Description
薬物療法としては、鎮痛剤、経口避妊薬などの対症療法、ホルモン療法などの本症治療薬による投与があげられ、外科的療法としては、保存手術、根治手術などがあげられる(非特許文献1)。しかしながら、外科的療法も不十分な症例があり、また子宮内膜症を根治させる薬剤はこれまでに開発されていない。

[図2]は、マウス子宮内膜症モデルにおける抗マウスIL−5抗体の子宮内膜症病変形成抑制作用を示した図である。縦軸は、病変サイズ(mm2)を示す。
IL−5とIL−5受容体との結合を阻害する抗体としては、IL−5に結合し、かつIL−5とIL−5受容体との結合を阻害する抗体、IL−5受容体に結合し、かつIL−5とIL−5受容体との結合を阻害する抗体などがあげられる。IL−5受容体としては、IL−5受容体α鎖、IL−5受容体β鎖等があげられる。
モノクローナル抗体としては、ハイブリドーマが産生する抗体、遺伝子組換え抗体などがあげられる。
ハイブリドーマとは、ヒト以外の哺乳動物に抗原を免疫して取得されたB細胞と、ミエローマ細胞とを細胞融合させて得られる所望の抗原特異性を有するモノクローナル抗体を生産する細胞をいう。
ヒト型キメラ抗体とは、ヒト以外の動物の抗体のVHおよびVLとヒト抗体のCHおよびCLとからなる抗体をいう。ヒト型キメラ抗体のCHとしては、ヒトイムノグロブリン(以下、hIgと表記する)に属すればいかなるものでもよいが、hIgGクラスのものが好適であり、さらにhIgGクラスに属するhIgG1、hIgG2、hIgG3、hIgG4といったサブクラスのいずれも用いることができる。また、ヒト型キメラ抗体のCLとしては、hIgに属すればいずれのものでもよく、κクラスあるいはλクラスのものを用いることができる。
ヒト化抗体とは、ヒト以外の動物の抗体のVHおよびVLのCDRをヒト抗体のVHおよびVLの適切な位置に移植した抗体をいい、ヒト型CDR移植抗体ともいう。
本発明のヒト化抗体は、ヒト以外の動物の抗体のVHおよびVLのCDRを任意のヒト抗体のVHおよびVLのフレームワーク(以下、FRと表記する)と連結したV領域をコードするcDNAを設計、構築し、ヒト抗体のCHおよびCLをコードするcDNAを有する動物細胞用発現ベクターにそれぞれ挿入してヒト化抗体発現ベクターを構築し、動物細胞へ導入することにより発現させ、製造することができる。
上述の抗体または抗体断片を構成するアミノ酸配列において、1以上のアミノ酸が欠失、付加、置換または挿入され、かつ上述の抗体または抗体断片と実質的に同一の活性を有する抗体または抗体断片も本発明で使用される抗体に包含される。

Fabは、IgG型抗体分子を蛋白質分解酵素パパインで処理して得られる断片のうち(H鎖の224番目のアミノ酸残基で切断される)、H鎖のN末端側約半分とL鎖全体がジスルフィド結合で結合した分子量約5万の抗原結合活性を有する抗体断片である。
本発明で使用されるFabは、抗体を蛋白質分解酵素パパインで処理して得ることができる。または、該抗体のFabをコードするDNAを原核生物用発現ベクターあるいは真核生物用発現ベクターに挿入し、該ベクターを原核生物あるいは真核生物へ導入することにより発現させ、製造することができる。
本発明で使用されるF(ab’)2は、抗体を蛋白質分解酵素ペプシンで処理して得ることができる。または、下記のFab’をチオエーテル結合あるいはジスルフィド結合させ、作製することができる。
本発明で使用されるFab’は、F(ab’)2を還元剤ジチオスレイトール処理して得ることができる。または、該抗体のFab’断片をコードするDNAを原核生物用発現ベクターあるいは真核生物用発現ベクターに挿入し、該ベクターを原核生物あるいは真核生物へ導入することにより発現させ、製造することができる。
本発明で使用されるscFvは、抗体のVHおよびVLをコードするcDNAを取得し、scFvをコードするDNAを構築し、該DNAを原核生物用発現ベクターあるいは真核生物用発現ベクターに挿入し、該発現ベクターを原核生物あるいは真核生物へ導入することにより発現させ、製造することができる。
本発明で使用されるdiabodyは、抗体のVHおよびVLをコードするcDNAを取得し、scFvをコードするDNAをリンカーのアミノ酸配列の長さが8残基以下となるように構築し、該DNAを原核生物用発現ベクターあるいは真核生物用発現ベクターに挿入し、該発現ベクターを原核生物あるいは真核生物へ導入することにより発現させ、製造することができる。
