Description
本発明は共有結合したオリゴヌクレオチド結合体に関する。特に本発明は、癌細胞において優先的に発現され、癌細胞の増殖に必要とされる酵素、テロメラーゼの活性を阻害するよう設計されたオリゴヌクレオチド共有結合体を目的とする。
本出願は、2002年9月25日に出願された米国特許出願第10/255,535号の優先権を主張するものである。この優先権出願は、その全体が参照として本明細書に組み入れられる。
オリゴヌクレオチド化学
核酸ポリマー化学は製薬、診断、および分析分野、特にアンチセンスおよび抗遺伝子療法、コンビナトリアルケミストリー、分枝DNAシグナル増幅、ならびにアレイ技術を用いた(array-based)DNA診断学および分析の下位分野における多くの発展中の科学技術において重大な役割を果たしている。この化学の多くは結合強度、特異性、およびDNAなどの天然核酸ポリマーのヌクレアーゼ耐性の改善を目的としている。残念ながら、ヌクレアーゼ耐性などの一つの性質を改善すると、結合強度などの他の性質に対するトレードオフを伴うことが多い。そのようなトレードオフの例は多数ある:ペプチド核酸(PNA)は良好なヌクレアーゼ耐性および結合強度を示すが、試験培養中の細胞取り込みは低い(例えば、Hanveyら、Science, 258:1481-1485, 1992);ホスホロチオエートは良好なヌクレアーゼ耐性および溶解性を示すが、典型的にP-キラル混合物として合成され、いくつかの配列非特異的な生物学的作用を示す(例えば、Steinら、Science, 261:1004-1012, 1993);ホスホン酸メチルは良好なヌクレアーゼ耐性および細胞取り込みを示すが、これも典型的にP-キラル混合物として合成され、二本鎖の安定性が低いなど。
テロメラーゼはテロメア反復配列の染色体末端への付加を触媒するリボ核蛋白質である。Blackburn, 1992, Annu. Rev. Biochem., 61:113-129を参照されたい。テロメア、テロメラーゼ、細胞老化、および癌の間のつながりを記載している文献は大量にある(総説としては、Oncogene、第21巻、2002年1月のテロメラーゼを特集した号を参照されたい)。したがって、テロメラーゼは癌治療薬の優れた標的として認められている(Lichsteinerら、Annals New York Acad. Sci. 886:1-11, 1999参照)。
本発明の組成物および方法は、オリゴヌクレオチドおよび少なくとも一つの共有結合した基、好ましくは芳香族系を含む化合物に関する。オリゴヌクレオチドの配列はテロメラーゼのRNA成分に相補的となるように選択され、共有結合した芳香族系は、例えば多芳香族炭化水素、単環式もしくは複素環式芳香族系、または核酸塩基(修飾された核酸塩基および核酸塩基類似体を含む)である。芳香族基は、必須ではないとしても典型的に、オリゴヌクレオチドの3'-位または5'-位のいずれかに共有結合し、特定の態様において、芳香族基は3'-および5'-位のそれぞれに結合していてもよい。最も単純な形において、本発明の化合物は下記の式で表すことができる:

(式中、Aは芳香族基を含み、Lは選択的なリンカー基であり(すなわち、リンカーまたは直接結合)、Oはオリゴヌクレオチドであり、nは1から1+m(mはオリゴヌクレオチドを構成するヌクレオシドの総数)の間の整数である)。この式において結合とは、従来通り、結合の各端の部分間における共有結合を示すよう用いられる。好ましい態様において、n=1または2であり、A部分は3'末端および5'末端の一つまたはそれぞれにおいてオリゴヌクレオチド成分と結合している。
A. 定義
