Description
本明細書で用いる「トランスフェクション」という用語は、核酸を細胞に導入することを意味する。核酸はいかなる起源のものであってもよく、受け取る細胞は、原核細胞または真核細胞である。
(i)核酸、特に問題の配列をコードする核酸、
(ii)インテグリン結合性成分、
(iii)ポリカチオン性核酸結合性成分、および
(iv)脂質成分
を含む複合体も開示する。その複合体は主にインテグリン媒介トランスフェクションベクターである。
a)X1SM(配列番号1);
b)LX2HK(配列番号2);
c)PSGX3ARA(配列番号9);
d)SX4RSMNF(配列番号16);および
e)LX5HKSMP(配列番号18),

実施例
実施例1
本研究で用いたペプチドライブラリー、C7C、はNew England Biolabs Incから得た。ファージ生長、滴定および増幅過程は製造者のハンドブックに記載のように行った。そのライブラリーは長さで7残基のランダムなペプチド配列からなり、シスチン残基に隣接させ、空気中での酸化により環化を可能にした。そのライブラリーは少なくとも1×109の異なるアミノ酸配列を含むらしい。
HAE細胞を24ウェルプレート中で集密まで増殖させた。用いたHAE細胞はカルホルニア大学、サンフランシスコのディエーター・グルエナート博士(現ベルモント大学)からの贈り物として得た1HAEo細胞であった。細胞をTris緩衝食塩水、pH7.4(TBS)で2度洗浄した後、4℃で30分間、ウェル当り2mlの2%マーベル、5%ウシ血清アルブミン(BSA)−TBSで細胞をブロックした。ブロッカーを除き、2×1011のファージを1mlの2%マーベル、5%BAS−TBSに加えた。そのファージを2時間振とうにより結合させた後、5回2%BSA−TBS洗浄し、4℃で5分振とうし、その後数秒間2%のBSA−TBSでさらに5回洗浄した。4℃で振とうしながら10分間ウェルに400μlの76mMクエン酸緩衝液pH2.5を添加して溶出させた。溶出液を除去し、600μlの1M Tris緩衝液pH7.5で中和し、溶出した画分として保持した。残った細胞を1mlの30mM Tris緩衝液pH8.0,1mMのEDTAで1時間氷上で溶菌した。細胞をプレートから捨て、溶出液をミクロ遠心管に移し、短く渦巻した。その溶出液は細胞関連画分として保持した。
組織培養HAE細胞へのファージの結合を、全細胞ELISAにより研究した。100mlのHanks Balanced Salt Solution(HBSS)中の約8×104HAE細胞を96ウェルプレートの各ウェルに添加し、細胞が付着するまで37℃でインキュベートした。細胞をHBSS中でおだやかに洗い、30分間HBSS中の0.5%BSAの添加によりブロッキングした。ブロッカー溶液中の1×1010のファージ粒子を各ウェルに加え、室温で40分間結合させた。結合していないファージを、HBSSで2回洗うことにより除き、結合したファージを3.7%パラホルムアルデヒド中で10分インキュベートすることにより細胞に固定した。細胞をPBS中で洗い、ブロッキング緩衝液中で45分インキュベートし、PBSで3回洗った。結合したファージは、1時間ブロッキング緩衝液中で1:5000に希釈した、ホースラディッシュパーオキシダーゼ(HRP)結合抗M13抗体の添加により検出した後、PBSで3回洗い、2,2’−アジノ−ビス(3−エチルベンズチアゾリン−6−スルホン酸(ABTS)基質溶液でELISAを発色させ、405nmでのプレートリーディング分光光度計における吸光度を読んだ。実験を3T3細胞を用いて繰返し、空のウェルを用い、結合親和性を比較することでHAE細胞に選択的に結合するファージの同定を可能にした。選択したペプチドについての結果を図2に示す。
次のオリゴリシン−ペプチドをトランスフェクション実験のために製造した。

K16:DNA結合部分、標的リガンドなし
望ましい細胞結合の特徴を示すファージから同定したペプチドを標準的な固相ペプチド合成化学を用いて合成し、16のリシンの尾を標準的な方法で付けた。標的ペプチドPと同じアミノ酸組成よりなるがランダムな順序である、対照ペプチド(S)はリポポリプレックス製剤への導入のため合成した。HAEおよび3T3細胞のトランスフェクションは、24時間前にプレートした20,000の細胞を含む96ウェルプレート中で行った。トランスフェクション複合体中で、DNA電荷に対するペプチドの割合(+/−)は、1.5:1、3:1および7:1で用いた。脂質成分は、DNAに対して重量で一定割合0.75:1に維持した。トランスフェクション複合体を作る前に、脂質成分を15μg/mlの濃度に希釈し、ペプチドは0.1mg/ml、DNAは20μg/mlで調製した。すべての希釈はOptiMEMレデュースド血清組織培養培地(Life Technologies)で行った。トランスフェクション複合体を、成分を、1)脂質、次に2)ペプチド、そして最後に3)DNAの順序で混合することにより作り、次にα25μgDNA/200μlのDNA成分に対応する濃度までOptiMEMで希釈し、その容積を各ウェルに加えた。各群は6レプリケートで行った。そのベクター複合体懸濁液を次に調製の5分以内に細胞に適用した。トランスフェクションインキュベーションは37℃で4時間行った。細胞を含まない抽出物中でのルシフェラーゼリポーター遺伝子検定を、Promegaのキットを用いて、製造者のプロトコールに従って48時間のインキュベーション後行った。光単位を各抽出以内のタンパク質濃度まで標準化した。トランスフェクション実験の結果を図3に示す。
実施例2
