Description
また、細胞内への粒子取込み方法としてリポソームを利用する試みがなされている。リポソームとは、生体膜の構成成分であるリン脂質により形成される、生体適合性に優れた小胞であり、その小胞内に様々な薬物を封入できることから、薬剤のキャリアーとして広く利用されている。さらに、リポソームの荷電、粒子径、脂質成分を変えたり、リポソーム表面に抗原、抗体、糖などの特異的リガンドを結合させたりすることで、細胞あるいは組織に対して特異性を持たせることが出来ることから、ターゲティング可能な薬剤キャリアーとして注目を集め、抗ガン剤をはじめとした副作用が強い化学療法剤の運び屋として臨床応用されている。

しかも、基本的にpHの変動や無機塩類の添加に対しても極めて安定である。さらに、表面上に存在するアミンによって表面電荷が正に帯電していることから、一般的に負の表面電荷をもつ細胞への親和性、取込み性が著しく高く、癌細胞の破壊を目的とした医療用材料として極めて有用である。これらの観点から、表面電位の最適範囲としては、良好な分散性と細胞取込み性を達成できる範囲にあればよく、+20mV以上あればよい。さらに望ましくは、一般に自主分散(粒子が沈殿しない状態)が十分に達成できる電位として+40mV以上あればよい。
二酸化チタン粒子へのポリエチレンイミンの導入(その1)
チタンテトライソプロポキシド3.6gとイソプロパノール3.6gを混合し、氷冷下で60mlの超純水に滴下して加水分解を行った。滴下後に室温で30分間攪拌した。攪拌後、12N硝酸1mlを滴下して80℃で8時間攪拌を行い、ペプチゼーションした。
ペプチゼーション終了後0.45μmのフィルターで濾過し、さらに脱塩カラムPD-10(アマシャム・ファルマシア・バイオサイエンス社製)を用いて溶液交換して固形成分1%の酸性二酸化チタンゾルを調製した。この分散液を100ml容のバイアル瓶に入れ、200Hzで30分間超音波処理を行った。超音波処理を行う前後の平均分散粒経はそれぞれ、36.4nm、20.2nmであった。超音波処理後、溶液を濃縮して固形成分20%の二酸化チタンゾルを調製した。得られた二酸化チタンゾル0.75mlを20mlのジメチルホルムアミド(DMF)に分散させ、ポリエチレンイミン(平均分子量:10000、和光純薬)450mgを溶解したDMF10mlを添加後、攪拌して混合した。
水熱反応容器(HU-50、三愛科学)に溶液を移し変え、150℃で6時間合成を行った。反応終了後、反応容器温度が50℃以下になるまで冷却し、反応液に対して2倍量のイソプロパノール(和光純薬)を添加した。室温で30分間静置後、遠心分離により沈殿を回収した。回収した沈殿を70%エタノールで洗浄後、2.5mlの水を加えてポリエチレンイミン結合二酸化チタン微粒子(アナターゼ型)の分散液を得た。作製したポリエチレンイミン結合二酸化チタン微粒子の分散粒径を、ゼータサイザーナノZS(シスメックス社製)を用いて、ゼータ電位測定セルにポリエチレンイミン結合二酸化チタン微粒子の分散液0.75mlを仕込み、溶媒の各種パラメーターを水と同値に設定し、25℃にて動的光散乱法により測定したところ、作製したポリエチレンイミン結合二酸化チタン微粒子の平均粒径は65.6nmであった。また、同様の条件でゼータサイザーナノZSを用いてゼータ電位を測定したところ、作製したポリエチレンイミン結合二酸化チタン微粒子のゼータ電位は+35.7mVであった。
二酸化チタン粒子へのポリエチレンイミンの導入(その2)
平均分子量7500のポリエチレンイミンを用いたこと以外、実施例1と全く同様の方法でポリエチレンイミン結合二酸化チタン微粒子を合成した。平均分子量7500のポリエチレンイミンを用いた場合でも、ポリエチレンイミン結合二酸化チタン微粒子(アナターゼ型)の分散液はいずれも良好な分散性を示し好適であった。