「アルキル基」とは、メチル、エチル、プロピルなどの1から20個の炭素原子を有するアルキルまたは置換アルキル基を意味する。低級アルキルは典型的にはC1からC5を意味する。中級アルキルは典型的にはC6からC10を意味する。「アシル基」とは、Rがアルキルである構造RCOを有する基を意味する。低級アシルはRが低級アルキルであるアシルである。
本発明は一般に下記の式を有する化合物を目的とする:
(A-L-)n-O
(式中、Aは芳香族基であり、Lは選択的なリンカー(すなわち、リンカーまたは直接結合)であり、Oはオリゴヌクレオチドであり、nは1から1+m(mはオリゴヌクレオチドを構成するヌクレオシドの総数)の間の整数である)。したがって、化合物の設計には三つの実体A、LおよびOの選択、ならびにこれらの実体の間の構造結合の決定が必要である。
Oのオリゴヌクレオチド配列は、標的テロメラーゼ、例えばヒトテロメラーゼのRNA配列に相補的となるように選択する。ヒトテロメラーゼのRNA成分は配列決定されている(Fengら、Science 269 (5228), 1236-1241, 1995参照、配列データはGenBankアクセッション番号U86046でも入手可能である)。ヒトテロメラーゼRNA(「hTR」)配列内はテロメラーゼが染色体末端に付加するテロメア反復配列の配列を特定する機能を果たす「鋳型領域」として同定されている領域であり、この鋳型領域はテロメラーゼ酵素の活性にとって必須である(Chenら、Cell 100:503-514, 2000;Kimら、Proc. Nat'l. Acad. Sci, USA 98(14): 7982-7987, 2001参照)。鋳型領域は、したがって阻害オリゴヌクレオチドに特に適した標的として同定されている、配列5'-CUAACCCUAAC-3'(配列番号:2)の11ヌクレオチド領域である。したがって、オリゴヌクレオチド結合体のO成分のオリゴヌクレオチド配列はhTR配列のいかなる領域からも選択することができるが、この鋳型および/またはhTR配列の隣接領域に相補的な配列を選択することが好ましい。選択した配列は好ましくは対応するhTR配列に厳密に相補的である。特定の場合にミスマッチが耐容されることもあるが、得られたオリゴヌクレオチド結合体の特異性および活性を低下させると予想される。したがって特定の態様において、オリゴヌクレオチドOの塩基配列は、ヒトテロメラーゼRNAの鋳型領域に正確に相補的な2〜11ヌクレオチドの配列を含むよう選択される。最適なテロメラーゼ阻害活性は、オリゴヌクレオチドOの全長がhTR鋳型領域配列の一部に相補的となるように選択される場合に得られる。
3'-[-Z6-P(=O)(-Z7R)-O-]-5'
(式中、Z6はOまたはNHであり;Z7はOまたはSであり;かつRは水素、アルキル、アリールおよびその塩からなる群より選択される)。
本発明の結合体は二つの局面でテロメラーゼに結合し、それによって酵素の機能を阻害すると考えられている。結合の第一の局面は、hTRのヌクレオシドと結合体のオリゴヌクレオチド(O)成分のヌクレオシドとの間の塩基対相補性である。結合の第二の局面は、テロメラーゼ蛋白質成分と結合体のA成分との間の相互作用である。広範な置換基が、本明細書において便宜上「芳香族」部分と称されるA成分によって提供されるこの第二の結合局面に役立つ可能性がある。この文脈において、「芳香族」なる用語は広く用いられ、例えば、多芳香族炭化水素、一および多環式ヘテロ芳香族系ならびに核酸塩基(修飾核酸塩基および核酸塩基類縁体を含む)を含む。

