Description
本発明は、ヒトに投与後に生物薬剤を免疫原性にさせ得るペプチドを単離および同定するための方法を提供する。本方法は、0.1〜5μgの量、好ましくは0.2〜3μgの量の潜在的免疫原性ペプチドとペプチド受容体の複合体を提供する、この量は、患者または健康なドナーの末梢血から得られるDC細胞から通常入手可能である物質の量に等しい。従来技術で必要な物質の最低量は、無制限の供与源(近交系マウス)に由来する約200μgのMHCクラスII分子である(Dongre A R et al., EJI 2001, 31, 1485〜94)。これは、ヒト末梢血から入手可能なものよりも約2桁多い物質である。

本発明(2)は、APRを発現する細胞がMHC IIを発現する細胞である、本発明(1)の方法である。
本発明(3)は、MHC IIを発現する細胞が樹状細胞である、本発明(2)の方法である。
本発明(4)は、樹状細胞が、樹状細胞に成熟するために誘導されるのと同時に、未成熟樹状細として胞免疫原の潜在的供与源に曝露される、本発明(3)の方法である。
本発明(5)は、潜在的免疫原の供与源が、サイトカイン、ケモカイン、成長因子、抗体、酵素、構造タンパク質、ホルモン、またはこれらの断片として治療的ポリペプチドを含むポリペプチドを含む群に属する、本発明(1)〜(4)のいずれか一発明の方法である。

本発明(11)は、単離された免疫原性ペプチドが、潜在的免疫原の供与源と接触させた細胞から同定されたペプチドを、その供与源と接触させなかった細胞から同定されたペプチドと比較することによって同定される、本発明(1)〜(10)のいずれか一発明の方法である。
本発明(12)は、免疫原性ペプチドは、天然にプロセスされた免疫原性ペプチドである、本発明(1)〜(11)のいずれか一発明の方法である。
本発明(13)は、以下の工程を含むポリペプチドの免疫原性を減少させるための方法である:
a)本発明(1)〜(12)のいずれか一発明の方法に従ってポリペプチドの免疫原性ペプチドを同定する工程、
b)抗原提示受容体の結合が、減少または消滅するように、ポリペプチドの対応するエピトープを修飾する工程、
c)これにより、免疫原性潜在能力が減少されたか、または免疫原性潜在能力がない修飾されたポリペプチドを作製する工程。
本発明の抗原ペプチドは、ヒトDGの表面上のMHCクラスII分子と会合するペプチドである。抗原ペプチドは、細胞内または細胞外MHCクラスII分子に結合していてもよい。本明細書に使用される「免疫原性ペプチド」という用語は、免疫応答を誘発し得る抗原性ペプチドをいう。免疫原性ペプチドは、DCと同時インキュベーション後にポリペプチドから誘導されてもよい。免疫原性ペプチドの潜在的供与源であるポリペプチドは、サイトカイン(すなわち、インターフェロン、インターロイキン、エリスロポエチン(Epo)、顆粒球/マクロファージコロニー刺激因子(GM-CSF)、または腫瘍壊死因子(TNF))、ケモカイン、成長因子、抗体(すなわち、モノクローナル、ポリクローナル、キメラ、およびヒト化抗体)、酵素、構造エレメント、ホルモン、およびこれらの断片などの治療的ポリペプチドを含むポリペプチドである。
本明細書に使用される「抗原提示受容体」または「APR」という用語は、抗原ペプチドに結合し、かつこれらをその他の免疫学的細胞に提示することにより、特異的な体液性免疫応答を媒介するペプチド受容体をいう。好ましい抗原提示受容体は、MHCクラスII分子である。MHCクラスII分子は、HLA-DR、HLA-DQ、およびHLA-DP分子を含むが、これらに限定されるわけではない。役割を果たし得る代替のAPRは、CD 1ファミリーの受容体またはCD4+ヘルパーT細胞に潜在的免疫原性ペプチドを提示する今までに未決定の他の受容体である。
