Description
a.13.5日齢マウス胚の嗅球から神経幹細胞(neural stem cells:NSC)を含
むニューロスフィアを産生する工程と、
b.生存ニューロスフィア細胞によって4ヶ月齢の雄のアルメニアンハムスターを免疫する工程と、
c.上記動物の脾臓を抽出してリンパ球を取得する工程と、
d.リンパ球と、非産生マウスミエローマ細胞とを融合して、ハイブリッド細胞またはハイブリドーマを生み出す工程と、
e.ヨウ化プロピジウムの存在下で、死細胞が分析から除外されるニューロスフィア細胞フローサイトメトリーを用いて、ハイブリドーマによって産生される抗体の決定および/または選択を行なう工程とを含む、前駆細胞の膜抗原を特異的に認識するモノクローナル抗体を産生する方法を提供する。

b)国際寄託当局デュスチェ サムラング フォン ミクルールガニスメン ウンド ゼルクルツレン GmbH(DSMZ)、ブラウンシュワイク、ドイツにアクセスDSM番号No.ACC2881で2008年2月4日に寄託されたハイブリドーマによって産生される、NILO2(クローン2 B7.10)と称されるモノクローナル抗体、
などから提供され、上述の処理によって取得される。
a)細胞集団と、モノクローナル抗体NILO1および/またはモノクローナル抗体NILO2によって認識される細胞表面マーカー中のエピトープまたは抗原決定基を認識する試薬とを接触させる工程と、
b)上記試薬と、上記エピトープまたは抗原決定基との接触がある細胞を選択する工程と、
を含む、幹細胞または前駆細胞が非常に豊富な集団を産生する方法について述べる。

本明細書において用いられる用語“抗体”は、免疫グロブリン分子および免疫グロブリン分子の免疫学的に活性な部分、すなわち表面マーカーまたはエピトープに対して特異的に結合する(免疫反応を伴って)抗原固定部位を含んでいる分子に関する。免疫グロブリン分子の免疫学的に活性な部位としては、例えば、ペプシンなどの酵素による処理抗体を産生し得る、フラグメントF(ab)およびF(ab’)2を含む。

以下、本発明者らによって行なわれるアッセイを用いて本発明について説明する。本発明者らは、モノクローナル抗体NILO1およびNILO2の特異性および有効性を明らかにしている。
本発明者らは、神経幹細胞に対するモノクローナル抗体を取得する方法を示す。当該方法は、抗原刺激源としての、13.5日胚細胞由来のニューロスフィアの利用、および免疫化のレセプターとしての、特異的なパターンを用いた3カ月齢未満のアルメニアンハムスターの利用を含む。ハムスター起源のモノクローナル抗体を分泌し、神経幹細胞に対して特異性を有する種属間ハイブリドーマの特異的な選択培地の選定の後、脾臓および脾臓とマウスミエローマ細胞との融合物を取得することは、胚起源および成体動物からの神経幹細胞を共に許容する。
ニューロスフィア由来の神経幹細胞(NSC)を用いた。ニューロスフィアとして成長するこの細胞は脱凝集することがあり、新たなニューロスフィアを形成する能力がある培養組織においてクローン化され得る。ニューロスフィアの一部を形成するこの細胞は、“インビトロ”の培養組織中で、ニューロンおよびグリア(アストロサイトおよびオリゴデンドロサイト)などの脳の様々な細胞型に分化する能力があるような神経前駆細胞と考えられる。さらに、これらの細胞は“ネスチン”などの規定された細胞内マーカーを有し、ニューロン、オリゴデンドロサイトまたはアストロサイトなどの任意の神経系統の細胞に分化し得る。
マウス神経前駆細胞に対するモノクローナル抗体を得るために、4ヶ月齢の雄のアルメニアンハムスターを用いる。独立した部屋および穏やかな環境であるが、他のマウスと同様に、通常の温度および湿度条件の飼育小屋で、動物を飼育する。また、アルメニアンハムスターが他のマウス抗原に対するモノクローナル抗体を取得するために用いられているとはいえ、今日、アルメニアンハムスターが神経成分に対する抗体を産生するために用いられているのでは決してない。重要なのは、アルメニアンハムスターが用いられて、他のハムスター種が同じように用いられないことである。
融合の手順は他の著者によってこれまでに示されるものに類似する。簡単に言うと、NSCによって免疫されたハムスターの脾臓由来のリンパ球を培地中で洗浄し、血清を含まないDMEMにおいて再懸濁する。マウスミエローマ細胞と、融合されることになる細胞とを50mlファルコンチューブの中で混合する。融合では、P3−X63.Ag8.653と称されるBalb.cから得た107非産生ミエローマ細胞と、免疫されたハムスターの脾臓から得た106細胞とを使用する。これらの細胞を室温で5分間、1500r.p.mで同時に遠心分離にかけた。浮遊物を吸引によって除去してすべて取り除く。細胞だけを含むチューブを37℃の湯浴中に置く。細胞懸濁液を時々ゆっくりとかき混ぜながら、1分間に0.4mlのポリエチレングリコール(水に40%PEG)を一滴ずつ添加する。その後、予め37℃に調節された、血清を含まないDMEM培地5mlを3分以下で一滴ずつゆっくりと希釈した。懸濁液を穏やかに混合し、DMEM−10%FCS完全培地で完成させて、37℃で30〜45分間インキュベートする。そして、細胞を平底96ウェルプレートに0.1ml/ウェルで分配し、37℃、5%CO2に設定されたインキュベーターにおいて保持する。融合から1日後、HA(2X)を含有しているDMEM−10%FCS培地を0.1ml/ウェルずつ加える。この培地を求め、週に一度交換する。ハイブリッドを目視により、または低分解能の倒立顕微鏡によって検出する。融合から3週間後、1ウェルにつき2mlの選択培地を有する24ウェルプレートにコロニーを移植する。