本発明で使用されるdsFvは、抗体のVHおよびVLをコードするcDNAを取得し、dsFvをコードするDNAを構築し、該DNAを原核生物用発現ベクターあるいは真核生物用発現ベクターに挿入し、該発現ベクターを原核生物あるいは真核生物へ導入することにより発現させ、製造することができる。
1.モノクローナル抗体の作製
(1)抗原の調製
抗原であるポリペプチドをコードするcDNAを含む発現ベクターを大腸菌、酵母、昆虫細胞、動物細胞などに導入、発現させ、組換え型蛋白質を得、これを抗原に用いることができる。あるいは、抗原のポリペプチドの部分アミノ酸配列を有する合成ペプチドを抗原に用いることもできる。
免疫に用いる動物としては、マウス、ラット、ハムスター、ラビットなどハイブリドーマを作製することが可能であれば、いかなるものでもよい。下記に、マウスおよびラットを用いる例について説明する。
骨髄腫細胞としては、マウスから得られた株化細胞である8−アザグアニン耐性骨髄腫細胞株P3−X63Ag8−U1(P3−U1)(European Journal of Immunology,6,511−519,1976)、SP2/0−Ag14(SP−2)(Nature,276,269−270,1978)、P3−X63−Ag8653(653)(Journal of Immunology,123,1548−1550,1979)、P3−X63−Ag8(X63)(Nature,256,495−497,1975)など、in vitroで増殖可能な骨髄腫細胞であればいかなるものでもよい。これらの細胞株の培養および継代については公知の方法(Antibodies−A Laboratory Manual,Cold Spring Harbor Laboratory,1988)に従い、細胞融合時までに2×107個以上の細胞数を確保する。
上記で得られた抗体産生細胞と骨髄腫細胞とを洗浄したのち、ポリエチレングリコール−100(以下、PEG−1000と表記する)などの細胞凝集性媒体を加え、細胞を融合させ、培地中に懸濁する。細胞の洗浄にはModified Eagle’s Medium(以下、MEMと表記する)またはPhosphate Buffered Saline(以下、PBSと表記する)などを用いる。また、融合細胞を懸濁する培地としては、目的の融合細胞のみを選択的に得られるように、HAT培地{通常培地[RPMI−1640培地に1.5mMグルタミン、50μM2−メルカプトエタノール、10μg/mLジェンタマイシンおよび10%牛胎児血清(以下、FBSと表記する)を加えた培地]に0.1mMヒポキサンチン、15μMチミジンおよび0.4μMアミノプテリンを加えた培地}を用いる。
抗原に反応する抗体を産生するハイブリドーマの選択は、公知の方法(Antibodies−A Laboratory Manual,Cold Spring Harbor Laboratory,1988)に従い、以下に述べるELISAにより選択することができる。本方法により、後述するヒト型キメラ抗体、CDR移植抗体またはそれらの抗体断片を産生する形質転換株の培養上清中に含まれる抗体あるいは培養上清より精製された抗体の抗原に対する結合活性を測定することができる。
抗原を96穴ELISAプレートに固定化し、ハイブリドーマなどの培養上清あるいは精製抗体を第一抗体として反応させる。第一抗体反応後、プレートを洗浄して第二抗体を添加する。第二抗体としては、第一抗体を認識することができる抗体を、ビオチン、酵素、化学発光物質あるいは放射性同位元素などで標識した抗体を用いる。具体的にはハイブリドーマ作製の際にマウスを用いたのであれば、第二抗体としてはマウス抗体を認識できる抗体を用いる。反応後、第二抗体の標識物質に応じた反応を行ない、抗原に特異的に反応するモノクローナル抗体を産生するハイブリドーマとして選択する。
0.5mLのプリスタン(2,6,10,14−テトラメチルペンタデカン)を腹腔内投与し、2週間飼育した8〜10週令のマウスまたはヌードマウスに、1(4)で得られたモノクローナル抗体産生ハイブリドーマ細胞5×106〜2×107細胞/匹を腹腔内に注射する。10〜21日間でハイブリドーマは腹水癌化する。該マウスまたはヌードマウスから腹水を採取し、遠心分離、40〜50%飽和硫酸アンモニウムによる塩析、カプリル酸沈殿法、DEAE−セファロースカラム、プロテインA−カラムあるいはセルロファインGSL2000(生化学工業社製)のカラムなどを用いて、IgGあるいはIgM画分を回収し、精製モノクローナル抗体とする。