OおよびA成分の間の結合は、リンカー配列を組み込むことによって促進することができる。そのような配列は、二つの部分の化学結合を促進するのに役立ちうるだけでなく、柔軟性を付与し、それにより各部分がテロメラーゼ酵素上の標的に結合する可能性を増大させることによって、結合体の活性を高めるのにも役立つと考えられる。リンカーはオリゴヌクレオチドOの糖環、またはヌクレオシド間ホスホジエステルもしくはホスホラミデート結合などのヌクレオシド間骨格、またはオリゴヌクレオチドの核酸塩基に直接結合してもよい。A成分の選択に応じて、様々なリンカー基を用いることができる。例示的リンカーは下記の構造である:

芳香族置換基は標準的化学合成を用いてオリゴヌクレオチドに共有結合することができる。典型的には、まずオリゴヌクレオチドの求核部位を保護し、所望の結合部位を選択的に脱保護する。次いで、下記のスキームに示すとおり、オリゴヌクレオチドを、例えば芳香族置換基がフルオレセインの場合、イソチオシアネート、イソシアネート、モノクロロトリアジン、ジクロロトリアジン、モノまたはジハロゲン置換ピリジン、モノまたはジハロゲン置換ジアジン、マレイミド、アジリジン、ハロゲン化スルホニル、酸ハロゲン化物、ヒドロキシスクシンイミドエステル、ヒドロキシスルホスクシンイミドエステル、イミドエステル、ヒドラジン、アジドニトロフェニル、アジド、3-(2-ピリジルジチオ)-プロプリオンアミド、グリオキサールまたはアルデヒドなどの反応性基を含むフルオレセインと反応させて、フルオレセインがオリゴヌクレオチドに、好ましくはチオ尿素またはアミド基を通じて結合した修飾オリゴヌクレオチドを得る。
適当な芳香族系Aに連結したリンカーLのいくつかの例を図3に示す。
テロメラーゼ酵素活性および/またはテロメラーゼ活性を有する細胞の増殖を阻害または低減するために、本発明の結合体を用いることができる。これらの状況において、酵素活性または細胞増殖の阻害または低減は、酵素または細胞を結合体で処理していない対照実験と比較して、測定した活性のレベルが低いことを意味する。特定の態様において、測定した活性の阻害または低減は、少なくとも10%の低減または阻害である。当業者であれば、特定の適用のためには少なくとも20%、50%、75%、90%または100%の測定活性の低減または阻害が好ましいことを理解すると思われる。
本発明は、テロメラーゼ活性を特異的かつ強力に阻害することができ、したがって腫瘍細胞などのテロメラーゼ陽性細胞の増殖を阻害するために用いることもできる、オリゴヌクレオチド結合体を提供する。皮膚、結合組織、脂肪、乳房、肺、胃、膵臓、卵巣、頚部、子宮、腎、膀胱、結腸、前立腺、中枢神経系(CNS)、網膜の癌および循環腫瘍(白血病およびリンパ腫など)由来の細胞を含む、非常に多様な癌細胞がテロメラーゼ陽性であることが明らかにされている。したがって、本明細書において提供されるオリゴヌクレオチド結合体は、広範な悪性腫瘍の治療において広く有用性を提供することができる。より重要なことに、本発明のオリゴヌクレオチド結合体は、悪性および正常細胞を高度に識別する治療を提供し、分裂細胞を無差別に死滅させる物質に頼るほとんどの現行の化学療法に伴う有害副作用の多くを避ける上で有効でありうる。したがって、本発明の一つの局面は患者における癌の治療法であって、患者に本発明のオリゴヌクレオチド結合体の治療上有効な用量を投与する段階を含む方法である。本発明のオリゴヌクレオチド結合体を含むテロメラーゼ阻害剤を、原発性腫瘍の外科的切除、化学療法剤および放射線治療を含む他の癌治療アプローチと共に用いてもよい。
一般法
31P NMRスペクトルはVarian 400 Mhz分光計で得た。31P NMRスペクトルは85%リン酸水溶液を基準にした。アニオン交換HPLCをDionex DX 500クロマトグラフィ装置で、Pharmacia Biotech Mono Q HR 5/5または10/16イオン交換カラムを用いて実施した。質量分析はMass Consortium、カリフォルニア州サンディエゴで実施した。オリゴヌクレオチドのMALDI-TOF分析は、PerSpective Biosystems Voyager Elite質量分析計を用いて遅延引き出しを行って得た。熱解離実験をCary Bio 100 UV-Vis分光計で実施した。
