本発明は、毛髪損傷の予防または修復に有用な毛髪処理剤に関する。

【従来の技術】
毛髪がパーマ処理等により損傷を受けキューティクルの捲れや枚数の減少が起こることが知られている。この際、毛髪から蛋白質が溶出されるという報告はあるが、その由来については不明であった。

また、損傷毛髪の修繕剤として従来シリコンなどの毛髪コート剤や毛髪ケラチン、その加水分解物またはその誘導体を補う試みが行われてきている。しかし、毛髪ケラチンは不溶性の蛋白質でパーマ処理等により溶出される蛋白質とは全く異なる物性を持つため充分な損傷毛髪の修繕効果を得ることは困難なのが実情である。

【発明が解決しようとする課題】
本発明者等はかかる事情に鑑み鋭意検討した結果、毛皮から抽出操作を行うことにより得られる、毛髪の表層を構成する特定の蛋白質が上記の毛髪の損傷により溶出される蛋白質の成分の一つであり、また該蛋白質が毛髪を還元剤を含む緩衝液で抽出することにより効率良く製造できること、さらには該蛋白質を含有せしめた毛髪処理剤として有用であることを見い出し本発明を完成するに至ったものであって、その目的は、毛髪損傷予防あるいは損傷毛髪修復に有用な毛髪処理剤を提供するにある。

【課題を解決するための手段】
従って、上述の目的は、哺乳類の毛小皮より抽出される、毛小皮由来抽出物、又はさらに蛋白質のSH基の全部又は一部を修飾することによって得られる毛小皮由来抽出物誘導体を配合することを特徴とする毛髪処理剤によって達成される。

【発明の実施の形態】
以下、本発明の構成について詳説する。

本発明で用いられる毛皮(キューティクル)の取得方法としては、公知の毛髪から分離して得る方法が挙げられる。例えば、毛髪を水中で振とう、攪拌あるいは超音波処理することにより、物理的に剥離することができる(Journal of the Society of Cosmetic Chemists,25巻,13頁,1974年参照)。剥離した毛皮は、ガーゼやメッシュで濾過することにより、毛髪から分離した後、遠心分離又は濾過することにより収集することができる。なお、本発明で用いる毛髪は人毛のほか羊毛、豚毛などの由来のものが挙げられる。

本発明の毛由来抽出物の製造方法としては、例えば収集した毛髪より分離して得た毛皮を、(1)還元剤のみを含む緩衝液, (2)還元剤及び可溶化剤を含む緩衝液、等の抽出液で抽出する方法等が挙げられる。特に(1)の還元剤のみを含む緩衝液中で抽出操作を行うと、より混在物の少ない毛由来抽出物を得ることができるので好ましい。

抽出操作は、毛皮の抽出緩衝液の液性をアルカリ性として行う。温度を25℃から 100℃の条件として行うことが好ましい。

また、毛皮から毛由来抽出物を製造する際には毛皮を抽出液中で攪拌するか、ホモジナイザー等を用いて毛皮を抽出液中擦り潰すと、効率良く本発明の毛由来抽出物が得られ好ましい。

本発明に用いられる還元剤としては、メルカプトエタノール、ジチオスレイトール、亜硫酸ナトリウム、チオグリコール酸アンモニウム等が挙げられる。

本発明に用いられる還元剤の、単独使用の濃度としては、緩衝液総量の1〜3重量%が好ましい。

抽出液に用いられる可溶化剤としては、周知公用の変性剤、界面活性剤等の可溶化剤等を用いることができ、例えば変性剤としては尿素、塩酸グアニジン等が挙げられ、界面活性剤としてはドデシル硫酸ナトリウム等が挙げられる。

本発明の毛由来抽出物誘導体の製造方法としては、本発明の毛由来抽出物に含有される蛋白質のSH基を例えば、モノヨード酢酸あるいはエチレンイミン等で修飾する通常の方法が挙げられる。

