Description
医療の現場でも容易に採取可能で測定できる生体試料として、血液、毛髪、唾液などの各生体試料を用いて、生体時刻を決定しようとする試みが最近活発になっている。しかし、血液を用いた方法(非特許文献1、特許文献1)の場合は、非侵襲的な測定試料採取方法とはいえない上に、メタボロミクスにより血液中に存在する概日変動する物質を選択したものであって、測定している物質と概日リズム調節機構の因果関係が科学的に証明されるには至っていない。さらに測定にはLC-MSやCE-MSといった特殊な測定装置が必要であり、簡単に測定することが難しい。
また、毛髪を用いた方法(非特許文献2)の場合は、時計遺伝子転写量の変動そのものをRNA量で測定するため生体から試料調整までの取り扱いが簡単ではなく、1日のうち異なる3点で試料採取する必要がある。
以上のことから、いずれの方法も普遍的な生体時刻測定法とはなっていない。
このように、唾液は生体より非侵襲的に採取でき、生物時計をモニターする上で有用な試料となりうる可能性が有るにもかかわらず、いまだ唾液を用いた生体時刻測定法の確立には至っていない。そのような原因の1つとして、唾液には以下に示す問題点を考慮する必要があることが考えられる。その1つは、唾液の分泌メカニズムが、大きく分けて(1)唾液腺細胞において合成された唾液の腺腔内への放出、及び(2)血清由来の水分、電解質等の管腔内への輸送、の2種類に起因することが知られているが、それぞれに対応する唾液の分泌腺は異なっており、それぞれの分泌腺から分泌される唾液の組成も異なっている点である。また、他の1つは、食事等、様々な刺激により唾液組成が変化することも知られており、さらに、口腔内に混在する細菌によっても影響を受けると考えられるため、採取時期、採取条件などでのデータのばらつきが懸念される。
本発明者らは、以前に、唾液腺細胞株HSG細胞が細胞生物学的に唾液腺細胞のモデルとして利用可能であること見出しており、当該HSG細胞を用いた研究により、以下の知見を得ている(非特許文献5)。
(1)唾液腺のような末梢組織には固有の概日リズム調節機構が存在していること、
(2)唾液腺腫瘍細胞HSGは正常唾液腺と同じような概日リズム調節機構を有していること、
(3)唾液腺概日リズム調節においては、負の調節因子Rev-erbαが中心となって調節をしており、正の調節因子RORαはほとんど発現をしていないこと。
すなわち、唾液腺細胞においては、主に負の転写調節因子Rev-erbαを中心に概日リズム転写調節が行われている。
そこで概日リズム機構により直接転写制御されうる遺伝子をクロマチンレベルで同定することを目的に、唾液腺細胞時計機構において鍵となるRev-erbα、ならびに一般的な時計機構において中心となるBmal1-Clockヘテロダイマーで転写調節されている遺伝子をChIP on chip法により同定した。Bmal1とRev-erbαの発現リズムは反対位相であることより、Bmal1を標的とする場合はHSG細胞をdexamethasone刺激36時間後、またRev-erbαを標的とする場合はdexamethasone刺激24時間後のクロマチンを用いて解析した。こうして500遺伝子が唾液腺細胞HSGにおいて時計機構により直接制御されていることが判明した。さらに転写産物(RNA)として発現量に明確な時刻依存性があるものを選択する為に、HSG細胞をdexamethasone刺激24時間後と36時間後においてRNA発現量の異なる遺伝子をマイクロアレイにより検討したところ、22遺伝子にまで絞り込むことができた。
さらにヒト唾液腺において実際の発現量を確認したところ、さらに9遺伝子にまで絞り込むことができた。このうちARRB1が、ヒト唾液を用いたウェスタンブロット解析において検出可能であり、時刻依存的発現をしていることを見出した。
以上の知見を得て本発明を完成した。
〔1〕 被検動物から採取された唾液中に含まれるβアレスチン−1(ARRB1)のmRNA転写量又はタンパク質発現量を、1日内での複数の時点で測定し、経時的な転写量又は発現量の変化パターンを観察することを特徴とする、被検動物の概日リズム測定方法。