本発明の方法は、抗原提示受容体を発現し、同時にCD4+ T細胞を初回刺激するか、または活性化することができる全ての細胞を包含する。これらの細胞はまた、抗原提示細胞(APC)と称される(Unanue, E. R.. Macrophages, antigen presenting cells and the phenomena of antigen handling and presentation. In: Fundamental Immunology, 第2版 (編集者 Paul, W. B) New York, Raven Press, 1989)。使用されるAPCは、ヒトB細胞、ヒトマクロファージ、好ましくはヒト樹状細胞を含む。これとは別に、末梢血単核細胞(PBMC)または末梢血リンパ球(PBL)などのAPCを含む細胞混合物を使用してもよい。
細胞からの抗原提示受容体-ペプチド複合体の精製のためには、細胞の膜を可溶化しなければならない。細胞溶解は、当技術分野において公知の方法、たとえば凍結解凍サイクルおよび界面活性剤の使用、ならびにこれらの組み合わせで実施してもよい。好ましい溶解方法は、界面活性剤、好ましくはTX-100、NP40、n-オクチルグルコシド、Zwittergent、ルブロール、CHAPS、最も好ましくはTX-100またはZwittergent3-12を使用する可溶化である。細胞細片および核は、遠心分離によって、可溶化された受容体-ペプチド複合体を含む細胞可溶化物から除去しなければならない。従って、本発明のさらなる態様において、免疫原性ペプチドと抗原提示受容体の複合体は、界面活性剤での可溶化を含む方法で細胞から単離される。
さらにまた、本発明は、免疫沈降または免疫アフィニティークロマトグラフィーを含む方法による、細胞可溶化物からのMHC-ペプチド複合体の精製を提供する。免疫沈降または免疫アフィニティークロマトグラフィーのために、MHCクラスII分子に特異的で、かつこれらの方法に適した抗体が使用される。特異的抗体は、好ましくはモノクローナル抗体であり、共有結合性に、または非共有結合的に、たとえばプロテインAを経て、ビーズ、たとえばセファロースまたはアガロースビーズに結合される。従来技術に使用される抗HLA抗体の広範なパネルの選択は、以下を含む:
抗HLA-DR抗体:L243、TU36、DA6.147、好ましくはL243;抗HLA-DQ抗体:SPVL3、TU22、TU169、好ましくはTU22、およびTU169;抗HLA-DP抗体B7/21。
受容体分子からペプチドを溶出することにより、潜在的免疫原の供与源に、および細胞内または細胞外起源のポリペプチドに由来する天然にプロセスされたペプチドの複合混合物が得られる。溶出後のみ、ペプチドを分画して、配列解析に供することができる。
本発明のさらなる態様において、単離された免疫原性ペプチドが分画され、配列決定され、および同定される。シーケンシングにより、単離された免疫原性ペプチドの混合物中の個々のペプチドのアミノ酸配列が、ペプチドのフェムトモル量を配列決定するのに十分な方法によって解明されることが理解される。同定により、これが、免疫原性ペプチドが由来するタンパク質またはポリペプチド、およびこれらのタンパク質またはポリペプチド内で構成する配列から確立されることが理解される。
溶出によって得られた全ペプチドレパートリーの定性分析のために、マトリックス支援レーザー脱離およびイオン化飛行時間型(MALDI-TOF)質量分析を行ってもよい。ペプチドを断片にしない設定を使用して、MALDI-TOF解析により、ペプチド混合物の複雑さおよび優性ペプチドの存在に関して大ざっぱな概要を提供する。
抗原提示受容体から溶出される単一ペプチドの量を推定するために、マイクロキャピラリーカラムを通る流れを、214nmの検出波長に操作されたフロースルーUV検出器によって解析してもよい。定量化のために、解析されるペプチドのピーク面積を段階量の合成標準ペプチドのピーク面積と比較する。
本発明のストラテジーでは、細胞培養におけるAPCの抗原提示受容体上にロードされた免疫原性ペプチドの同定を予見する(インビトロアプローチ、図1)。
