実験1の実施例1,2及び比較例1,2により得られた培養容器を、クリップを用いて、それぞれ培養部が、5cm×11cmとなるように区切った。
次いで、ヒト末梢血単核球(Cell Applications社製)5.8×104個を、10%ウシ胎児血清(ライフテクノロジーズジャパン株式会社製)入りのALyS505N-7培地(株式会社細胞科学研究所製)に懸濁し、4mlを培養容器に封入した。そして、そのまま37℃で75時間静置して、リンパ球の活性化工程を行った。
このとき、実施例1、2及び比較例2は、培養容器の固相化面を下にして行った。なお、比較例1の培養容器は、上記の通り、その内側の上下両面に同量の抗CD3抗体が固相化されており、上下の区別、すなわち、固相化面、非固相化面の区別はない。

培養開始からの上記各操作の時点(75時間後、75+26(=101)時間後、75+26+62(=163)時間後)、及びクリップを外してから18時間後(75+26+18(=119))に、各培養容器におけるリンパ球の数を計数した。その結果を図7に示す。

図7に示すように、培養容器内の片面のみにそれぞれ40ng/cm2、11ng/cm2の抗CD3抗体を固相化した実施例1,2では、増殖工程において、リンパ球を顕著に増殖することが可能になっている。このとき、固相化濃度が40ng/cm2の場合(実施例1)と11ng/cm2の場合(実施例2)との間で、増殖効率に大きな差異は見られなかった。
一方、培養容器内の両面に抗CD3抗体を固相化した比較例1では、実施例1,2と比較して、増殖工程におけるリンパ球の増殖効率が大幅に低くなっていることがわかる。
また、固相化する抗CD3抗体の濃度を、実施例2よりもわずかに減らした比較例2では、増殖工程においてリンパ球をほとんど増殖することができていない。このことから、培養容器の固相化面に固相化する抗CD3抗体の濃度は、約10ng/cm2以上にすることが好ましいことがわかる。