抗体のサブクラスとは、クラス内のアイソタイプのことで、マウスでは、IgG1、IgG2a、IgG2b、IgG3、ヒトでは、IgG1、IgG2、IgG3、IgG4があげられる。
(7−1)抗原への結合活性評価
培養上清中あるいは培養上清から精製されたモノクローナル抗体の抗原に対する結合活性は、上記1(5)のELISAおよび表面プラズモン共鳴(Journal of Immunological Methods,145,229−240,1991)などにより測定することができる。また、抗原ペプチドまたはその部分ペプチドを用いた競合ELISAにより、抗原との反応性および抗原エピトープを解析することができる。抗体が抗原である蛋白質の立体構造を認識しているか否かは、通常行われる立体構造的解析法、あるいは種々の免疫学的測定法を組み合わせることにより、推測することができる。立体構造解析法としては、例えば、X線結晶解析、核磁気共鳴法などがあげられる。種々の免疫学的測定法を組み合わせる方法としては、例えば、非変性状態の抗原に対するELISA法と変性状態の抗原に対するELISA法を組み合わせる方法があげられる。このとき、非変性状態の抗原にのみ反応性を示す抗体は、抗原の立体構造を認識している可能性が高いものと推測できる。非変性状態の抗原に対するELISA法とは、液層中で非変性抗原と抗体を反応させるELISA法などがあげられる。変性状態の抗原に対するELISA法としては、抗原がもとの立体構造を保持していない状態で抗体を反応させるELISA法であればいずれでもよく、例えば、疎水性の反応プレート上に直接固定化した抗原や、適当な長さに消化した部分ペプチドなどに対するELISA法があげられる。
ポリクローナル抗体は、上記1.(2)に示された方法で免疫を施した動物のうち、その血清が十分な抗体価を示した動物の血清から調製することができる。
(1)ヒト型キメラ抗体およびヒト化抗体発現用ベクターの構築
ヒト型キメラ抗体およびヒト化抗体発現用ベクター(以下、両者をまとめてヒト化抗体発現用ベクターという)としては、ヒト抗体のCHおよび/またはCLをコードする遺伝子が組み込まれた動物細胞用発現ベクターであればいかなるものでもよい。ヒト化抗体発現用ベクターは、動物細胞用発現ベクターにヒト抗体のCHおよびCLをコードする遺伝子をそれぞれクローニングすることにより構築することができる。
ヒト以外の動物の抗体、例えば、マウス抗体のVHおよびVLをコードするcDNAは以下のようにして取得することができる。
ハイブリドーマから全RNAを調製する方法としては、チオシアン酸グアニジン−トリフルオロ酢酸セシウム法(Methods in Enzymology,154,3−28,1987)、また全RNAからmRNAを調製する方法としては、オリゴ(dT)固定化セルロースカラム法(Molecular Cloning:ALaboratory Manual,Cold Spring Harbor Lab.Press New York,1989)などがあげられる。また、ハイブリドーマからmRNAを調製するキットとしては、Fast Track mRNA Isolation Kit(Invitrogen社製)、Quick Prep mRNA Purification Kit(Pharmacia社製)などがあげられる。
上記3(1)に記載のヒト化抗体発現用ベクターのヒト抗体のCHおよびCLをコードする遺伝子の上流に、ヒト以外の動物の抗体のVHおよびVLをコードするcDNAをクローニングし、ヒト型キメラ抗体発現ベクターを構築することができる。例えば、ヒト以外の動物の抗体のVHおよびVLをコードするcDNAを、ヒト以外の動物の抗体のVHおよびVLの3’末端側の塩基配列とヒト抗体のCHおよびCLの5’末端側の塩基配列とから成り、かつ適当な制限酵素の認識配列を両端に有する合成DNAとそれぞれ連結し、それぞれを上記3(1)に記載のヒト化抗体発現用ベクターのヒト抗体のCHおよびCLをコードする遺伝子の上流にそれらが適切な形で発現するようにクローニングし、ヒト型キメラ抗体発現ベクターを構築することができる。また、ヒト以外の動物の抗体のVHおよびVLをコードするcDNAを含むプラスミドを鋳型として、5’末端に適当な制限酵素の認識配列を有するプライマーを用いてPCR法によりVHおよびVLをコードするcDNAを増幅し、それぞれを上記3(1)に記載のヒト化抗体発現用ベクターのヒト抗体のCHおよびCLをコードする遺伝子の上流にそれらが適切な形で発現するようにクローニングし、ヒト型キメラ抗体発現ベクターを構築することができる。
