Description
1)本発明は、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法であって、下記の工程:
a)少なくとも1つの候補化合物を準備する工程;
b)少なくとも1つの候補化合物を、ERK MAPKタンパク質およびMyD88タンパク質と、ERK MAPKおよびMyD88を前記候補分子の不存在下で相互作用させるのに好適な条件下で接触させる工程;
c)少なくとも1つの候補化合物の存在下および不存在下で測定した場合のERK MAPKおよびMyD88の相互作用を決定する工程;および
d)ERK MAPKおよびMyD88の相互作用を阻害する候補化合物を選択する工程
を含んでなる方法に関する。
a)少なくとも1つの候補化合物を準備する工程;
b)誘導プロモーターの制御下でMyD88を安定発現し、かつ、SREプロモーター、ELKプロモーターおよびMyCプロモーターから選択されるMAPK経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
c)少なくとも1つの候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
d)前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害する少なくとも1つの候補化合物を選択する工程;
e)MyD88およびMyc癌遺伝子でのトランスフェクションにより形質転換された不死化線維芽細胞を準備する工程;
f)工程d)で選択された少なくとも1つの候補化合物の存在下および少なくとも1つのDNA損傷誘導化学療法薬の存在下でフォーカス形成をモニタリングし、フォーカス形成に対する前記DNA損傷誘導化学療法薬の阻害効果を増強する候補化合物を選択する工程
を含んでなる、方法。
・誘導プロモーターの制御下でMyD88を安定発現し、かつ、Nf−kBプロモーターおよびISREプロモーターから選択される炎症経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
・少なくとも1つの候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
・前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害しない候補化合物を選択する工程
をさらに含んでなる、6または7に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
a)候補化合物を準備する工程;
b)誘導プロモーターの制御下でMyD88を安定発現し、かつ、SREプロモーター、ELKプロモーターおよびMyCプロモーターから選択されるMAPK経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
c)前記候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
d)前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害する候補化合物を選択する工程;
e)MyD88およびMyc癌遺伝子でのトランスフェクションにより形質転換された不死化線維芽細胞を準備する工程;
f)工程d)で選択された候補化合物の存在下でフォーカス形成をモニタリングし、フォーカス形成が阻害されていれば前記化合物を選択する工程
を含んでなる方法に関する。
・誘導プロモーターの制御下でMyD88を安定発現し、かつ、Nf−kBプロモーターおよびISREプロモーターから選択される炎症経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
・候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
・前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害しない候補化合物を選択する工程
をさらに含んでなる。

炎症は発癌の促進因子であるということは十分に確立されている。従って、TLRシグナル伝達経路のアダプタータンパク質であるMyD88は、炎症におけるその役割を介して発癌に関連づけられている。本発明はここで、驚くことに、MyD88が、in vitroおよびin vivoにおいてras依存性発癌に本質的役割を果たしていることを示す。MyD88の阻害は、癌細胞においてアポトーシスを誘導することにより、in vitroおよびin vivoで発癌を軽減する。DNA損傷誘導化学療法との相乗作用が見られる。
a)少なくとも1つの候補化合物を準備する工程;
b)少なくとも1つの候補化合物を、ERK MAPKタンパク質およびMyD88タンパク質と、ERK MAPKおよびMyD88を前記候補分子の不存在下で相互作用させるのに好適な条件下で接触させる工程;
