Description
フェノール分解活性が良好な、7種類の異なる安水処理活性汚泥(A〜G)、及びフェノール分解活性が不調な3種類の異なる安水処理活性汚泥(H〜J)を、それぞれ製鉄所の安水処理活性汚泥槽から採取した。採取した各安水処理活性汚泥を遠心分離により沈降した活性汚泥を回収した。
図2に示した容積2.7m3の活性汚泥槽で流量1.3L/minでHRT36時間の条件で、表1の組成の模擬安水の処理を行った。経時的に処理水中の残存CODを測定した。図3に示すように、運転開始後126日目から154日目にかけて処理水の残存CODが徐々に増加し、活性汚泥の性能の悪化が起こった。そこで、運転開始後154日目の、活性汚泥に含まれる、安水処理活性汚泥に特徴的なカテコール分解酵素の遺伝子の数量を以下のようにして調べた。

Claims (2)
- 安水処理活性汚泥から抽出、精製したDNAを鋳型として、配列表の配列番号1に示される塩基配列からなる核酸分子、及び、配列番号2に示される塩基配列からなる核酸分子をプライマーとして用いたPCRにより、前記安水処理活性汚泥に含まれるカテコール分解酵素の遺伝子の数量を調べ、当該遺伝子の数量が、少なくとも10 6 コピー/mLである場合に、安水処理活性汚泥の状態を良好であると診断することを特徴とする安水処理活性汚泥の診断方法。
- 請求項1に記載の安水処理活性汚泥の診断方法を用いる安水処理活性汚泥の操業管理方法であって、前記カテコール分解酵素の遺伝子の数量が10 6 コピー/mL未満である場合に、当該案水処理活性汚泥に、カテコール分解酵素の遺伝子の数量が10 6 コピー/mL以上の安水処理活性汚泥を加えることを特徴とする安水処理活性汚泥の操業管理方法。