オリゴリボヌクレオチドN3'→P5'ホスホロアミデートの合成
ホモプリンおよびホモピリミジンオリゴリボヌクレオチドN3'→P5'ホスホロアミデートはホスホロアミダイト転移反応を用いて固相支持体上で効率よく構築しうることが以前に報告されている(Gryaznovら、(1998) Nucleic Acids Res., 26:4160-4167)。発明者らは、この方法が4種の天然塩基すべて、ならびにチミジンおよび2,4-ジアミノプリン(D)を含む異種ホスホロアミデートオリゴリボヌクレオチドの合成にも、同じようにうまくはたらくことを見いだした。調製したオリゴリボヌクレオチドN3'→P5'ホスホロアミダイトはそれぞれ、5'-末端残基として支持体に結合した2'-デオキシ-3'-アミノヌクレオシドを用いて5'-末端から出発して合成した。カップリング段階は、接近する5'-O-ホスホロアミダイトのジイソプロピルアミノ基と、支持体に結合したヌクレオシドの3'-アミノ基との交換を含む。標準的RNA合成のカップリング時間(10分間)および活性化剤(1H-テトラゾール)を各合成サイクルに用いた。次いで、未反応の3'-アミノ基を無水イソ酪酸でキャッピングし、その後ヌクレオチド間ホスホロアミダイトジエステル結合のホスホロアミデート基への酸化を水性ヨウ素により実施した。続いて、付加した残基の3'-アミノ基を脱トリチル化することにより、所望のオリゴリボヌクレオチドホスホロアミデート構築のために、さらに鎖伸長段階を繰り返すことが可能となった。次いで、アンモニア/エタノール溶液で処理して、樹脂に結合した化合物を脱保護し、支持体から切断した。2'-O-ブチルジメチルシリル基の除去は、THF中1M TBAFを用いて行い、その後、完全に脱保護したオリゴリボヌクレオチドホスホロアミデートをIEを用いて分析し、単離した。混合塩基の9〜13量体オリゴリボヌクレオチドホスホロアミデートを、MMTrアッセイ法により判断して約96〜98%の段階的カップリング収率で合成した。オリゴヌクレオチドは31P NMRおよびMALDI-TOF質量分析の両方で特徴分析した。
オリゴヌクレオチドN3'→P5'チオホスホロアミデートの合成
オリゴヌクレオチドN3'→P5'チオホスホロアミデートを、ABI 394合成機でアミダイト転移反応により調製した。完全に保護した構築ブロックモノマーは、ヌクレオシドが3'-デオキシチミジン、2',3'-ジデオキシ-N2-イソブチリル-グアノシン、2',3'-ジデオキシ-N6-ベンゾイル-アデノシンまたは2',3'-ジデオキシ-N4-ベンゾイル-シチジンである3'-アミノトリチルヌクレオシド-5'-(2-シアノエチル-N,N-ジイソプロピル)ホスホロアミダイトであった。5'-スクシニル-3'-アミノトリチル-2',3'-ジデオキシヌクレオシドをアミノ基含有長鎖細孔性ガラス(LCAA-CPG)とカップリングさせ、固体支持体として用いた。合成は、5'から3'の方向に行った。オリゴヌクレオチドN3'→P5'チオホスホロアミデートの構築のために下記のプロトコルを用いた:(i)脱トリチル化、ジクロロメタン中3%トリクロロ酢酸;(ii)カップリング、アセトニトリル中0.1Mホスホロアミダイトおよび0.45Mテトラゾール、25秒間;(iii)キャッピング、Cap Aおよび標準Cap B溶液として無水イソ酪酸/2,6-ルチジン/THF(1/1/8、v/v/v);(iv)硫化、アセトニトリル:2,6-ルチジンの1:1混合物に溶解したフェニルアセチルジスルフィド(PADS)の0.1M溶液、5分。