二酸化チタン粒子へのポリエチレンイミンの導入(その3)
酸性二酸化チタンゾルの代わりにアルカリ性二酸化チタンゾル(タイノックAL―6、多木化学社製)を用いたこと以外、実施例1と全く同様の方法でポリエチレンイミン結合二酸化チタン微粒子を合成した。アルカリ性二酸化チタンゾルを用いた場合でも、ポリエチレンイミン結合二酸化チタン微粒子(アナターゼ型)の分散液はいずれも良好な分散性を示し好適であった。
磁性材/酸化チタン複合微粒子へのポリエチレンイミンの導入
セパラブルフラスコ内にポリオキシエチレン(15)セチルエーテル(C-15:日本サーファクタント工業社製)を45.16gを溶解させて5分間窒素置換した後、シクロヘキセン(和光純薬社製)75mlを添加、0.67MのFeCl2(和光純薬社製)水溶液3.6mlを添加し、250rpmで攪拌しながら30%アンモニア水溶液5.4mlを添加し、1時間反応させた。その後、50mMテトラエチルオルソシリケイト水溶液(和光純薬社製)を0.4ml滴下して1時間反応させた。その後、チタンテトライソプロポキシド(和光純薬社製)を最終濃度5mMになるように加えた。50%(w/v)エタノール水溶液10mlを1mlずつ10分間隔で添加した。水溶液を遠心分離し、沈殿物を350℃で2時間焼成した。焼成後、10mM硝酸水溶液に分散させて超音波処理後、0.1μmのフィルターでろ過した。得られた磁性材/酸化チタン複合体ゾル0.75mlを20mlのジメチルホルムアミド(DMF)に分散させ、ポリエチレンイミン(平均分子量:10000、和光純薬社製)450mgを溶解したDMF10mlを添加後、攪拌して混合した。水熱反応容器(HU-50、三愛科学社製)に溶液を移し変え、150℃で6時間合成を行った。反応終了後、反応容器温度が50℃以下になるまで冷却し、反応液に対して2倍量のイソプロパノール(和光純薬社製)を添加した。室温で30分間静置後、遠心分離により沈殿を回収した。回収した沈殿を70%エタノールで洗浄後、2.5mlの水を加えてポリエチレンイミン結合磁性材/二酸化チタン複合微粒子(アナターゼ型)の分散液を得た。本分散液は白濁も生ぜず、微粒子が良好に分散しており単一の二酸化チタンの場合と同様に好適な分散液であった。
ポリエチレンイミン結合二酸化チタン微粒子のイソプロパノールに対する溶解性
ポリエチレンイミン200mgを10mlのDMFに溶解したものを、溶液(A)とした。実施例1の工程中で得られた固形成分20%の二酸化チタンゾル0.25mlを10mlのDMFに分散させたものを、溶液(B)とした。また、実施例1の工程中で得られた固形成分20%の二酸化チタンゾル0.25mlとポリエチレンイミン200mgを10mlのDMFに溶解したものを混合させたものを、溶液(C)とした。さらに、溶液(C)を150℃で6時間反応させたポリエチレンイミン結合二酸化チタン微粒子の分散液を、溶液(D)とした。(A)〜(D)の各溶液に2倍量のイソプロパノールを添加して撹拌後静置し、沈殿の生成を確認した。その結果、溶液(A)〜(C)はいずれもイソプロパノール中で沈殿のない透明な分散体であったが、溶液(D)のみが沈殿を生じたことが確認され、単にポリエチレンイミンと二酸化チタン微粒子を混合した状態に比べてポリエチレンイミン結合二酸化チタン微粒子は、ポリエチレンイミンと二酸化チタン微粒子の間により強い結合を有しているためにイソプロパノールによる影響を受けて沈殿を形成したことが示唆された。
ポリエチレンイミン結合二酸化チタン微粒子の中性溶液における安定性
