Description
TRAF6(TNF受容体結合因子6)はNF−κB活性化受容体(RANK)シグナリングに対する重要なアダプタータンパク質である。本発明は、細胞膜を横切ることを可能にするペプチドリーダー配列を有する、または、有していない新規なTRAF6おとり(decoy)ペプチド(T6DP)を開示する。ここにおいて、TRAF6おとりペプチドは、リーダー配列が付着したときのみに、RANKLシグナリングおよびRANKL媒介破骨細胞分化に関連する初期の出来事を阻害することを示している証拠が開示されている。このデータは、NF−κB活性化受容体およびTRAF6の間の相互作用の標的化分断が、代謝性骨障害、白血病、関節炎、および骨の転移性がんの治療に有効であることを示している。
ヒト胎児腎臓293細胞系およびマウスマクロファージ細胞系RAW264.7はアメリカン・タイプ・カルチャー・コレクション(Rockville, MD)から入手した。293細胞は10%ウシ胎仔血清および抗生物質で補強したMEMで培養した。RAW264.7細胞は10%ウシ胎仔血清および抗生物質で補強したDMEM−F12において培養した。リン酸ERK、p38、およびJNKに対するモノクローナル抗体はNew England Biolabsから購入した。西洋ワサビパーオキシターゼ複合ヤギ抗ウサギIgGはBioRad Laboratories (Hercules, CA) から購入した。抗JNK1および抗IκBはSanta Cruz Biotechnology (Santa Cruz, CA) から購入した。西洋ワサビパーオキシダーゼ複合ヤギ抗マウスIgGはTransduction Laboratories (Lexington, KY) から購入した。タンパク質A/GセファローズビーズはPierce (Rockford, IL)から購入し、抗FLAGはSigma (St. Louis, MO)から購入した。酒石酸塩抵抗酸性ホスファターゼ(TRAP)陽性破骨細胞の染色は、実質的に(30)に記載されたやり方、またはシグマから入手した酸性ホスファターゼキットを使用することにより行った。
マウスFLAG付着TRAF5およびTRAF6をコードする発現プラスミド(31)はナカノ(H. Nakano)(順天堂大学、東京、日本)から提供され、FLAG付着TRAF2をコードする発現プラスミドはニ(J. Ni)(Human Genome Sciences, Inc.)から提供された。発現ベクター、並びに、GST、GST−NF−κB活性化受容体の細胞質ドメイン、およびGST−Jun(1−79)に対するGST融合タンパク質の精製については、以前記載してきた(7,8)。完全長マウスRANKL(また、TNF関連活性化誘導サイトカイン(TRANCE)としても知られている)の発現ベクター(pcDNA3.1−TRANCE)はチョイ(Y. Choi)(Rockefeller University, New York, NY)から提供された。
293細胞は、6ウェルプレートに0.6 x 106細胞/ウェルで入れられ、次の日に、(8)に記載されたようにトランスフェクトされた。プラスミドDNAの全量は、空のpCMVFLAGIベクターを加えることにより、一定に保たれた。細胞および調節された上清は、トランスフェクションの24〜36時間後に収穫された。溶解物は(8)の記載に従って調製した。ウエスタンブロット分析のために、全細胞溶解物(15〜30μg)またはGST親和性沈降により得られたタンパク質を8.5%SDS−PAGEで分離し、ニトロセルロース膜に電気泳動的転写を行い、示された抗体とインキュベートした。膜はその後、促進化学発光(ECL)システム(Amersham, Chicago, IL)を使用して現像した。
一次骨髄単球(BM)またはRAW264.7細胞は、48ウェル皿において、1 x 105細胞/ウェルまたは2 x 103細胞/ウェルの密度で培養した。それから、培養の開始時に示された要素により処理され、3日目に培地を交換した。1ウェルに存在する多核(>3核)で、TRAP陽性細胞の総数を、7〜10日目の間(BM)、または、5日目(RAW264.7)に計測することで、破骨細胞形成を評価した(30)。
20μlのグルタチオンアガロースビーズに接着された、等量のGSTまたはGST−RANK細胞質ドメイン(GST−RANKcd)融合タンパク質は、結合バッファー(20mM TRIS、pH8、150mM NaCl、1mM DTT、2mM EDTA、および0.1% NP−40)中で、エピトープ付着TRAFタンパク質が発現するようにプログラムされた293細胞の溶解物(50μg)および示されたペプチドと混合され、4℃で1時間、回転させられた。ビーズは遠心分離により収集され、結合バッファーで3回洗浄し、その後、低塩バファー(20mM TRIS、pH8、50mM NaCl、および1mM DTT)で1回洗浄した。結合タンパク質はSDS添加サンプルバッファーにより溶出し、煮沸した。溶出タンパク質は、7.5%SDS−PAGEされ、抗FLAG抗体によるウエスタンブロット分析が行われた。