オリゴヌクレオチドチオホスホロアミデートを固体支持体から切断し、濃アンモニア水で脱保護した。化合物をHPLCで分析し、精製した。イオン交換(IE)HPLCを商標DIONEX DNAPacイオン交換カラムを用いて、pH12(10mM NaOH)で、10mM NaOH/1.5M NaClの1%/分直線勾配、流速1ml/分により行った。生成物をSephadex NAP-5ゲルろ過カラム(Pharmacia)で脱塩し、減圧下で凍結乾燥した。硫化分析の程度を解析するために、31P NMR実験を重水中で実施した(31P NMR δ、ppm 58、Rp-、Sp-異性体の60の幅広いシグナル)。
を下記のとおりに合成した:ABI Model 394合成機を3'-トリチルアミノ-2',3'-ジデオキシ-N6-ベンゾイル-アデノシン(N2-イソブチリル-グアノシン、およびチミジン)5'-(2-シアノエチル-N,N-ジイソプロピル)ホスホロアミダイトの0.1M溶液で調整した。ステーション#10の試薬ビンにニート二硫化炭素を充填し、ステーション#15の試薬ビンに1%トリエチルアミンを含む15%S8/二硫化炭素を充填した。活性化剤として、市販の0.45Mテトラゾール/アセトニトリル溶液を用いた。Cap A溶液(ステーション#11)をテトラヒドロフラン/無水イソ酪酸/2,6-ルチジン(8/1/1、v/v/v)溶液で置き換えた。Cap Bも市販の試薬であった。ステーション#10からカラムに二硫化炭素を送るために新しい機能を作成した。デフォルトの硫黄合成サイクルを下記のとおりに変更した:硫化時間を5分に設定した。合成カラムに1μモルの固体支持体、N2-イソブチリル-3'-(トリチル)アミノ-2',3'-ジデオキシグアノシン-5'-スクシニル装填CPG(細孔性ガラス)を充填した。化合物の配列は
とプログラムした。トリチル基を合成の最後に除去した。固体支持体をカラムから取り出し、密封したガラスバイアル中、濃アンモニア水(1ml)により55℃で6時間処理した。ろ過後、ほとんどのアンモニアを蒸発させ、残った溶液を商標Sephadex NAP-5ゲルろ過カラム(Pharmacia)を用いて脱塩した後、減圧下で凍結乾燥した。生成物を前述のとおりに分析し、精製した。
フルオレセインの3'-アミノまたは5'-アミノオリゴヌクレオチドへの結合
反応は、1.5mLのエッペンドルフ試験管内で、遊離アミノ基を有するオリゴヌクレオチド約10〜20 ODを0.1重炭酸ナトリウム緩衝液(pH8、100uL)およびDMSO(50uL)に溶解して行った。次いでFITC(Aldrichより、約1mg)を溶液に加え、反応混合物をボルテックスで1〜2分間撹拌した。反応混合物を55℃で30分間加熱し、次いで室温の暗所で終夜放置した。同じ試験管内でEtOH(1.2mL)を加え、-18℃で1時間置いて、オリゴヌクレオチド生成物を沈殿させた。沈殿を上清から分離し、1M NaCl(100uL)からEtOHで二回再沈殿させた。
ガンシクロビルの3'-アミノまたは5'-アミノオリゴヌクレオチドへの結合
無水アセトニトリル中のモノ-O-ジメトキシトリチルガンシクロビル-O-CE-ホスホラミダイトの0.1M溶液を、ABI Model 394合成機内に、ホスホラミダイト用の追加ステーションの一つを用いてセットした。合成とその後の後処理は実施例2および3に記載したものと同様に実施した。結合したオリゴヌクレオチドの純度をイオン交換HPLCで評価した。
テロメラーゼ活性を有するアフィニティ精製抽出物の調製
