この方法は、普通の蛍光体を使用し、蛍光体の大半を三重項等の準安定暗状態に「切り替え」、残った蛍光体又は自然に基底状態に戻った蛍光体の位置を計算することに基づく遠視野蛍光ナノ顕微鏡法に関する。(用語「切り替え」は本明細書において非常に広い意味で使用されるため、より特定的な意味での切り替え可能であることが既知の蛍光体を特に指すものではない)。単一のレーザによる連続した広視野照明及び連続動作するカメラが、ローダミン及び蛍光タンパク質で標識された(生きている)試料の二色画像をもたらし、単純であるが、それでも強力な超高分解能手法を証明する。

図5は、PVA内のAtto532(黒い四角)及び酸素スカベンジャー系を含む水性緩衝液内に埋め込まれたPtK2細胞内のAtto532(白い丸)−免疫染色された微小管の基底状態抑制後に回復した蛍光信号の部分を示す。プローブ−ポンプ−プローブモードでの蛍光回復を測定した。第1のプローブパルス(532nm)は、抑制前に参照信号を確立した。抑制は、PVAの場合では、焦点強度I=1kW/cm2、水溶液の場合では焦点強度I=100kW/cm2の10msパルス(532nm)により行い、その後、第2のプローブパルス(532nm)が0.5ms間にわたって続き、I=100W/cm2を生成する。第2のプローブパルスは、抑制パルスから10msだけ遅れた。ポンプと第2のプローブパルスとの間の期間中の強度I405nm(a)の405nm光照明及び強度I671nmの671nm光照明(b)により、上記でグラフ化したように、抑制と第2のプローブパルスとの間の時間期間中の蛍光回復部分が増大した。暗状態での染料の405nm光又は671nm光の吸収は、逆系間交差の場合に観測されるように1,2、より高い励起状態を介して暗状態低減を誘導し得る。暗状態吸収及び一重項系への逆交差がより強いことにより、且つ/又は複数の長く続く暗状態の存在により、効果はPVAにおいてより強力であり得る。
暗状態数[D]及び回復時間τは、ポンプ−プローブ測定において染料アンサンブルで検出された蛍光信号から特定された。回復データの時間依存性は拡張指数関数(〜exp(−(t/τ)α))に従い、指数α=0.6〜0.8は、単一指数(mono−exponential)回復(α=1)から有意に逸脱しており、いくつかの暗状態を示す3,4.有機染料(Bodipyまで表を参照)の場合、PVA内の純粋な染料の測定及び酸素スカベンジャー系を含む緩衝液を有する水性環境内のPtK2細胞内の免疫染色された微小管の測定を実行した。蛍光タンパク質EGFP、EYFP、Citrine、及びPhiYFPの場合、測定は、ポリ−L−リシンによりガラス表面上に固定され、PBS緩衝液(水性)で覆われた精製タンパク質に対して実行された。Citrineの場合、細胞内で実行された測定の結果は、図2f、gに示される試料に適用して追加された。励起は1532nm、2488nm、及び3561nmであった。純水環境と比較して、細胞内のCitrineのτの甚大な増大の理由は、pH又は分子酸素濃度の変更に起因し得、これらは両方とも、三重項状態又は異なるプロトン付加状態等の蛍光タンパク質の暗状態の性質に影響する4.細胞環境内のCitrineの暗状態の寿命τがより長いことにより、退色の確率も増大する。退色の顕著な経路は、蛍光体の暗状態において光子が吸収され、それにより、反応性の高い、より高い励起エネルギーレベルに励起することである5,6。
蛍光染料。蛍光染料ローダミン6G(Rh6G)、ローダミン110(Rh110)、ローダミン123(Rh123)、Atto532、Atto565、Alexa488、Oregon Green 488、Texas Red、蛍光イソチオシアネート(FI)TC)、及びBodipy(FL)は、異なる供給業者(MoBiTec、Goettingen、Germany;AttoTec、Siegen、Germany;Sigma−Aldrich;Invitrogen;又はRadiant Dyes、Wermelskirchen、Germany)から購入された。ローダミン誘導体Rh―sart3b及びRh―sart3f(1,2,3,4―テトラヒドロキノリン−7−オールから得られる、強化キサンタン断片を有するスルホン化ローダミン)は、化合物3b及び3fとして参照[7]に提示された。ポリ(ビニル−アルコール)(PVA)でのアンサンブル測定の場合、染料の緩衝貯蔵液を(5%)PVA内で希釈して、〜10−6Mの濃度の最終染料にし、PVA試料を顕微鏡のカバーガラス上にスピンコートした。マーキングした場所に、10%(v/v)のβ−メルカプトエタノール(Fluka)をスピンコート前に添加した。