a)0.1〜5μgの受容体を提供する数の、好ましくは0.2〜3μgを提供する数の抗原提示受容体(APR)を発現する細胞を提供する工程、
b)(a)からの細胞を免疫原性ペプチドの供与源と接触させる工程、
c)細胞からAPR-免疫原性ペプチド複合体を単離する工程、
d)APRから会合したペプチドを溶出する工程、
e)免疫原性ペプチドを同定する工程、
f)エピトープとして同定された免疫原性ペプチドを検証する工程。
本発明の方法によって同定されたペプチド配列は、MHC結合モチーフ、MHC結合能、およびCD4+ Tリンパ球による認識を含むいくつかの基準のうちの1つによって検証してもよい。
本発明の方法は、任意の生物薬剤の免疫原性に関与するペプチド、特に容認できない効力喪失が抗薬物抗体を中和することによるものを同定するために、または臨床試験における有害もしくは重篤な有害事象が免疫原性に依存すると考えられる場合に、適用することができる。
細胞株および培養
本研究は、後述するように、単球から分化したヒト樹状細胞で行った。単球は、ヒト末梢血から精製した。全ての細胞は、1mM Pyruvat、2mM グルタミン、および10% 熱不活性化ウシ胎児血清(Gibco BRL, Rockville, Md.)を補ったRPMI 1640培地(短く:RPMI)中で培養した。
末梢血は、健康なドナーからの標準的バフィーコート製剤として、地域の血液バンクから得た。ヘパリン(200I.U./ml血液、Liquemine, Roche)を使用して凝固を防止した。末梢血単核細胞(PBMC)は、LSM(登録商標)(1.077-1.080g/ml;ICN, Aurora, Ohio)で800g(室温)にて30分間遠心分離することによって単離した。PBMCを間期から収集し、20mM Hepesを含むRPMI中で2回洗浄した(500gを15分間、300gを5分間)。赤血球を除去するために、PBMCをALT緩衝液(140mM塩化アンモニウム、20mM Tris、pH 7.2)で37℃にて3分間処理した。PBMCを20mM Hepesを含むRPMIで2回洗浄した(200gを5分間)。
単球の単離および樹状細胞の分化のために使用したPBMCのHLA-DR遺伝子型は、Roche Molecular Systems(Alameda, CA, USA)によって決定した。
単球は、抗CD 14磁気ビーズ(Miltenyi Biotech, Auburn, Calif.)を使用して製造業者プロトコールに従ってポジティブソーティングによってPBMCから単離した。単球を1%の非必須アミノ酸(Gibco, BRL, Rockville, Md.)、50ng/mlの組換えヒト顆粒球マクロファージ-コロニー刺激因子(GM-CSF;S.A. 1.1×107U/mg)(Leucomax; Novartis, Basel Switzerland)、および3ng/mlの組換えヒトIL-4(S.A. 2.9×104U/mg)(R&D Systems, Minneapolis, Minn.)を補ったRPMI中で培養した。単球を6-ウェルプレート(Costar)に0.3×106/mlで5日間播種し、未成熟樹状細胞を得た。
樹状細胞による薬学的タンパク質の取り込みを容易にするために、6×106個の未成熟樹状細胞を5〜50μgの生物薬剤に曝露した。同時に、10μg/mlの組換えヒト腫瘍壊死因子(TNFα;S.A. 1.1×105U/mg)を添加することによって、樹状細胞の成熟を誘導した。対照として、6×106個の樹状細胞をTNFα単独と共にインキュベートした(図1)。
抗HLA-DRモノクローナル抗体(mAb)L243(ATCC, Manassas, Va.)は、それぞれのマウスハイブリドーマ細胞株を培養することによって作製した。mAb L243は、製造業者のプロトコールに従って、プロテインAセファロース(Pharmacia, Uppsala, Sweden)を使用し、精製されかつ2.5mg/mlの終濃度でCNBr活性化セファロースビーズ(Pharmacia)に固定された。L243ビーズは、0.1%のZwittergent 3-12(Calbiochem, La Jolla, Calif.)を含むPBS溶液に貯蔵した。