テロメラーゼ阻害剤をスクリーニングするために用いる抽出物を、テロメラーゼの蛋白質触媒サブユニット(hTERT)を過剰発現する293細胞から常法により調製した。これらの細胞は親293細胞の2〜5倍のテロメラーゼ活性を有することが判明した。パック細胞(培養物約100リットルから回収、200ml)を等量の低張緩衝液(10mM Hepes pH7.9、1mM MgCl2、1mM DTT、20mM KCl、1mM PMSF)に再懸濁し、ダンス型ホモジェナイザーを用いて溶解した。グリセロール濃度を10%に調節し、NaClをゆっくり加えて最終濃度最終濃度0.3Mとした。溶解した細胞を30分間撹拌し、次いで100,000×gで1時間ペレット化した。固体硫化アンモニウムをS100上清に42%飽和に達するまで加えた。この材料を遠心分離し、ペレットを元の量の5分の1に再懸濁し、50mM NaClを含む緩衝液「A」に対して透析した。透析後、抽出物を25,000×gで30分間遠心分離した。アフィニティクロマトグラフィの前に、商標Triton X-lOO(0.5%)、KCI(0.3M)およびtRNA(50μg/ml)を加えた。親和性オリゴヌクレオチド(
;下の場合は2'O-メチルリボヌクレオチド、上の場合はデオキシヌクレオチドを表す)を抽出物に加えた(抽出物10mlあたり1nmol)。30℃で10分間インキュベートした後、Neutravidinビーズ(Pierce;50%懸濁液を250μl)を加え、混合物を4℃で終夜回転させた。ビーズをペレット化し、0.3M KClを含む緩衝液「B」で三回、0.6M KClを含む緩衝液「B」で二回、0.3M KClを含む緩衝液「B」でさらに二回洗浄した。テロメラーゼを、0.3M KCl、0.15%商標Triton X-100および2.5モル過剰の置換オリゴヌクレオチド(
充填Neutravidinビーズ125μlあたり0.5ml)を含む緩衝液「B」中、室温で30分間溶出した。二回目の溶出を行い、一回目のものと合わせた。精製した抽出物は典型的に、抽出物1μlあたり1分間に取り込まれたヌクレオチド10fmol、または全蛋白質1mgあたり1分間に200ヌクレオチドの比活性を有していた。
オリゴヌクレオチド結合体によるテロメラーゼ阻害
Bio-Tel FlashPlateアッセイ法
テロメラーゼ活性の検出および/または測定に適したアッセイ法はTTAGGG(配列番号:8)テロメア反復配列のビオチン化テロメラーゼ基質プライマーへの付加、すなわちテロメラーゼによって触媒される反応の測定に基づいている。ビオチン化生成物をストレプトアビジンでコーティングしたマイクロタイタープレート中で捕捉する。下記のとおり、テロメラーゼ生成物を測定するために、33Pで標識した3.5テロメア反復配列に相補的なオリゴヌクレオチドプローブを用いる。未結合のプローブを洗浄によって除去し、捕捉テロメラーゼ生成物にアニールしたプローブの量をシンチレーション計数により定量する。
1. オリゴヌクレオチド結合体を濃縮保存液として保存し、PBS中に溶解した。
2. 試験のために、オリゴヌクレオチド結合体をPBS中で希釈して15倍標準保存液とし、96穴マイクロタイタープレートの二つのウェルに2μlを分配した(二回ずつアッセイした)。
3. テロメラーゼ抽出物を、テロメラーゼ希釈緩衝液1μlあたり1分間に取り込まれたdNTPの比活性0.04〜0.09fmolに希釈し、18μlを各試料のウェルに加えて、化合物と共に室温で30分間予備インキュベートした。
4. テロメラーゼ抽出物および試験するオリゴヌクレオチド化合物を含むウェルに混合原液10μlを加えることにより、テロメラーゼ反応を開始した。プレートを密封し、37℃で90分間インキュベートした。
5. 10μlのHCSを加えて反応を停止した。
6. 反応混合物25μlを96穴のストレプトアビジンでコーティングした商標FlashPlate(NEN)に移し、ゆっくり撹拌しながら室温で2時間インキュベートした。
7. ウェルを180μlの2×SSCでインキュベートせずに洗浄した。
