本発明はまた、少なくとも40℃の温度を有する水性または水含有システムにおいて微生物を抑制するのに有用である保護された抗微生物化合物を提供する。

N,N’−(ペンタン−l,5−ジイリデン)ビス(プロパン−2−アミンオキシド)の製造(IPHA付加物)
窒素入口、磁気攪拌装置、および添加漏斗を備えた250mlの丸底フラスコ(RBF)に、19.6重量%のイソプロピルヒドロキシルアミンの水溶液(IPHA、56.1g、11.0g活性物質、0.147モル)を入れる。これは、ドライアイス浴によって冷却され(3℃)、白色スラリーを提供する。50%グルタルアルデヒド水溶液(13.3g、6.66g活性物質、0.066モル、0.45当量)が添加漏斗によって添加される。グルタルアルデヒド添加が完了した直後に、グルタルアルデヒドが消費されたことを確認するために、サンプルが採取され、GCによって分析される。
グルタルアルデヒド放出の分析
IPHA−付加物のサンプルがグルタルアルデヒド含有量について分析される。サンプルは、滅菌脱イオン水中で、2000ppmグルタルアルデヒドのモル当量で調製される。標準の500ppmグルタルアルデヒドも調製される。当初測定はサンプル調製の直後に行われる。次いで、サンプルは55℃で、2時間または24時間にわたって熱老化させられ、そして再び分析される。グルタルアルデヒド濃度は直接にGCによって、およびプレカラム誘導体化の後にHPLCによって測定される。GCによってグルタルアルデヒドは検出されない。HPLCは低レベルのグルタルアルデヒドを示す。これら結果は、反応生成物が高温に対して安定であるが、誘導体化およびHPLC分析に必要な酸性条件の存在下でわずかに分解することに一致する。
殺生物有効性のアッセイ
室温での好気性生物のプールに対する、および40℃での硫酸還元バクテリア(SRB)に対する殺生物活性について、実施例1からの精製された付加物、付加物反応混合物(粗付加物)、および保護成分のみ(IPHA)が試験される。試験は以下のように行われる:
a.好気性バクテリア.
リン酸塩緩衝生理食塩水中で約5×106CFU/mLの6種類のバクテリアの混合プールが96−ウェルプレート(1mL/ウェル)に導入される。各ウェルは独立した化学物質処理(すなわち、様々な濃度での付加物、保護成分、グルタルアルデヒドなど)を受ける。次いで、各セルにおける残存細胞密度が、所定の時点で、指示薬としてレサズリン染料を含む媒体中で消滅までの希釈によって測定される。
これら付加物または保護基のいずれもグルタルアルデヒド300ppm(重量)までに相当する濃度で殺生物性ではないことが見いだされる。
T.サーモフィラス(thermophilus)の48−72時間の経過した培養物が2000gでの遠心分離によってペレットにされ、そしてそのペレットは、培養物容積の10倍の緩衝液(PBSまたは炭酸塩−緩衝合成淡水)に再懸濁される。この懸濁物はスクリューキャップガラスチューブに10mLアリコートで分配された。次いで、それぞれのチューブはグルタルアルデヒドまたは付加物で処理され、そして70℃でインキュベートされた。サンプルを逐次希釈するおよび固体培地上でのプレート希釈による消滅までの希釈によって、示された時点での各チューブにおける細胞密度が測定される。
Claims (5)
- 各出現におけるR3がC1−C4アルキルである請求項1の方法。
- 前記式Iの化合物が、N,N’−(ペンタン−l,5−ジイリデン)ビス(メタンアミンオキシド)、N,N’−(ペンタン−l,5−ジイリデン)ビス(エタンアミンオキシド)、N,N’−(ペンタン−l,5−ジイリデン)ビス(プロピルアミンオキシド)、N,N’−(ペンタン−l,5−ジイリデン)ビス(プロパン−2−アミンオキシド)、N,N’−(ペンタン−l,5−ジイリデン)ビス(2−メチルプロパン−2−アミンオキシド)、またはこれらの2種以上の混合物である、請求項1の方法。
- 前記水性または水含有システムが、油田またはガス田流体、抄紙機白水、産業用循環水、デンプン溶液、ラテックスエマルション、高温回転機械加工液、または産業用食器洗浄もしくは洗濯液である請求項1の方法。
- 前記水性または水含有システムが、破砕流体、掘削流体、水攻システムまたは油田水である請求項1の方法。
