【発明の属する技術分野】
本発明は、二次元的に放射される微弱な電磁波や粒子線を検出する二次元微弱放射検出装置に関する。

【従来の技術】
近年、病気の予防や診断を有効に行うために、DNAチップによるハイブリッド形成法が注目されている。これは、DNAの特定部位の断片(セル)をガラス基板の上にマトリックス状に配置したものである。そして、DNAチップは、検査対象の人から採取した血液などの検体に蛍光物質を添加して検体のDNAを標識化し、この検体とDNAチップ上のセルとを接触させてハイブリダイゼージョンを行い、検査対象者が特定の病気に係る遺伝子を有するか否かを判定する、などに用いられる。

従来、検体中のDNAがDNAチップのどのセルとハイブリッドを形成したかを検知する場合、図11に示したようなDNAチップリーダと称する装置を用いて行っている。このDNAチップリーダ10は、いわゆる共焦点方式となっていて、図示しない検査ステージ上に配置したDNAチップ(試料)12にレーザ光を照射するレーザ光源14を備えている。レーザ光源14の下方には、集光レンズ16とピンホール18とが直列に配置してあって、レーザ光源14の出射したレーザビーム20を絞るようになっている。

ピンホール18を通過したレーザビーム20は、DNAチップ12から放射された蛍光とレーザビーム20とを分離するためのダイクロイックミラー22を透過したのち、コリメートレンズ24によって平行光にされる。コリメートレンズ24によって平行光となったレーザビーム20は、一対のガルバノミラー26、28によって反射されたのち、対物レンズ30に入射し、DNAチップ12上に収束してセルに照射される。ガルバノミラー26、28は、レーザビーム20をDNAチップ12の面に沿って走査するためのもので、例えばガルバノミラー26を回転させるとレーザビーム20がDNAチップ12の面上をX方向に移動し、ガルバノミラー28を回転させるとレーザビーム20がY方向に移動する。従って、ガルバノミラー26、28の回転を制御することにより、DNAチップ12にマトリックス状に配置した任意のセルにレーザビーム20を照射することができる。

DNAチップ12のセルは、蛍光物質によって標識化された検体中のDNAとハイブリッドを形成すると、レーザビーム20が照射されると蛍光を発する。このセルから放射された蛍光は、対物レンズ30、ガルバノミラー28、26、コリメートレンズ24を介してダイクロイックミラー22に入射する。そして、ダイクロイックミラー22は、入射した蛍光のみを選択的に90度偏向し、ピンホール32を介して光電子倍増管34に入射する。光電子倍増管34は、入射した蛍光によって光電子を生成し、それを増幅して電圧パルスとして出力する。従って、光電子倍増管34の出力を監視することにより、DNAチップ12のどのセルが蛍光を放射したか、すなわち検体中にどのセルとハイブリッドを形成する遺伝子が含まれているかを知ることができる。

【発明が解決しようとする課題】
しかし、上記した従来のDNAチップリーダ10は、レーザビーム20をステップ状に移動させてDNAチップ12の面を走査する必要がある。このため、N×N個のセルをマトリックス状に配置したDNAチップ12の面に沿ってレーザビーム20を走査すると、Nの数が100〜1000(セル数が1万個〜100万個)と大きくなると、走査時間が指数関数的に増大し、セルの情報を読み取るのに多大な時間を必要とする。このため、複数の光電子倍増管34を平面的に配置し、DNAチップ12の全体にレーザビームを照射し、各光電子倍増管34の出力パルスを並列的に読み出して二次元情報を得る試みもあるが、光電子倍増管34は高価であるとともに、大きな設置スペースを必要とするために現実的でない。