c)少なくとも1つの候補化合物の存在下および不存在下で測定した場合のERK MAPKおよびMyD88の相互作用を決定する工程;および
d)ERK MAPKおよびMyD88の相互作用を阻害する候補化合物を選択する工程
を含んでなる方法に関する。
a)少なくとも1つの候補化合物を準備する工程;
b)誘導プロモーターの制御下でMyD88を安定発現し、かつ、SREプロモーター、ELKプロモーターおよびMyCプロモーターから選択されるMAPK経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
c)少なくとも1つの候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
d)前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害する少なくとも1つの候補化合物を選択する工程;
e)MyD88およびMyc癌遺伝子でのトランスフェクションにより形質転換された不死化線維芽細胞を準備する工程;
f)工程d)で選択された少なくとも1つの候補化合物の存在下および少なくとも1つのDNA損傷誘導化学療法薬の存在下でフォーカス形成をモニタリングし、フォーカス形成に対する前記DNA損傷誘導化学療法薬の阻害効果を増強する候補化合物を選択する工程
を含んでなる方法に関する。
・誘導プロモーターの制御下でMyD88を安定発現し、かつ、Nf−kBプロモーターおよびISREプロモーターから選択される炎症経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
・少なくとも1つの候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
・前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害しない候補化合物を選択する工程
をさらに含んでなり得る。
a)有効量の上記化合物、および
b)不活性であっても生理学的に活性であってもよい薬学的に許容される担体
を含んでなる医薬組成物も提供する。
MyD88は腫瘍の発生に関連することがこれまでに示されていたので、本発明者らはMyD88が定着腫瘍の腫瘍表現型の維持に役割を果たし得るかどうかを調べた。特異的siRNAによるMyD88の消衰時、細胞はアポトーシスを受ける。このアポトーシスにおける腫瘍サプレッサータンパク質p53の役割を解明するために、本発明者らは、HCT116 p53+/+またはp53−/−細胞株を用いた。本発明者らは、p53タンパク質の安定化とともに、p53−/−細胞株に比べてp53+/+細胞株においてMyD88阻害時の大規模なアポトーシスを観測した。本発明者らは、種々のヒト結腸癌細胞株においても同じ実験を行い、MyD88阻害はこれらの細胞株においてp53依存性アポトーシスを誘導することを確認し、これが少なくとも結腸においては一般的な機構であり得ることが示唆された。
MyD88は、Ras/ERKシグナル伝達経路に必要である。MAPK経路はDNA修復機構の誘導に関与していることが知られている。従って、本発明者らは、MyD88阻害時に、ERKリン酸化の低下がDNA修復機構の欠如をもたらし、DNA損傷の蓄積を誘導し、p53依存性アポトーシスを誘発するとの仮説を立てた。本発明者らのデータによりこの仮説が確認された。実際に、HCT116 p53+/+ shMyD88細胞を、活性化MEKキナーゼを構成的に発現するプラスミドでトランスフェクトすると、ドキシサイクリン処置時に誘導されるそれらのアポトーシスが減少し、このことは、MyD88消衰により誘導されるアポトーシスがMAPK経路の阻害に基づくことを示す。MAPK経路はDNA修復機構、特に、ERCClタンパク質などのDNA修復に関連するいくつかの遺伝子の調節に関連づけられており、従って、本発明者らは、MyD88阻害時のERCClタンパク質レベルを定量した。本発明者らは、ERCClタンパク質レベルの低下およびpH2AXの蓄積を見出し、このことは二本鎖DNA損傷の蓄積を示す。
本発明者らの目的は、MyD88およびホスホ−ERKの相互作用を阻害することのできる小分子またはペプチドを種々のライブラリーから選択することである。本発明者らのデータに基づけば、細胞におけるMyd88の過剰発現は、ERKのリン酸化、NF−kBの核移行、および下流転写因子の活性化により示されるERKおよびNf−kBの両経路の活性化を誘導することが分かる。よって、本発明者らは、二段階スクリーニング技術を設計した。
1)SREプロモーター(ERK経路)またはNf−kBプロモーター(Nf−kB経路)の制御下のルシフェラーゼcDNA
2)抗生物質誘導プロモーター(ドキシサイクリン)の制御下のMyD88 cDNA
を同時に安定発現する細胞株(HCT116、A375、Hela)を用いている。
