【発明の属する技術分野】
本発明は、質量分析やイオン選別、イオン開裂等に用いられるイオントラップ装置に関する。

【従来の技術】
イオントラップ装置の一つの利用例として、イオントラップTOF質量分析装置を説明する。図1に示すように、イオントラップTOF質量分析装置10では、TOF(Time Of Flight;飛行時間)型質量分析装置11のイオン導入口にイオントラップ装置12が設けられ、そこにおいてイオンのアイソレーション(選別)や開裂等が行われる。イオントラップ装置12自体も質量分離の機能を有するが、イオントラップTOF質量分析装置10ではイオントラップ装置12は単にイオンをアイソレーション及び開裂するためだけに用いられ、イオンの質量分析は次段のTOF11で行われる。これにより、イオントラップ装置12を設けない場合と比較して、TOF11ではより高い質量分解能で質量分析ができるようになるとともに、分子構造についてより多くの情報が得られるようになる。

イオントラップ装置12では、図2に示すように、入口及び出口の両エンドキャップ電極21,22とその間に位置するリング電極23とで囲まれるイオントラップ空間24中に1×10-3Torr(0.1Pa)程度のヘリウム(He)ガスを定常的に導入する。これは一つには、イオンとヘリウムガスの衝突によってイオンをイオントラップ空間24の中心近傍に集めることにより、感度及び質量分解能を向上させる作用を行う。これをクーリング作用と呼ぶ。また、イオントラップ装置12の両エンドキャップ電極21,22にイオンの共振周波数と同一の周波数を含む波形を印加した場合には、これによりイオンの運動エネルギーが高まり、その状態でイオンがヘリウム分子と衝突することにより、イオンの開裂が行われるという作用も有する。これをCID(Collision Induced Dissociation;衝突誘起開裂)作用と呼ぶ。

イオントラップ装置を用いた質量分析装置では、多段のMS/MS分析が可能であるという特徴を有する。すなわち、特定のイオンのみをイオントラップ空間内に残し、これをCID作用で開裂し、それにより生じたイオン(プロダクトイオン)を更にCID作用により開裂する。このような開裂を多段で繰り返すことにより、分光構造について多くの情報が得られるようになる。

【発明が解決しようとする課題】
上記の通り、CIDは、通常はイオントラップ装置の両エンドキャップ電極21,22に、開裂したい目的のイオンの共振周波数と同一の周波数成分を有する高周波信号を印加して目的イオンを共振させることで運動エネルギを高め、イオントラップ空間内に存在するヘリウムガス等の不活性ガス(以下、これをCIDガスと呼ぶ)との衝突により目的イオンを開裂することにより行われる。

ここで、両エンドキャップ電極21,22に印加する電圧(CID電圧)を高くすることにより開裂が促進されるが、その値を過度に高くすると、親イオンは開裂する前に電極21,22,23の壁に衝突したり、中央の穴から外に排出されたりして消失してしまう。

これを詳しく説明すると次の通りである。イオントラップ空間24内では、次式で表されるような閉じこめポテンシャルφが定義される。

ここで、qはイオントラップ装置12により定まるパラメータであり、リング電極23のRF電圧V、イオンの質量m、リング電極23の内接半径r0、リング電圧の周波数Ω、素電荷eより次式で定義される。

CID電圧を上げすぎると、親イオンの運動エネルギがこの閉じこめポテンシャルφを超えてしまうために、イオントラップ空間24内にトラップされ得なくなる。また、できるだけ低質量数のプロダクトイオンまで観測できるようにするためには、CID電圧のq値をできるだけ下げることが必要であるが、そうすると閉じこめポテンシャルφも低くなり、親イオンが消失しやすくなってしまう。

親イオンが開裂される前に消失してしまうと、プロダクトイオンの強度が弱くなる。特に多段のMS/MS分析では、段数が進むほどイオンの数が少なくなってゆくため、段数毎に感度が悪化する。

本発明はこのような課題を解決するために成されたものであり、その目的とするところは、親イオンの消失をできるだけ抑えながら、CID電圧を高くしてCIDの効率を上げることのできるイオントラップ装置を提供することにある。

【課題を解決するための手段】
上記課題を解決するために成された本発明に係るイオン開裂方法は、リング電極と、それを挟んで対向する一対のエンドキャップ電極を備え、それらの電極で囲まれたイオントラップ空間にガスを導入しつつそれらの電極に適宜の電圧を印加することによりイオンを開裂するイオントラップ装置において、該ガスをパルス状に繰り返し導入すると共に、各ガスパルスの導入直後にCID電圧を印加することを特徴とする。

【発明の実施の形態及び効果】
本発明に係るイオントラップ装置では、ヘリウムやアルゴン等のCIDガスをパルス状にイオントラップ空間に導入する。このパルスの幅(ガスの導入時間)は、イオントラップ装置のサイズ(イオントラップ空間の容積)にも依存するが、リング電極の内径が10mm程度の通常のイオントラップ装置の場合、数百μsec程度の非常に短いものとし、繰り返し速度は数十Hz以下とする。なお、CIDガスをパルス状に導入するためには、パルスガスバルブ25を使用することができる(図2)。

イオントラップ装置を排気するポンプの排気能力にも依存するが、イオントラップ装置にCIDガスをこのようなパルス状で導入すると、導入直後の数十msec間はCIDガスはかなり高濃度(10-2Torr台)にイオントラップ空間内に存在する。CID電圧は、CIDガスを導入してから数msec後程度で両エンドキャップ電極21,22に印加する。CIDガスパルスの導入とCID電圧印加のタイミングを図3に示す。

CIDガスをパルス状に導入した場合、その直後に多少高いCID電圧を印加しても、高濃度に存在するCIDガスとの衝突の影響により、親イオンはすぐにクーリングされ、イオントラップ内から消失する確率は大幅に低減される。また、低いq値においても有効に機能するので、このような条件でも開裂効率が高い。

また従来、最適CID電圧は開裂目的イオンの質量数に依存する傾向があった。すなわち、低い質量数のイオンに対しては低いCID電圧、高い質量数のイオンに対しては高いCID電圧が必要である。しかし、イオン種によっては例外があることもあった。このため、CIDの目的とするイオンに対してその都度CID電圧を変える必要性があり、また、そのように設定をしてもうまくCIDが行われない場合があった。それに対し、本発明では、そのような質量数依存性が殆どなくなり、低い質量数の目的イオンに対して高いCID電圧を使用しても親イオンの損失が少ないという特長を有する。

図4に、従来のイオントラップ装置と本発明に係るイオントラップ装置における親イオン及びプロダクトイオンのイオン強度の変化のグラフを示す。CIDガスを連続的に導入する従来の装置では(a)、CID電圧を上げるに従い、プロダクトイオンの強度は、ピーク以降徐々に低下してゆく。これは、CID電圧を上げることにより、プロダクトイオンが生成される前に親イオンが失われてしまうためである。
それに対し、CIDガスをパルス状に導入する本発明の装置では(b)、CID電圧を上げてもプロダクトイオンの強度はあまり低下しない。