タンパク質精製。蛍光タンパク質EGFP9、EYFP(Clontech、Mountain View、Ca)、Citrine10、及びPhiYFP(Evrogen、Moscow、Russia)のプラスミドコードを大腸菌BL21DodonPlus RIL(Stratagene、La Jolla、CA、USA)内に転移させた。タンパク質発現のため、プラスミドを保持する細胞を、100μg/mlのアンピシリンを含有するLB培地内で、OD600が0.6になるまで成長させ、1mMのイソプロピル−1−チオ−L−D−ガラクトプラノシドを使用して誘導した。25°Cで6時間インキュベートした後、生成された生物量を遠心分離によりペレット化し、プロテアーゼ阻害剤と共にPBS内に再懸濁させ、超音波処理した。遠心分離により溶菌液を除去し、Ni−ニトリロ三酢酸(NTA)−アガロースカラム(Qiagen、Hilden、Germany)の上澄みから蛍光タンパク質を精製した。アンサンブル測定のために、100μlのポリ−L−リシン溶液(Sigma−Aldrich、Steinheim、Germany)を顕微鏡カバースリップ上にコーティングした。水で手短にすすいだ後、50μlのタンパク質溶液(PBS pH7.4内)を添加した。1時間後、未結合タンパク質を水で手短にすすいで除去し、10μlのPBS(pH7.4)を添加した。最後に、試料をシリコーン接着剤で封入した。

Claims (18)
- 試料内の対象構造を高空間分解能で撮像する方法であって、
励起して蛍光可能な第1の蛍光性を有する第1の状態及び暗状態である第2の蛍光性を有する第2の状態を有し、一波長の光により励起して自然に蛍光を発することができ、前記一波長の光により前記第1の状態から前記第2の状態に変換することができ、且つ、前記第2の状態から前記第1の状態に回復することができる物質群から物質を選択するステップと、
前記試料内の対象構造を前記物質の分子でマーキングするステップと、
前記試料を、前記試料内の前記物質の最近傍分子の平均間隔よりも大きい空間分解能限界で、センサアレイ上に結像するステップと、
前記センサアレイ上への前記試料の結像の空間分解能限界よりも大きな寸法を有する前記試料の少なくとも1つの領域を前記少なくとも一つの領域内で略一定強度の前記一波長の光に露出させ、順次前記分子の一部を前記一波長の光で励起し自然に蛍光を放射させて前記第2の状態に変換することにより、前記第1の状態にある前記分子の割合を減らし、前記第1の状態にある前記分子のうちの少なくとも10%が前記第1の状態にある他の分子から前記空間分解能限界よりも離れているようにする、前記分子の一部を前記第2の状態に変換するステップと、
前記試料を前記一波長の光に連続露出させている間、前記分子の前記領域から自然に発せられた蛍光を、前記センサアレイにより記録される複数の画像に記録するステップと、
前記第1の状態にあり、且つ、前記第1の状態にある他の分子から前記空間分解能限界よりも離れている分子の前記試料内の位置を、前記センサアレイにより記録された前記画像のそれぞれから特定するステップと、
を有し、
前記第1の状態は前記物質の一重項状態であり、前記第2の状態は前記物質の三重項状態であり、
前記分子の一部を前記第2の状態に変換するステップと、前記分子の一部から自然に発せられた蛍光を前記画像に記録するステップは同時に行われる、
ことを特徴とする、方法。 - 前記一波長の光への前記試料の露出開始時において、前記物質の分子の90%を超える部分が前記第2の状態に変換されるように、前記一波長の光の強度を高く設定することを特徴とする、請求項1に記載の方法。