凍結樹状細胞のペレットを、10倍体積の氷冷溶解緩衝液(プロテアーゼ阻害剤キルノスタチン(chyrnostatin)、ペプスタチン、PMSFおよびロイペプチン(Roche, Mannheim, Germany)を含む1% トリトン-X-100、20mM Tris、pH 7.8、5mM MgCl2)に再懸濁して、水平シェーカー内で1000rpm、4℃で1時間溶解した。細胞可溶化液を2000g、4℃で10分間の遠心分離によって細胞片および核から除いた。可溶化液をL243ビーズ(100μl細胞可溶化液につき5〜10μl L243ビーズ)と共に水平シェーカー内で1000rpm、4℃で2時間、共インキュベーションした。L243ビーズに結合した免疫沈降したHLA-DR-ペプチド複合体を2000g、4℃で5分間遠心分離することによって沈殿させ、PBS溶液内で300μlの0.1%Zwittergent 3-12(Calbiocbem)で3回洗浄した。
L243ビーズに結合したHLA-DR-ペプチド複合体を400μl H2O(HPLC等級;Merck, Darmstadt, Germany)に再懸濁し、限外濾過チューブ、Ultrafree MC、30kDカットオフ(Millipore, Bedford, Mass.)に移して、14000rpmで4℃において2〜4分間遠心分離することによって400μl H2O(HPLC等級)で10回洗浄した。結合したペプチドを溶出するために、50μlの 0.1% トリフルオロ酢酸(Fluka, Buchs, Switzerland)H2O(HPLC等級)溶液を添加して、インキュベーションを37℃で30分間行った。溶出されたペプチドを14000rpmにて3分間RTで限外濾過チューブを遠心分離することによって新たなエッペンドルフチューブに収集し、Speed-Vac(登録商標)真空遠心で直ちに凍結乾燥した。
HLA-DR分子から溶出された凍結乾燥されたペプチドを0.05%のトリフルオロ酢酸、5% アセトニトリル(Merck, Darmstadt, Germany)のH2O溶液(HPLC-等級)に溶解し、FAMOS(登録商標)オートサンプラーおよびULTIMATE(登録商標)ナノフローHPLC(Dionex, Olten, Switzerland)に接続した75μm×15 cm C18 PepMapキャピラリー(C18;3μm;100Å)(LC-Packings, Amsterdam, Netherlands)上で分離した。以下の200nl/分の一定流速での非直線勾配を使用した:0〜40分5〜50%系B;40〜50分50-90%系B。系Aは、0.05% トリフルオロ酢酸、5% アセトニトリル/H2Oであり、および系Bは、0.04% トリフルオロ酢酸、80% アセトニトリル/H2Oであった。分離は、214nmおよび280nmの二重UV吸収を経てモニターした。画分(400nl)は、フラクションコレクターPROBOTT(BAI, Weiterstadt, Germany)を使用して収集し、AnchorChip 600/384 MALDI-MS target(Bruker, Bremen, Germany)上にスポットした。
複合体ペプチド混合物の高スループットシーケンシングを行うために、液体クロマトグラフィーでの分画、続く質量分析によるシーケンシングに基づいたMudPIT(多次元的タンパク質同定技術)を使用した(Washburn M P et al., Nat Biotechnol 19(2001), 242-247)。