また、DNAチップ12のハイブリッドを形成したセルの位置を二次元的に得るために、CCD方式光子計数ビデオカメラを用いることが考えられる。このCCD光子計数ビデオカメラは、入射した光子を光電変換して光電子を発生させ、この光電子を多数のキャピラリー(毛細管)からなるマイクロチャンネルプレート(MCP)と称する二次電子倍増管によって、各キャピラリー(チャンネル)ごとに電子数を増幅し、これを再び蛍光体に入射して電子→光変換し、変換した光をCCDビデオカメラで受信するようになっている。しかし、このCCD方式光子計数ビデオカメラを用いる場合、次のような課題がある。

CCDビデオカメラからの信号に対して光子計数モードの処理を行う。すなわち、CCDビデオカメラの各画素(ピクセル)に相当するCCD素子の出力信号を2値化し、単位時間当りの出力信号(入射した光子の数)を計数する。しかし、CCDビデオカメラの各素子に対する出力信号の読み出しは、せいぜい100回/s程度である。

一方、DNAチップ12のセルから発生する蛍光は極めて微弱であって、MCPの各チャンネル(キャピラリー)には、光子が希にしか入射しない。しかも、MCPからCCD素子に入射する電子のパルスの持続時間は、0.1〜10nsと桁外れに短い。このため、CCDビデオカメラの各素子は、希にしかこない入射光子に対応した電子のパルスを、電子のパルス持続時間に比べて106 〜108 倍も長い、信号の読み出し周期となる10ms程度の間積分することになる。ところが、CCDには暗電流というノイズがあり、このノイズも読み出し周期の間積分されることになり、パルスのS/Nが106 〜108 以上ないと検出システムが実現できないという非現実的なこととなる。

本発明は、前記従来技術の欠点を解消するためになされたもので、二次元的な微弱な放射を高速、かつ高精度で検出できるようにすることを目的としている。また、本発明は、微弱な放射に基づいた二次元カラー画像を容易に得られるようにすることを目的としている。

【課題を解決するための手段】
上記の目的を達成するために、本発明に係る二次元微弱放射検出装置は、光子の入射によって電子を放出する光電変換部と、この光電変換部に対面配置されて光電変換部が放出した電子を増幅する電子増幅部が複数設けられた増幅モジュールと、この増幅モジュールを構成している前記各電子増幅部に対応して設けられ、電子増幅部からの電子が入射する電子検出部が複数設けられた検出モジュールと、この検出モジュールを構成している前記各電子検出部を直交変調パターンに基づいて作動させる動作制御部と、この動作制御部の制御信号と前記各電子検出部の出力信号とに基づいて、前記光電変換部に入射した前記光子の位置を求める光入射位置演算部と、を有することを特徴としている。

また、本発明に係る二次元微弱放射検出装置は、光子の入射によって電子を放出する光電変換部と、この光電変換部に対面配置されて光電変換部が放出した電子を増幅する電子増幅部が複数設けられた増幅モジュールと、この増幅モジュールを構成している前記各電子増幅部に対応して設けられ、電子増幅部からの電子が入射する電子検出部が複数設けられた検出モジュールと、この検出モジュールを構成している前記各電子検出部を直交変調パターンに基づいて作動させる動作制御部と、この動作制御部の制御信号と前記各電子検出部の出力信号とに基づいて、前記光電変換部に入射した前記光子の位置を求める光入射位置演算部と、前記各電子検出部の出力信号の大きさに基づいて前記光子のエネルギーを求め、予め与えられた色信号に変換する波長演算部と、を有することを特徴としている。

【作用】
上記のごとく構成した本発明は、微弱な放射による光子が光電変換部によって光電子に変換され、この光電子(電子)が増幅モジュールにおいて数を増幅されたのち、検出モジュールに入射する。検出モジュールは、増幅モジュールの複数の電子増幅部に対応して電子検出部が設けてあって、これらの電子検出部が直交変調パターン(例えば、2値直交変調パターンであるアダマール行列の各行に対応するパターン)に従って作動される。従って、常にn×n個の電子検出部の1/4から出力信号(データ)が得られる。そして、この得られた出力信号について、光入射位置演算部において得られたデータの逆変換(例えば、アダマール逆変換)を行うことにより、出力信号を出力した電子検出部を特定することができ、光電変換部に入射した光子の位置を求めることができる。従って、二次元の微弱な放射を高速、かつ高精度で二次元的に検出することができ、微弱な放射による二次元映像を得ることができる。

