Claims (21)
- 単離された樹状細胞前駆体集団(DC.com)であって、分葉状の核およびCD11b+/CD11c−/CD48−/MHC I−/MHC II−の特徴的な表面表現型を有し;ここで、DC.comがGM−CSFの存在下で培養された場合、そのDC.comは特徴的な樹状突起の形態を示すDCに分化し、好中球細胞および樹状細胞の両方の表現型を維持する、前記樹状細胞前駆体集団。
- 表面マーカーCD48およびGr−1を用いることにより同定される、請求項1に記載の単離された樹状細胞前駆体集団(DC.com)。
- CD48−/MHC I−/MHC II−/CD1a−/CD1d−/CD11c−の特徴的な表面表現型を有する、単離されたヒト樹状細胞前駆体集団(DC.com)。
- 請求項3に記載の樹状細胞前駆体集団であって、DC.comがGM−CSFの存在下で培養された場合に、そのDC.comが特徴的な樹状突起の形態を示す樹状細胞(DC)に分化する、前記樹状細胞前駆体集団。
- CD11c+/MHC II+/CD86+/DEC205+の表面表現型を有する顆粒球由来DC(gr−DC)を含む、請求項1に記載のDC.comに由来する単離された樹状細胞集団。
- gr−DCが、CD11c、MHC II、CD86およびDEC205の発現に関してスクリーニングすることにより同定される、請求項5に記載の樹状細胞集団。
- gr−DCが、インビトロまたはエキソビボ系において免疫応答を誘導するように抗原を提示することができる、ここで当該抗原は以下:外来性抗原、内因性抗原または自己抗原;インビトロまたはエキソビボ系において感染性病原体に対する免疫応答を誘導するように微生物抗原を提示することができる抗原;インビトロまたはエキソビボ系において腫瘍細胞に対する免疫応答を誘導するように腫瘍抗原を提示することができる抗原;インビトロまたはエキソビボ系においてウイルス病原体に対する免疫応答を誘導するようにウイルス抗原を提示することができる抗原;および、インビトロまたはエキソビボ系において免疫応答を誘導するように非微生物性外来性抗原を提示することができる抗原;の1またはそれ多くである、請求項5または6に記載の樹状細胞集団。
- gr−DCがLy6Gの表面発現、または好中球細胞および樹状細胞の両方の表面発現を示す、請求項5ないし7のいずれか1項に記載の樹状細胞集団。
- 活性化された樹状細胞を得るための方法であって、以下:
請求項1〜4のいずれか1項に記載のDC.com、およびCD11c+/MHC II+/CD86+/DEC205+の表面表現型を有するgr−DCの少なくとも一方を含む細胞の集団を提供する、および
細胞の少なくとも一部の成熟化を誘発するためにその集団の中の細胞の少なくとも一部を活性化させる、
を含む、前記方法。 - 少なくとも1個のDC.comまたはgr−DCが、以下:少なくとも1種類のウイルスまたはその派生物;少なくとも1種類の細菌またはその派生物;少なくとも1種類の寄生生物;少なくとも1種類の菌類またはその派生物;少なくとも1種類のサイトカインまたは少なくとも1種類のリガンド;の1又はそれより多くにより活性化される、請求項9に記載の方法。
- Tリンパ球を含む生物学的試料に対して実施される、請求項9に記載の方法。
- その試料が、自家血液を含む血液である、請求項11に記載の方法。
- その樹状細胞がヒトのものである、請求項9に記載の方法。
- 樹状細胞系を単離するためのインビトロの方法であって、以下:
対象から単離した血液試料を有効量のGM−CSFを含む適切な培地の中で培養状態に置く、
単離した血液試料から、請求項1〜4のいずれか1項に記載のDC.comを生成する、
および
対象に特有の樹状細胞系を得るために、DC.comを連続的な細胞分裂により増殖させる;
を含む、前記方法。 - 生成されたDC.comがCD48陰性/MHCクラスI陰性の表現型を有する、請求項14に記載の方法。
- 樹状細胞へ方向付けされた請求項1〜4のいずれか1項に記載のDC.comの拡張のための方法であって、単離されたDC.comをGM−CSF中で培養する、および場合によりその培養物に骨髄(BM)フィーダー細胞を添加する工程を含む、前記方法。
- 哺乳類の前駆体樹状細胞から請求項5または6に記載の樹状細胞集団を生成するための方法であって、以下:
i)哺乳類の血液試料から前駆体樹状細胞を含む細胞画分を用意する;
ii)次のことにより、工程(i)の細胞画分の少なくとも1サブセットを単離する:
工程i)からの集団を、CD45.1+である新しく単離したBM細胞とGM−CSFの存在下で共培養する;
抗CD45.2および抗CD45.1抗体で分染することにより識別された請求項1に記載のDC.comに由来する細胞を識別する;そして
CD45.2+細胞を1種類以上の示されたマーカーの表面発現に関して分析する;ならびに
iii)工程(ii)により得られた細胞を集める;
を含む、前記方法。 - 樹状細胞(DC)のT細胞との相互作用の作用因子を同定する方法であって、以下:
i)請求項5または6に記載の樹状細胞集団をT細胞および試験薬剤と混合する、ならびに
ii)候補物質が樹状細胞集団のT細胞との相互作用を変化させるかどうかを決定する、
ここで、樹状細胞集団のT細胞との相互作用を変化させる試験薬剤は樹状細胞の相互作用の作用因子である;
を含む、前記方法。 - 試験薬剤が、樹状細胞集団のT細胞への結合における変化、および樹状細胞集団に仲介されるT細胞の活性化における変化、の1又はそれより多くにより同定される、請求項18に記載の方法。
- 対象における炎症性の疾患を検出する、当該障害を発症するリスクを検出する、または当該障害の予後を検出する方法であって、以下:
対象からの検査試料においてCD11c、MHC II、CD86およびDEC205のマーカーのレベルを測定する、
ここで、検査試料におけるそのマーカーのレベルの、対照試料における対応するマーカーのレベルと比較した変化は、対象における当該障害の存在、または当該障害を発症するリスクの指標である、
を含む、前記方法。 - 炎症性障害を処置するための療法薬のための候補化合物に関するスクリーニングのためのキットであって、次のもの:CD11c、MHC II、CD86およびDEC205のマーカーの試薬;ならびに、該マーカーを発現する細胞、を含む前記キット。