ヒト化抗体のVHおよびVLをコードするcDNAは、以下のようにして構築することができる。まず、目的のヒト以外の動物の抗体のVHおよびVLのCDRのアミノ酸配列を移植するヒト抗体のVHおよびVLのFRのアミノ酸配列を選択する。ヒト抗体のVHおよびVLのFRのアミノ酸配列としては、ヒト抗体由来のものであれば、いかなるものでも用いることができる。例えば、Protein Data Bankなどのデータベースに登録されているヒト抗体のVHおよびVLのFRのアミノ酸配列、ヒト抗体のVHおよびVLのFRの各サブグループの共通アミノ酸配列(Sequence of Protein of Immunological Interest,US Dept.Health and HumanServices,1991)などがあげられるが、その中でも、十分な活性を有するヒト化抗体を作製するためには、目的のヒト以外の動物の抗体のVHおよびVLのFRのアミノ酸配列とできるだけ高い相同性(少なくとも60%以上)を有するアミノ酸配列を選択することが望ましい。次に、選択したヒト抗体のVHおよびVLのFRのアミノ酸配列に目的のヒト以外の動物の抗体のVHおよびVLのCDRのアミノ酸配列を移植し、ヒト化抗体のVHおよびVLのアミノ酸配列を設計する。設計したアミノ酸配列を抗体の遺伝子の塩基配列に見られるコドンの使用頻度(Sequences of Proteins of Immunological Interest,US Dept.Health and Human Services,1991)を考慮して塩基配列に変換し、ヒト化抗体のVHおよびVLのアミノ酸配列をコードする塩基配列を設計する。設計した塩基配列に基づき、100塩基前後の長さからなる数本の合成DNAを合成し、それらを用いてPCRを行う。この場合、PCRでの反応効率および合成可能なDNAの長さから、VH、VLとも6本の合成DNAを設計することが好ましい。
ヒト化抗体は、目的のヒト以外の動物の抗体のVHおよびVLのCDRのみをヒト抗体のVHおよびVLのFRに移植しただけでは、その抗原結合活性は元のヒト以外の動物の抗体に比べて低下してしまうことが知られている(BIO/TECHNOLOGY,9,266−271,1991)。この原因としては、元のヒト以外の動物の抗体のVHおよびVLでは、CDRのみならず、FRのいくつかのアミノ酸残基が直接的あるいは間接的に抗原結合活性に関与しており、それらアミノ酸残基がCDRの移植に伴い、ヒト抗体のVHおよびVLのFRの異なるアミノ酸残基へと変化してしまうことが考えられている。この問題を解決するため、ヒト化抗体では、ヒト抗体のVHおよびVLのFRのアミノ酸配列の中で、直接抗原との結合に関与しているアミノ酸残基やCDRのアミノ酸残基と相互作用したり、抗体の立体構造を維持し、間接的に抗原との結合に関与しているアミノ酸残基を同定し、それらを元のヒト以外の動物の抗体に見出されるアミノ酸残基に改変し、低下した抗原結合活性を上昇させることが行われている(BIO/TECHNOLOGY,9,266−271,1991)。ヒト化抗体の作製においては、それら抗原結合活性に関わるFRのアミノ酸残基を如何に効率よく同定するかが、最も重要な点であり、そのためにX線結晶解析(Journal of Molecular Biology,112,535−542,1977)あるいはコンピューターモデリング(Protein Engineering,7,1501−1507,1994)などによる抗体の立体構造の構築および解析が行われている。これら抗体の立体構造の情報は、ヒト化抗体の作製に多くの有益な情報をもたらして来たが、その一方、あらゆる抗体に適応可能なヒト化抗体の作製法は未だ確立されておらず、現状ではそれぞれの抗体について数種の改変体を作製し、それぞれの抗原結合活性との相関を検討するなどの種々の試行錯誤が必要である。