また、本発明においては、電子検出部の出力信号の大きさに基づいて入射した光子のエネルギーを求め、このエネルギーに対応して予め与えられている色信号に変換するようになっているため、複数の波長の光に基づく光子が入力した場合に、二次元的に求めた微弱放射の画像を通常のカラー画像とすることが可能で、二次元微弱放射の状態をより容易に認識、理解することができる。また、カラーの暗視カメラなどを容易に形成することができる。

電子検出部の出力信号の大きさは、電子検出部が出力する出力信号の頻度に基づいて求めるようにすると、計測の揺らぎによる誤りを避けることができ、検出すべき出力信号の大きさを容易、確実に求めることができる。そして、光電変換部の前面に、電磁波または粒子線の入射によって光子を放出する発光部を設けると、微弱なX線やガンマ線、α線などの検出が可能となる。

【発明の実施の形態】
本発明に係る二次元微弱放射検出装置の好ましい実施の形態を、添付図面に従って詳細に説明する。
図1は、本発明に係る二次元微弱放射検出装置の実施形態の概略ブロック図であって、DNAチップリーダに適用した例を示したものである。図1において、二次元微弱放射検出装置である微弱光検出装置40は、レーザ照射ユニット42を有している。このレーザ照射ユニット42は、検査ステージ44の上に配置した試料となるDNAチップ46にレンズ系48を介してレーザビーム50を照射できるようにしてある。また、微弱光検出装置40は、検査ステージ44の上方に配設した検出ユニット52と、この検出ユニット52の出力信号に基づいハイブリッドを形成したDNAチップ46内のセルの位置などを求める信号処理ユニット54を備えている。

検出ユニット52は、詳細を後述するように、微弱な光を検出する複数のマイクロキャピラリーを備えた検出部56と、この検出部56を作動させる動作制御部58とからなっている。また、信号処理ユニット54は、検出部56の出力信号を読み込むデータ読取り部60を有する。さらに、信号処理ユニット54には、データ読取り部60の出力側に接続した振幅検出部62、振幅検出部62の出力側に設けた計数部64、計数部64からの信号が入力する画像作成部68、画像作成部68の出力側の設けた出力部70を有している。そして、画像作成部68は、その作用を後述する画像演算部68aとスペクトル作成部86bとから形成してある。また、出力部70には、出力装置となっている表示装置70やプリンタ72、外部記憶装置74などが接続されている。

検出ユニット52の検出部56は、図2に示したように、レンズ80、光電変換部82、増幅モジュールとなるマイクロチャンネルプレート84、検出モジュール86を備えていて、レンズ80が検出窓となっている。そして、検出部56は、図3に示したように、レーザビーム50や外来光を遮蔽する遮光容器56aが設けてあって、この遮光容器56aの前端面にレンズ80が取り付けてある。さらに、遮光容器56a内には、レンズ80の後方に光電変換部82、マイクロチャンネルプレート84、検出モジュール86をこの順に配置してある。これらの光電変換部82、マイクロチャンネルプレート84、検出モジュール86は、超高速フォトカウンティング方式の二次元受信器56bを構成している。また、レンズ80と二次元受信器56bとの間には、光学フィルタ56cが配置してある。この光学フィルタ56cは、レーザ光の周波数帯域を遮断するとともに、蛍光に対応した周波数帯域の光を選択的に透過して二次元受信器56bに入射させるようになっている。

二次元受光器56bは、図4に示したように、真空容器56dを有していて、真空容器56dの前面に光電変換部82が取り付けてある。そして、マイクロチャンネルプレート84と検出モジュール86とは、真空容器56dの内部に配設してある。また、二次元受信器56bは、光電変換部82とマイクロチャンネルプレート84と検出モジュール86とが相互に密接するように配置してある。