ヒト気道上皮細胞ライン(HAEo−)を、10%胎児子牛血清(FCS)、ペニシリンおよびストレプトマイシン、並びにグルタミン酸を含むEagleの最小必須培地(MEM)HEPES改変(Sigma,Poole)の中に保持した。マウス線維芽細胞ライン373およびヒト神経芽腫細胞ラインIMR32を、グルタマックス−1を有し、ピルビン酸ナトリウムを有せず、4500mg/Lグルコースを有し、ピリドキシン(Gibco BRL)、10%FCSを有し、ペニシリンおよびストレプトマイシンを添加したDulbecco's MEM中で増殖させた。Neuro−2A細胞をグルタマックス−1(Gibco BRL)、10%FCS、ピルピン酸ナトリウム、ペニシリンおよびストレプトマイシンおよび非必須アミノ酸をDulbecco's MEM中に保持した。
HAEo−細胞を24ウェルプレート中で集密まで増殖させた。細胞をTBS中で2度洗った後、細胞を4℃で30分間ウェル当り2mlの2%Marrel,5%BSA−TBSでブロックした。ブロッカーを除去し、2×1011のファージを1mlの2%Marvel、5%BSA−TBSに加えた。ファージを4℃で2時間振とうしながら結合させた後、4℃で振とうしながら5分間2%BSA−TBSで5回洗浄した後、2%BSA−TBSでわずか数秒間さらに5回洗浄した。400μlの76mMのクエン酸緩衝液pH2.5をウェルに加え、4℃で10分間振とうすることによってファージを溶出させた。溶出液を除去し、残った細胞を1mlの30mM Tris pH8.0、1mMのEDTAで1時間、氷上で溶菌した。細胞をプレートから捨て、溶出液をエッペンドルフに移し、短時間渦巻かせた。この溶出液を細胞関連画分として取っておいた。この溶出液からのファージを、プラーク形成単位(PFV)として、NEBによるライブラリーで提供される文献に記載のように滴定した後、文献に記載のようにE.coli ER2738細胞中でファージを増殖させた。パニングの第2ラウンドのために、前のラウンドからの増殖させたファージの2×1011をインプットファージとして用いた。しかしながら、単離された塑性でブロッキング分子結合性ファージの数を減少させるために、ファージを細胞のないブロックされたウェルに30分間、4℃で添加する4つの前選択を行った後、ファージをHAEo細胞に加えた。1mlの76mMクエン酸緩衝液pH3.5を用いて4℃で10分洗浄の添加による第2および第3ラウンドにおいて洗浄のストリンジェンシーも増加させた。第3ラウンドのために、第2ラウンドからの2×1011の増幅したファージを30分間細胞を含まない5のブロックされたウェル中で前選択した後、4℃で1時間1ウェルで行った。ファージ結合および溶出は第2ラウンドについて記載した通りである。第3ラウンドの溶出液の滴定後、単一ウェルの単離プラークを取り、増幅させ、配列決定および全細胞ELISAによるクローン結合キャラクタリゼーションのために精製した。
ファージを小スケールのPEG生産物から精製し(供給者の方法を参照)、一本鎖ファージDNAを、Brian K.Kay,Jill WinterおよびJohn McCaffertyにより編集されたPhage display of Peptides and Proteinsに記載の方法を用いて配列決定のために調製した。簡単にいうと、タンパク質のコートを、フェノール・クロロホルム抽出により試料から除去し、DNAをエタノール沈殿によりペレットにした。微量の塩を氷冷70%エタノールでペレットから洗浄した後、DNAをTE中に再懸濁した。
100mlのHBSS中の約8×104HAE細胞を96ウェルプレートの各ウェルに加え、細胞が接着するまで、37℃でインキュベートした。細胞をHBSS中でおだやかに洗浄した後、30分間HBSS中の0.5%BSAの添加によりブロックした。ブロックカー中の1×1010ファージ粒子を各ウェルに添加し、室温で40分結合させた。結合しなかったファージをHBSSで2度洗浄することにより除去し、結合ファージを3.7%のパラホルムアルデヒド中で10分間インキュベートすることにより細胞に固定した。細胞をPBS中で洗浄し、ブロッキング緩衝液中で45分インキュベートした後、PBSで3回洗浄した。結合したファージは、ブロッキング緩衝液中で1時間、1:5000に希釈したHRP結合抗M13抗体の添加により検出した後、PBS中で3回洗浄し、ABTS溶液でELISAを発色させ、405nmでの吸収を読んだ。
リポポリプレックス形成
次の成分を次の順序で加えることにより、試験管中で複合体を静電的に形成させた。50μlのリポフェクチン(Life Technologies Ltd)をOptiMEM中で30μg/mlの濃度まで希釈、70μlのペプチド(複合体中のペプチド:DNA電荷の最適化のためにOptiMEM中でさまざまな濃度で)、50μlのルシフェラーゼリポータープラスミドpCILucを最後に加えたOptimem中40μg/mlの濃度で加えた。複合体を短くピペッティングすることに混合した後、Optimem中で最終容積1.57mlまで希釈した。
96ウェルプレート中で一晩2×104細胞/ウェルでプレートした半集密HAEo細胞から培地を除去し、200μlの複合体(約0.25ngのプラスミドDNA)を各ウェルに加え、複合体を作り、細胞に加える最小の時間を残した。すべてのトランスフェクションはそれぞれ6ウェル中で行った。細胞を複合体と共に4時間インキュベートした後、48時間正常培地で置換え、その後リポーター遺伝子発現をルシフェラーゼ検定(Promega)により分析した。
細胞をPBSで2度濯いだ後、100μlのリポーター溶解緩衝液(Pregma、dH2O中5中1に希釈)を細胞に20分の間4℃で加えた後、凍結−解凍を行った。20μlの溶解物を白色プレートに移し、ルシフェラーゼを100μlの試薬の添加後、ルシルルミノメーターで測定した。