〔2〕 βアレスチン−1(ARRB1)に対する抗ARRB1抗体を有効成分として含むことを特徴とする、唾液中の概日リズム測定用キット。
〔3〕 配列番号1に示される塩基配列中の連続した14塩基以上の部分配列を含むポリヌクレオチドからなるARRB1遺伝子増幅用プライマー及び/又はARRB1遺伝子用のプローブを有効成分として含むことを特徴とする、唾液中の概日リズム測定用キット。
〔4〕 被検動物の概日リズムの変調の判定方法であって、前記〔1〕に記載の測定方法により得られた被検動物のβアレスチン−1(ARRB1)のmRNA転写量又はタンパク質発現量の経時的な変化のパターンが、24時間周期パターンから外れている場合に、被検動物の概日リズム周期長が変調していると判定する方法。
〔5〕 被検動物の概日リズムの変調の判定方法であって、下記の(1)〜(4)の工程を含む方法;
(1)被検動物から唾液を採取し、採取された唾液中に含まれるARRB1のmRNA転写量又はタンパク質発現量を、1日内での複数の時点で測定し、転写量又は発現量の経時的な変化パターンを観察する工程、
(2)概日リズムが正常に働いている同種の動物から唾液を採取し、採取された唾液中に含まれるARRB1のmRNA転写量又はタンパク質発現量を、1日内での複数の時点で測定し、転写量又は発現量の経時的な変化パターンを観察する工程、
(3)工程(1)の経時的変化パターンを工程(2)の経時的変化パターンと比較する工程、
(4)両者の経時的変化パターンの周期長もしくは位相のいずれかにずれが生じている場合に、被検動物の概日リズムの周期長もしくは位相が変調していると判定する工程。
〔6〕 βアレスチン−1(ARRB1)に対する抗ARRB1抗体を有効成分として含むことを特徴とする、概日リズムの変調の判定用キット。
〔7〕 配列番号1に示される塩基配列中の連続した14塩基以上の部分配列を含むポリヌクレオチドからなるARRB1遺伝子増幅用プライマー及び/又はARRB1遺伝子用のプローブを有効成分として含むことを特徴とする、概日リズムの変調の判定用キット。
〔8〕 概日リズムの変調を改善する物質のスクリーニング方法であって、下記の(1)〜(4)の工程を含む方法;
(1)請求項2又は3に記載された判定方法によって、概日リズムの周期長もしくは位相が変調していると判定されたヒト以外の動物を用意する工程、
(2)工程(1)の動物に対して、被検物質を投与する工程、
(3)工程(2)の動物に対して、請求項2又は3に記載の判定方法を適用する工程、
(4)工程(3)の判定により概日リズムの周期長もしくは位相変調が改善された場合に、被検物質を概日リズムの変調を改善する物質であると評価する工程。
本発明により同定された、マーカータンパクARRB1は、唾液腺細胞において体内時計により調節されている物質であり、またラジオアイソトープを用いない通常のウェスタンブロット解析やELISAにより測定可能であり、簡便に生体時刻決定が可能となる。
これらの方法により生体時刻を測定することにより、クロノセラピー(時間治療)等の医療において、より効率のよい治療を行うことが可能となる。また、時差ぼけ等の概日リズム障害を伴う疾患の治療方針の指標となりうる。
東京歯科大学生科学教室木崎治俊教授より分与された唾液腺細胞株HSG細胞(Shirasunaらが樹立)は、細胞生物学的に唾液腺細胞のモデルとして以前から注目されており、本発明者らの最近の当該HSG細胞を用いた研究によって、唾液腺細胞での唾液腺特異的概日リズム調節機構の存在が明らかになった。唾液腺細胞においては、時計遺伝子の中で最も中心となるBmal1遺伝子プロモーター領域は低メチル化状態のCpGアイランド中に存在しておりBmal1遺伝子は正常に機能しているが、Bmal1遺伝子の概日リズム転写調節に重要な正の転写調節因子RORαは発現していない。唾液腺細胞では視交叉上核(SCN)や肝臓などと異なり、主に負の転写調節因子Rev-erbαを中心に概日リズム転写調節が行われている(非特許文献5)。
本発明の「生体リズムマーカー」として同定された「βアレスチン−1(ARRB1)」は、シグナル伝達の制御に重要なタンパク質ファミリーのアレスチン類に属する。(アクセッション番号:mRNA:NM_004041.4、タンパク質:NP_004032.2)