本発明の毛由来抽出物誘導体は、還元剤や可溶化剤が存在しなくても水に溶解するので広い範囲に応用することができる。

本発明の毛由来抽出物、並びに毛由来抽出物誘導体自体もS−S架橋又はSH基を多量に含有した蛋白質であるので、直接シャンプー、リンス、ヘアートリートメント等の毛髪処理剤に配合することによって、毛髪の保護成分として使用することも可能である。

本発明の毛由来抽出物、毛由来抽出物誘導体を含む毛髪処理剤の剤形としてはトリートメント、リンス、シャンプー、ヘアクリーム、ヘアーローション、ヘアーパック、パーマネント用剤等、毛髪に対して用いる各種のものが挙げられる。

発明の毛由来抽出物および毛由来抽出物誘導体の含有量は、組成物の総量を基準として0.01重量%〜5重量%が好ましく0.1 重量%〜1重量%が特に好ましい。

この他、本発明の毛由来抽出物、並びに毛由来抽出物誘導体は、例えば毛皮を特異的に認識する抗体を作製する際の抗原としても有用である。

【実施例】
以下、実施例、比較例によって本発明を更に詳細に説明する。

製造例1、2及び参考例1、2(ヒト毛皮からの毛由来抽出物、毛由来抽出物誘導体、毛皮特異蛋白質及び毛皮特異蛋白質誘導体の製造)
(1)ヒト毛皮 800mgを、8M尿素及び 0.2M 2-メルカプトエタノールを含む0.2Mトリス(pH 9.5)3ml に加え、50℃1時間インキュベートした後、ガラスホモジナイザーを用いて擦り潰した。更に50℃1時間インキュベートした後、遠心分離(10,000 g)して上清を集め、毛由来抽出物(蛋白質量として21mg;製造例1)を得た。

(2)また、この毛由来抽出物製造例1)の半量にモノヨード酢酸75mgを加え、トリスでpH 9.0 とし、遮光下室温で1.5 hr攪拌した。10mMトリス−塩酸緩衝液(pH 8.0)により透析して毛由来抽出物(カルボキシメチル化)誘導体製造例2)を得た。

(3)毛皮特異蛋白質ならびにその毛皮特異蛋白質(カルボキシメチル化)誘導体はSDS-ポリアクリルアミド電気泳動により分離した。上記によって得られた製造例1又は2の抽出物を、6M尿素を含む15%ゲルを用いて電気泳動した後、ポリビニリデンジフルオライド(PVDF)膜に転写し、得られた毛皮抽出物の分子量約7,000 のバンドならびにその毛皮抽出物誘導体の分子量約15,000のバンドを切り取り、酢酸/アセトニトリル/水(7:2:1)の混合溶液で抽出し、毛皮特異蛋白質(参考例1)及び毛皮特異蛋白質(カルボキシメチル化)誘導体(参考例2)を得た。

PVDF膜上の毛皮特異蛋白質(カルボキシメチル化)誘導体を0.5%ポリビニルピロリドンで処理した後、トリプシン(プロメガ社製)を加え24時間37℃で反応させた。得られた分解物を下記条件の逆相液体クロマトグラフィーに付し、保持時間14.2分及び18.3分の画分を分取した。

ヒト毛皮800 mgを、8M尿素及び 0.2M 2-メルカプトエタノールを含む0.2Mトリス(pH 9.5)3ml に加え、50℃1時間インキュベートした後、ガラスホモジナイザーを用いて擦り潰した。更に50℃1時間インキュベートした後、遠心分離(10,000 g)して上清を収集した。残渣をガラスホモジナイザーを用いて擦り潰し、50℃1時間インキュベートした後、遠心分離(10,000 g)し上清を収集する操作を、計6回繰り返し、得られた上清を集めて毛由来抽出物(蛋白質量として21mg)を得た。