RANKLで刺激されたRAW細胞から溶解物が、図面に示された説明に従って調製された。約30μgを、その後、示された抗体およびタンパク質A/Gセファローズビーズとともに、1時間の免疫沈降のために用いた。ビーズは遠心分離により収集され、溶解バッファーで3回洗浄し、その後、低塩バファーで1回洗浄した。JNK活性は、先の記載(8)に従って、基質として外因性GST−Jun(1−79)を用いて分析された。キナーゼ活性は、ホスホイメージャー(PhosphoImager)およびイメージクアントソフトウエア(Imagequant Software)(Molecular Dynamics, Sunnyvale, CA)を用いて定量された。
(34)の記載に本質的に従って、トランスフェクト細胞から核抽出物を調製した。等量の核タンパク質を、(34)の記載に従い、HIV−LTR由来の32P標識NF−κBオリゴヌクレオチドとのEMSA反応に用いた。NF−κB活性はホスホイメージャーおよびイメージクアントソフトウエアを用いて定量された。
TRAF6のみに結合し、TRAF2またはTRAF5には結合しない、RANKにおける新規なTRAF6結合ドメインは以前に確認されていた(8)。293細胞でトランスフェクトされた場合、NF−κB活性化受容体のこの領域は、NF−κBを活性化するには十分なものであった(8)。ヒトおよびマウスのCD40およびRANKにおけるTRAF6結合部位の構造を基礎として配列を整列させることにより、TRAF6結合モチーフPxExx(Ar/Ac)(Arは芳香族残基およびAcは酸性残基を示す)(図1A)が明らかになった。RANK配列の注意深い調査の結果、3つの潜在的TRAF6結合部位が存在することが示された(図1A)。
マウスマクロファージ細胞系RAW264.7は、細胞表面にNF−κB活性化受容体を発現し、NF−κB活性化受容体リガンドで刺激した場合、4〜5日後に、多核の酒石酸塩抵抗リン酸(TRAP)陽性破骨細胞に分化する(図2A)。L−T6DP−1がRANKL媒介破骨細胞分化を阻害することができるかを決定するために、RAW264.7細胞を、徐々に濃度を増加させたL−T6DP−1またはT6DP−1のいずれか、および、30ng/mlのRANKLと、4日間、共存培養した。
RAW264.7細胞系で得られた結果をさらに補強するために、1次マウス由来単球におけるRANKL媒介破骨細胞分化阻害に対するこれらのペプチドの能力を検査した。多核、TRAP陽性破骨細胞を染色することで、骨髄由来単球をRANKLおよびM−CSFで共刺激すると、7〜10日後に破骨細胞分化が起こるものと決定された(30)。RAW264.7細胞での結果と同様に、L−T6DP−1およびL−T6DP−2の両方がTRAP陽性破骨細胞の発生を、量依存的に阻害し、その効果は、L−T6DP−1のほうが高かった(図3)。リーダー配列を欠いたTRAFおとりペプチドは破骨細胞分化を阻害することができず(図3)、このペプチドは細胞に対して毒性がないことが示唆される。30μMのL−T6DP−1で処理された後、細胞はTRAP陽性となるが、多核化破骨細胞は100μMのL−T6DP−1では観察されず、破骨細胞は観察されなかった(図3B)。これらの結果を合わせるとTRAF6とRANKの相互作用は、RANKL媒介破骨細胞分化に必須のものであることが示される。
1個のTRAF6結合ドメインを有するNF−κB活性化受容体の突然変異体はNF−κBを活性化するのに十分であり(8)、優性ネガティブTRAF6は、293細胞においてRANK媒介NF−κB活性化を阻害する(9)。RANKL誘導NF−κB活性化に対するTRAF6結合ペプチドの効果を調査するために、RANKLで刺激したときにNF−κBを活性化するRAW264.7細胞においてNF−κB活性化を調査した(37)。RANKL刺激RAW264.7細胞は、ゲル移動度シフトアッセイで示されるように、NF−κBを活性化する(図4A)。NF−κB活性化は、L−T6DP−1による前処理のみで量依存的に抑制される(図4A)。NF−κBの活性化および抑制を同時に起こさせるIκBαの水準は、図4Bに示されるとおりである。これらのデータは、L−T6DP−1が、RAW264.7細胞において特異的にRANKL媒介NF−κB活性化を阻害することを示す。
RANKの細胞質ドメインは、TRAF1、2、3、5および6を含む多くのTRAF分子と相互作用する(7−9、11、12)。TRAF1、2、3および5はNF−κB活性化受容体のC末端テールと相互作用するが、TRAF6はNF−κB活性化受容体の細胞質ドメインの膜近位領域と相互作用する。ここで開示されたTRAF6結合ペプチドが、TRAF6のRANKとの相互作用を阻害することを確認するために、競合GSTプルダウンアッセイを行った。