アレスチンとして最初に見出された「アレスチン−1」は視細胞などで特異的に発現されているが「βアレスチン−1(ARRB1)」は、非視覚アレスチンであり、種々の組織中に広く分布している。G−タンパク質共役型受容体のシグナル伝達や核内での足場タンパク質として作用するといわれており(Y.Shiら、Nat.Immunol.8.817-824(2007))、CD4陽性T細胞の恒常性に重要な役割を果たすという報告(E.M.Adler,Sci.STKE2007,tw268(2007))もなされているが、概日リズム関連物質に関する報告はない。
また、βアレスチン−1(ARRB1)遺伝子は、ヒトのみならず哺乳動物一般で広く保存されているので、本発明で「βアレスチン−1(ARRB1)」というとき、ヒト、サルなどの霊長類、ウシ、ウマなどの家畜類、イヌ、ネコなどの愛玩動物、マウス、ラットなどの実験動物を含め、哺乳動物一般に由来する「βアレスチン−1(ARRB1)」を意味する。
(1)測定対象
本発明における、唾液中での「βアレスチン−1(ARRB1)」の概日変化を量を観察することによる概日リズム測定方法は、主にはヒトを対象とするが、ヒトには限られず唾液を分泌する動物であればよく、サル、ウシ、ウマ、イヌ、ネコ、マウス、ラットなどの哺乳動物一般に適用できる。
すなわち、それぞれの哺乳動物の唾液中の「βアレスチン−1(ARRB1)」mRNA転写量、タンパク質発現量の概日変化を測定することで、その概日リズム変化を簡便に測定できる。特に、実験動物を用いて概日リズムが乱れたモデル動物を作製して適用することで、概日リズム改善剤のスクリーニングが簡便にできる。
自然分泌される全唾液を約1ml遠沈管などに採取し、そのうちの1部(1〜10μl、好ましくは2〜5μl)を、細胞等の夾雑物を遠心して除いて唾液試料とする。なおフィルター等を用いて夾雑物をろ過して除くことも可能である。
唾液の採取は、1日中の経時的変化パターンが観察できる程度の期間内で、異なる複数の時点で行う。具体的には、10時間〜48時間、好ましくは12時間〜30時間、典型的には24時間の間の複数の時点で唾液採取を行う。好ましくは、経時的に30分〜4時間おき、より好ましくは1時間〜3時間おきに採取する。
得られた唾液試料中のARRB1タンパク質の発現量を異なる複数の時点で測定して、経時的な変化のパターンを観察する。
採取のたびに直ちに唾液試料を調整して測定してもよいが、採取した唾液を−20℃冷凍の状態で保存しておき、全唾液試料を同時に測定してもよい。
ARRB1タンパク質の測定方法としては、抗ARRB1抗体を用いたウェスタンブロッティング法、またはELISA法により試料中のARRB1タンパク質量を定量的に測定することが好ましいがそれには限られない。
ウェスタンブロッティング法の場合、11%ポリアクリルアミドゲルを用いて電気泳動した後、PVDF膜に転写して抗ARRB1抗体によるタンパク質の検出を行う。なお、抗ARRB1抗体はAbcam社などから市販されている。その際の唾液のタンパク内部標準としてアクチンなどを検出しておき数値補正する。標識したHRP抗体などの二次抗体を用いて標識量を測定すれば、正確にARRB1タンパク量を定量可能である。
典型的な他の方法としてELISA法がある。唾液試料を96ウェルプレートに吸着させ、上記各抗体を用いてELISAにて定量することも可能である。
測定された唾液試料中のARRB1タンパク質量を経時的にプロットし、Cosinor法(Acta Med. Romana,18,399-440,1980)により変動曲線を引くことができる。本発明では、24時間あたりの当該変動曲線の形状を「経時的変化パターン」という。
得られた唾液試料中のARRB1のmRNA転写量を、異なる複数の時点で測定して、経時的な変化のパターンを観察する。採取のたびに直ちに唾液試料を調整して測定してもよいが、採取した唾液を−20℃冷凍の状態で保存しておき、全唾液試料を同時に測定してもよい。
唾液試料をAPGC法(P.Chomczynskiら、Anal. Biochem.,162,156-159 (1987))によりRNAを調製した後、1μgのRNAよりcDNAを合成する。