検出部56は、レンズ80がDNAチップ46と対面するように配置される。そして、レンズ80は、DNAチップ46のセルが放射する微弱放射である微弱光な蛍光(入射光)を光電変換部82上に結像させる。そして、光電変換部82は、レンズ80と光学フィルタ56cとを透過した蛍光(光子)が88入射すると、電子(光電子)90を放出する。また、光電変換部82が放出した電子90は、詳細を後述するマイクロチャンネルプレート84において数が107 〜107 倍程度に増幅され、増幅電子92となって検出モジュール86に入射し、検出モジュール86によって検出される。

マイクロチャンネルプレート84は、二次電子倍増管である複数のマイクロキャピラリー94を、DNAチップ46のセルに対応させてマトリックス状に配設した構造を有している。マイクロチャンネルプレート84を構成しているマイクロキャピラリー94は、図5に示したように、直径が6μm、長さが1cm程度の加速管96と、この加速管96の両端に設けたカソード98、アノード100からなっている。そして、マイクロキャピラリー94は、カソード98とアノード100とが直流電源102に接続され、1000〜10000Vの直流高電圧がカソード98とアノード100との間に印加してある。このため、カソード98側から加速管96内に入射した電子90は、カソード98とアノード100との間に印加してある高電圧によって加速され、加速管96の内壁に衝突するたびに二次電子を生じて雪崩的に数が増幅され、増幅電子92としてアノード100側から出射される。

検出モジュール86は、図6に示したようになっている。すなわち、検出モジュール86は、実施形態の場合、MOSトランジスタからなる複数の検出トランジスタ104(104ij)と、MOSトランジスタからなる複数の読み出しトランジスタ106(106a、106b、106c……… )とを備えている。検出トランジスタ104ij(i=1、2、3、………n、j=1、2、3、………n)は、マイクロチャンネルプレート84を構成しているマイクロキャピラリー94に対応してn×n個がマトリックス状に配置してある。また、読み出しトランジスタ106は、マトリックス状に配置した検出トランジスタ104の各列に対応してn個設けてある。

検出トランジスタ104は、各行ごとにゲートがゲート制御線108(108a、108b、108c、………)に接続され、これらのゲート制御線108が動作制御部58を構成しているゲート切替回路110に接続してある。また、検出トランジスタ104は、ドレインが各列ごとにデータ線112(112a、112b、112c、………)を介して読み出しトランジスタ106のソースに接続してある。そして、各読み出しトランジスタ106は、ドレインが信号処理ユニット54のデータ読取り部60に接続してあり、それぞれのゲートが対応する読み出し線114(114a、114b、114c、………)に接続してある。
また、検出トランジスタ104のソースには、マイクロキャピラリー94の出力側に対面して設けた検出電極120が接続してある。

各読み出し線114は、動作制御部58を構成している読み出し線切替回路116に接続してある。動作制御部58は、ゲート切替回路110、読み出し線切替回路116、切替制御部118などから構成してある。そして、切替制御部118は、詳細を後述するように、2値直交変調パターン(直交変調パターン)に基づいて切替制御信号を生成し、この切替制御信号をゲート切替回路110と読み出し線切替回路116とに与え、各検出トランジスタ104を直交変調パターンに基づいて切替動作させるようになっている。

このように構成した実施形態に係る微弱光検出装置40の作用は、次のとおりである。まず、図示しない検体とハイブリッドを形成させたDNAチップ46を検査ステージ44の上に配置し、レーザ照射ユニット42によってレーザビーム50をDNAチップ46の全体に照射する。DNAチップ46のセルが蛍光物質によって標識されている検体のDNAとハイブリッドを形成していると、そのセルから蛍光が放射される。この蛍光(光子)は、検査ステージ44の上方に設けた検査ユニット52の検出部56に入射する。

