Description
共焦点レーザ走査顕微鏡観察(LSM)は、本願発明によって使用可能なパーフルオロカーボンナノカプセルの大きさの測定及び均一構造の証明に使用される。このために、ナノカプセルは、ナイルレッド蛍光染色(9−ジエチルアミノ−5H−ベンゾ[α]フェノキサジン−5−ワン)で染色され、その親油性によってカプセル壁に蓄積される。LSM測定は、ヘリウム/ネオンレーザ及び高解像度レンズを使用して実施される。染色されたパーフルオロカーボンナノカプセル及び生理的バッファによって希釈されたパーフルオロカーボンナノカプセルは、ポリ−L−リジンで処理されたスライド表面で改質されたペンツ室に沈着し、蛍光発光が、液柱のいろいろな焦点面に表示される。共焦点システムの膜厚は、光学的に大変小さいので、パーフルオロカーボンナノカプセルの少数の空間表現を示すことも可能である。それらの大きさ及び均一性は、LSM510システムの対応する「生理学的評価」ソフトウェアを使用して検出された信号に基づいて数量化される。
Claims (8)
- 輸血のための代用血液としての貯蔵可能な人工酸素キャリアの分散において、前記分散が、可逆的酸素貯蔵能力を有するカプセルを含むこと、
該カプセルが、フッ素化炭化水素、パーフルオロカーボンを含有する及び/若しくは包含する酸素透過性カプセル材料を具備し、
前記カプセルが50〜1000nmの範囲内の直径を有し、
前記酸素透過性カプセル材料は、酸素透過性有機ポリマーを具備すること若しくはそれからなること、
前記ポリマーは、ポリ(ラクチド−コ−グリコリド)、ポリアルキルシアノアクリレート及びそれらの混合物からなる群から選択され、
前記酸素透過性カプセル材料が、硫化物架橋を含まず、
前記カプセル内のフッ素化炭化水素に対する酸素透過性カプセル材料の重量比が、75:25〜10:90の範囲内であり、
前記フッ素化炭化水素が、250〜2000g/molの範囲内の分子量を有し、且つ、液体形状であり、
前記分散が、分散に対して、1〜70容量%の容量に関連した量でカプセルを含み、
前記分散が、生理学的pHに調整され、
前記分散は、10℃〜40℃の範囲内の温度に対して、0.1〜1.8sの範囲内の特別な粘度ηを有することをし、
前記分散は、水性ベースの分散の形であること、且つ又は、前記分散が連続相として水を含むものであり、生理学的及び/若しくは生理食塩水の形であることを特徴とする分散。 - 前記カプセルが、コア/シェルカプセルとして構成され、酸素透過性カプセル材料が、カプセルシェルを形成すること、フッ素化炭化水素が酸素透過性カプセル材料によって囲まれること若しくは包含されること、及びフッ素化炭化水素がコア/シェルカプセルの内部を形成すること、又は、
前記カプセルが、マトリクスカプセルの形であること、前記酸素透過性カプセル材料がマトリクスを形成すること、及び、フッ素化炭化水素が、酸素透過性カプセル材料に基づくマトリクスに、均質及び/若しくは均等に分布されるように沈着されることを特徴とする請求項1記載の分散。 - 前記酸素透過性カプセル材料が、酸素透過性有機ポリマーを具備すること若しくはそれからなること、
前記ポリマーは、全フッ素置換化合物で及び/若しくはポリアルキレングリコールで誘導体化されるポリ(ラクチド−コ−グリコリド)、及び、フッ素化されるポリアルキルシアノアクリレート及びそれらの混合物からなる群から選択されることを特徴とする請求項1又は2記載の分散。 - 前記酸素透過性カプセル材料が、酸素透過性有機ポリマーを具備する若しくはそれからなること、該有機ポリマーが、エマルジョン重合、界面重合若しくは界面析出によって得られる若しくは製造されること、を特徴とする請求項1〜3のいずれか1つに記載の分散。
- 前記カプセル内のフッ素化炭化水素に対する酸素透過性カプセル材料の重量比が、75:10以下であること、且つ/又は、フッ素化炭化水素に対する酸素透過性カプセル材料の重量比が、70:30〜15:85の範囲内で変化することを特徴とする請求項1〜4のいずれか1つに記載の分散。
- 前記分散が、分散に対して、5〜65容量%に関連した量でカプセルを含むことを特徴とする請求項1〜5のいずれか1つに記載の分散。
- 前記分散が、分散に対して、5〜80重量%の質量に関連した量でカプセルを含むことを特徴とする請求項1〜6のいずれか1つに記載の分散。
- 前記分散が、生理学的イオン強度及び/若しくは生理学的モル浸透圧濃度及び/若しくは生理学的イオン構成に調整されることを特徴とする請求項1〜7のいずれか1つに記載の分散。