ヒト毛皮を抽出する溶液として、2%ドデシル硫酸ナトリウム及び 10mM ジチオスレイトールを含む0.2Mトリス(pH 9.0)3ml を用いる以外は製造例5と同様にして、毛由来抽出物(蛋白質量として15mg)を得た。

参考例5(全毛髪のパーマ液による毛皮特異蛋白質画分の製造)
ヒト毛髪2.5gをパーマ第1液(12%チオグリコール酸アンモニウム、1%エタノールアミン、0.05%エチレンジアミン四酢酸ナトリウム、0.42%アンモニア水溶液)50mlに室温15分間浸した。5回パーマ第1液を交換して溶出液を集め、限外濾過膜を用いて濃縮し毛皮特異蛋白質画分(蛋白質量として0.13mg)を得た。

製造例8、9及び参考例6、7(羊毛皮からの毛由来抽出物、毛由来抽出物誘導体、毛皮特異蛋白質及び毛皮特異蛋白質誘導体の製造)
羊毛皮 200mgを用い、製造例1、2及び参考例1、2の(1)と同様の方法により、毛由来抽出物(蛋白質量として5.2 mg;製造例8)を得た。

皮特異蛋白質ならびにその毛皮特異蛋白質(カルボキシメチル化)誘導体はSDS-ポリアクリルアミド電気泳動により分離した。上記によって得られた製造例8又は抽出物を、6M尿素を含む15%ゲルを用いて電気泳動した後、得られた毛皮抽出物の分子量約7,000 のバンドならびにその毛皮抽出物誘導体の分子量約15,000のバンドを切り取り、蛋白質溶出装置(ISCO社製、1750)を用いて抽出し、毛皮特異蛋白質(参考例6)及び毛皮特異蛋白質(カルボキシメチル化)誘導体(参考例7)を得た。

得られた画分を6M塩酸で加水分解後、日立製アミノ酸分析機でアミノ酸を分析した、下記の結果を得た。なお、システイン含量は参考例6では過ギ酸酸化後加水分解して得たシステイン酸、参考例7ではS-カルボキシメチルシステインとしての定量値である。

予め、パーマ処理を3回施した毛髪7束を、実施例1〜5の毛由来抽出物含有毛髪損傷修復剤に各々室温で30分間浸した後、水洗した。無処理毛髪(健常毛),修復剤処理前及び処理後のパーマ処理毛髪の3種の毛髪に、キューティクルの捲れが観察し易い様にそれぞれ結び目を作り、走査型電子顕微鏡観察を行った。パーマ処理毛髪に観察されるキューティクルの捲れが、いずれの処理剤で処理することによっても健常毛程度に修復した様子が観察された。

ヒト毛束(黒髪5g)を実施例6〜11の毛由来抽出物含有毛髪トリートメント剤に1時間浸漬後、流水で10分間水洗して風乾した。処理後毛束のくし通り、しなやかさ、風合について処理前の毛束とどちらがよいか官能評価を行った結果、いずれも毛由来抽出物含有毛髪トリートメント剤で処理した毛束の方がくし通り、しなやかさ、風合が良好であった。

以下に記載した4種の溶液よりなる毛髪診断キットを作製し、ヒト毛髪で損傷度を診断した。

以上の結果から、毛由来抽出物を抗原として得た抗体で、毛髪の損傷状態あるいは健康状態を知ることができることは明らかである。

尚、A液を加え37℃、1時間インキュベートした時点で、傷んだ毛髪の方は、ハリが戻る等の損傷修復効果も見られた。このことから、本発明の毛由来抽出物を抗原として得た抗体は、毛髪の損傷の修復・予防にも有用であることが明らかとなった。

【発明の効果】
以上の結果から、本発明により、毛髪の損傷状態あるいは健康状態を知るマーカーとして,あるいは損傷毛の修復および予防用素材として有用な、毛由来抽出物、由来抽出物誘導体、及び該抽出物を配合した毛髪処理剤を提供できることは明らかである。