検出部56に入射した光子88は、図2に示したように、レンズ80を透過して光電変換部82によって電子(光電子)90に変換される。そして、電子90は、マイクロチャンネルプレート84を構成しているマイクロキャピラリー94の増幅管96に入射する(図5参照)。電子90は、増幅管96に入ると、増幅管96の両端に印加されている高圧直流電圧によって加速され、加速管96の内壁に多数回衝突して二次電子を生成し、個数が106 〜107 倍程度に増幅される。この増幅電子92は、検出モジュール86の検出電極120に入射し、検出電極120を帯電させる。従って、検出トランジスタ104を順次切り替えて動作させることにより、どの検出トランジスタ104の検出電極120に増幅電子92が入射したかを知ることができる。

ところで、DNAチップ46のセルが放射する蛍光は、極めて微弱であって、マイクロチャンネルプレート84の各マイクロキャピラリー94には、光電変換により生成された電子90が希にしか入射しない。すなわち、検出モジュール86の各検出電極120には、増幅電子92が希にしか入射しない。このため、ゲート1チャンネル(1つのゲート制御線108)、読み出し1チャンネル(1つの読み出し線114)を選択してデータを読み出す場合、図7(a)に示したように、データ読取り部60は、まばらにしか検出パルス122を出力しない。しかも、ゲート制御線108と読み出し線114とを順次切り替えるためにn×n回の切り替えが必要で、nの数が100〜1000(セル数が1万個〜100万個)となると、多大な読み出し時間を必要とする。

そこで、複数のゲート制御線108と複数の読み出し線114とを選択して複数の検出トランジスタ104を同時に駆動すると、各データ線112には、図7(b)に示したように、検出トランジスタ104を個々に駆動したときより多くのパルス124が得られる。そして、データ読取り部60は、これらを合成したパルスを出力するため、同図(c)に示したように、時系列における検出パルス122の密度を高くすることができる。しかし、このままでは、どの検出トランジスタ104の出力によって得られた検出パルス122であるかを知ることができず、DNAチップ46のどのセルがハイブリッドを形成しているかを特定することができない。このため、この実施形態においては、直交変調パターンに基づいた駆動信号を生成して検出トランジスタ104を駆動するようにしている。

以上に述べてきた直交変調パターンとしては、2値直交変調パターンであるアダマール行列の各行に対応した変調パターンが適している。アダマール行列は、要素が「+1」と「−1」とからなっていて、対角線に沿って対象位置にある要素が同じである対称行列となっている。例えば、一次のアダマール行列H(1) を具体的に書くと、

そこで、実施形態においては、検出ユニット52の動作制御部58を構成している切替制御部118が、スイッチをオンにする場合を「+1」、スイッチをオフにする場合を「−1」に対応したアダマール行列に基づいて、検出トランジスタ104の切替動作信号を作成し、切替制御信号としてゲート切替回路110と読み出し線切替回路116とに与える。例えば、検出モジュール86が8×8個の検出トランジスタ104で構成した場合、検出トランジスタ104の動作信号は図8のようになる。この図8において斜線を施した部分が動作電圧を与えられてオンとなる+1に相当し、白抜きの部分がオフである−1に相当している。

すなわち、切替制御部118は、ゲート切替回路110にアダマール行列に従った切替制御信号を与え、ゲート制御線108を介して各検出トランジスタ104のゲートにアダマール行列に従ってゲート電圧を切り替えて印加するとともに、読み出し線切替回路116にアダマール行列に基づいた切替信号を与えて読み出しトランジスタ106をアダマール行列に従って切り替えて動作させる。例えば、検出トランジスタ104が8×8個である場合、切替制御部118は、アダマール行列に基づいて図8の下部に示したn個の変調モードを生成し、ゲート切替回路110に図8の下部右側に示した0次の動作信号(切替信号)を与え、すべてのゲート制御線108をゲート電源に接続し、すべての検出トランジスタ104のゲートにゲート電圧を印加するとともに、読み出し線切替回路116に図8の下部左側の0次から7次の切替信号を与え、読み出しトランジスタ106をアダマール行列に基づいて順次切り替えて駆動する。