実施例5で用いた(A)〜(D)と同組成の各溶液を用いて、中性溶液におけるそれぞれの安定性を評価した。すなわち、(A)〜(D)の各溶液を200mMリン酸緩衝液(pH7.0)で10倍に希釈して撹拌後静置し、沈殿生成の有無を観察した。その結果、二酸化チタンゾルを含む溶液(B)と(C)は沈殿を生じたが、溶液(A)と(D)では沈殿を生じなかった。これは二酸化チタンの等電点が中性付近にあるため、溶液(B)と(C)では二酸化チタンが凝集してしまい沈殿を生じたと考えられる。一方、溶液(D)では、二酸化チタン表面は無数のアミンにより修飾されているため、中性付近では粒子全体が正電荷を帯びており、均一に分散された状態を保持している。すなわち、溶液(C)と(D)の比較において実施例7と8の結果から、ポリエチレンイミン結合二酸化チタン微粒子は、ポリエチレンイミンの添加で単に分散性を高められた状態の二酸化チタン微粒子とは、全く異なる物性を示すことが分かった。
ポリエチレンイミン結合二酸化チタン微粒子分散液の二酸化チタン含量の測定
実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子の分散液を110℃で1時間加熱乾燥し、さらに4時間強熱して完全に灰化した。これをシリカゲルデシケータ中で冷却し、前記分散液中の正味の二酸化チタン量として質量を測定した。その結果、前記分散液は、0.25%(w/v)の二酸化チタンを含むことが示された。
ポリエチレンイミン結合二酸化チタン微粒子分散液のアミノ基含量の測定
実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子のアミノ基をフルオレスカミン(東京化成工業社製)との反応により確認と定量を行った。フルオレスカミンはアミノ基と反応し、蛍光物質を生成するので、ポリエチレンイミン結合二酸化チタン微粒子とフルオレスカミンとの反応での生成物の蛍光強度を測定することにより確認と定量を行うことができる。100mMホウ酸緩衝液(pH9.0)によって所定濃度に調製したグルコサミン溶液を作製して、励起波長395nmおよび蛍光波長480nmでの蛍光強度について検量線を作成し、この検量線を用いてポリエチレンイミン結合二酸化チタン微粒子上のアミノ基含量を求めた。その結果、前記分散液は4.01×10-2Mの濃度でアミノ基を含むことが示された。実施例7の結果から、前記分散液のアミノ基/二酸化チタン量比は、1.63×10-2(mol/g)であった。
ポリエチレンイミン結合二酸化チタン微粒子(アナターゼ型)の光触媒活性の評価
実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子(アナターゼ型)を固形成分が0.02%になる様に50mMリン酸緩衝液(pH7.0)で希釈した。メチレンブルー三水和物(和光純薬社製)を40μMになる様に水溶液に添加した。攪拌しながら、本水溶液に波長340nmの紫外光を1.5mW/cm2になるように照射し、580nmにおける波長の吸収を紫外-可視光分光光度計により測定した。結果を図2に示した。紫外線を照射しなかった混合液に比べ、紫外線照射を行った混合液は照射時間の経過と共にメチレンブルーの分解にともなう吸光度の減少が認められることから、実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子(アナターゼ型)が光触媒活性を保持していることは明らかである。
ポリエチレンイミン二酸化チタン微粒子のpH安定性の評価
50mMの異なるpHを持つ緩衝液(pH3=グリシン塩酸緩衝液、pH4および5=酢酸緩衝液、pH6=2-モルフォリノエタンスルホン酸緩衝液)、pH7および8=2-[4-(2-ヒドロキシエチル)-1-ピペラジニル]エタンスルホン酸緩衝液、pH9=ホウ酸緩衝液、pH10=グリシン水酸化ナトリウム緩衝液)を作成し、終濃度0.025(w/v)%になるように実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子を含む分散液を添加し、1時間室温にて静置した。その後、ゼータサイザーナノZSにて実施例1と同様に平均分散粒径の測定を行った。結果を図3に示す。pHが3から10の間で粒径の変化は認められるものの70から85nm程度であり、安定した分散性を示した。