8. ビオチン化テロメラーゼ生成物にアニールしたプローブの量をシンチレーション計数器で検出した。

オリゴヌクレオチド結合体の抗腫瘍活性
細胞アッセイ法
a. 細胞におけるテロメラーゼ活性の阻害および腫瘍細胞増殖の阻害
下記は試験結合体の腫瘍細胞増殖に対する効果を調べるために用いることができる、一般的細胞アッセイ法の説明である。ヒト胸部上皮細胞のコロニー(自発的に不死化)を、標準的方法および材料を用いて調製する。コロニーを、15センチメートルのプレートに各プレート約106細胞を播種することにより調製する。プレートをインキュベートして、細胞コロニーを約80%コンフルエンスまで増殖させ、その時点で各コロニーを2群に分ける。分割し、播種した後、1群は実験9のチオホスホロアミデートポリヌクレオチド結合体の亜急性用量に、所定の濃度(例えば、約100nMから約20μMの間)で、約4〜8時間暴露した。第2細胞群は未結合の対照オリゴヌクレオチドに同様に曝露する。
本発明の短いオリゴヌクレオチド結合体を、標準の技法を用いてハイブリダイゼーション試験または酵素阻害アッセイ法を行うことにより、テロメラーゼおよびRNA成分を有することが知られている他の酵素に対する活性(IC50)についてスクリーニングする。テロメラーゼに対するIC50値がスクリーニングにかけた他の酵素に対するIC50値に比べて低いオリゴヌクレオチドを、テロメラーゼに対する特異性を有するとする。
細胞毒性についての細胞死(XTT)アッセイ法を、HME50-5E、Caki-1、A431、ACHN、およびA549細胞タイプを用いて実施する。アッセイ法に用いる細胞株を、短いオリゴヌクレオチド結合体の一つに、約1μMから約100μMの範囲の濃度で、脂質存在下および非存在下、72時間暴露する。この期間中に、試料の光学密度(OD)を540ナノメートル(nm)の光線について求める。様々な細胞タイプで得られるIC50値は一般に1μM未満である。したがって、約100μM未満の濃度では有意な細胞毒性効果は観察されないと予想される。細胞毒性を調べるために、正常ヒトBJ細胞などの対照細胞株に加えて、卵巣腫瘍細胞株OVCAR-5およびSK-OV-3などの他の腫瘍細胞株を用いることもできることが理解されると思われる。MTTアッセイ法(Berridgeら、Biochemica 4:14-19, 1996)および商標alamarBlueアッセイ法(米国特許第5,501,959号)などの他の細胞毒性についてのアッセイ法も同様に用いることができる。

Claims (13)
- n=1であり、かつXがオリゴヌクレオチドOの3'核酸塩基の糖環に結合している、請求項1記載の化合物。
- Aがグアニンである、請求項1〜3のいずれか一項記載の化合物。
- イン・ビトロでテロメラーゼ酵素の活性を阻害する方法であって、テロメラーゼ酵素を請求項1〜6のいずれか一項記載の化合物とイン・ビトロで接触させる段階を含む方法。
- イン・ビトロで細胞の増殖を阻害する方法であって、細胞を請求項1〜6のいずれか一項記載の化合物とイン・ビトロで接触させる段階を含む方法。
- 薬学的組成物の製剤化法であって、
(a)請求項1〜6のいずれか一項記載の化合物を提供する段階と;
(b)化合物を薬学的に許容される賦形剤と混合する段階とを含む方法。 - 薬学的に許容される賦形剤中で製剤化された請求項1〜6のいずれか一項記載の化合物を含む薬学的組成物。
- 薬品における請求項1〜6のいずれか一項記載の化合物の使用。
- テロメラーゼ酵素の活性を阻害するため、または癌を治療するための薬剤の調製における請求項1〜6のいずれか一項記載の化合物の使用。
- テロメラーゼ酵素の活性を阻害するため、または癌を治療するための請求項1〜6のいずれか一項記載の化合物。