βアレスチン−1(ARRB1)の標準的な塩基配列(NM_004041.4:配列番号1)に基づいて設計されたプライマー、例えば5’-agaagcctctctggataagg-3’(配列番号2)ならびに5’-gtagaccttgcagaacgtcg-3’(配列番号3)のプライマーを用いるRT-PCRによりARRB1遺伝子の定量を行う。
ARRB1mRNAの転写量からの変動曲線も、上記(3)で述べたARRB1タンパク質量の変動曲線と同様に引くことができ、「経時的変化パターン」が観察できる。
その際用いるプローブ及び/又はプライマーは、βアレスチン−1(ARRB1)遺伝子の標準的な塩基配列(配列番号1)の部分配列であって、14塩基以上、好ましくは16塩基以上、より好ましくは20塩基以上、さらに好ましくは25塩基以上の部分配列を含むポリヌクレオチドからなるARRB1遺伝子増幅用プライマー及び/又はARRB1遺伝子用のプローブ(例えば配列番号2及び配列番号3のプライマー)と表現することもできる。
唾液細胞中のARRB1遺伝子の発現量は、中心的時計遺伝子のBmal1遺伝子に対する負の転写調節因子Rev-erbαの支配下にあるため、Bmal1遺伝子発現量の経時的変化パターンとは逆のパターンを示し、Rev-erbα遺伝子発現量の経時的変化パターンとは一致することになる。すなわち、正確な24時間周期の概日リズム(生体時刻)を刻むことになる。
したがって、被検動物における唾液中のARRB1タンパク質発現量又はARRB1mRNA転写量の経時的変化パターンが、24時間周期を外れた場合には概日リズム周期障害であると判定でき、また、正常動物本来のBmal1遺伝子発現量もしくはRev-erbα遺伝子発現量の経時的変化パターンと位相がずれた場合には概日リズムの位相の障害があると判定できる。
また、正常な昼型活動を行っている哺乳動物、ヒトの場合であれば、正常の昼型活動(例えば、就寝時間午後9〜10時、睡眠時間6〜8時間)を行っている正常人から唾液を採取して、唾液中のARRB1発現量又はARRB1mRNA転写量の標準的な経時的パターンを測定し、唾液中のARRB1概日リズムの正常パターンを決定する。当該正常パターンからのずれを観察することで、被検動物(被験者)の概日リズムの周期長又は位相の変調を正確に判定できる。
ARRB1タンパク質発現量又はARRB1mRNA転写量の標準的な経時的パターンの測定は、前もって行っておいても良いし、被検動物(被験者)の測定と同時期に行っても良い。
概日リズムの変調を判定するためのキットとしては、前記3.(3)又は(4)で述べたARRB1タンパク質の測定用キット又はARRB1mRNAの転写量の測定用キットを用いることができる。
ヒト以外の哺乳動物(例えばマウス又はラット)に対して、概日リズムを変調させた概日リズム変調動物を作製しておくか、天然のヒト以外の哺乳動物から、前記4.に記載の概日リズムの変調を判定する方法により概日リズムが変調していると判定された概日リズム変調動物を選択することにより、概日リズム変調モデル動物を用意する。
次いで、当該概日リズム変調モデル動物に対して、被検物質を経口投与又は静脈注射、皮膚への塗布などの非経口投与を施す。なお、被検物質の投与は、1回のみの投与でも複数回の継続的な投与でも良い。
投与から一定期間経過後(例えば、1昼夜の後)、前記3.に記載の概日リズムの変調の判定方法を適用し、ARRB1タンパク質発現量又はARRB1mRNA転写量の標準的な経時的パターンに近づいているか否かを判定し、当該標準的な経時的パターンと一致した場合、又は近づいた場合に、被検物質を概日リズム変調の改善剤候補とする。

また、本発明で引用した先行文献又は特許出願明細書の記載内容は参照して、本明細書の記載として組み入れるものとする。
(1−1)ChIP on Chip解析