上記3(1)に記載のヒト化抗体発現用ベクターのヒト抗体のCHおよびCLをコードする遺伝子の上流に、上記3(4)および(5)で構築したヒト化抗体のVHおよびVLをコードするcDNAをクローニングし、ヒト化抗体発現ベクターを構築することができる。
作製した多種類のヒト型キメラ抗体およびヒト化抗体の抗原結合活性を効率的に評価するために、上記3(3)および(6)に記載のヒト型キメラ抗体発現ベクターおよびヒト化抗体発現ベクター、あるいはそれらを改変した発現ベクターを用いてヒト型キメラ抗体およびヒト化抗体の一過性発現を行うことができる。発現ベクターを導入する宿主細胞としては、ヒト型キメラ抗体およびヒト化抗体を発現できる宿主細胞であれば、いかなる細胞でも用いることができるが、その発現量の高さから、COS−7細胞(ATCC CRL1651)が一般に用いられる(Methods in Nucleic Acids Research,CRC press,283,1991)。COS−7細胞への発現ベクターの導入法としては、DEAE−デキストラン法(Methods in Nucleic Acids Research,CRC press,283,1991)、リポフェクション法(Proceedings of the National Academy of Sciences of the United States of America,84,7413−7417,1987)などがあげられる。
上記3(3)および(6)に記載のヒト型キメラ抗体発現ベクターおよびヒト化抗体発現ベクターを適当な宿主細胞に導入することによりヒト型キメラ抗体およびヒト化抗体を安定に発現する形質転換細胞を得ることができる。宿主細胞への発現ベクターの導入法としては、エレクトロポレーション法(Cytotechnology,3,133−140,1990)などがあげられる。ヒト型キメラ抗体発現ベクターおよびヒト化抗体発現ベクターを導入する宿主細胞としては、ヒト型キメラ抗体およびヒト化抗体を発現させることができる宿主細胞であれば、いかなる細胞でも用いることができる。例えば、マウスSP2/0−Ag14細胞(ATCC CRL1581)、マウスP3X63−Ag8.653細胞(ATCC CRL1580)、ジヒドロ葉酸還元酵素遺伝子(以下、dhfrと表記する)が欠損したCHO細胞(Proceedings of the National Academy of Sciences of the United States of America,77,4216−4220,1980)、ラットYB2/3HL.P2.G11.16Ag.20細胞(ATCC CRL1662、以下、YB2/0細胞と表記する)などがあげられる。
ヒト型キメラ抗体およびヒト化抗体と抗原との結合活性評価は、上記に記載のELISAを用いて行うことができる。
抗体断片は、上記1および3に記載の抗体をもとに遺伝子工学的手法あるいは蛋白質化学的手法により、作製することができる。
蛋白質化学的手法としては、ペプシン、パパインなどの蛋白質分解酵素を用いた部位特異的切断、精製などの方法があげられる。
Fabは、蛋白質化学的にはIgGを蛋白質分解酵素パパインで処理することにより、作製することができる。パパインの処理後は、元の抗体がプロテインA結合性を有するIgGサブクラスであれば、プロテインAカラムに通すことで、IgG分子やFc断片と分離し、均一なFabとして回収することができる(Monoclonal Antibodies:Principles and Practice,third edition,1995)。プロテインA結合性を持たないIgGサブクラスの抗体の場合は、イオン交換クロマトグラフィーにより、Fabは低塩濃度で溶出される画分中に回収することができる(Monoclonal Antibodies:Principles and Practice,third edition,1995)。