選択は、NSC細胞に対するフローサイトメトリーにおける、本発明の抗体によって陽性標識された細胞の検出によってなされる。NSC細胞の誘導体は;1)13.5日齢胚から得た嗅球ニューロスフィア、2)成体マウスのニューロスフィア、3)BW5147マウスから得た胸腺腫瘍細胞、3)成体マウスの骨髄細胞である。
NILO1およびNILO2は、ハムスターのBリンパ球と、Balb/cマウス由来の非産生ミエローマP3− P3−X63.Ag8.653.653とのハイブリドーマによって産生される、ハムスター免疫グロブリンIgGである。種属間の融合(ハムスター×マウス)であるにも関わらず、ハイブリドーマの安定性は選択およびクローニングを保証するほど十分高い。NILO1ハイブリドーマおよびNILO2ハイブリドーマによって産生されるモノクローナル抗体は、G−タンパク質またはA−タンパク質とのセファロースカラム結合によって精製する。これにより、抗体の純度を保証し、様々な色素(フルオレセイン、フィコエリスリンなど)によって直接標識することが可能になる。現在、NILO抗体はFITCによって、およびPEによって標識される。
NILO抗体は、成体マウス脳の神経原性領域における限定的に存在することによって証明されるように、神経幹細胞に特異的である。主に、NILOに対してポジティブな細胞は、成体マウスのSVZ領域および海馬の歯状回において検出されている。
これら抗体のうちNILO抗体免疫沈降によって認識される抗体を同定することは、神経前駆細胞において、NILO抗体によって認識される抗原のおおよその大きさを知るために行なわれている。神経前駆細胞は、ビオチンによって、またはNILO2およびNILO2抗体によって認識される代替可能なC2C12系統によって、細胞表面において標識されて用いられる。ビオチンによる細胞標識の後、抗体は結合され、続いてBriji界面活性剤を用いる材料および方法(Material and Methods using the Briji detergent)において述べられているように溶解される。ニューロスフィア細胞およびC2C12系統のいずれにおいても、NILO2によるタンパク質免疫沈降では140kDおよび170kDaの2つに大多数のバンドがある。一方、NILO1は“ウェスタンブロット”に用いられなくてよい。これらのデータは、免疫組織化学に基づくこれら抗体の類似の位置に依存せず、各モノクローナル抗体が前駆細胞の細胞表面タンパク質の異なる型を同定することを証明する。極めて関連性のある状況では、前駆細胞の細胞表面タンパク質の異なる型がNILOによって同定される表面抗原(細胞レセプター)により同定される。
NILOの細胞分布は、神経幹細胞において均一である。神経幹細胞は、その前駆細胞の状態で維持される。この状態は、繊維芽細胞成長因子2(fibroblast growth factor 2:FGF2)および表皮成長因子(epidermal growth factor:EGF)などの成長因子の存在下における厳密な細胞培養条件のもと、ウシ胎児血清(fetal calf serum:FCS)の非存在下で保持されることでもたらされる。これらの因子、すなわちFGFおよびEGFの非存在下で、且つ、FCSの存在下において細胞は増殖を停止し、任意の神経系統細胞:すなわち、ニューロン、オリゴデンドロサイト、またはアストロサイトに分化する。
NILOモノクローナル抗体を有する他の組織の分析は、神経領域の外側にこれら抗体によって同様に標識される他の少数集団が存在することを証明した。特に、NILO1抗体によって同定される2つの前駆細胞集団は、骨髄の間葉前駆細胞および乳房組織の前駆細胞である。
6週齢マウスの乳房組織の脱凝集状態で、NILO1にわずかな細胞(<3%)を同定する能力があることが観察されている。マンモスフィアを形成する能力を有するこれらの細胞は、Sca−1によって弱く標識されていることから、前駆細胞であり得ることを示す(図11b)。この議論を証明するために、脱凝集した乳房組織細胞をNILO1抗体によって標識し、無菌状態で分離した。続いて、NILO1陽性細胞およびNILO1陰性細胞を実験用マウスの胸腺脂肪中の乳房前駆細胞の移植に用いた。そして、3次元乳房構造の有無を移植後8週目に分析した(図11c)。観察されるように、サイトメトリーを用いて選択した1200セルのNILO1細胞は、3次元乳房構造を再構築する能力があった。これらのデータは、NILO1が乳房組織前駆細胞の構造を認識することを示すと思われる。