HCT116 p53+/+またはp53−/− shMyD88細胞株で併用試験を行った(なお、MyD88の消衰はドキシサイクリン処理の48時間後に得られる)。4つの異なる化学療法薬を用いた(シスプラチン、オキサリプラチン、エトポシド、およびパクリタキセル(Pacitaxel))。各組合せアッセイは3つのセットから構成される。
セット2=細胞+化学療法単独
セット3=細胞+ドキシサイクリン+化学療法
各セットは下記のように5種類の異なる濃度範囲を含む(二反復で実施)。
・シスプラチン処理:0−6.25−12.5−25−50−100μΜ
・オキサリプラチン処理:0−8.75−17.5−35−70−140μΜ
・エトポシド処理:0−520.5−1041−2082−4164−8328nM
・パクリタキセル処理:0−4.5−9−18−36−72nM
0日目に、HCT116 shMyd88 p53+/+またはp53−/−細胞株を96ウェルプレートに10000細胞/ウェルで播種した。
HCT116 p53+/+ shMyD88細胞をヌードマウスの側腹部に皮下注射した。腫瘍発生(200mm3)後、4群のマウスを設定した。
・ドキシサイクリン群:飲用水中のグルコース3%+ドキシサイクリン2mg/mLを受容するマウス
・シスプラチン群:飲用水中のグルコース3%と、2日ごとの用量0.5mg/kgのシスプラチンの腹腔内注射とを受容するマウス
・併用群:飲用水中のグルコース3%+ドキシサイクリン2mg/mLと、2日ごとの用量0.5mg/kgのシスプラチンの腹腔内注射とを受容するマウス
プロトコールの終了まで3日ごとに腫瘍体積を測定し、モニタリングを行った。
1)近接ライゲーションアッセイ
近接ライゲーションアッセイは、標的抗原を認識する異なる種で生じた2つの一次抗体に基づき、内因性のタンパク質−タンパク質相互作用を決定するために使用する。それぞれユニークな短いDNA鎖が結合している種特異的二次抗体(PLAプローブと呼ぶ)が前記の一次抗体に結合する。
・細胞を5分および10分間10%FCSで刺激し、4%PFAで固定し、透過処理を施し、PBS中0.3%サポニン、10%BSAでブロッキングした。
・次に、細胞をMyD88に対する一次抗体(Assay Designs)およびp−ERK(Sigma− Aldrich)とともに一晩インキュベートした。
・1倍PBSで3回洗浄した後、細胞を37℃で2時間、適当なDNA結合二次抗体(PLA)とともにインキュベートした。
・1倍PBSで2回洗浄した後、ハイブリダイゼーション溶液を37℃で15分加える。
・TBS−Tで2回洗浄した後、Duolinkポリメラーゼを37℃で90分加える。
・TBS−Tで2回洗浄した後、検出溶液を37℃で60分加える。
・その後、2分の洗浄を示したように順次行う:SSC 2倍、SSC 1倍、SSC 0,2倍、SSC 0.02倍およびEtOH70%。
・細胞をDuolink封入剤を用いてマウントする。
免疫沈降アッセイは、タンパク質−タンパク質相互作用を可視化するために使用する。
・24時間後、HEK293Tを3μgの目的プラスミドでトランスフェクトする。
・トランスフェクション48時間後に、細胞を採取し、1%NP40、20mM Tris(pH7.5)、150mM NaCl、および2mM EDTAを含有する溶解バッファーを用いて溶解する。
・溶解およびタンパク質投与の後、各条件につき1〜2mgのタンパク質を用いて免疫沈降実験を行う。
・溶解液を30μLのプロテインAセファロースビーズとともに、4℃にてローター上で30分間インキュベートする。
・これらのチューブを4℃、2000rpmで5分間遠心分離する。
・上清を新しいチューブに移す。
・予め明澄化した溶解液に2〜5μgの抗体を加える。4℃にてローター上で2〜4時間インキュベートする。
・50μLのプロテインAでファロースビーズを加える。チューブを4℃にてローター上で1時間インキュベートする。
・4℃、2000rpmで5分間遠心分離する。上清を捨て、ビーズを保持する。
・前記ビーズを1mLの溶解バッファーで3〜4回洗浄する。4℃、2000rpmで5分間遠心分離する。
・タンパク質を溶出させるために前記ビーズに50μLのLaemelli2倍バッファーを加える。
・前記2つのタンパク質間の相互作用をWBで可視化する。

Claims (15)
- 癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法であって、下記の工程:
a)少なくとも1つの候補化合物を準備する工程;
b)少なくとも1つの候補化合物を、ERK MAPKタンパク質およびMyD88タンパク質と、ERK MAPKおよびMyD88を前記候補分子の不存在下で相互作用させるのに好適な条件下で接触させる工程;
c)少なくとも1つの候補化合物の存在下および不存在下で測定した場合のERK MAPKおよびMyD88の相互作用を決定する工程;および
d)ERK MAPKおよびMyD88の相互作用を阻害する候補化合物を選択する工程