F(ab’)2は、蛋白質化学的にはIgGを蛋白質分解酵素ペプシンで処理することにより、作製することができる。ペプシンの処理後は、Fabと同様の精製操作により、均一なF(ab’)2として回収することができる(Monoclonal Antibodies:Principles and Practice,third edition,Academic Press,1995)。また、下記4(3)に記載のFab’をo−PDMやビスマレイミドヘキサンなどのようなマレイミドで処理し、チオエーテル結合させる方法や、DTNB[5,5’−dithiobis(2−nitrobenzoic acid)]で処理し、S−S結合させる方法によっても作製することができる(Antibody Engineering,A Practical Approach,IRL PRESS,1996)。
Fab’は、上記4(2)に記載のF(ab’)2をジチオスレイトールなどの還元剤で処理して得ることができる。また、Fab’は遺伝子工学的には、多くは大腸菌、また、昆虫細胞や動物細胞などを用いて作製することができる。例えば、上記3(2)、3(4)および3(5)に記載の抗体のV領域をコードするDNAを、Fab’発現用ベクターにクローニングし、Fab’発現ベクターを作製することができる。Fab’発現用ベクターとしては、Fab’用のDNAを組み込み発現できるものであればいかなるものも用いることができる。例えば、pAK19(BIO/TECHNOLOGY,10,163−167,1992)などがあげられる。Fab’発現ベクターを適当な大腸菌に導入し、封入体あるいはペリプラズムにFab’を生成蓄積させることができる。封入体からは、通常蛋白質で用いられるリフォールディング法により、活性のあるFab’とすることができ、また、ペリプラズムに発現させた場合は、リゾチームによる部分消化、浸透圧ショック、ソニケーションなどの処理により菌を破砕し、菌体外へ回収させることができる。リフォールディング後あるいは菌の破砕液からは、プロテインGカラムなどを用いることにより、均一なFab’を精製することができる(Antibody Engineering,A Practical Approach,IRL PRESS,1996)。
scFvは遺伝子工学的には、ファージまたは大腸菌、また、昆虫細胞や動物細胞などを用いて作製することができる。例えば、上記3(2)、3(4)および3(5)に記載の抗体のV領域をコードするDNAを、scFv発現用ベクターにクローニングし、scFv発現ベクターを作製することができる。scFv発現用ベクターとしては、scFvのDNAを組み込み発現できるものであればいかなるものも用いることができる。例えば、pCANTAB5E(Pharmacia社製)、pHFA(Human Antibodies &Hybridomas,5,48−56,1994)などがあげられる。scFv発現ベクターを適当な大腸菌に導入し、ヘルパーファージを感染させることで、ファージ表面にscFvがファージ表面蛋白質と融合した形で発現するファージを得ることができる。また、scFv発現ベクターを導入した大腸菌の封入体あるいはペリプラズムにscFvを生成蓄積させることができる。封入体からは、通常蛋白質で用いられるリフォールディング法により、活性のあるscFvとすることができ、また、ペリプラズムに発現させた場合は、リゾチームによる部分消化、浸透圧ショック、ソニケーションなどの処理により菌を破砕し、菌体外へ回収することができる。リフォールディング後あるいは菌の破砕液からは、陽イオン交換クロマトグラフィーなどを用いることにより、均一なscFvを精製することができる(Antibody Engineering,A Practical Approach,IRL PRESS,1996)。
diabodyは遺伝子工学的には、多くは大腸菌、また、昆虫細胞や動物細胞などを用いて作製することができる。例えば、上記3(2)、3(4)および3(5)に記載の抗体のVHとVLをリンカーがコードするアミノ酸残基が8残基以下となるように連結したDNAを作製し、diabody発現用ベクターにクローニングし、diabody発現ベクターを作製することができる。diabody発現用ベクターとしては、diabodyのDNAを組み込み発現できるものであればいかなるものも用いることができる。例えば、pCANTAB5E(Pharmacia社製)、pHFA(Human Antibodies Hybridomas,5,48,1994)などがあげられる。diabody発現ベクターを導入した大腸菌の封入体あるいはペリプラズムにdiabodyを生成蓄積させることができる。封入体からは、通常蛋白質で用いられるリフォールディング法により、活性のあるdiabodyとすることができ、また、ペリプラズムに発現させた場合は、リゾチームによる部分消化、浸透圧ショック、ソニケーションなどの処理により菌を破砕し、菌体外へ回収することができる。リフォールディング後あるいは菌の破砕液からは、陽イオン交換クロマトグラフィーなどを用いることにより、均一なscFvを精製することができる(Antibody Engineering,A Practical Approach,IRL PRESS,1996)。
