ざ瘡に罹患している患者の非病変皮膚と比較した、これらの患者の病変皮膚におけるTH17分子プロファイルのモジュレーション:IL17A、IL17F、IL26およびCCL20ならびに提案されたマーカーIL-12Rβ1/IL-23R、CCR6、BATF、AHR、STAT3およびIRF4の発現の分析。
ざ瘡患者の皮膚生検を、有効な臨床実務に従って、12人のざ瘡を有する患者において、炎症性丘疹からおよび非病変皮膚から得た(ざ瘡サブタイプの臨床的記述は、Wilkinら、2002年、J. Am. Acad. Dermatol. 46巻、584〜587頁の分類に従って実施した)。
RNeasy抽出キット(Quigen Inc.、Valencia、CA)を使用してmRNAを皮膚から単離し、Agilentの2100 Bioanalyserを使用して質を評価した。T7-オリゴプライマーおよびAffymetrixの1サイクルcDNA合成キットを使用した二本鎖cDNAの生成(即ち、in vitro転写プロセス)の後に、Gene Chip IVT標識化キットによって、mRNA発現を評価した。RNAをエタノール沈澱してサンプルを濃縮し、次いで、分光光度計を使用して定量した。各サンプル由来の、およそ200ngの良好な品質の総RNA[RNA指標数(RNA indication number)(RIN)≧7]を使用し、T7-オリゴ(dt)プライマー(1サイクルcDNA合成キット、Affymetrix)を使用して二本鎖cDNAを生成した。in vitro転写(Gene Chip IVT標識化キット、Affymetrix)によって生成されたビオチン化cRNAを、断片化し、AffymetrixヒトU133A 2.0 plusマイクロアレイにハイブリダイズさせた。これらのアレイを、Gene Chip Fluidics Station 450で処理し、Affymetrix Gene Chip Scanner(Santa Clara、CA)でスキャンした。
Affymetrix Gene Chipからの発現データを、RMA(Robust Multi-array Analysis)法を用いて標準化する。生の強度値を、バックグラウンド補正し、log2変換し、次いで分位数標準化(quantile normalize)する。次に、線形モデルを、標準化したデータにフィッティングして、各アレイ上の各プローブセットについて発現測定を得る。異なるざ瘡サブタイプのサンプルにおいて有意にモジュレートされた遺伝子を同定するために、JMP 7.0.1(SAS Institute)およびirMF 3.5(National Institute of Statistical Sciences、NISS)ソフトウェアを使用して、Benjamini-Hochberg多重補正と共に一元配置ANOVAを実施した。
Th17分化プロファイルの発現もまた、qRT-PCRによって測定した。
タンパク質を、12人のざ瘡を有する患者における炎症性丘疹および非病変皮膚から抽出した。Luminexアッセイ(Millipore & Procartaサイトカイン投薬キット)を使用して、サイトカインを、タンパク質抽出物中に投与した。サイトカイン量を、タンパク質の総濃度に対して標準化した。ペアードP値(paired P-value)を、各サイトカインについて計算した。
免疫組織化学分析:
正常な皮膚において、本発明者らは、リンパ球が浸潤することは殆どないが、病変領域由来の生検では、より多い数でリンパ球が見出されることを観察した。この文脈において、免疫組織化学技術を使用したIL-17検出を、病変領域由来のこれらの浸潤物において実施した。
Claims (6)
- IL-12Rβ1、CCR6をコードするDNAまたはmRNAあるいは対応するタンパク質、ならびに、IL-6、IL-17A、IL-17F、IL-21、IL-22、IL-26、TNFαおよびCCL20から選択されるマーカーのうち少なくとも1つの、ざ瘡についてのマーカーとしての使用。
- a)個体からのサンプル中の、IL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質、ならびに、IL-6、IL-17A、IL-17F、IL-22、CCL20から選択されるマーカーのうち少なくとも1つの発現レベルを検出する工程、
b)健康な個体からのサンプル中の、IL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質、ならびに、IL-6、IL-17A、IL-17F、IL-22およびCCL20から選択されるマーカーのうち少なくとも1つの発現レベルを検出する工程、
c)工程b)の健康な個体において、および、工程a)の個体において、それぞれ検出された少なくとも1つのマーカーの発現レベルを比較する工程であって、
工程a)の個体におけるマーカーのうち少なくとも1つの過剰発現がざ瘡の指標であり、これによりざ瘡を同定する、工程、
を含む、ざ瘡の同定のための方法。 - 生物学的サンプル中の、IL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質のうち少なくとも1つ、ならびに、IL-6、IL-17A、IL-17F、IL-22およびCCL20から選択されるマーカーのうち少なくとも1つの発現レベルを分析する工程であって、マーカーのうち少なくとも1つの発現における異なる時点におけるバリエーションが、ざ瘡の進行またはバリエーションの指標である、工程、
を含む、個体におけるざ瘡の進行またはバリエーションをモニタリングする方法。 - ざ瘡の1つまたは複数の症状を有すると同定された個体から得られた生物学的サンプルにおいて、IL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質、ならびに、IL-6、IL-17A、IL-17F、IL-22、および、CCL20から選択されるマーカーのうち少なくとも1つの発現レベルを分析する工程であって、ざ瘡の処置の個体への投与の前後でのマーカーのうち少なくとも1つの発現におけるバリエーションが、ざ瘡の処置における効力の指標である、工程、
を含む、個体におけるざ瘡を処置するための処置の効力をモニタリングする方法。 - IL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質の発現を阻害もしくは下方調節する、ならびに/あるいは、IL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質の、トランス活性化活性を含めた生物学的機能を阻害する、候補の能力を決定する工程を含む、Th-17細胞分化インヒビターのin vitroスクリーニング方法。
- a)ざ瘡病変を模倣する少なくとも1つの生物学的サンプルまたはざ瘡病変を模倣する生物学的サンプルの混合物を、試験される1つまたは複数の薬物候補と接触させる工程;
b)a)で得られた生物学的サンプルまたは生物学的サンプルの混合物中の、IL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質のうち少なくとも1つ、ならびに、IL-6、IL-17A、IL-17F、IL-22およびCCL20から選択されるマーカーのうち少なくとも1つの発現または生物学的機能を検出する工程;
c) 工程b)で検出された発現または生物学的機能と、薬物候補と接触していないざ瘡病変を模倣するサンプルで検出された発現または生物学的機能とを比較する工程、
d) 工程b)において検出されたような、工程a)の生物学的サンプルまたは混合物中のIL-12Rβ1またはCCR6をコードするDNAまたはmRNAあるいは対応するタンパク質、ならびに、IL-6、IL-17A、IL-17F、IL-22、および、CCL20から選択された少なくとも1つのマーカーの発現または生物学的機能を阻害する薬物候補を選択する工程であって、前記選択された候補がTh17細胞分化インヒビターとして同定される工程
を含む、Th17細胞分化インヒビターの同定のためのin vitroスクリーニング方法。