潜在的T細胞エピトープの予測は、TEPITOPEアルゴリズムを使用することによって達成した。以下の検索基準を適用した:閾値(最高のスコアリングについて1〜3%および天然リガンドの中程度スコアリングについて4〜6%)、ペプチド長(15アミノ酸残基)、および乱交雑(9対立遺伝子のうちの少なくとも6つと結合することが予測される)。乱交雑の程度を決定するために、以下の9対立遺伝子を白人集団におけるこれらの頻繁な出現と一致するように選択した:HLA-DRB1*0101、*0301、*0401、*0701、*0801、*1101、*1305、*1501、およびDRB5*0101。膜貫通ドメインおよびシグナルペプチドは、エピトープ検索に含めなかった。
新鮮なPBMCからのCD4+ T細胞の調製は、Miltenyi Biotech(Auburn, CA, USA)からのCD4+ T細胞単離キットを使用してネガティブ選択によって行った。T細胞群は、トリパンブルー染色(Sigma-Aldrich)で判断されたように、>75%純粋で、かつ>95%生存可能であった。T細胞を2×106細胞/mlでAIM V培地(Gibco BRL, Rockville, MD)に再懸濁した。樹状細胞(DC)を記載したようにPBMCから分化して、完全マクロファージSFM培地(Gibco BRL, Rockville, MD)中で培養した。4日目に、未成熟DCを10μg/ml LPS(Sigma-Aldrich)で刺激した。6日目に、成熟DCを洗浄し、AIM V培地に2×105細胞/mlで再懸濁した。共培養のために、0.1mlのCD4+ T細胞(2×105)および0.1mlの自己DC(2×104)(両方ともAIM V培地中に)を丸底96ウェル形式プレート内で混合した。OKT3 mAbおよびInflexal V(登録商標)を、それぞれ20μg/mlおよび1μg/mlの終濃度に添加した。合成ペプチドを20μMの終濃度に添加した。それぞれの抗原を3回試験した。共培養の5日目に、10μMの5-ブロモ-2’-デオキシウリジン(BrdU)(Roche, Basel, Switzerland)をそれぞれのウェルに添加した。24時間インキュベーション後、培養液を収集して、製造業者のプロトコールに従ってプロセスした。抗原を添加していない培養液のT細胞増殖を参照として使用し、平均刺激指数(SI)を1にセットした。
OKT3は、最初の治療的抗体であった。これは、1986年にFDAの承認を得た。これは、CD3特異的マウスIgG2a抗体であり、移植(L. Chatenaud, 2003)、1型糖尿病(E. Masteller & J. Bluestone, 2002)および乾癬(T. Udset et al., 2002)において免疫抑制剤として、臨床において広く使用されている。OKT3で誘導される十分な免疫抑制にもかかわらず、異種タンパク質に対する抗OKT3反応の発生は、OKT3の迅速なクリアランスおよび中和を促進する初期の臨床試験において主な欠点のうちの1つであった(G. Goldstein, 1987)。免疫原性の発生率は、OKT3処理した個体に関する研究において、およそ85%であることが報告された(C. Pendley et al., 2003)。
図1に概説したストラテジーを、HLA-DR遺伝子型HLA-DRB1*0701/1601を示す樹状細胞によって提示されたOKT3のペプチドエピトープを同定するために使用した。
図1に概説したストラテジーを、HLA-DR遺伝子型HLA-DRB1*1101/1202によって制限されたT細胞によって認識されるように、OKT3のペプチドエピトープを同定するために使用した。
図1に概説したストラテジーを、HLA-DR遺伝子型HLA-DRB1*0301/0401を示す樹状細胞によって提示されるOKT3のペプチドエピトープを同定するために使用した。