dsFvは遺伝子工学的には、多くは大腸菌、また、昆虫細胞や動物細胞などを用いて作製することができる。まず、上記3(2)、3(4)および3(5)に記載の抗体のVHおよびVLをコードするDNAの適当な位置に変異を導入し、コードするアミノ酸残基がシステインに置換されたDNAを作製する。作製した各DNAをdsFv発現用ベクターにクローニングし、VHおよびVLの発現ベクターを作製することができる。dsFv発現用ベクターとしては、dsFv用のDNAを組み込み発現できるものであればいかなるものも用いることができる。例えば、pULI9(Protein Engineering,7,697−704,1994)などがあげられる。VHおよびVLの発現ベクターを適当な大腸菌に導入し、封入体あるいはペリプラズムにdsFvを生成蓄積させることができる。封入体あるいはペリプラズムからVHおよびVLを得、混合し、通常蛋白質で用いられるリフォールディング法により、活性のあるdsFvとすることができる。リフォールディング後は、イオン交換クロマトグラフィーおよびゲル濾過などにより、さらに精製することができる(Protein Engineering,7,697−704,1994)。
CDRを含むペプチドは、Fmoc法あるいはtBoc法等の化学合成法によって作製することができる。また、CDRを含むペプチドをコードするDNAを作製し、作製したDNAを適当な発現用ベクターにクローニングし、CDRペプチド発現ベクターを作製することができる。発現用ベクターとしては、CDRペプチドをコードするDNAを組み込み発現できるものであればいかなるものも用いることができる。例えば、pLEX(Invitrogen社製)、pAX4a+(Invitrogen社製)などがあげられる。発現ベクターを適当な大腸菌に導入し、封入体あるいはペリプラズムにを生成蓄積させることができる。封入体あるいはペリプラズムからCDRペプチドを得、イオン交換クロマトグラフィーおよびゲル濾過などにより、精製することができる(Protein Engineering,7,697−704,1994)。
精製した抗体断片と抗原との結合活性評価は、上記1(7)に記載のELISAを用いて行うことができる。
本発明の子宮内膜症の治療剤としては、IL−5アンタゴニストを有効成分として含む医薬であればいかなるものでもよいが、通常は薬理学的に許容される一つあるいはそれ以上の担体と一緒に混合し、製剤学の技術分野においてよく知られる任意の方法により製造した医薬製剤として提供するのが望ましい。好ましくは水、あるいは食塩、グリシン、グルコース、ヒトアルブミン等の水溶液等の水性担体に溶解した無菌的な溶液が用いられる。また、製剤溶液を生理的条件に近づけるための緩衝化剤や等張化剤のような、薬理学的に許容される添加剤、例えば、酢酸ナトリウム、塩化ナトリウム、乳酸ナトリウム、塩化カリウム、クエン酸ナトリウム等を添加することもできる。また、凍結乾燥して貯蔵し、使用時に適当な溶媒に溶解させて用いることもできる。
経口投与に適当な製剤としては、乳剤、シロップ剤、カプセル剤、錠剤、散剤、顆粒剤等があげられる。例えば乳剤およびシロップ剤のような液体調製物は、水、ショ糖、ソルビトール、果糖等の糖類、ポリエチレングリコール、プロピレングリコール等のグリコール類、ごま油、オリーブ油、大豆油などの油類、p−ヒドロキシ安息香酸エステル類等の防腐剤、ストロベリーフレーバー、ペパーミント等のフレーバー類等を添加剤として用いて製造できる。カプセル剤、錠剤、散剤、顆粒剤等は、乳糖、ブドウ糖、ショ糖、マンニトール等の賦形剤、デンプン、アルギン酸ナトリウム等の崩壊剤、ステアリン酸マグネシウム、タルク等の滑沢剤、ポリビニルアルコール、ヒドロキシプロピルセルロース、ゼラチン等の結合剤、脂肪酸エステル等の界面活性剤、グリセリン等の可塑剤等を添加剤として用いて製造できる。
