liposome suspensions A and B containing CPC did not show cytotoxicity in all CPC concentration regions. For reference, liposomes C and D containing no CPC were also evaluated in the high concentration region, but none of them showed cytotoxicity.