No.ACC2881で寄託されたハイブリドーマによって産生される、NILO2と称されるモノクローナル抗体、
から選択されるモノクローナル抗体。 - 請求項2に記載のいずれかのモノクローナル抗体の抗原結合フラグメント。
- 請求項2に記載の抗体、または請求項3に記載の抗原結合フラグメントに転写する能力がある、DNAの遺伝子構築物。
- 請求項2に記載のいずれかのモノクローナル抗体の生成ハイブリドーマ。
- 前駆細胞が豊富な集団を産生する方法であって、
a.細胞集団と、試薬とを接触させる工程であって、
上記試薬は、
i.国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2887で寄託されたハイブリドーマによって産生される、NILO1と称される抗体またはその抗原結合フラグメント、および
ii.国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2881で寄託されたハイブリドーマによって産生される、NILO2と称される抗体またはその抗原結合フラグメント:
を含むリストから選択され、
b.上記試薬と、エピトープまたは抗原決定基との接触がある細胞を選択する工程とを含む方法。 - 前駆細胞が豊富な上記集団は、神経前駆細胞または神経誘導細胞を含む、請求項6に記載の方法。
- 神経前駆細胞または神経誘導細胞を含んでいる上記集団は、ニューロスフィアの培養組織由来である、請求項7に記載の方法。
- 上記試薬は、酵素、発色団、化学発光材料、放射性核酸またはナノ粒子などのマーカーと結合される、国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2887で寄託されたハイブリドーマによって産生される、NILO1と称されるモノクローナル抗体および/または国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2881で寄託されたハイブリドーマによって産生される、NILO2と称されるモノクローナル抗体を含む、請求項6〜8のいずれか一項に記載の方法。
- 前駆細胞の選択は、フローサイトメトリー技術(FACSフローサイトメトリーを含む)もしくは顕微鏡検査を用いて、免疫細胞化学技術(細胞)もしくは免疫組織化学(組織)を用いて、または磁性選択法によって行なう、請求項6〜9のいずれか一項に記載の方法。
- 薬剤をスクリーニングまたは発見する方法であって、以下の工程:
a.国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2887で寄託されたハイブリドーマによって産生される、NILO1と称されるモノクローナル抗体および/もしくは国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2881で寄託されたハイブリドーマによって産生される、NILO2と称されるモノクローナル抗体、またはそれらの、1つの抗原結合フラグメントもしくは複数の抗原結合フラグメントと結合される、幹細胞または前駆細胞を含む集団から選択し、幹細胞に関する濃縮された集団を産生する工程と、
b.上記濃縮された集団を非ヒト哺乳動物に接種する工程と、
c.上記哺乳動物、および上記集団を接種されていない別の非ヒト哺乳動物(対照被験体)に対して、薬学的な潜在性を有する組成物を投与する工程と、
d.両哺乳動物における上記投与の作用を比較する工程とを含む方法。 - 薬剤をスクリーニングまたは発見する方法であって、以下の工程:
a.国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2887で寄託されたハイブリドーマによって産生される、NILO1と称されるモノクローナル抗体またはその抗原結合フラグメントと結合される、幹細胞または前駆細胞を含んでいる集団から選択し、国際寄託当局DSMZ、ブラウンシュワイク、ドイツにDSMアクセス番号No.ACC2881で寄託されたハイブリドーマによって産生される、NILO2と称されるモノクローナル抗体またはその抗原結合フラグメントと結合される、幹細胞または前駆細胞のさらなる選択によって上記集団をさらに濃縮し、原始細胞の集団と比較した場合、幹細胞に関する濃縮された集団を産生する工程と、
b.幹細胞または前駆細胞に関する上記濃縮された集団を、非ヒト哺乳動物に接種する工程と、
c.上記哺乳動物、および上記集団を接種されていない別の非ヒト哺乳動物(対照被験体)に対して、薬学的な潜在性を有する組成物を投与する工程と、
d.両哺乳動物における上記投与の作用を比較する工程とを含む方法。 - 上記非ヒト哺乳動物はげっ歯動物である、請求項11または12に記載の方法。
- 上記幹細胞または前駆細胞は、神経幹細胞または神経前駆細胞である、請求項11または12に記載の方法。