以下に本発明の実施例を示す。
ラット子宮内膜症自家移植モデルを用いた抗IL−5受容体抗体および抗IL−5抗体の抑制効果について、以下の実験を行った。
方法としては、公知の方法[ファーティリティ・アンド・ステリリティ(Fertility and sterility)、44巻、684〜694ページ(1985年)]を改変して行った。
被験薬物をラットに静脈内投与した後、ペントバルビタール(ソムノペンチル(R)、武田シェリング・プラウアニマルヘルス社製)の腹腔内投与により全身麻酔したラットの腹部を剃毛し、正中線に沿って約6センチメートル開腹した。双角に存在する子宮のうち、右側の子宮の根元部位より約1センチメートル上部ならびに卵巣より約2センチメートル下部を結索し、その間にある子宮を摘出した。摘出子宮(管状)はペニシリン・ストレプトマイシン(INVITROGEN社製)を含有するダルベッコ・モディファイド・イーグル・メディウム(INVITROGEN社製)中で洗いながら、縦に開列しシート状にした。このシートから約2ミリメートル四方の子宮内膜片を作製した。作製した子宮内膜片は、左右腹膜にそれぞれ滅菌糸付縫合針(外科用弱弯角針No.12−黒ナイロンNo.6−0、夏目製作所製)を用いて自家移植した。この際、子宮内膜面が腹膜に接するように移植した。また、対照として、子宮内膜片と同サイズの脂肪組織を同一個体から摘出し、腹膜に同様に自家移植した。最後に、移植部位ならびに子宮摘出部位をミクロノマイシン(サガミシン(R)注射液、協和発酵工業社製)を含有する生理食塩水で洗い、直ちに腹膜および皮膚を滅菌糸付縫合針により丁寧に縫合した。
移植片サイズについての結果を表1に示した。移植片サイズは縦径と横径を乗じて算出した値とし、左右の移植片サイズを平均して示した。
ラット子宮内膜症自家移植モデルを用いて、抗IL−5受容体抗体が子宮内膜の癒着を抑制していることを確認するために、以下の実験を行った。実験方法は、実施例1に記載された方法に準じて行った。

癒着が認められないもの;スコア0
癒着が認められるが軽度のもの;スコア1
強い癒着が認められるが剥離性であるもの;スコア2
強い癒着が認められ非剥離性であるもの;スコア3
試験は一群5匹で実施した。また、必要に応じ、移植部位の病理標本を定法に従って作製し、病理組織学的な解析を行った。
病変部位における癒着スコアについての結果を表2に示した。癒着スコアはそれぞれを点数とし、左右の移植片の癒着スコアを平均化して示した。
抗IL−5受容体抗体の子宮内膜症に対する有効性を検証するために、以下の実験を行った。
方法としては、公知の方法[ヒューマン・リプロダクション(Human Reproduction)、14巻、2944〜2950ページ(1999年)]を改変して行った。
病変サイズについての結果を図1に示した。
実施例3に記載された方法に準じ、抗IL−5抗体の子宮内膜症に対する有効性を検証した。

Claims (7)
- インターロイキン−5受容体に結合する抗体またはFab、Fab’、F(ab’) 2 、一本鎖抗体(scFv)、二量体化可変領域(Diabody)、ジスルフィド安定化可変領域(dsFv)若しくはCDRを含むペプチドから選ばれるその抗体断片を有効成分として含有する子宮内膜症の治療剤。
- インターロイキン−5受容体がインターロイキン−5受容体α鎖である請求項1記載の治療剤。
- インターロイキン−5受容体がヒトインターロイキン5受容体である請求項1または2記載の治療剤。
- 抗体がモノクローナル抗体である、請求項1〜3のいずれか1項に記載の治療剤。
- モノクローナル抗体が遺伝子組換え抗体である、請求項4項に記載の治療剤。
- 遺伝子組換え抗体が、ヒト型キメラ抗体、ヒト化抗体およびヒト抗体からなる群から選ばれる遺伝子組換え抗体である、請求項5記載の治療剤。
- 子宮内膜症治療剤の製造のための請求項1〜6のいずれか1項に記載のインターロイキン−5受容体に結合する抗体またはFab、Fab’、F(ab’) 2 、一本鎖抗体(scFv)、二量体化可変領域(Diabody)、ジスルフィド安定化可変領域(dsFv)若しくはCDRを含むペプチドから選ばれるその抗体断片の使用。