ポリエチレンイミン二酸化チタン微粒子の塩強度安定性の評価
0.05〜5Mの異なる塩化ナトリウムを含む10mMリン酸緩衝液に実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子を終濃度0.025%になるように添加し、1時間室温にて静置した。その後、ゼータサイザーナノZSにて実施例1と同様に平均分散粒径の測定を行った。結果を図4に示す。
系中の塩濃度が0.05から1Mの間はほとんど平均分散粒径の変化は認められず、安定した分散性を示すことが明らかになった。
二酸化チタン複合微粒子の均一性(透明度)の評価
0.1Mの塩化ナトリウムを含む10mMリン酸緩衝液を用いて、実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子を含む分散液を終濃度0.1%になるように調整し、1時間室温にて静置した。また、二酸化チタン微粒子としてP25(日本アエロジル)を0.1Mの塩化ナトリウムを含む10mMリン酸緩衝液を用いて、同様に終濃度0.1%になるように調整し、1時間室温にて静置した。その後、シャーレに5ml移し上方から撮影し、確認した。その結果を図5に示す。P25水溶液に対してポリエチレンイミン結合二酸化チタン微粒子を含む分散液は明らかに透明度が高く、均一に分散していることが確認された。また、分光光度計(UV-1600、島津製作所)を用いて波長660nmにおける吸光度の測定を行った結果、P25水溶液は吸光度が1を大きく上回り測定不能であったのに対して、ポリエチレンイミン結合二酸化チタン微粒子を含む分散液は吸光度が0.044であり、また沈殿の形成は起きていなかった。更に、これらの溶液を室温暗所にて2週間静置した後に、同様に波長660nmにおける吸光度の測定を行った結果、P25水溶液は吸光度が1を大きく上回り測定不能であったのに対して、ポリエチレンイミン結合二酸化チタン微粒子を含む分散液は吸光度が0.051であった。このことから、水溶液中において二酸化チタン複合微粒子の分散液が透明度の高い、均一な分散性を示し、かつ安定していることが明らかになった。
ポリエチレンイミン二酸化チタン微粒子の細胞毒性の評価
実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子を含む分散液を、固形分が1.0%になるように10%血清を含むRPMI1640培地(GIBCO社製)で調整した。培養ガン細胞(Jurkat)を、10%血清を含むRPMI1640培地(GIBCO社製)で37℃、5%二酸化炭素雰囲気下で培養し、5.0×104 細胞数/mlとなるように調製した。これを再度20時間同条件で培養した。この細胞培養液に、上記ポリエチレンイミン結合二酸化チタン微粒子を含む分散液を終濃度で0.1%、0.01%、0.001%、0.0001%になるように96穴プレート上で調整し、200μlの試験用細胞培養液とした。この試験用細胞培養液を37℃、5%二酸化炭素雰囲気下で20時間培養した後、それぞれ100μlを用いてCelltiter-Glo Luminescent Cell Viability Assay(Promega社製)により生細胞由来の発光反応を行い、イメージアナライザLAS-3000UVmini(富士フィルム社製)を用いてその発光量測定を行うことで細胞毒性の評価を行った。その結果を図6に示す。何も添加していないコントロールの培養細胞における発光量に比べ、どの分散液濃度においても同等の発光量を確認したことから、この濃度域のポリエチレンイミン結合二酸化チタン微粒子を含む分散液は細胞毒性が認められないことが明らかになった。
ポリエチレンイミン二酸化チタン微粒子の細胞取込み性の評価