唾液腺細胞における概日リズム機構の鍵となる遺伝子Rev-erbαのように、コアの時計遺伝子Bmal1/Clockにより転写調節されるのみならず、唾液腺特異的な機構としてRev-erbαにより転写調節されている遺伝子が唾液腺固有マーカーとしての可能性が高いと考えられる。そこで、非特許文献5に従い、ヒト唾液腺腫瘍細胞HSGにMyc-Rev-erbαおよびFlag-Bmal1を発現させた後、100nM Dexamethasoneで2時間処理することにより当該細胞の概日時計をリセットした。次いで、12時間後に抗Flag抗体(Sigma社製)を用いて、24時間後に抗Myc抗体(Roche Diagnostics社製)を用いて、本発明者らの開発した「ChIP (Chromatin immunoprecipitation):(非特許文献5)」を行い、Rev-erbαおよびBmal1により転写調節されている遺伝子領域を回収した。具体的には、前記HSG細胞を1%ホルマリンによる室温15分処理により、クロマチンのタンパク質とDNAをクロスリンクした後、マイクロコッカルヌクレアーゼによりクロスリンクされたクロマチンを断片化し、断片化されたクロマチンに含まれるMyc-Rev-erbαおよびFlag-Bmal1と結合しているDNA断片を、抗Myc抗体ならびに抗Flag抗体を免疫沈降法により回収、精製した。ChIPに供したHSG細胞の遺伝子DNAと上記ChIPにより回収されたDNAをそれぞれCy3、Cy5でラベルし、プロモーターDNAアレイ(NimbleGen社製)を用いて競合ハイブリダイゼーションすることによりRev-erbαならびにBmal1により転写調節されうる遺伝子の同定を試みた。
その結果、Bmal1により転写調節されうる遺伝子が797、Rev-erbαにより転写調節されうる遺伝子が2076あり、そのうち両方に調節されている遺伝子は500であった。これら遺伝子群は、唾液腺固有の時計システムで制御されうる遺伝子群(図1)である。なお、この500遺伝子中にRev-erbαは含まれていた。
前記(1−1)で得られた唾液腺固有の時計システムで制御されうる遺伝子群のうち、マーカーとして用いるためには、発現変動(RNA転写量変動)の大きいものを用いるほうが利点が大きい。そこでRev-erbαと同様なRNA転写量の変動を示す遺伝子を選択する為に、HSG細胞を100nM Dexamethasoneで2時間処理して時計遺伝子をリセットし、24時間後ならびに36時間後のRNAを用いてAgilent社製マイクロアレイ解析を行った。両時刻において変動が0.5(底が2の対数)以上の遺伝子を選択したところ1368遺伝子が選択され、このうち先のChIP on Chip解析と重複する遺伝子は22となった(図2)。なお、この22遺伝子中にRev-erbαは含まれていた。
前記(1−1)及び(1−2)の解析実験より選択された22遺伝子のうち、DNAマイクロアレイ(図2)において遺伝子発現が非常に弱い、シグナル強度が50以下の遺伝子を除いた16遺伝子について、ヒト正常唾液腺細胞においての遺伝子発現の有無を市販のヒト正常唾液腺RNA(TAKARA)を用いてRT-PCRにて検討した。
その結果、ヒト正常唾液腺細胞においてNR1D1(Rev-erbα)と匹敵する発現量を示す遺伝子として、COBRA1,ARRB1,KDELR1,SLC25A1,DBNL,TRIM28,PLCB3,TFDP1の8遺伝子を同定した(図3)。
前記(1−3)で選択された8遺伝子及びNR1D1(Rev-erbα)における概日リズムについて、100nM Dexamethasone2時間処理によりリセットしたHSG細胞を用いて、RT-PCRにより検討した。
その結果、NR1D1(Rev-erbα)と同様の転写量の周期性が観察された遺伝子は、ARRB1,KDELR1,TRIM28の3遺伝子に絞られ、これらの3遺伝子を、唾液中での概日リズム検出用マーカー候補とした(図4)。
(2−1)唾液中でのタンパク質発現量の測定
前記(実施例1)で得られた3遺伝子は、いずれもRNA転写量も充分に高く、かつその転写量がNR1D1(Rev-erbα)と同様の概日リズムを刻むことから、いずれも遺伝子レベルでは優れた概日リズム検出用マーカーとして用いることができる。しかし、これら遺伝子のうちで、唾液細胞内でタンパク質発現がなされ、かつ発現されるタンパク質量もNR1D1(Rev-erbα)の概日リズムと同期する遺伝子であれば、唾液に対して簡便な免疫学的検出法を適用することで、簡単に概日リズム変化を測定することができる。
