Affinity chromatography (Blue Sepharose FF)

アフィニティーカラムの調製:50 mlのアミノヘキサン酸 Sepharoseを5 Lの水で洗浄する。フィルターケーキを40 mlの水にて再懸濁する。4.2 gの4-アセトキシ安息香酸(保護基を導入したヒドロキシ安息香酸)を50 mlのDMFに溶解し、pH 4.7に調整(10 M水酸化ナトリウム)する。3.9 gのEDCを10 mlの水に溶解し、15分以内に結合混合液に加える。pHは塩酸により4.7に保持する。その後、さらに50 mlのDMFを加え、該混合物は室温で16時間撹拌する。該カラム材料はG2ガラス吸引フィルターを介した吸引処理により回収し、1 Lの50%水/DMF溶液により洗浄する。その後1Lの水で洗浄する。該カラム材料はその後200 mlの冷炭酸水素ナトリウム溶液に再懸濁し、2 mlの無水酢酸を加える。脱保護は氷上で90分間実施するが、これにより二酸化炭素が激しく発生する。反応溶液を吸引処理により除き、カラム材料を水によって洗浄する。次いで、該カラム材料は1 Mイミダゾール溶液中、pH 7.9で1時間撹拌し、その後再び吸引処理により除き、水で洗浄し、この状態で保存する。タンパク質はこのカラムには吸着しない。

The column material is collected by suction through a G2 glass suction filter and washed with 1 L of 50% water / DMF solution. Then wash with 1 L of water. The column material is then resuspended in 200 ml of cold sodium bicarbonate solution and 2 ml of acetic anhydride is added. Deprotection is carried out on ice for 90 minutes, which causes a violent evolution of carbon dioxide. The reaction solution is removed by suction and the column material is washed with water. The column material is then stirred in 1 M imidazole solution at pH 7.9 for 1 hour and then removed again by suction, washed with water and stored in this state. Protein does not adsorb to this column.