実施例1で得られた二酸化チタンゾル0.75mlを20mlのジメチルホルムアミド(DMF)に分散させ、ポリアクリル酸(平均分子量:5000、和光純薬社製)0.2gを溶解したDMFを10ml添加後、攪拌して混合した。水熱反応容器に溶液を移し変え、180℃で6時間水熱合成を行った。反応終了後、反応容器温度が50℃以下になるまで冷却し、溶液を取り出した後に水80mlを添加して攪拌混合した。エバポレータでDMFおよび水を除去した後に、再度、水20mlを添加してポリアクリル酸結合二酸化チタン微粒子の水溶液とした。2N塩酸1mlを添加して二酸化チタン粒子を沈殿させて、遠心後に上清を除去することにより未反応のポリアクリル酸を分離した。再度水を添加して洗浄を行い、遠心後に水を除去した。50mMリン酸緩衝液(pH7.0)を10ml添加後、200Hzで30分間超音波処理を行い、二酸化チタン粒子を分散させた。超音波処理後、0.45μmのフィルターで濾過し、重量百分率で0.25%のポリアクリル酸結合二酸化チタン微粒子の分散液を得た。ゼータサイザーナノZS(シスメックス社製)を用いて、ゼータ電位測定セルにポリエチレンイミン結合二酸化チタン微粒子の分散液0.75mlを仕込み、溶媒の各種パラメーターを水と同値に設定し、25℃にて動的光散乱法により測定したところ、作製したポリアクリル酸結合二酸化チタン微粒子の平均粒径は45.9nmであった。このポリアクリル酸結合二酸化チタン微粒子の分散液2mlに対して、0.8M 1-Ethyl-3-[3-Dimethylaminopropyl]Carbodiimide Hydrochlorideを250μlおよびN-Hydroxysuccinimideを250μl加えて、撹拌しながら室温で1時間反応させた。10mM 酢酸緩衝液(pH5.0)で平衡化した脱塩カラムNAP-10(アマシャム・ファルマシア・バイオサイエンス社製)を用いてゲル濾過を行って溶液交換し、その後に10mM酢酸緩衝液(pH5.0)を用いて全量を9.5mlとした。そこへ、DMFに溶解させた100mM 5-amino fluorescein(NCI社製)を5μl加え、遮光下で撹拌しながら室温で1時間反応させた。次に、0.1Mのエタノールアミン(和光純薬工業社製)水溶液を500μl加え、遮光下で撹拌しながら室温で30分間反応させた。この溶液を100mMリン酸緩衝食塩水(pH7.5)で平衡化した脱塩カラムPD-10を用いてゲル濾過を行って溶液交換し、未反応の5-amino fluoresceinを分離し、その後、溶液を2mlにまで濃縮した。これを蛍光色素標識ポリアクリル酸結合二酸化チタン微粒子の分散液とした。
また、実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子の分散液を500μlを、100mMリン酸緩衝食塩水(pH7.5)で平衡化した脱塩カラムNAP-10を用いてゲル濾過を行って溶液交換し、そこへ、最終濃度が0.8mMになるようにDMSOに溶解したフルオレセインイソチオシアネート(ピアース社製)を加え、30分間室温で穏やかに攪拌した。反応終了後、あらかじめPBSにて平衡化したPD-10(アマシャム・ファルマシア・バイオサイエンス社製)により溶液交換を行い、その後、溶液を2mlにまで濃縮した。これを蛍光標識ポリエチレンイミン結合二酸化チタン微粒子の分散液とした。
