Description
(1) K7株一倍体の選抜
ランダム胞子分離法により、実用醸造酵母株きょうかい7号(K7株)二倍体から、二倍体と同等の醸造能力を示す一倍体を選抜した。具体的方法は以下のとおりである。
抗生物質nourseothricin(商品名cloNAT)耐性遺伝子を用いた公知の薬剤耐性遺伝子カセットnatMX4(Yeast, 15(14):1541-1553 (1999))を用いて、Fis1遺伝子破壊カセットを調製し、これを用いて868株のFis1遺伝子を破壊した。
60gのアルファ化米、23gの乾燥麹、200mlの水、45μLの乳酸に、2×109個のFis1遺伝子破壊酵母を添加して、15℃で15日間発酵させることにより、清酒を製造した。比較として、Fis1非破壊の親株868株を同様に用いて清酒を製造した。清酒中の各種有機酸含有量を調べた結果を図1に示す。試験は、3つの独立したFis1遺伝子破壊株を用いて行ない、有機酸の含有量は3回の実験の平均値として示した(図1)。
Claims (3)
- ミトコンドリア外膜タンパク質Fis1をコードするFis1遺伝子が破壊された醸造酵母を用いてアルコール発酵を行うことを含む、リンゴ酸の含有量が高い酒類の製造方法。
- 前記酒類はコハク酸及び/又は酢酸の含有量が低い酒類である請求項1記載の方法。
- 前記Fis1遺伝子は配列表の配列番号1に示される塩基配列から成る請求項1又は2記載の方法。