インターフェロンβ(IFN-β)は、現在多発性硬化症の治療のための第一線の治療法である(Deisenhammer et al., 2000)。3つの異なるIFN-β製剤:Avonex、Rebif(両方ともIFN-β-1a)およびBetaseron(IFN-β-1b)が現在市販されている。これらのBetaseronは、最初に市販されたものであり、促進された承認規則下で1993年にFDAに承認された。AvonexおよびRebifとは対照的に、Betaseronは、例外的に免疫原となることが公知である。Betaseronでの治療後、28〜47%程度の患者が、抗IFN-βの中和抗体を産生するが、一方、Avonexで治療した患者の2〜6%だけが、中和抗薬物抗体を示す(Deisenhammer et al., 2000; Bertolotto et al., 2004)。この状況において、AvonexおよびRebifは、チャイニーズハムスター卵巣細胞において天然のアミノ酸配列を持つグリコシル化されたタンパク質として発現されているが、Betaseronは、大腸菌(E.coli)においてMet-1欠失およびCys17からSerへの点突然変異をもつ非グリコシル化形態で発現されることに言及することが重要である(Mark et al., 1984; Holliday and Benfield, 1997)。これまで、これらの相違が様々な免疫原性の原因となるかどうかは不明である。
図1に概説したストラテジーを、HLA-DR遺伝子型HLA-DRB1*1101/1404を示す樹状細胞によって提示されるIFN-β-1bのペプチドエピトープを同定するために使用した。

Claims (11)
- 以下の工程を含む治療的ポリペプチドの免疫原性を減少させるための方法:
a) 治療的ポリペプチドの免疫原性ペプチドを同定する工程であって、
i) 0.1〜5μgの分子を提供する数のMHC II分子を発現する細胞を提供すること、
ii) i)からの細胞を免疫原性ペプチドの供与源と接触させること、
iii) 細胞からのMHC II分子-免疫原性ペプチド複合体を単離すること、
iv) MHC II分子から会合したペプチドを溶出すること、
v) 免疫原性ペプチドを同定すること、および
vi) 同定された免疫原性ペプチドをエピトープとして検証すること
を含む工程、
b) MHC II分子の結合が、減少または消失するように、ポリペプチドの対応するエピトープを修飾する工程、並びに
c) これにより、免疫原性潜在能力が減少されたか、または免疫原性潜在能力がない修飾されたポリペプチドを作製する工程。 - MHC IIを発現する細胞が樹状細胞である、請求項1記載の方法。
- 樹状細胞が、樹状細胞に成熟するために誘導されるのと同時に、未成熟樹状細胞として免疫原の潜在的供与源に曝露される、請求項2記載の方法。
- 治療的ポリペプチドが、サイトカイン、ケモカイン、成長因子、抗体、酵素、構造タンパク質、ホルモン、およびこれらの断片から選択される、請求項1〜3のいずれか一項記載の方法。
- 免疫原性ペプチドとMHC分子の複合体が、界面活性剤による細胞の可溶化、および免疫沈降または免疫アフィニティークロマトグラフィーによるMHC II分子と免疫原性ペプチドの複合体の隔離を含む方法で細胞から単離される、請求項1〜4のいずれか一項記載の方法。
- 隔離された免疫原性ペプチドをもつMHC分子の複合体が、ペプチドを溶出する前に限外濾過チューブ内において水で洗浄される、請求項1〜5のいずれか一項記載の方法。
- 免疫原性ペプチドが、希釈された酸を使用してMHC分子から溶出される、請求項1〜6のいずれか一項記載の方法。
- 工程(a)(iv)の単離された免疫原性ペプチドが、分画および配列決定される、請求項1〜7のいずれか一項記載の方法。
- 単離された免疫原性ペプチドが、液体クロマトグラフィーおよび質量分析を含む方法によって分画、ならびに配列決定される、請求項8記載の方法。
- 単離された免疫原性ペプチドが、潜在的免疫原の供与源と接触させた細胞から同定されたペプチドを、その供与源と接触させなかった細胞から同定されたペプチドと比較することによって同定される、請求項1〜9のいずれか一項記載の方法。
- 免疫原性ペプチドは、天然にプロセスされた免疫原性ペプチドである、請求項1〜10のいずれか一項記載の方法。