そして、切替制御部118は、読み出しトランジスタ106に対して0次から7次までの切り替えが終了したならば、ゲート切替回路110に1次の駆動信号を与え、第1行、第3行、第5行、第7行のゲート制御線108aに接続した検出トランジスタ104のゲートに電圧を印加し、この状態で読み出しトランジスタ106を0次から7次まで切り替える。このようにして、切替制御部118は、検出トランジスタ104のゲートに対して印加する電圧を、0次から7次まで順に切り替えるごとに、読み出しトランジスタ106を0次から7次まで切り替える。これにより、検出トランジスタ104は、常に半分がゲート電圧を印加され、半分のデータ線112がオン状態となり、全体の1/4の検出トランジスタ104から出力信号がデータ読取り部60に入力する。

データ読取り部60は、検出トランジスタ104の出力信号として入力する電流を電圧に変換して増幅し、図6に示したような出力パルス130を電圧として出力する。この出力パルス130は、図1に示したように、信号処理ユニット54の振幅検出部62に入力される。なお、データ読取り部60の出力パルス130は、A/D変換するようにしてもよい。

検出トランジスタ104のゲートをアダマール行列に基づいて切り替えて電圧を印加し、読み出しトランジスタ106をアダマール行列によって切り替えて動作させた場合、データ読取り部60の出力するパルス130(アダマール行列に基づいて変調したデータ)から、アダマール逆変換をすることにより復調することにより、パルス130を出力した検出トランジスタ104を求めることができる。すなわち、ゲート制御線108を行、読み出し線114(データ線112)を列とする頻度データ行列を考えた場合、この行方向と列方向とにアダマール逆変換(二次元アダマール逆変換)を行うことにより、任意に選択した1つのゲート制御線108と1つの読み出し線114との組み合わせによる検出トランジスタ104を特定することができる。従って、画像演算部68aは、詳細を後述するように入射位置演算部となっていて、計数部64の係数値に基づいてアダマール逆変換をすることにより、増幅電子92が入射した検出トランジスタ104を求めることができ、光電子90が入力したマイクロキャピラリー94を特定することができ、光電変換部82のどの位置に光子88が入射したかを知ることができる。よって、DNAチップ46のどのセルが蛍光を放射したかを知ることができる。

信号処理ユニット54の振幅検出部62は、データ読取り部60の出力するパルス130を振幅(電圧の大きさ)を求めて計数部64に入力する。すなわち、DNAチップ46のセルに結合させる図示しない検体に複数の蛍光物質が添加してある場合、セルがハイブリッドを形成すると、DNAチップ46からは複数の蛍光が放射される。蛍光物質の放射する蛍光は、その蛍光物質に特有の波長を有する。従って、蛍光物質が異なると、異なる波長の蛍光が放射され、光電変換部82に入射する光子88のエネルギーがそれぞれ異なる。このため、光電変換部82において生成される電子(光電子)90は、入射した光子88のエネルギーの違いによって運動エネルギーが異なり、波長の短い蛍光が入射するとより運動エネルギーの大きな電子90が生成される。そして、より運動エネルギーの大きな電子90がマイクロキャピラリー94に入射すると、より多くの二次電子が生成され、増幅電子92の数が多くなる。従って、波長の短い蛍光による光子88に基づく電子90が入射したマイクロキャピラリー94に対応した検出トランジスタ104の出力が大きくなり、データ読取り部60の出力するパルス130の大きさとなる振幅(パルス130の高さ)がより大きくなる。つまり、データ読取り部60の出力パルス130は、DNAチップ60が複数の蛍光を放射する場合、図9に示したように振幅の異なったものとなる。