NF−κB活性化受容体の刺激は、JNK、ERKおよびp38キナーゼを含むMAPKファミリーの構成員を活性化する。ペプチド結合TRAF6による、NF−κB活性化受容体リガンド誘導JNK、ERKおよびp38キナーゼの活性化に対する阻害能力をRAW264.7細胞において調査した。インビトロJNKキナーゼアッセイの結果は、JNKはRANKLによって活性化され、L−T6DP−1ペプチドによる処理だけがRANKL媒介JNK活性化を阻害することができることを示した(図5A)。JNKの結果と同様に、L−T6DP−1だけが、RAW264.7細胞においてNF−κB活性化受容体リガンドにより誘導されるERK(図5B)およびp38キナーゼ(図5C)の活性化を阻止することができた。前記の結果を考慮すると、本発明は、L−T6DP−1がNF−κBリガンド媒介破骨細胞分化、並びに、NF−κB、JNK、ERKおよびp38キナーゼ活性化を含むNF−κB活性化受容体リガンド開始初期シグナリングを抑制できることを示している。
乳がんは、アメリカ合衆国における女性の最も一般的な悪性腫瘍であり、女性のがん死亡の2番目に多い原因である。乳がんを患った女性は、骨転移の危険性を有する。乳がんを患った患者の5から10%は、初期の骨の転移性疾患になる。転移性乳がん由来の骨溶解性骨疾患を有する患者は、病理学的骨折、骨の痛み、索状組織圧縮およびハイパーカルセミアの危険性が大きい。骨転移の現在の標準的な治療方法は、骨に起こる進行を遅らせるが、完全には防ぐことができないビスホスホネート治療である。さらに、全ての患者にこの治療の効果があるわけではない。より効果的な治療方法が望まれており、この病気のさらなる生物学的および分子的な詳細な分析が必要とされる。実際に、近年、乳がん細胞を注射されたヌードマウスモデルにおいて、オステオプロテゲリン(OPG)が骨溶解を阻害し、腫瘍の負担を減少させることが示された。
以前に記載された方法(J. Biol. Chem., 276: 12235-12240,2001)と同様のELISAを基本とする方法がRANKおよびTRAF6の相互作用を阻害する小さな分子を発見するために用いることができる。簡単に説明すると、T6DPを含むペプチドをビオチニル化し、TRIS緩衝塩溶液(50mM TRIS pH7.5、150mM NaCI)に溶解し、ニュウートリアビシン(NeutrAvidin)でコートした96ウェルマイクロティッタープレートのウェルに入れた。プレートは4℃で一晩振とうさせ、TBS−BT(TBS含有0.1%ウシ胎仔血清アルブミン(BSA)および0.1%トゥーイン(Tween)20)に続いてTBSですすいだ。ライブラリーからの検査小分子を含む溶液が、その後ウェルに加えられた。6個のヒスチジン付着TRAF6(309−522)がその後各ウェルに加えられた。プレートを室温で1時間インキュベートし、その後TBS−BTで3回洗浄した。TRAF6のC末端に対する抗体がその後ウェルに加え、プレートを室温で1時間インキュベートし、その後TBS−BTで3回洗浄する。ヤギ抗マウスアルカリ性ホスファターゼからなる第二抗体をその後各ウェルに加えられ、プレートを室温で1時間インキュベートし、その後TBS−BTで3回洗浄した。プレートは以前記載されたように(Darnay et al., J. Biol. Chem. 274: 77247731, 1999)、蛍光プレートリーダーを使用し、アルカリ性ホスファターゼ活性を検査した。

Claims (8)
- TNF受容体結合因子6(TRAF6)媒介シグナリングを阻害するポリペプチドであって、
a) PxExx(Ar/Ac)で示されるコンセンサスモチーフを含むTRAF6結合ドメイン、および
b) カポシ繊維芽細胞成長因子シグナル配列であるリーダーシグナル配列
を含むことを特徴とするポリペプチド。
(但し、上記PxExxのxは任意のアミノ酸を示し、上記Ar/AcのArは芳香族残基、Acは酸性残基を示す。) - 前記TRAF6結合ドメインが、CD40、NF−κB活性化受容体、IL−1受容体結合キナーゼ1(IRAK1)、IL−1受容体結合キナーゼ2(IRAK2)、IRAK−M、およびRIP2からなる群から選択されるタンパク質からのTRAF6結合ドメインであり、かつ配列番号1〜18からなる群から選択される配列を含む請求項1記載のポリペプチド。
- 前記ポリプチドが、配列番号19および20からなる群から選択される配列を含む請求項1記載のポリペプチド。
- 請求項1記載のポリペプチドによるNF−κB活性化受容体およびTRAF6間の相互作用の阻害の結果、NF−κB活性化受容体リガンド誘導破骨細胞分化を阻害するポリペプチド。
- 前記ポリペプチドが、リポソーム、ウイルス、および遺伝子伝達ベクターからなる群から選択された手段により細胞に伝達される請求項4記載のポリペプチド。
- 前記破骨細胞分化が、乳がん細胞によって誘導される請求項4記載のポリペプチド。
- 代謝性骨障害、白血病、多発性骨髄腫、関節炎、または骨の転移性がんからなる群から選択される疾患を治療するための請求項4記載のポリペプチド。
- 請求項1〜7のいずれか1項に記載のポリペプチドおよび薬学的に許容される担体を含む製薬組成物。