- 前記一波長の光への前記試料の露出開始時において、前記物質の略すべての分子が前記第2の状態に変換されるように、前記一波長の光の強度を高く設定することを特徴とする、請求項2に記載の方法。
- 前記物質が、
前記第2の状態から前記第1の状態に、自然に戻る物質、
前記第2の状態から前記第1の状態に、前記一波長の光の動作により戻る物質、及び、
前記第2の状態から前記第1の状態に、自然にも戻るし、前記一波長の光の動作によっても戻る物質、
を含む物質の部分集合から選択されることを特徴とする、請求項1〜3のいずれか一項に記載の方法。 - 前記試料内の前記物質の前記第2の状態の寿命を変更する少なくとも1つの対策が実施されることを特徴とする、請求項1〜4のいずれか一項に記載の方法。
- 前記試料内の前記物質の前記第2の状態の寿命を延ばす少なくとも1つの対策が実施されることを特徴とする、請求項5に記載の方法。
- 前記画像が記録される前に、前記物質の部分が、前記一波長の光及び別の波長の光から選択される高強度光による退色により、前記第1の状態及び前記第2の状態と異なる永久的な暗状態に変換されることを特徴とする、請求項1〜6のいずれか一項に記載の方法。
- 前記一波長の光の強度が、前記画像の記録中、一定値に設定されることを特徴とする、請求項1〜7のいずれか一項に記載の方法。
- 前記一波長の光の強度が、前記画像の記録中、前記画像の記録順に伴って時間変調された強度プロファイルに設定されることを特徴とする、請求項1〜7のいずれか一項に記載の方法。
- 前記一波長の光が、前記試料の領域上に連続して向けられることを特徴とする、請求項1〜9のいずれか一項に記載の方法。
- 前記一波長の光は、前記画像の記録中に消失しない高速パルスで前記試料の領域に向けられることを特徴とする、請求項1〜9のいずれか一項に記載の方法。
- 記録された個々の前記画像の前記第1の状態にある前記分子の各々が、前記物質の分離不可能な蛍光分子、すなわち、前記第1の状態にある他の分子から前記空間分解能限界よりも離れていない蛍光分子であるか否かに関してオンラインで評価されること、及び前記分離不可能な蛍光分子の密度が特定の閾値を下回るまで、前記一波長の光の強度が変更されることを特徴とする、請求項1〜11のいずれか一項に記載の方法。
- 記録された個々の前記画像の前記第1の状態にある前記分子の各々が、前記物質内の分離可能な蛍光分子、すなわち、前記第1の状態にある他の分子から前記空間分解能限界よりも離れている蛍光分子であるか否かに関してオンラインで評価されること、及び前記分離可能な蛍光分子の密度が閾値に達するまで、前記一波長の光の強度が変更されることを特徴とする、請求項1〜11のいずれか一項に記載の方法。
- 前記一波長の光への前記試料の露出開始時に、前記試料内の前記物質全体の蛍光の強度分布が、前記空間分解能で前記センサアレイにより記録されることを特徴とする、請求項1〜13のいずれか一項に記載の方法。
- 前記試料の同じ領域のさらなる画像の記録の終了基準が、前記試料内の前記物質全体の蛍光の前記強度分布に基づいて定義されることを特徴とする、請求項14に記載の方法。
- 前記複数の画像の1つに登録された前記物質の分子の各位置が、前記試料を前記センサアレイ上に結像する点拡がり関数、又はそこから導出される関数を使用して畳み込まれること、及び前記畳み込みによって再構築された結果が初期記録強度分布と比較されることを特徴とする、請求項15に記載の方法。
- 前記試料が生物学的試料であって、前記試料を遺伝子技術を使用して改変することにより、前記試料自体が前記物質の性質を発現し、前記試料の対象となる構造が前記物質でマーキングされることを特徴とする、請求項1〜16のいずれか一項に記載の方法。
- 前記試料が生物学的試料であって、前記物質用または前記物質に結合するリンカ用の特定の結合部位を有するタンパク質を発現するように、遺伝子技術を使用して前記試料を改変することにより、前記試料の対象となる構造が、前記物質でマーキングされることを特徴とする、請求項1〜16のいずれか一項に記載の方法。