を含んでなる、方法。 - ERK MAPKおよびMyD88の一方または両方が検出可能な標識と結合されている、請求項1に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- ERK MAPKおよびMyD88の相互作用が、ツーハイブリッドシステム、アフィニティークロマトグラフィー、共免疫沈降法、細胞下分画および大分子複合体の単離、免疫ブロット法、免疫標識法、近接ライゲーションアッセイ、免疫沈降アッセイ、biacoreアッセイまたはGSTプルダウンアッセイを用いて決定される、請求項1または2に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 前記DNA損傷誘導化学療法薬がオキサリプラチン、シスプラチン、カルボプラチン、ドキソルビシン、エトポシド、ブレオマイシン、およびそれらの混合物からなる群から選択される、請求項1〜3のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 前記癌が結腸直腸癌、腸癌、黒色腫、肺癌および頸癌からなる群から選択される、請求項1〜4のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法であって、下記の工程:
a)少なくとも1つの候補化合物を準備する工程;
b)誘導プロモーターの制御下でMyD88を安定発現し、かつ、SREプロモーター、ELKプロモーターおよびMyCプロモーターから選択されるMAPK経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
c)少なくとも1つの候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
d)前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害する少なくとも1つの候補化合物を選択する工程;
e)MyD88およびMyc癌遺伝子でのトランスフェクションにより形質転換された不死化線維芽細胞を準備する工程;
f)工程d)で選択された少なくとも1つの候補化合物の存在下および少なくとも1つのDNA損傷誘導化学療法薬の存在下でフォーカス形成をモニタリングし、フォーカス形成に対する前記DNA損傷誘導化学療法薬の阻害効果を増強する候補化合物を選択する工程
を含んでなる、方法。 - 工程b)において、MAPK経路由来のプロモーターがSREプロモーターである、請求項6に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 下記の工程:
・誘導プロモーターの制御下でMyD88を安定発現し、かつ、Nf−kBプロモーターおよびISREプロモーターから選択される炎症経路由来のプロモーターの制御下でリポーター遺伝子を発現する形質転換細胞を準備する工程;
・少なくとも1つの候補化合物の存在下で前記形質転換細胞においてMYD88の過剰発現を誘導する工程;
・前記リポーター遺伝子の発現を測定し、前記リポーター遺伝子の発現を阻害しない候補化合物を選択する工程
をさらに含んでなる、請求項6または7に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。 - 前記炎症経路由来のプロモーターがNf−kBである、請求項8に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 工程b)において、前記細胞がHCT116細胞株、A375細胞株およびHeLa細胞株から選択される、請求項6〜9のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 前記誘導プロモーターが抗生物質誘導プロモーターである、請求項6〜10のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 前記不死化線維芽細胞がNIH3T3不死化線維芽細胞である、請求項6〜11のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 前記癌が結腸直腸癌、腸癌、黒色腫、肺癌および頸癌からなる群から選択される、請求項6〜12のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 前記DNA損傷誘導化学療法薬がオキサリプラチン、シスプラチン、カルボプラチン、ドキソルビシン、エトポシド、ブレオマイシンおよびそれらの混合物から選択される、請求項6〜13のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
- 前記工程a)の候補化合物が請求項1〜5のいずれか一項に記載の方法に従って選択される、請求項6〜14のいずれか一項に記載の、癌治療のためのDNA損傷誘導化学療法薬の効果を増強することができる化合物をin vitroにおいて選択するための方法。