次に、メラノーマ細胞株T-24を10%血清を含むF12培地(ギブコ社製)で100%コンフルエントにになるまで培養し、フラスコを100mMリン酸緩衝食塩水(pH7.4)で2回洗浄し、100mMトリプシン-エチレンジアミン三酢酸溶液を1ml添加し、10分静置後、フラスコ壁面より剥離した細胞を回収し、9mlの10%血清を含むF12培地で希釈した。細胞数を血球計算盤により計測し、5×104個の細胞を含む培地500μlをそれぞれ24穴マイクロタイタープレートに接種し、最終濃度0.01%になるように分注した。そこに、先の蛍光色素標識ポリアクリル酸結合二酸化チタン微粒子の分散液および蛍光色素標識ポリエチレンイミン結合二酸化チタン微粒子の分散液をそれぞれ最終濃度0.01%となるよう100μl加え、24時間CO2インキュベータ内で培養した。その後、細胞のフラスコへの接着を確認し、フラスコを100mMリン酸緩衝食塩水にて洗浄し、200μlの10%血清を含むF12培地を添加し、蛍光顕微鏡により観察を行った(図7)。蛍光視野像を観察した結果、蛍光色素標識ポリアクリル酸結合二酸化チタン微粒子よりも蛍光色素標識ポリエチレンイミン結合二酸化チタン微粒子のほうが明らかに細胞に対して高い親和性と細胞取込み性をもつことが確認された。
ポリエチレンイミン二酸化チタン微粒子の細胞殺傷性の評価
メラノーマ細胞株T-24を10%血清を含むF12培地(ギブコ社製)で100%コンフルエントにになるまで培養し、フラスコを100mMリン酸緩衝食塩水(pH7.4)で2回洗浄し、100mMトリプシン-エチレンジアミン三酢酸溶液を1ml添加し、10分静置後、フラスコ壁面より剥離した細胞を回収し、9mlの10%血清を含むF12培地で希釈した。細胞数を血球計算盤により計測し、5×104個の細胞を含む培地500μlをそれぞれ24穴マイクロタイタープレートに接種した。そこに、実施例1で得られたポリエチレンイミン結合二酸化チタン微粒子の分散液を100mMリン酸緩衝食塩水(pH7.4)で調整し、最終濃度0%および0.01%となるよう100μl加え、ブラックライト(東芝社製)により波長340nmの紫外光を2.5mW/cm2で0分間および60分間照射し、24時間CO2インキュベータ内で培養した。Cell counting kit-8(同人化学社製)を試薬のマニュアルに従い調整して加え、96穴プレート上にて吸光度計Benchmark(Bio-Rad社製)を用い、波長450nmの吸光度測定を行った。その結果を表1に示す。

- 光触媒性二酸化チタン微粒子と、該光触媒性二酸化チタン微粒子の表面に修飾される親水性のカチオンポリマーとを含んでなり、前記親水性カチオンポリマーと光触媒性二酸化チタンが結合されてなる、殺細胞剤。
- 前記光触媒性二酸化チタンが、アナターゼ型、またはルチル型である、請求項1に記載の殺細胞剤。
- 前記光触媒性二酸化チタンの粒径が、2〜200nmである、請求項1または2に記載の殺細胞剤。

- 前記カチオンポリマーが、水溶性高分子である、請求項1〜4のいずれか一項に記載の殺細胞剤。
- 前記水溶性高分子がアミンである、請求項5に記載の殺細胞剤。
- 前記水溶性高分子が、ポリエチレンイミンを含む、請求項5に記載の殺細胞剤。
- 前記水溶性高分子が、分子中に複数のアミン単位を有する共重合体を含む、請求項5に記載の殺細胞剤。
- 前記光触媒性二酸化チタン微粒子の表面電位が+20mV以上である、請求項1〜8のいずれか一項に記載の殺細胞剤。
- 水系溶媒に分散していることを特徴とする、請求項1〜9のいずれか一項に記載の殺細胞剤。
- 前記水系溶媒のpHが3〜9である、請求項10に記載の殺細胞剤。
- 前記水系溶媒がpH緩衝液である、請求項10に記載の殺細胞剤。
- 前記水系溶媒の塩濃度が1M以下である、請求項10〜12のいずれか一項に記載の殺細胞剤。
- 前記水系溶媒が、生理食塩水である、請求項13に記載の殺細胞剤。
- 前記光触媒性二酸化チタン微粒子の分散液が、前記光触媒性二酸化チタン微粒子を重量百分率で0.0001〜0.1%含んでなる、請求項10〜14のいずれか一項に記載の殺細胞剤。
- 生体の皮膚表面、生体内の表層部に存在する患部に適用する、請求項1〜15のいずれか一項に記載の殺細胞剤。
- 前記細胞が癌細胞である、請求項1〜16のいずれか一項に記載の殺細胞剤。