そこで、本実施例では、NR1D1(Rev-erbα)抗体(Abcam社)と共に、マーカー候補それぞれの抗体のARRB1抗体(Abcam社),KDELR1抗体(Enzo Life Science社)及びTRIM28抗体(Santa Cruz Biotechnology社)を用いて、1mlを採取したヒト唾液のうち3μlを用いてタンパク質検出を試みた。その結果、タンパク質の検出ができたのは、ARRB1のみであった(図示せず)。
正常の昼型活動を行っている被験者(就寝時間午後9時)からの自然発生したヒト唾液を約1ml遠沈管で3時間おきに採取し、そのうちの3μlを遠心して細胞等の夾雑物を除き唾液試料とした。それぞれの試料についてのウェスタンブロット解析を行うために、11%ポリアクリルアミドゲルを用いて電気泳動した後、PVDF膜に転写し、3%スキムミルクで非特異反応をブロッキングした後、抗ARRB1抗体(Abcam社製)によりタンパク質の検出を行った。その際、同一PVDF膜をEzReprobe (アトー社製)にて抗ARRB1抗体をはがした後、抗アクチン抗体(Santa Cruz Biotechnology社製)により、唾液のタンパク内部標準としてアクチンを検出する。
これらのタンパクの定量は、HRP二次抗体とClarity Western ECL Substrate (Bio-Rad社製)による発光をLumiVisionPRO 400EX(アイシン精機社製)により測定した。
上記ウェスタンブロット解析の結果、概日リズム発現が観察された(図5)。図5では、Cosinor法(Acta Med. Romana,18,399-440,1980)により求められた値を実線で示した。
Claims (6)
- 被検動物から採取された唾液中に含まれるβアレスチン−1(ARRB1)のmRNA転写量又はタンパク質発現量を、1日内での複数の時点で測定し、経時的な転写量又は発現量の変化パターンを観察することを特徴とする、被検動物の概日リズム測定方法。
- βアレスチン−1(ARRB1)に対する抗ARRB1抗体を有効成分として含むことを特徴とする、唾液中の概日リズム測定用キット。
- 配列番号1に示される塩基配列中の連続した14塩基以上の部分配列を含むポリヌクレオチドからなるARRB1遺伝子増幅用プライマー及び/又はARRB1遺伝子用のプローブを有効成分として含むことを特徴とする、唾液中の概日リズム測定用キット。
- 被検動物の概日リズムの変調の判定のための方法であって、請求項1に記載の測定方法により得られた被検動物のβアレスチン−1(ARRB1)のmRNA転写量又はタンパク質発現量の経時的な変化のパターンが、24時間周期パターンから外れている場合に、被検動物の概日リズム周期長が変調しているとの基準に基づき、請求項1に記載の測定を行う方法。
- 被検動物の概日リズムの変調の判定のための測定方法であって、
(1)被検動物から唾液を採取し、採取された唾液中に含まれるARRB1のmRNA転写量又はタンパク質発現量を、1日内での複数の時点で測定し、転写量又は発現量の経時的な変化パターンを観察する工程と、
(2)概日リズムが正常に働いている同種の動物から唾液を採取し、採取された唾液中に含まれるARRB1のmRNA転写量又はタンパク質発現量を、1日内での複数の時点で測定し、転写量又は発現量の経時的な変化パターンを観察する工程と、
(3)工程(1)の経時的変化パターンを工程(2)の経時的変化パターンと比較する工程、
とを含む、
両者の経時的変化パターンの周期長もしくは位相のいずれかにずれが生じている場合に、被検動物の概日リズムの周期長もしくは位相が変調しているとの基準に基づく判定のための測定方法。 - 概日リズムの変調を改善する物質のスクリーニング方法であって、下記の(1)〜(4)の工程を含む方法;
(1)請求項1または4に記載の測定方法によって、概日リズムの周期長もしくは位相が変調していると判定されたヒト以外の動物を用意する工程、
(2)工程(1)の動物に対して、被検物質を投与する工程、
(3)工程(2)の動物に対して、請求項1又は4に記載の測定方法を適用する工程、
(4)工程(3)の測定により概日リズムの周期長もしくは位相変調が改善された場合に、被検物質を概日リズムの変調を改善する物質であると評価する工程。
